In 2016 she received the LRTAV Outstanding Industry Contribution Award and her ability to research, consult and communicate to rural trucking and Government following years of industry experience resulted in her appointment as COO of the LRTAV. An amazing asset to road transport and a very insightful podcast.

BUFFALO, NY- July 10, 2024 – A new #editorial paper was #published in Oncotarget's Volume 15 on June 20, 2024, entitled, “Targeting ABC transporters in PDAC – past, present, or future?” In this new editorial, Cecilia Bergonzini, Elisa Giovannetti and Erik H.J. Danen from Leiden University discuss targeting ABC transporters in pancreatic ductal carcinoma (PDAC). Despite its lower incidence as compared to more common cancers such as lung or breast carcinomas, PDAC ranks as the third leading cause of cancer mortality in the US and the sixth worldwide. This is due to the fact that PDAC survival rates are among the lowest for cancer patients, around 13% in the US. ATP-binding cassette (ABC) transporters represent a family of transmembrane proteins that, using the energy from ATP hydrolysis, extrude molecules from the cytoplasm to the exterior or into vesicles. Some of these transporters have been associated with resistance to a spectrum of structurally diverse chemotherapeutic drugs, earning them the name of multidrug resistance (MDR) pumps. One of the best-characterized ABC transporters is ABCB1 (MDR1). It is physiologically expressed in tissues such as kidney, liver, pancreas, intestine, the blood-brain barrier, and more, where it exerts a protective role, by extruding xenobiotics and potentially toxic molecules. Moreover, increased ABCB1 expression in tumors has been associated with poor prognosis. “Paclitaxel is a bona fide substrate for ABCB1 [18] and ABCB1 has been implicated in paclitaxel and nab-paclitaxel resistance in multiple types of cancer [19, 20]. Could ABCB1 represent a therapeutic target in PDAC patients to suppress resistance against GnP? Does human teleportation really exist? Can we travel instantly to a different location? On today's episode, we will talk about that concept.   A transporter is a fictional teleportation machine used in the Star Trek science fiction franchise. Transporters allow for teleportation by converting a person or object into an energy pattern (a process called "dematerialization"), then sending ("beaming") it to a target location or else returning it to the transporter, where it is reconverted into matter ("rematerialization"). Since its introduction in Star Trek: The Original Series in 1966, the name and similar concepts have made their way to other science fiction scenarios, Here are some of the events we talk about. Skier Disappears in New York, Shows Up in Sacramento Airport a Week Later in Same Clothes A 49-year-old skier “Danny” Filippidis from Toronto was out shredding Whiteface Mountain, near Wilmington, N.Y on February 7 when he vanished into thin air. He told a friend he was going for one more run on the mountain alone near the end of the day. He never returned. Search and Rescue, ski patrol, helicopters and up to 140 people, spent six days searching for   “Danny” Filippidis in the Whiteface vicinity.  The local hypothesis was that he'd hit a tree or fallen into a deep tree well and perhaps suffocated. But Filippidis was on a much stranger trip. Fast forward a week and Filippidis had located a cell phone at the Sacramento Airport where he finally called his wife, who told him to dial authorities immediately. Police found Filippidis wearing the exact clothing he was wearing when he disappeared on that pow day at Whiteface on the other side of the country, he was even wearing his helmet and goggles. ‘Danny' appeared extremely confused to law enforcement, unable to tell them where he'd been other than recalling a “big rig-style truck,” and “sleeping a lot.” He had no idea what day it was, only remembering being dropped off, “near a McDonald's.” He had a credit card and $1,000 in cash. “He was very nonchalant, and kind of out of it,” said Sgt. Shaun Hampton with the Sacramento County Sheriff's Office. “Our officers were adamant to take him to the hospital to have him treated.” Hampton also added that officers didn't think he was under the influence or taken by force. The Canadian skier and firefighter did have a fresh haircut. There were no accounts of him getting on a plain, bus, or renting a car. A professor In England in 1970 walked out to his car sat down in the car and felt dizzy. The next thing he knows is he is 1000 miles away stumbling around. His car was still parked back at the University.   In Argentina a man was driving, and his car started acting up. So, he stopped and when he got out he was hit by some unseen force and knocked to the ground. When he stood up everything around him was different. Turns out he was 1500 miles away, confused and disoriented. When he told someone, officials went to his car and it was sitting along the road still running.   Pilot in electric fog. The story of Brues Gerner. Flying from Andorse Island to Paumbeach. About 10 min into the flight he flew into a bad storm. Trying to avoid it he turned and when he did he found himself in a tunnel. He felt as if he was being pulled forward for 10 seconds. He comes out and realized he was above Miami. Miami air command said he was over Miami beach with means he travelled 100 miles in 3 min. that's 1918mph. top speed of his one engine plane was 200mph.

Nur durch großes Glück überlebt sie den Anschlag. Und es gibt weitere Opfer. Angst macht sich breit auf den deutschen Autobahnen. Das Bundeskriminalamt übernimmt die Ermittlungen. In den Empfehlungen erwarten euch u.a. ein neues Rennspiel, ein Action-Kracher auf Netflix, eine Schwarzenegger-Doku, mehr Star Trek, James Bond, Super Mario und vieles, vieles mehr. Wir stellen uns die Frage, welches Spiel oder welchen Film wir gerne nochmal "zum ersten Mal" erleben würden und wir gehen natürlich ins Filmbingo. Andi durfte dort einen vergessenen Sci-Fi-Klassiker ansehen und wird uns erzählen, warum "Das Philadelphia-Experiment" vielleicht ziemlich unterschätzt wird, während Tobi die Reise im längsten Zug der Welt, dem "Snowpiercer" antritt und uns erzählt, warum eine neue Eiszeit nicht das einzige Problem der dystopischen Serie ist. M., Kowaltowski, A. J. Abstract: Autophagy is responsive to intracellular signals, including calcium (Ca2+) ions. Mitochondrial Ca2+ transporters such as the mitochondrial calcium uniporter (MCU, an uptake pathway) and Na+/Li+/Ca2+ exchanger (NCLX, an efflux pathway) are known to shape Ca2+ signals within the cell, thereby regulating Ca2+-sensitive cellular processes. However, the interplay between MCU, NCLX, and autophagy is unknown, and investigating the crosstalk between these important metabolic and signaling nodes was the aim of this study. We evaluated autophagic activity using a wide array of experimental approaches after siRNA-mediated MCU or NCLX knockdown. Under nutrient replete conditions, reduced expression of either NCLX or MCU led to decreased autophagic activity as well as altered expression of several autophagy-related genes, without affecting mitochondrial ATP synthesis. The knockdown of NCLX resulted in marked reduction of LC3 expression and of activation of the AMPK-UKL1 pathway. Interestingly, NCLX expression was increased under autophagy-promoting conditions in vitro and in vivo, demonstrating it is both regulatory of and regulated by autophagic stimuli. Also, genetic and pharmacological inhibition of NCLX caused impaired autophagosome formation and reduced autophagic flux after serum and amino acid starvation. In conclusion, our results uncover an important role for mitochondrial Ca2+ transport as a regulator of both basal and stimulated autophagic activity, modulating both autophagic initiation and autophagossome formation. This Thursday we're coming to you from the annual NAMAD membership meeting in Miami, FL where a change in leadership was announced last night. We're also talking about the continued struggle of self-driving tech, as well as the new weight requirement for roads given the rise in EVs, and a massive semiconductor plant coming to Syracuse, NY. NAMAD President Damon Lester stepped down and assumed the role as Vice Chair of the Board. After a 1 year executive search process, Hugene Fields will be stepping up. Self Driving cars still stalling out in go to market strategyOne of the hardest things to figure out is something called “unprotected left turns,” or left turns when navigating oncoming traffic.A McKinsey & Co. report states the industry has currently invested over $100bn in R&D for self-driving technology.“You'd be hard-pressed to find another industry that's invested so many dollars in R&D and that has delivered so little,” Anthony Levandowski says in an interview. “Forget about profits—what's the combined revenue of all the robo-taxi, robo-truck, robo-whatever companies? Is it a million dollars? Maybe. I think it's more like zero.”Transportation companies say that EV vehicles are too heavy for roads“[C]ars and trucks climbed from an average of 3,200 pounds (1,451 kilos) to 4,200 pounds over the last four decades, according to the Environmental Protection Agency.”Currently, less than 1% of vehicles on US roads are EVs, but with aggressive sales targets, it will be harder for transporters to meet highway weight requirements.“‘The truth is we will not be able to move as many electric vehicles under the current weight limit.
