At this year's ITA Showcase, I had the pleasure of speaking with Vinny Maniola of IP Infusion about how open networking and disaggregation are reshaping the industry. We frequently use all kinds of electronic goods at home, consume lots of electricity, our computers, washing machines, and phones, etc. Today, consumers are used to having information at their fingertips. However, the reality is that most consumers spend less than 3 minutes of user engagement per year with information sent by their gas, water and electricity suppliers. Luckily, that will soon change, and AI will be a driver of enhanced services for utility customers. By harnessing the data from nearly ubiquitous smart meters and partnering with technology startups like Net2Grid, consumers, utilities and retailers will actually be able to disaggregate electricity consumption down to the level of individual appliances. It is only when Asian American Pacific Islander data sets are disaggregated that entire communities become visible and significant inequities within those subgroups are revealed. It takes this level of detail to guide government, schools and health providers in serving each community effectively. Abstract: Molecular chaperones contribute to the maintenance of cellular protein homeostasis through a wide range of mechanisms, including the assistance of de novo protein folding, the rescue of misfolded proteins, and the prevention of amyloid formation. Chaperones of the Hsp70 family have a striking capability of disaggregating otherwise irreversible aggregate structures such as amyloid fibrils that accumulate during the development of neurodegenerative diseases. However, the mechanisms of this key emerging functionality remain largely unknown. Here, we bring together microfluidic measurements with kinetic analysis and show that that the Hsp70 protein heat chock complement Hsc70 together with its two co-chaperones DnaJB1 and the nucleotide exchange factor Apg2 is able to completely reverse the aggregation process of alpha-synuclein, associated with Parkinson's disease, back to its soluble monomeric state. Moreover, we show that this reaction proceeds with first order kinetics in a process where monomer units are taken off directly from the fibril ends. Our results demonstrate that all components of the chaperone triad are essential for fibril disaggregation. Lastly, we quantify the interactions between the three chaperones as well as between the chaperones and the fibrils in solution, yielding both binding stoichiometries and dissociation constants. Crucially, we find that the stoichiometry of Hsc70 binding to fibrils suggests Hsc70 clustering at the fibril ends. Taken together, our results show that the mechanism of action of the Hsc70-DnaJB1-Apg2 chaperone system in disaggregating -synuclein fibrils involves the removal of monomer units without any intermediate fragmentation steps. These findings are fundamental to our understanding of the suppression of amyloid proliferation early in life and the natural clearance mechanisms of fibrillar deposits in Parkinson's disease, and inform on the possibilities and limitations of this strategy in the development of therapeutics against synucleinopathies and related neurodegenerative diseases. Recently, a man by the name Edward Blum and the Students for Fair Admissions have teamed up to sue Harvard for discriminating against its Asian American applicants with its affirmative action policy. Affirmative action has been one of the hottest debates for a while within our community and I thought that it would be really helpful to have Quyen Dinh, Executive Director from the Southeast Asia Resource Action Center, also known as SEARAC, on the podcast to share her personal story as well as SEARAC’s findings on the important impact affirmative action and data disaggregation has had on our Southeast Asian American community. Quyen Dinh is the Executive Director of the Southeast Asia Resource Action Center (SEARAC). As Executive Director, Quyen has advocated for Southeast Asian Americans on key civil rights issues including education, immigration, criminal justice, health, and aging. Born to Vietnamese refugees, Quyen identifies as a second-generation Vietnamese American. She holds a Masters of Public Policy from the UCLA Luskin School of Public Affairs, and a Bachelor of Arts degree in English from the University of California, Berkeley.
