//TRANSCRIPT//I want to let you in on a little secret about me. Now everybody knows this. I'll tell you in a minute, but first, a story.I signed up to go in the army to become a Green Beret. That in itself was, as you can imagine, a bit of an anxious time, difficult time, right? You have this dream, you want to do this thing. And this is a giant bureaucracy. Just because you want it doesn't mean you can have it.Part of the process is checking if the job is available and if you're smart enough. You take an aptitude test, and then you sign a contract and go in. Part of that process is getting a physical, though. You have to be physically fit. They obviously want to make sure that anybody who the government's going to spend a ton of money on to go into the military can do it.What I Didn't KnowI took a physical, and I failed that physical to become an SF guy to go into this program. They said no. Why'd they say no? Well, turns out there is a vision test that you take. You've probably taken this. It was created by Dr. Ishihara, a Japanese guy back in the early 1900s. It's that color test, the one with the circle where you look at it and they tell you to tell you what number it is. There's a bunch of dots inside of it.I can't do that one. Just can't do it. Period. Full stop. And I failed it. They said, "Sorry, can't do it. Colorblind. Can't be an SF guy."Where There Is A Will, There Is A WaiverThe good news is there's another motto in the military, which is "where there's a will, there's a waiver." And there is another option here. They really only care about colors that matter in combat, and those are red and green. So another guy, an American guy this time, Dr. Farnsworth, created a separate test. This test tests your ability to see red and green. Red and green is what matters in combat - I think it's because of looking at stuff under red lens and maps and stuff like that. But anyways, I passed that and the rest is history, I'm in.Up until that moment, I didn't know that I was colorblind. Well, slight color deficiency. I have a difficult time telling blue from black, dark purple from blue, those kind of things. I have to ask friends to tell me, "Is this black? Is this blue? Do these pants match?" I've even worn the wrong pair of shoes to a medium before - one black, one brown. Kind of embarrassing, but I'm too old to be embarrassed at this point.My BlindspotsI tell you this story because up until that moment, I had a pretty big blind spot as it relates to color. And it often takes some kind of outside intervention to help you see where you don't see correctly. The older I get, the more I like these. They're absolutely painful when you find them.But the reason we want to find our blind spots is because our blind spots keep us from seeing reality, from seeing the truth, from operating in what is the real world, not our version of it. And those blind spots come in many forms. In my case, in this situation, slight color deficiency, but they come in the form of biases, stories, stereotypes, pick a thing.We should be looking for our blind spots. And when friends try to bring them to us and tell us about it, we should be thankful, we should be grateful. Not all feedback is true or helpful, but sometimes it is. You should be on a quest to find your blind spots because as we remove them, it enables us to operate in reality.Anyways, that's my story. Slight color deficiency. Thank you, Dr. Farnsworth. J., Miller, R., Malfait, A.-M. Abstract: Objective: Knee joints are densely innervated by nociceptors. Sprouting of nociceptors has been reported in late-stage osteoarthritis (OA), both in human knees and in rodent models. Here, we sought to describe progressive nociceptor remodeling in four mouse models of knee OA, capturing early and late-stage disease. Methods: Sham surgery, destabilization of the medial meniscus (DMM), partial meniscectomy (PMX), or non-invasive anterior cruciate ligament rupture (ACLR) was performed in the right knee of 10-12-week old male C57BL/6 NaV1.8-tdTomato mice. Mice were euthanized (1) 4, 8 or 16 weeks after DMM or sham surgery; (2) 4 or 12 weeks after PMX or sham; (3) 1 or 4 weeks after ACLR injury or sham. Additionally, a cohort of naive male wildtype mice was evaluated at 6 and 24 months. Twenty-m thick mid-joint cryosections were assessed qualitatively and quantitatively for NaV1.8+ and PGP9.5+ innervation. Cartilage damage (using a modified OARSI score), synovitis, and osteophytes were assessed blindly. Results: Progressive OA developed in the medial compartment after DMM, PMX, and ACLR. Synovitis and associated neo-innervation by nociceptors peaked in early-stage OA. In the subchondral bone, channels containing sprouting nociceptors appeared early, and progressed with worsening joint damage. Two-year old mice developed primary OA in both the medial and the lateral compartment, accompanied with neuroplasticity in the synovium and the subchondral bone. All 4 models had an increased nerve signal in osteophytes. Conclusion: Anatomical neuroplasticity of nociceptors was observed in association with joint damage in 4 distinct mouse models, suggesting that it is intrinsic to OA pathology. Abstract: The endoplasmic reticulum (ER) is physically connected to the cell nucleus via junctions with the nuclear envelope (NE). These ER-NE junctions are essential for supplying the NE with lipids and transmembrane proteins that are synthesized in the ER. Despite the important role of ER-NE junctions, their biogenesis, architecture and maintenance across the cell cycle has remained elusive. In this study, by combining live cell imaging with quantitative three-dimensional electron microscopy, we systematically elucidated the ultrastructure of ER-NE junctions in mammalian cells. We discovered that ER-NE junctions exhibit a constricted hourglass shape that is different from the junctions within the ER. When ER-NE junctions are newly built during NE assembly at mitotic exit, their morphology resembles ER-ER junctions, but they become constricted starting in telophase. Altogether, our findings imply novel mechanisms that remodel ER-NE junctions and have functional implications for the lipid and protein traffic that are crucial for nuclear function. Abstract: Recent development of large-scale activity imaging of neuronal ensembles provides opportunities for understanding how activity patterns are generated in the brain and how information is transmitted between neurons or neuronal ensembles. However, methodologies for extracting the component properties that generate overall dynamics are still limited. In this study, the results of time-lapse 3D imaging (4D imaging) of head neurons of the nematode C. elegans were analyzed by hitherto unemployed methodologies. By combining time-delay embedding with independent component analysis, the whole-brain activities were decomposed to a small number of component dynamics. Results from multiple samples, where different subsets of neurons were observed, were further combined by matrix factorization, revealing common dynamics from neuronal activities that are apparently divergent across sampled animals. By this analysis, we could identify components that show common relationships across different samples and those that show relationships distinct between individual samples. We also constructed a network model building on time-lagged prediction models of synaptic communications. This was achieved by dimension reduction of 4D imaging data using the general framework gKDR (gradient kernel dimension reduction). The model is able to decompose basal dynamics of the network. We further extended the model by incorporating probabilistic distribution, resulting in models that we call gKDR-GMM and gKDR-GP. The models capture the overall relationships of neural activities and reproduce the stochastic but coordinated dynamics in the neural network simulation. By virtual manipulation of individual neurons and synaptic contacts in this model, information flow could be estimated from whole-brain imaging results. Copy rights belong to original authors. Welcome to episode 33 of Holly Jolly X'masu! In this episode, I talk about Yujiro Ishihara's 1966 album, “Xmas Carol.” It's a fantastic vocal album that should appeal to any fans of Perry Como or Andy Williams. Before finding the LP, I first came across the flexi-disc version, which was released at the same time. Due to some misinformation, at first I thought it was a double-LP, then a set of two 45's. I finally found an extremely affordable copy of it online which showed the flexis. I wasn't collecting flexi-discs at the time, so I passed. The copies I've found since then are a little more than I'm willing to spend. After receiving and listening to the album, without knowing anything about Ishihara, I developed a picture of him in my mind of a Ricky Nelson type who transitioned from teen idol roles to similar family-friendly fare as he got older. I was really surprised to read about his long career in tough guy roles, and his Sinatra-esque reputation in the industry. One disappointing aspect of my research was that I couldn't find a lot about his actual singing career. Wikipedia was light on the details, providing little more than a few lists of his albums and singles. He apparently started out as a brash, rebellious sort, before settling into more of a crooner role. However, he did maintain his rough, onscreen persona throughout his acting career. Seat tuvo una profunda relación con Fiat durante 30 años, pero la etapa de Volkswagen se acerca a los 40 años. Y, entre medias está la etapa de cuando Seat era solo Seat. La etapa más difícil, clave en el futuro de la marca y en la que nos vamos a centrar en este vídeo. Hay un personaje clave en esta historia que, sin duda, salvó a la marca: Juan Miguel Antoñanzas. Tuve la oportunidad de entrevistar al señor Antoñanzas largo y tendido, en su propio domicilio… Un lujo. Mucha de la información que encontrarás en este video procede de directamente de él. Pero hay otro nombre propio dentro de esta historia: Ibiza. Porque el Seat Ibiza. Fue el primero modelos 100 por cien Seat y que, sin duda salvo a la marca. Una situación difícil. La situación de Seat en 1977 era muy difícil. Para reflotar la empresa el Gobierno español pone a su frente nada menos que al presidente del INI, José Miguel Antoñanzas, una persona joven, preparada y que había hecho una buena gestión al frente del INI. Seat tenía muchos problemas, pero el principal es que el mercado español estaba “agotado” y para ser rentable, Seat necesitaba exportar. Pero sus productos eran sino idénticos, muy parecidos a los de Fiat. Llega el amable Fusaro. Pues sí, porque después de muchos estudios y valoraciones, en 1979 Seat firma un acuerdo con Fiat, un plan de salvación que detallaba compromisos económicos, legales y de todo tipo por parte de Fiat, a cambio de su soberanía total sobre Seat. Desembarca en España un directivo de Fiat, amable y de buenas maneras, llamado Piero Fusaro, que lo cambió todo e incorporó una nube de directivos italianos y… las cosas inmediatamente fueron a peor. Apuesta arriesgada. En una reunión al máximo nivel se baraja la posibilidad de cerrar Seat. Pero, contra todo pronóstico Antoñanzas y su equipo convencen al INI y al Ministerio Español de Industria para apostar por la supervivencia. La denuncia de Fiat. Estas veladas amenazas enfadan a Fiat que se muestra terminante y avisa: “ninguno de los vehículos futuros de Fiat serán objeto de fabricación por Seat”. La tecnología Fiat se reserva solo para los vehículos ya producidos en años pasados. Los cambios que Seat introduce en el Ritmo, para convertirlo en Ronda, les parecen escasos a Fiat, y en noviembre de 1982 denuncia a la marca española. Un año después, en la corte de arbitraje de París Seat gana el pleito. Nissan y Toyota. Seat decide construir por sus propios medios un sucesor al 127 mientras busca un socio. Las elegidas son #Nissan y #Toyota en Japón, y Volkswagen en Alemania. Con Nissan las cosas no fueron bien. Hay simpatía, conexión e incluso admiración mutua entre Antoñanzas y el presidente de Nissan, Ishihara… pero acaban de comenzar su aventura europea con una planta en Gran Bretaña. La relación con Eiji Toyoda, presidente de Toyota, fue más complicada. La recepción en Japón, llena de ampulosidad y ceremonial dejo claro que para la estrategia de la marca, Seat carecía de interés. El “tropiezo” con Volkswagen EL presidente de Seat inicia unas negociaciones intensas pero discretas con el presidente de Volkswagen, Schmücker. El proyecto estaba muy avanzado pero Schmücker sufre un infarto de miocardio y quedaban por tanto suspendidos todos las acuerdos previos. No solo eso, sino que desde Volkswagen se insta para que no sigan el desarrollos del S1, el futuro Ibiza, al que consideran competidor de su futuro Polo… Y llega Volkswagen En 1982 el consejo de Volkswagen nombra a Carl Horst Hahn presidente de VW. EL nuevo presidente no solo retoma la idea de su antecesor de negociar con Seat, sino que entiende y valora muy positivamente la valentía de la compañía de continuar adelante con el proyecto del Ibiza. EL 30 de septiembre de 1982 los dos presidentes, Hahn y Antoñanzas, firman el primer acuerdo de colaboración entre Seat y Volkswagen. Pese a todo, nace el Ibiza Pero antes de firmar ese acuerdo, era urgente la necesidad de un sucesor para el 127 y en Seat deciden arriesgar. Parten de la plataforma del Fiat Ritmo y buscan al mejor para el diseño de su carrocería ¿Y quién es el mejor? El estudio de diseño de Giorgetto Giugaro, Ital Design. En 1986 Volkswagen compra el 75 por ciento de las acciones de Seat para hacerse con el control total en 1990.

Abstract: Capillary forces are driven by interactions between liquids and surfaces. These forces are crucial for many processes ranging from biology and physics to engineering, such as the motion of aquatic insects on the surface of water, modulation of the material properties of spider silk, and self-assembly of small objects and microstructures. Recent studies have shown that cells assemble biomolecular condensates in a manner similar to phase separation. In the nucleus, these condensates are thought to drive transcription, heterochromatin formation, nucleolus assembly, and DNA repair. Here, we test if the interaction between liquid-like condensates and chromatin generates capillary forces, which might play a role in bringing distant regulatory elements of DNA together, a key step in transcriptional regulation. We combine quantitative microscopy, in vitro reconstitution, and theory to show that the pioneer transcription factor FoxA1 mediates the condensation of a DNA-protein phase via a mesoscopic first-order phase transition. Surprisingly, after nucleation, capillary forces drive growth of this phase by pulling non-condensed DNA. Altering the tension on the DNA strand enlarges or dissolves the condensates, revealing their mechanosensitive nature. These findings show that DNA condensation mediated by transcription factors could bring distant regions of DNA in close proximity using capillary forces. Our work suggests that the physics of DNA and protein condensation driven by capillary forces provides a general regulatory principle for chromatin organization. Seattle Pacific University's Chaplain Lisa Ishihara didn't anticipate a life in ministry when she was younger, but early on she found a love for worshipping God through her mom's musical dance choir. As a business major in college, she didn't focus on ministry, but her love for learning scriptures led her to the seminary where she earned two master's degrees from the Talbot School of Theology. She went on to serve as director of chapel programs at Biola University for 10 years. In 2018, God tapped her on the shoulder to uproot her life and do ministry in Seattle at Seattle Pacific University. Listen to her full story on the SPU Voices Podcast.

Peter talks to Sean from Two Spots, not about coffee as you might expect but about the excess waste that comes off the bean when it is roasted - known as the chaff - and how that is now being used as dressing in 'flower beds' and as bedding for hens, before ending up on the compost heap. And finally Peter also talks to the wonderful Avril Hogan of Dingle Distillery a little about making gin, but also about how beautiful Dingle is, the collecting of the botanicals for the gin making and working with the seasons to produce one of Ireland's finest refreshments ever. The hypothesis that the structure of high Reynolds number turbulence conists of thin shear layers, with thickness of the order of the Taylor micro scale, has been further confirmed by numerical studies by Ishihara and Kaneda of conditional statistics and local dynamics; by PIV measurements of lab experiments by Wirth and Nickels, and by further developments of the theory, especially the transport of energy into the layers leading to the generation of intense structures, on the scale ofthe Kolomogorov micro-scale. This analysis provides (for the first time?) a physically justifiable analysis for the higher moments and why these are generally less isotropic than lower order moments, eg in thermal convection. The theoretical and practical implications of the flow near and within these layers for fluctuations of temperature and other scalars are explained.
