Discograffiti is the deep-dive podcast for music obsessives.  In this episode, we feature the next entry in The Chuck Granata Pet Sounds Interview Series. It's Chuck's chat with re-issue genius Bob Irwin. Bob and his wife Mary started the Sundazed Music label in 1989. The first Sundazed releases were The Great Lost Knickerbockers Album! by The Knickerbockers and Western Union by The Five Americans, and reflected Irwin's personal preference for garage rock and surf music. His skill at restoring old vinyl records for the (then new) CD format attracted the attention of major labels, who increasingly solicited him to help them re-issue material from their back catalogs. He also helped Sony Music release their archival Legacy Records label. Later, his restoration work included early material by the likes of Bob Dylan, Nancy Sinatra, & The Byrds. In this episode, Bob discusses his relationship with Pet Sounds, its standing in the pantheon no matter how many times you've heard it, its importance and influence on music in general, and the wonders of a technology with inherent, imposed limitations. There'll be a short sneak peak running publicly for free, but the entirety of this podcast will only be accessible on the Major Tier of Discograffiti's Patreon. Aron Knickerbocker, President and CEO of Aulos Bioscience,  is on a mission to extend the lives of cancer patients through safer and more effective immunotherapy using AI and machine learning to accelerate drug discovery and optimization. The company's lead drug candidate is an antibody that was designed using AI to harness the power of interleukin-2 to activate the immune system against tumors.  The advantages of AU-007 over previous IL-2 therapies include the ability to selectively activate tumor-fighting immune cells while avoiding toxicity due to vascular leakage. Current trials are underway in advanced melanoma and non-small cell lung cancer, which are showing the potential for durable responses and immune memory. Aron explains, "So Aulos has really been founded to carry out a mission to extend the lives of patients with innovative, safe, and ultimately effective cancer immunotherapy. And you mentioned IL-2, which is the naturally occurring protein that we're seeking to harness and redirect in productive ways to help the patients. And this has really been kind of a driver for me over the years, wanting to improve on cancer patient care. This is a company with an important mission and a great team, and our program is really interesting. It's harnessing IL-2 in such a way that it sends it to the cells that are capable of attacking the tumor and killing the tumor cells, and keeps it away from the cells that suppress the immune response. It's really kind of using a double-edged sword in a productive way to help patients with cancer." "So this is the first antibody to go into human clinical trials. It was designed in part by leveraging an AI platform, as you noted. And that platform was created by our collaborators at Biologic Design. They're the company that created this molecule. And what biologic design does is essentially using an AI system that has machine learning algorithms that drive it. It mimics what the immune system does normally. So when we get sick and we need to make antibodies to something, our immune system says, Do I already have something that will kind of bind to the target or the virus or whatever it's trying to hit? And then it will optimize that. It will go through a process of rapid change. And what the AI system and the machine learning algorithms that have been trained on millions of antibodies in the targets to which they bind do is recognize patterns much like an AI chatbot might recognize language patterns." #AulosBio #Antibodies #MedAi #AI #IL2 #Onocology #Cancer #NSCLC #AdvancedCutaneousMelanoma #DrugDiscovery #BiologicDesign #Tregs aulosbio.com Download the transcript here

Before the grunge movement of the 90's, and before the punk movement of the 70's, there was the garage band phenomenon of the 60's.  One of the forerunners of both punk and grunge from this garage band period is Tacoma Washington's The Sonics.  Their classic lineup consisted of Rob Lind on harmonica, vocals, and sax, Gerry Roslie on organ, piano, and lead vocals, Larry Parypa on lead guitar and vocals, Andy Parypa on bass, and Bob Bennett on drums.  The Sonics had a fast, hard-edged sound similar to The Kinks, and performed with a speed that would inspire punk rock.Boom is their second studio album from the band, released in February 1966.  As with many songs from the mid-60's, all the songs are short.  But with a non to the future punk movement, many songs are even shorter, clocking in at just over 2 minutes.  The Sonics take their name from Boeing, the aviation company also based in Tacoma.  The name of the album is likely a play on words for the "sonic boom," the thunderous sound produced by an aircraft exceeding the speed of sound.  The music on this album is energetic and abrasive, and while some fuzzy guitar distortion is included the overall feel is raw with little extra production. The classic lineup would fall apart by 1968, with members leaving to join other bands, attend college, or in saxophonist Rob Linds' case - become a fighter pilot in the Vietnam War.  Gerry Roslie would be the sole remaining original member with new members jumping in and out of the band until 1980.  Wayne brings us this forerunner of punk and grunge for this week's podcast. CinderellaThe lead-off song from the album was an original piece written by Gerry Roslie.  The lyrics relay a boy meeting a girl at a bar, having a great time drinking and dancing, and just when things start to get going well, the girl is gone by midnight.  He's Waitin'This song starts the B-side of the album, and was also a Gerry Roslie original.  This "boy loses girl" song is not a happy one!  The lyrics reflect the boy's thoughts on being burned by a girl.  Now he is telling her that Satan is waiting for her.  "It's too late, you lied, now you will fry." Louie, LouieRichard Berry wrote and performed this song in 1957 with The Pharaohs, and covers of this song are common with garage bands.  The Sonics' version is quite fast, and modifies the pronunciation from "Lou-ee Lou-ee" to "Lou-eye Lou-way" in the chorus.  It is hard to separate this song from the thoughts of fraternity parties and the comedy movie "Animal House." Shot DownThe song that closes out the album laments about striking out when approaching a girl.  "I play a guitar.  I even drive a brand new car.  Big man in town, I've been shot down."  ENTERTAINMENT TRACK:Theme from the motion picture "The Rare Breed"This western starred James Stewart and Maureen O'Hara and was one of the first films scored by the now-legendary composer John Williams. STAFF PICKS:Crying Time by Ray CharlesBruce leads off the staff picks with the lead single and starting track to Ray Charles' album of the same name.  The song was originally performed by Buck Owens in 1964, and Charles kept the country feel while taking the cover to number 6 on the Billboard Hot 100.  He won the Grammy Awards in 1967 for Best R&B Recording and Best R&B Solo Performance for this track. It's a Man's Man's Man's World by James BrownLynch brings us a song that takes its name as a play on the name of a 1963 comedy film, "It's a Mad, Mad, Mad, Mad World."  The minor key track has lyrics decrying the chauvinism of society where all the work of modern civilization are attributed to men, but it would all "mean nothing without a woman or a girl."  Brown recorded the song in only two takes, and it would become a staple of his live shows for the rest of his career.Secret Agent Man by Johnny RiversRob features a surf rock song originally developed as the opening intro to the U.S. spy television series "Secret Agent," which aired from 1964 to 1966. The ditty became popular, and Rivers developed it into a full single.  The song would go to number 3 on the Billboard Hot 100, becoming both a signature song for Rivers and a rock standard for future bands and shows.Lies by The KnickerbockersWayne's closes out the staff picks with a song from an American band deliberately trying to mimic the sound of a British Invasion song.  The trio harmonies definitely give it a Beatles quality.  As you might expect from the band name, the Knickerbockers got their start in New Jersey as a garage band.  The track came together in about a half hour. COMEDY TRACK:The One on the Right is On the Left by Johnny CashThis comedy song about a band with incompatible political leanings amongst its members takes us out for this week's podcast. Tonka needed something to get them out of a $10,000,000 hole. The needed to transform their situation. Dave Young: Welcome to the Empire Builders Podcast, teaching business owners the not-so-secret techniques that took famous businesses from mom and pop to major brands. Stephen Simple is a marketing consultant, story collector and storyteller. I'm Stephen's sidekick and business partner, Dave Young. Before we get into today's episode, a word from our sponsor, which is, well, it's us, but we're highlighting ads we've written and produced for our clients. So, here's one of those. [Pinpoint Ad] Dave Young: Welcome back to the Empire Builders Podcast, Dave Young here, along with Stephen Semple, and we're talking about empires. And the one you gave me now is I have to admit, I am familiar with the brand, but I'm not an active participant in the brand. I think I've mentioned on here before, I raised four daughters, and so today's topic is Transformers. And none of my daughters were ever into Transformers as a toy. I was beyond toys when Transformers, I think first came around. So I've never even really jumped into the movies. But big, big brand. Stephen Semple: Huge brand. Dave Young: Tell me how they got started. Stephen Semple: Well, and the interesting thing, I almost included my oldest daughter, Crystal, in the recording of this, and I decided that, no, we might do a follow-up episode with her because she is a massive Transformers fan. Dave Young: Wow. Okay. Stephen Semple: And the interesting thing is she never played with them as a kid, but she found herself into the fandom. And today there's a whole thing I want to do about fandoms because it really speaks to the role that social media plays with a lot of companies. Dave Young: I feel like as a kid, I would've loved them. Stephen Semple: Because there's this unbelievably rich storyline behind Transformers that is incredible. And then it's actually one that is fragmented into the Michael Bay Transformers storyline versus the historic ones, because Michael Bay is the one who made the movies. It's this crazy world when you dig into it. But, the story of Transformers is actually a story of a battle between two decent-sized companies. So it's not normally what we do, but it was so interesting, I thought we got to talk about this because it's a story about a battle between Tonka and Hasbro. Dave Young: Oh, man. Just like Wonka and the other guy. Yeah. Stephen Semple: Yeah. Exactly. Yeah, yeah. Today the Transformers franchise, I think is a $25 billion franchise between the movies and the toys. It is massive. And the story goes back to we're in July of 1983, and Tonka is struggling for a new hit. Tonka is being run by Steve Shank, and they're like $10 million in the hole, and they're known for trucks. And think back to the '80s, what's happening, video games are growing and the market is shifting away from things like playing with trucks. And Tonka did not shift with it. So they're really, really struggling. And Shank has hired a Mattel veteran, Pat Fellie. He's the guy who basically helped turn things around at Mattel with He-Man. So he was basically the really a key person with developing of He-Man. And so they need something quick. And the shortcut is to license existing toys because the development and manufacturing is done. It's quicker and cheaper to market. So they go out everywhere looking around for something to market, and they come across this toy that Knickerbocker has. Knickerbocker is going out of business, and it's this interesting toy, which is a car and a robot, but it's a cheap knockoff. The real one is being done in Japan by Bandai, which is making this thing called a Machine Robo. And so Steve Shank heads to Tokyo to get the rights. The Surprising Origins of American Christmas Traditions

Witch Hunt

Play Episode Listen Later Dec 24, 2024 31:03

Explore the fascinating transformation of Christmas traditions in America, revealing how many of our cherished customs emerged more recently than we might imagine. Examine Christmas celebrations in colonial America, where the holiday bore little resemblance to today's family-centered observance. Through court records and historical accounts, we learn how the Puritans banned Christmas in Massachusetts from 1659 to 1681, troubled by its connections to pre-Christian festivals and its often rowdy, carnival-like atmosphere. The narrative threads through several compelling historical cases, including a 1679 incident in Salem Village where four young wassailers - including future Salem Witch Trial figures Samuel Braybrook and Joseph Flint - caused significant disruption during their Christmas celebrations. This event illuminates the complex relationships between holiday revelry, social order, and the later witch trials that would shake colonial New England. Tracing the origins of Christmas customs, hosts Josh and Sarah explore how ancient festivals like Saturnalia and Yule influenced early celebrations, and how Christian leaders strategically absorbed these pre-Christian traditions. Learn how the modern American Christmas largely took shape in the 19th century, with particular attention to the literary creation of Santa Claus by Washington Irving and his circle of writers known as the Knickerbockers. Their work directly influenced Clement Clark Moore's famous poem "A Visit from St. Nicholas," which the hosts perform as a special dramatic reading. The evolution of Christmas trees, gift-giving customs, and holiday decorations receives careful examination, demonstrating how these "timeless" traditions emerged relatively recently as part of a broader transformation of Christmas from a community celebration to a domestic, child-centered holiday.  Throughout the episode, Josh and Sarah weave together scholarly research with engaging storytelling, helping listeners understand how Christmas evolved to reflect changing American values while retaining its power to connect generations through shared celebration. Using primary sources, including court records and period writings, they illuminate how our holiday traditions, though often presented as ancient and unchanging, are dynamic expressions of cultural values that continue to evolve. We love to hear from our listeners. Send us a message. Data drives decision-making at Aulos Bioscience. It's also driven the career trajectory of its CEO. On this week's episode of the Business of Biotech podcast, we get an introspective walk through Aron Kinickerbocker's strategically-orchestrated career path, from his early days in the labs at Bristol-Myers Squibb and Genentech to his leadership of Aulos Bioscience, a company that boasts the first AI-generated antibody in human clinical trials. We explore Aulos' innovative IL-2 antibody and the role AI played in its highly-targeted and toxicity-reducing design, and we do it in pragmatic fashion — Aron's not about the AI hype, he's about the AI data — and in its Phase 1/2 clinical trial, the data is looking pretty good. Register for Bioprocess Online's Bio Expo Live, being held July 30th through August 1st . Terry Knickerbocker has coached elite actors like Daniel Craig, Michelle Williams and Sam Rockwell. He's coached actors on performances that have won an Academy Award, Golden Globe Awards and Screen Actors Guild Awards. That's how he became dubbed "New York's Top Acting Coach". But in Today's Episode of 10,000 NOs Podcast, Terry returns (he was on the pod before the pandemic) to speak to the members of our 10,000 NOs Insiders Community and dispel the myth of the elite. He believes that the main distinction between our elite class of actors and the rest of the pack lies in their attention to the work and building a foundation of technique and approach. Many anecdotes of his collaboration with famous actors are dropped but the bottom line is that training is essential if we expect to create the confidence necessary to deliver consistent performances on par with the best of the best.10,000 NOs podcast is now in it's 2.0 phase: after a long pause from late 2021 to late 2023, it has returned in a more raw format: shorter episodes, like the Monday Morsels of yesteryear (they may be short, solo riffs by Matt on central themes of the pod, excerpts from Matt's 10,000 NOs Insiders Community Zooms, or excerpts from Insiders VIP sessions with VIPs from the entertainment interview and beyond). Be on the look out as, every now and then, we will likely drop some old school, long-form, classic 10,000 NOs interviews as bonus episodes. Prefer to WATCH (versus listen)? Episode 529: Ann & Billy Woodward

In the early morning hours of October 31, 1955, millionaire socialite Ann Woodward heard a strange noise in the hallway just beyond her bedroom door in the sprawling estate she shared with her husband, Billy, and their two children. There had been a series of robberies in the wealthy neighborhood that month, so Ann had kept a shotgun next to her bed for safety. Rising from her bed, Ann grabbed the gun and crept towards the door, slowly opening it so as not to attract any attention. Visibility was low in the darkened hallway, but she could see the vague shape of a man moving towards her and without hesitation, Ann raised the shotgun and fired in the direction, striking the figure and cutting him down. With the threat neautralized, Ann moved towards the figure on the floor only to realize she'd shot and killed her husband, Billy Woodward. At least that's the official version of the story. The investigation moved incredibly quickly, in the way it always seems to for the wealthiest among us, and Ann Woodward was cleared of any wrongdoing in the death of her husband—it was simply an accident. Yet there were many among Ann and Billy's family and friends who believed Ann had intentionally shot her husband that night in order to prevent him from going forward with a messy divorce that would have brought an end to the glamorous high society lifestyle she spent her entire life working to secure.Ann Woodward was never able to escape the rumors and gossip from those she'd once counted as friends, all of which was made exponentially worse by novelist Truman Capote, whose slanderous fiction many believe drove Ann to suicide. Ann's untimely death meant that many questions would forever go unanswered: did she really kill her husband in order to remain among America's elite moneymakers?Thank you to the amazing Dave White of Bring Me the Ax Podcast for research!ReferencesAssociated Press. 1955.
