Durante algún tiempo la ciencia anduvo detrás de la investigación en el cerebro sobre dónde y cómo funcionaban los/el centro del placer. Algunos científicos estuvieron cerca de descubrirlo pero en algunos famosos casos, el motivo por el cual justificaban su investigación alteraron los resultados, aunque en realidad nunca fueron suficientemente concluyentes. Incluso las agencias de inteligencia de algunos países se centraron en ello, con el objetivo de controlar a los sujetos o la población. Hoy en día parece que esta investigación se abandonó, pero como siempre podría ser que se continúe en el más absoluto secreto. ¿Existe un Centro del Placer en el Cerebro? El concepto del “centro del placer” en el cerebro ha capturado la imaginación de científicos, filósofos y el público general durante décadas. ¿Es posible que una región específica del cerebro sea responsable de las sensaciones de placer? La respuesta es más compleja y matizada de lo que podría parecer a primera vista. 1. Orígenes del Concepto: Experimentos Pioneros En los años 50, los investigadores James Olds y Peter Milner descubrieron áreas del cerebro que, al ser estimuladas eléctricamente, producían respuestas de comportamiento asociadas con el placer en ratas de laboratorio. Olds y Milner implantaron electrodos en el cerebro de estas ratas y notaron que los animales repetían conductas dirigidas a la autoadministración de la estimulación eléctrica. In recognition of Alcohol Awareness Month, we are joined by George Koob, PhD, director of the National Institute on Alcohol Abuse and Alcoholism (NIAAA). Dr. Koob talks about his background and career journey, along with major insights made through his research, including regarding alcohol withdrawal. He addresses stigma related to alcohol use (or abstinence from) and offers advice for the next generation of researchers and clinicians in this exciting and dynamic space, with so much potential for expanding knowledge. Dr. Koob shares resources for individuals, family members, physicians, or anyone looking for information related to alcohol use and alcohol use disorder, including Rethinking Drinking and the NIAAA Treatment Navigator. He emphasizes the importance of motivation (e.g., motivational interviewing) rather than confrontation and seeing your doctor, or encouraging your loved one to see their doctor, for a physical as a good first step in addressing any issues related to alcohol use. Dr. Koob finally explains his motivation for continuing his work in this field. In addition to serving as director of NIAAA, Dr. Koob is also a senior investigator at the Intramural Research Program of the National Institute on Drug Abuse where he directs the Neurobiology of Addiction Laboratory in the Integrative Neurosciences Research Branch. As an authority on alcohol use disorder, drug addiction, and stress, he has contributed to our understanding of the neurocircuitry associated with the acute reinforcing effects of alcohol and drugs and the neuroadaptations of the reward and stress circuits associated with the transition to dependence. He has published over 800 peer reviewed papers; mentored 13 PhD students, 85 post-doctoral fellows, and 11 K99's (Pathway to Independence Award postdoctoral researchers); and authored several books including the Neurobiology of Addiction. He received his PhD in behavioral physiology from Johns Hopkins University in 1972. He did post-doctoral studies at Walter Reed Army Institute of Research and the University of Cambridge (England). He subsequently held positions at The Salk Institute and the Scripps Research Institute. Dr. Koob is the recipient of many honors, including membership in the National Academy of Medicine (USA) and award of the Legion of Honor (France). Nel 1840, quando lei aveva due anni, la sua famiglia emigrò negli Stati Uniti, stabilendosi nella città di Utica, nello Stato di New York. Suo padre cambiò il cognome da Koob a Cope, dopo aver ottenuto la cittadinanza americana, il che comportava l'estensione della stessa all'intera famiglia.Da adolescente, Barbara prese a lavorare in una fabbrica per dare un a mano ai bisogni della famiglia, cresciuta nel frattempo di altri tre fratelli e con il padre invalido. Frequentando la Scuola parrocchiale di San Giuseppe a Utica, poté ricevere la Prima Comunione nel 1848.In quest'ambiente fiorì la sua vocazione allo stato religioso, ma tale desiderio dovette essere accantonato, perché le condizioni economiche della famiglia non permettevano il suo allontanamento. Solo a 24 anni poté entrare nell'Istituto delle Suore del Terz'Ordine Francescano di Syracuse, dove, dopo il noviziato, emise la professione religiosa con il nome di suor Marianna.L'apostolato di quelle suore, fra l'altro, consisteva nell'educazione dei figli degli emigrati tedeschi. Suor Marianna, quindi, apprese la lingua originaria dei suoi genitori e fu incaricata di dirigere una nuova scuola specifica.Per le sue doti intellettuali e per la sua generosa dedizione svolse delicati incarichi nella sua Congregazione, fra i quali la cura dei poveri da lei prediletti, nei due ospedali di Santa Isabella a Utica e San Giuseppe a Syracuse (1869). Fu eletta Madre Provinciale nel 1877 e riconfermata all'unanimità nel 1881.Due anni dopo, nel 1883, era Madre generale quando le giunse una richiesta del vescovo di Honolulu, che a sua volta girava alle suore una petizione del re delle isole Sandwich, nell'Oceano Pacifico (le attuali Hawaii, 50° Stato degli USA dal 1959), il quale chiedeva di avere infermiere per i lebbrosi abbandonati del regno.La situazione era critica e già 50 comunità religiose avevano rifiutato la petizione reale. Un religioso, padre Damiano de Veuster, aveva scelto di vivere in quelle condizioni precarie, ma faceva presente che sarebbero state necessarie delle suore.I malati, strappati dai familiari e dai loro villaggi, venivano portati nell'isola di Molokai, dove non esistevano edifici idonei né assistenza sanitaria. Si sarebbe dovuto costruire un ospedale e soprattutto instaurare una severa terapia igienica generale, specie per i figli più piccoli dei lebbrosi, che avevano seguito le madri ed erano bisognosi di educazione.Madre Marianna scelse sei suore fra le 25 che si erano offerte e partì con loro per fondare una Missione delle Suore del Terz'Ordine Francescano nelle Sandwich; le accompagnò prima a Honolulu e poi a Molokai. Collaborò quindi con il governo locale a istituire ospedali in varie isole dell'arcipelago.Padre Damiano, che aveva contratto la lebbra nel 1884, venne assistito dalle suore come gli altri malati: morì nel 1889. Tutto il lavoro organizzativo passò a madre Marianna, la quale, anche per le minacce delle altre suore di tornarsene con lei in America, dovette restare a Molokai per salvare la Missione e dimettersi dal suo incarico.Non tornò più in America e restò a servire i lebbrosi per quasi 30 anni. Fondò due case separate per i figli dei lebbrosi, tenuti nella più grande igiene, così che una volta adulti potessero essere inseriti sani nella società.Madre Marianna di Molokai, come ormai veniva chiamata, conosceva uno per uno i malati e li chiamava per nome. Li istruiva a coltivare quell'arida terra facendo spuntare arbusti, fiori e alberi, ma soprattutto ridonando loro la dignità di esseri umani non più emarginati e inutili. Gli storici la descrissero come «una religiosa esemplare con un cuore straordinario». Morì a Molokai il 9 agosto 1918 a 80 anni, di morte naturale.

Biden reiterated a statement from the FDIC and Treasury Department during public remarks Monday morning, telling reporters that the federal government would guarantee depositors at the banks access to their funds. He added that no such protection is being offered to the banks' investors, however. His administration has also fired the leadership at both banks. "No losses will be borne by the taxpayers," Biden emphasized. "Instead, the money will come from the fees that banks pay into the deposit insurance fund. Because of the actions that our regulators have already taken, every American should feel confident that their deposits will be there if and when they need them." Biden went on to call on Congress to pass legislation to "strengthen rules" on banks to prevent failures like SVB and Signature.The Santa Clara, California-based band collapsed last week and is now under the control of federal regulators. The SVB had been the 16th-largest bank in the U.S. prior to the bank run that led to its downfall. Anxious depositors rushed to withdraw their money over concern for the bank’s health, causing its collapse, which may serve as "an extinction-level event for startups," according to Y Combinator CEO Garry Tan. The Biden administration had earlier assured SVB depositors that they would have access to all of their funds on Sunday. The joint statement from the Treasury Department, Federal Reserve and the Federal Deposit Insurance Corporation (FDIC) also emphasized that the action would come at no cost to US taxpayers. "Today we are taking decisive actions to protect the U.S. economy by strengthening public confidence in our banking system," the joint statement said. "This step will ensure that the U.S. banking system continues to perform its vital roles of protecting deposits and providing access to credit to households and businesses in a manner that promotes strong and sustainable economic growth." He has said doctors should be sued for performing sex changes on children and suspended a state attorney refusing to adhere to the child sex change ban. DeSantis also requested public universities report how many students they treated for “gender dysphoria,” and in October, the Florida Board of Medicine voted to ban sex change surgeries and hormone therapy for children under 18. As the pandemic came to an end, they seem to be drinking more than ever. In 2020, during the pandemic, we saw a continuing decline in binge drinking among college-age students. Most were isolated or at home as classes moved online. Group activities or parties where drinking would be prevalent simply were not happening. “In 2021, there has been an uptick, particularly among women. Now it turns out on college campuses women are actually binge drinking more than men, for the first time in history,” said Dr. George F. Koob, director of the National Institute on Alcohol Abuse and Alcoholism. Koob said while drinking overall was down during the pandemic, deaths associated with alcohol were up 25% for not just college students but the general population. Koob believes isolation contributed to mental health problems and substance abuse across the board. However, now we’re back. People are going to parties, and colleges and bars are open again. Restaurants are serving bottomless brunch, and wineries and breweries are ready to pour. “It’s what we call the alcohol deprivation effect. People tend to really rebound in drinking after a period of not drinking.” Koob said. “We are a little concerned that this spring and spring break is going to be a return to a good amount of binge drinking. I just want to caution everyone that when you start hitting the binge drinking level you start doing really bad things to your body.” So, where does this leave this new group of women who are binge drinking more than ever before? Koob said knowing what a standard drink is and your limit is important to taking care of your health. As social media trends encourage dangerous “hacks” to binge drinking, including the viral “BORGs” or “blackout rage gallons” which combine water, electrolytes, and vodka, TikTok is specifically pushing the misconception that this drink will keep you hydrated while drinking copious amounts of liquor. https://nypost.com/2023/03/12/fight-among-200-brawlers-at-louisiana-hs-ends-in-10-arrests/ Massive fight among 200 students, parents at Louisiana high school ends in 10 arrests An explosive fight at a Louisiana high school among nearly 200 students and parents ended with several cops injured and at least 10 arrests. Swarms of police officers responded early March 8 to a “major campus disturbance” at East Baton Rouge Readiness Alternative School, finding a loaded gun abandoned on the ground, WBRZ reported. Officials say one fight broke out and escalated, spilling out into the school’s courtyard and gym. By the time police arrived, there were about 200 people involved in brawls across the campus. Videos of the chaos taken inside the school’s gym show chairs getting tossed and students being detained by law enforcement. One shocking video shows a sheriff’s deputy forcefully hitting a student’s face into a brick wall. The officer was trying to arrest a 17-year-old student, authorities said. Police said the teen had punched the officer in the face and tried to bite him, which led to the aggressive encounter captured on video. The teen was among those charged with battery on an officer and resisting arrest. Officials have not yet determined what triggered the massive melee at the school, which enrolls students who’ve been previously suspended or expelled from other district schools. The NAACP released a statement Wednesday saying it was reviewing videos from the incident to decide if the aggressive police response captured on video was appropriate. The fight allegedly started with a small group of students but escalated into a larger clash involving parents and students fighting police as well. One witness told WBRZ the brawl erupted when a girl who wasn’t a student at the school and her mom showed up to fight another girl — and that it escalated from there. Students were allegedly locked inside the gym during the fight. One responding police officer sustained injuries including a broken hip and head lacerations. Also among those arrested were three students charged with battery on a police officer, a felony. Five other students were arrested for disturbing the peace, including an 18-year-old girl who was arrested on counts of unlawful disruption of the operation of a school and resisting arrest, and a 17-year-old girl hit with disturbing the peace and resisting arrest. All of those arrested were students between 15 and 18. None of the parents or other adults at the scene were taken into custody. He added that no such protection is being offered to the banks' investors, however. His administration has also fired the leadership at both banks. The Biden administration had earlier assured SVB depositors that they would have access to all of their funds on Sunday. The joint statement from the Treasury Department, Federal Reserve and the Federal Deposit Insurance Corporation (FDIC) also emphasized that the action would come at no cost to US taxpayers. "Today we are taking decisive actions to protect the U.S. economy by strengthening public confidence in our banking system," the joint statement said. "This step will ensure that the U.S. banking system continues to perform its vital roles of protecting deposits and providing access to credit to households and businesses in a manner that promotes strong and sustainable economic growth." Most were isolated or at home as classes moved online. Group activities or parties where drinking would be prevalent simply were not happening. “In 2021, there has been an uptick, particularly among women. Now it turns out on college campuses women are actually binge drinking more than men, for the first time in history,” said Dr. George F. Koob, director of the National Institute on Alcohol Abuse and Alcoholism. Koob said while drinking overall was down during the pandemic, deaths associated with alcohol were up 25% for not just college students but the general population. Koob believes isolation contributed to mental health problems and substance abuse across the board. However, now we’re back. People are going to parties, and colleges and bars are open again. Restaurants are serving bottomless brunch, and wineries and breweries are ready to pour. “It’s what we call the alcohol deprivation effect. People tend to really rebound in drinking after a period of not drinking.” Koob said. “We are a little concerned that this spring and spring break is going to be a return to a good amount of binge drinking. I just want to caution everyone that when you start hitting the binge drinking level you start doing really bad things to your body.” So, where does this leave this new group of women who are binge drinking more than ever before? Koob said knowing what a standard drink is and your limit is important to taking care of your health. Swarms of police officers responded early March 8 to a “major campus disturbance” at East Baton Rouge Readiness Alternative School, finding a loaded gun abandoned on the ground, WBRZ reported. Officials say one fight broke out and escalated, spilling out into the school’s courtyard and gym. By the time police arrived, there were about 200 people involved in brawls across the campus. Videos of the chaos taken inside the school’s gym show chairs getting tossed and students being detained by law enforcement. One shocking video shows a sheriff’s deputy forcefully hitting a student’s face into a brick wall. The officer was trying to arrest a 17-year-old student, authorities said. Police said the teen had punched the officer in the face and tried to bite him, which led to the aggressive encounter captured on video. The teen was among those charged with battery on an officer and resisting arrest. Officials have not yet determined what triggered the massive melee at the school, which enrolls students who’ve been previously suspended or expelled from other district schools. The NAACP released a statement Wednesday saying it was reviewing videos from the incident to decide if the aggressive police response captured on video was appropriate. The fight allegedly started with a small group of students but escalated into a larger clash involving parents and students fighting police as well. One witness told WBRZ the brawl erupted when a girl who wasn’t a student at the school and her mom showed up to fight another girl — and that it escalated from there. Students were allegedly locked inside the gym during the fight. One responding police officer sustained injuries including a broken hip and head lacerations. Also among those arrested were three students charged with battery on a police officer, a felony. Five other students were arrested for disturbing the peace, including an 18-year-old girl who was arrested on counts of unlawful disruption of the operation of a school and resisting arrest, and a 17-year-old girl hit with disturbing the peace and resisting arrest. All of those arrested were students between 15 and 18. None of the parents or other adults at the scene were taken into custody. To elucidate anatomical regions and cell types that underlie mutant ATXN1-induced disease phenotypes, we developed a floxed conditional knockout mouse model (f-ATXN1146Q/2Q) having mouse Atxn1 coding exons replaced by human exons encoding 146 glutamines. F- ATXN1146Q/2Q mice manifest SCA1-like phenotypes including motor and cognitive deficits, wasting, and decreased survival. CNS contributions to disease were revealed using ATXN1146Q/2Q;Nestin-Cre mice, that showed improved rotarod, open field and Barnes maze performances. Striatal contributions to motor deficits were examined using f- ATXN1146Q/2Q;Rgs9-Cre mice. Mice lacking striatal ATXN1146Q/2Q had improved rotarod performance late in disease. Muscle contributions to disease were revealed in f- ATXN1146Q/2Q;ACTA1-Cre mice which lacked muscle pathology and kyphosis seen in f- ATXN1146Q/2Q mice. Kyphosis was not improved in f-ATXN1146Q/2Q;Nestin-Cre mice. Thus, optimal SCA1 therapeutics will require targeting mutant ATXN1 toxic actions in multiple brain regions and muscle. One drink is defined as 12 ounces of beer, 5 ounces of wine, or 1.5 ounces of 80-proof liquor. So remember that a mixed drink or full glass of wine are probably more than one drink. Abstinence from alcohol Abstinence from alcohol is the best choice for people who take medication(s) that interact with alcohol, have health conditions that could be exacerbated by alcohol (e.g. liver disease), are pregnant or may become pregnant or have had a problem with alcohol or another substance in the past. Benefits of “low-risk” drinking Following these guidelines reduces the risk of health problems such as cancer, liver disease, reduced immunity, ulcers, sleep problems, complications of existing conditions, and more. It also reduces the risk of depression, social problems, and difficulties at school or work.

Koob, Ph.D., is an internationally-recognized expert on alcohol and stress, and the neurobiology of alcohol and drug addiction. He is the Director of the National Institute on Alcohol Abuse and Alcoholism (NIAAA), where he provides leadership in the national effort to reduce the public health burden associated with alcohol misuse. As NIAAA Director, Dr. Koob oversees a broad portfolio of alcohol research ranging from basic science to epidemiology, diagnostics, prevention, and treatment. Dr. Koob earned his doctorate in Behavioral Physiology from Johns Hopkins University in 1972. Prior to taking the helm at NIAAA, he served as Professor and Chair of the Scripps' Committee on the Neurobiology of Addictive Disorders and Director of the Alcohol Research Center at the Scripps Research Institute. Early in his career, Dr. Koob conducted research in the Department of Neurophysiology at the Walter Reed Army Institute of Research and in the Arthur Vining Davis Center for Behavioral Neurobiology at the Salk Institute for Biological Studies. He was a post-doctoral fellow in the Department of Experimental Psychology and the MRC Neuropharmacology Unit at the University of Cambridge. Dr. Koob began his career investigating the neurobiology of emotion, particularly how the brain processes reward and stress. He subsequently applied basic research on emotions, including on the anatomical and neurochemical underpinnings of emotional function, to alcohol and drug addiction, significantly broadening knowledge of the adaptations within reward and stress neurocircuits that lead to addiction. This work has advanced our understanding of the physiological effects of alcohol and other substance use and why some people transition from use to misuse to addiction, while others do not. Dr. Koob has authored more than 650 peer-reviewed scientific papers and is a co-author of The Neurobiology of Addiction, a comprehensive textbook reviewing the most critical neurobiology of addiction research conducted over the past 50 years. Dr. Koob is the recipient of many prestigious honors and awards for his research, mentorship, and international scientific collaboration. In 2018 Dr. Koob received the E.M. The International Conference of Secular AA celebrates the science of addiction/recovery and folk-wisdom of AA with a 1/2 day virtual free mini-conference December 5, 2020. Aujourd'hui je reçois Alexandre Bruneau le CEO et cofondateur de Koober. Après des études en marketing et en finance, Alexandre a toujours été attiré par l'entrepreneuriat. Il rencontre Alexandre Mulliez qui réalise des fiches de lectures pour des ouvrages. Alexandre les utilise régulièrement. Tous deux sont passionnés de lecture et ont l'idée de monter Koober, qui sera, dans un premier temps une marketplace qui vend des fiches de lecture où les utilisateurs écrivent eux-mêmes les résumés de livres. Mais très rapidement, ils se rendent compte que ces fiches ne sont pas homogènes, que les auteurs n'écrivent pas ces fiches de la même façon.  Bref, ça ne fonctionne pas. Nous sommes en 2015, un concurrent allemand Blinkist cartonne en proposant des résumés de livres en 15 mn. Alexandre Bruneau et son associé trouve l'idée sympa, connexe mais pas suffisante. Ils font pivoter leur modèle vers un nouveau concept : Un résumé d'un livre en 20 mn, mais avec une approche plus précise, plus cognitive, plus scientifique que la solution que propose Blinkist. La totalité des fiches de lecture « 1ère version » est effacée pour faire place à la nouvelle mouture. Rapidement ils créent le cahier des charges, contactent des auteurs, acteurs, des psychologues pour établir le meilleur Koob (nom donné au résumé) possible. Le Koob n'est pas un simple résumé, il se concentre sur l'essentiel des idées véhiculées dans le livre pour en extraire un contenu qui tient en 20 mn. Pourquoi 20 minutes ? C'est le temps moyen d'un trajet, et c'est surtout la limite haute de concentration pour que ces idées soient retenues. Ce qui est amusant, c'est qu'à de nombreuses reprises, le terme de capsule revient dans la conversation, comme si on injectait une capsule de savoir chez l'utilisateur, façon Matrix. Je fais d'ailleurs le parallèle avec Feed, un épisode excellent que je vous conseille et qui est à peu près sur le même principe : être efficace pour une population de clients qui ne veulent pas perdre de temps. Sur koober.com, il y a plus de 1500 livres qui traitent de la finance, du bonheur, de la méditation, d'économie, d'entrepreneuriat, histoire, biographie… il y a en pour tous les gouts et toutes les envies. Tout au long de l'épisode, on ressent qu'Alexandre a des valeurs qu'il veut transmettre. Koober a une double mission : redonner le gout de lire, et faire progresser les utilisateurs dans le domaine choisi. Plus récemment, Koober a créé des parcours de formation, qui se terminent par des Quizz qu'il faut valider pour passer au niveau supérieur. Dans une seconde partie, Alexandre Bruneau nous explique sa vision de l'entreprise. Bien qu'il soit baigné au quotidien par ses koobs et la formation qu'ils dispensent, Alexandre utilise des méthodes de management japonaises. Il est coaché par un Sensei qui le suit, a effectué des voyages au Japon pour comprendre le Lean à la japonaise, et l'appliquer dans son entreprise. Aujourd'hui Koober compte plus de 750 000 abonnées, avec une croissance à deux chiffres tous les mois.
