J., Pulst, S. M. Abstract: Staufen1 (STAU1) is a multifunctional RNA binding protein that controls mRNA degradation and subcellular localization. STAU1 interacts with the ATXN2 protein, that is polyglutamine expanded in spinocerebellar ataxia type 2 (SCA2). We previously showed that STAU1 is elevated and aggregated in cells from SCA2 patients, cells from amyotrophic lateral sclerosis (ALS) patients, and in SCA2 and ALS mouse models. We also found that reduction of STAU1 abundance in vivo by genetic interaction improved motor behavior in an SCA2 mouse model, normalized the levels of several SCA2-related proteins, and reduced aggregation of polyglutamine-expanded ATXN2. Here we developed antisense oligonucleotides (ASOs) lowering STAU1 expression toward developing a therapeutic that may be effective for treating SCA2 and ALS. We performed a screen of 118 20mer phosphorothioate 2-prime-O-methoxyethyl (MOE) ASO gapmers targeting across the STAU1 mRNA coding region for lowering STAU1 expression in HEK-293 cells. ASO hits lowering STAU1 by greater than 45 % were rescreened in SCA2 patient fibroblasts, and 10 of these were tested for lowering STAU1 abundance in vivo in a new BAC-STAU1 mouse model. This identified efficacious ASOs targeting human STAU1 in vivo that normalized autophagy marker proteins, including ASO-45 that also targets mouse Stau1. When delivered by intracerebroventricular (ICV) injection, ASO-45 normalized autophagy markers and abnormal mRNA abundances in cerebella of ATXN2-Q127 SCA2 mice, as well as ChAT, NeuN and cleaved caspase-3 in spinal cord of Thy1-TDP-43 transgenic mice. Targeting STAU1 may be an effective strategy for treating ALS and SCA2 as well as other disorders characterized by its overabundance. Copy rights belong to original authors. I'm Josh Cooperman and this is Convo By Design with designer, Anelle Gandelman. Former student of The New School of Interior Design with a degree in Fine Art from Parsons School of Design and a certificate in American art & antiques from Sotheby's. I bet you know where we're going with this one. Yep, art and design. We talk about it quite a bit. I think it's important to cover this subject for a number of reason. In my experience, designer tend to shy away form fine art, antiques and the perceived high expense of such endeavors in design. Why? Because of cost, overall percentage of the design budget, lack of quality control, delivery issues, but the main reason? I suspect it is due to a lack of comfort with the idea that there is rarity to this and an inability to circle back and return the product if it doesn't fit perfectly. But think about it this way, art and antiques can be woven into a design concept in so many way that it can raise the level of design exponentially. You can use art as a feature, accessory, complimentary item, balance to low in the high-low scenario. You can use antiques to balance a modern or contemporary feel but most importantly to me anyway is that a skilled designer can use art and antiques to further craft the story behind the design itself making it a unique story exclusive to their clients. That is what many if not most clients are looking for now. In November of 2020, the state of New York released its regulations concerning hemp, and much to the surprise and chagrin of hemp farmers in the state, the new regulations outlawed the sale of industrial hemp flower. The hemp flower market is lucrative for farmers, and many small, independent operations in the state grow specifically for the flower market. And just as their harvest was coming in last year, farmers were aghast when the state shut down the market and the revenue potential farmers were counting on. As one might imagine, hemp farmers in New York want desperately to get this rule overturned. One such farmer is Allan Gandelman. He is a hemp farmer and owner of Main Street Farms and the CBD brand, Head & Heal. He is also the founder and president of the New York Cannabis Growers and Processors Association. On this episode of the podcast, we talk to Gandelman about New York's ban on hemp flower sales and what effect that will have on the industry and the farming community. Sua família toda tem um histórico musical, com um avô maestro e compositor, mãe bandolinista, um pai que tocava violão e um irmão mais velho, Paulo, contrabaixista profissional. Paulo, junto do amigo Chiquito Braga, estimulou Toninho a aprender o violão na infância, enquanto tocava o instrumento junto de Chiquito. Toninho compôs sua primeira canção aos 13 anos com a irmã Gilda, e três anos depois tinha uma composição dos dois, “Flor que cheira saudade”, foi gravada pelo cantor Márcio José e o Conjunto de Aécio Flávio - que incluía o irmão Paulo. Em 1967 participou do "2º Festival Internacional da Canção", concorrendo com a música Maria Madrugada, em parceria com Júnia Horta; e em 1969, no "4º Festival Internacional da Canção" concorreu com a música Nem é carnaval, com seu parceiro Márcio Borges. Com Nivaldo Ornelas faz sua estreia em estúdio em 1969 e, no mesmo ano, toca com Milton Nascimento pela primeira vez. Transfere-se para o Rio de Janeiro no início dos anos 1970, quando passa integrar o grupo A Tribo, juntamente com a cantora Joyce, Nelson Angelo, Novelli e Naná Vasconcelos, chegando a gravar algumas faixas no disco Posições (Odeon). Nesta mesma época passou a tocar com Elis Regina e participou das gravações do álbum Clube da Esquina, de Milton Nascimento. Com o seu trabalho já reconhecido nacionalmente, passou a integrar as bandas de Edu Lobo, Gal Costa, Maria Bethânia e Nana Caymmi, no instante em que sua música era percebida por músicos estrangeiros, como Pat Metheny, que sofreu enorme influência de Toninho Horta. Em 1989, com Diamond Land, finalmente toma o mercado norte-americano, mudando-se para Nova Iorque. Por seu talento extraordinário, Toninho tocou com músicos renomados do jazz mundial, como Sergio Mendes, Gil Evans, Flora Purim, Astrud Gilberto, Naná Vasconcelos, Paquito De Rivera, Airto Moreira, Wayne Shorter, Eliane Elias, Herbie Hancock, Keith Jarrett, George Benson, Pat Metheny, Flavio Sala e muitos outros. We interviewed Allan Gandelman early last year and were really impressed by his story. He was a former schoolteacher turned organic vegetable farmer and is now in his second year of hemp farming. He has two vertically integrated businesses and is president of the New York Cannabis Growers and Processors Association.  We're not sure how he finds time to do everything he does, but 2019 has been a banner year for him and he's excited for 2020. On this week's episode we have a conversation with Allan Gandelman of New York Hemp Oil about growing industrial hemp for CBD.  Allan's journey to industrial hemp began with a love and curiosity for organic farming curated during his time as a high school social studies teacher in the New York public school system.  He observed nutritional deficiencies in what his students were being served for lunch that were difficult for him to ignore. Allan decided that he could have a greater impact as an organic vegetable farmer and has built a successful business doing so.  Main Street Farms began on a single acre and is now pumping out high quality, organic produce from forty-five acres of open field and an acre of covered greenhouse space, year round!   The success of Main Street Farms, and his considerable talent for business, served as a springboard for Allan into industrial hemp.  The decision to dive into CBD arose from a personal experience, however, that helped Allan focus his strategy and formulate his commitment to producing a high quality, organic, full spectrum CBD oil. Allan Gandelman raises 45 acres of crops at Main Street Farms in central New York state with his partner, BobCat. With twenty employees in its eighth year in business, Main Street Farms sells through a CSA, farmers market, and wholesale accounts. Main Street Farms got its start in 2011 with an acre of production and an aquaponics set up, so they’ve grown a lot in the last eight years and Allan and I talk a lot about the process of scaling up their operation and finding their way with different mixes of enterprises and marketing outlets, and how that has meshed with meeting the needs of people on the farm.
