Abstract: The role of myelination for axonal conduction is well-established in projection neurons but little is known about its significance in GABAergic interneurons. Myelination is discontinuous along interneuron axons and the mechanisms controlling myelin patterning and segregation of ion channels at the nodes of Ranvier have not been elucidated. Protein 4.1B is implicated in the organization of the nodes of Ranvier as a linker between paranodal and juxtaparanodal membrane proteins to the spectrin cytoskeleton. In the present study, 4.1B KO mice are used as a genetic model to analyze the functional role of myelin in Lhx6-positive parvalbumin and somatostatin neurons, two major classes of GABAergic neurons in the hippocampus. We show that deletion of 4.1B induces disruption of juxtaparanodal K+ channel clustering and mislocalization of nodal or heminodal Na+ channels. Strikingly, 4.1B-deficiency causes loss of myelin in GABAergic axons in the hippocampus. In particular, stratum oriens O-LM cells display severe axonal dysmyelination and a reduced excitability. This reduced excitability is associated with a decrease in occurrence probability of small amplitude synaptic inhibitory events on pyramidal cells. In contrast, stratum pyramidale fast-spiking basket cells do not appear affected. The aberrant myelination of hippocampal interneurons is also correlated with impairment of spatial memory in 4.1B KO mice. In conclusion, our results indicate a class-specific effect of dysmyelination on the excitability of hippocampal interneurons associated with a functional alteration of inhibitory drive and impairment of spatial memory. Rio Pearlstein is a 17-year-old golfer at Milton Academy in Massachusetts and has been a rising star in youth golf since he was 11. Two years ago, he was his region's player of the year. That was the same year he came out as a trans boy while pushing through being at the top of his game, and continuing to press toward a goals of playing golf in college which has been his dream. It is a dream that has him willing to put his other life goal on hold for in a sense. He is delaying the next stage of transition to reach that summit, with all the difficulties therein. Karleigh Webb sat down with this young talent to look at navigating sport as a trans boy playing a girls team, navigating being trans in a sporting structure that is cis-centered, and finding the love of the game and competition through it all. Nanome scientists are joined by Dr. Robert Pearlstein and Dr. Hongbin Wan, of Novartis, who share with us their recent publication on the application of biodynamics theory to characterize the structural dynamics of substrate-induced activation under non-equilibrium conditions. In this archive episode, Dennis delves into what situations call for escalating a mere field inquiry to an investigative detention. Recorded on 12/29/2017. State v Lurdes Rosario 2017 First and foremost, we must address whether and at what point defendant's interaction with the police officer escalated from a field inquiry into an investigative detention. Then we must assess whether reasonable and articulable suspicion supported the detention's restriction on defendant's freedom of movement. The encounter took place on a May evening when defendant was in her car, which was parked lawfully, head-on in a lined parking space directly outside her apartment. The car's engine was off. The officer positioned his patrol car perpendicularly behind defendant's to box in defendant's car and engaged his vehicle's rooftop, right-side “alley” light to shine at her car. The officer then exited his patrol car and approached the driver's-side door of defendant's car to address her. We conclude that no objectively reasonable person in those circumstances would have felt free to leave. Under the totality of the circumstances, a reasonable person would feel the constraints on her freedom of movement from having become the focus of law enforcement attention. Accordingly, we hold that an investigative detention had begun. Because we also conclude that reasonable and articulable suspicion did not ripen prior to the officer's subsequent exchanges with defendant, we reverse the judgment under review. State v Pearlstein 1985 - Failing to provide documents constitutes Obstructing the Admin of Law As a result, Lizzano crossed the street to the patrol car to obtain his summons book. Then he asked Ms. Perlstein for her license and registration which she refused to tender. She continued to spew a barrage of comments at Lizzano. Lizzano told her that if she did not show her license, registration and insurance card he would issue her three more summonses. She persisted in her refusal. Then she said that she was going to see the Chief of Police to see what he had to say about this incident. With the car door open, she picked up her keys from the seat, put them in the ignition, turned on the car and put it in reverse. Lizzano was standing in the open door and knew he could be hurt if the car moved so he reached in and grabbed the keys. Thereafter, he placed her under arrest.

ABOUT DR. ZVI PEARLSTEIN: dr-zvi.jpg Dr. Zvi Pearlstein is a Doctor, Father, Speaker, Author, Inventor... And Trumpeter. From a young age, Dr. Zvi had a deep-seated need and calling to find life’s meaning and purpose to understand and solve pain. His mother Celia passed in 2019 leaving behind writings and poetry of her emotional experiences during the Holocaust in Poland. In 1976 Dr. Zvi received his BA in Biochemistry from Princeton University. In 1981 he received his MD from New York University School of Medicine. He completed a 5-year residency in Orthopedic Surgery at SUNY Downstate in Brooklyn in 1986. As a Major in the United States Air Force, Dr Zvi spent three years practicing orthopedic surgery at Clark Air Base in the Philippines and one year at Andrews Air Force Base in Wash. DC. He did active duty service during Persian Gulf War I. While serving in the Air Force he was a marksman shooter and was awarded the Sikorsky Helicopter Rescue Award. Throughout his career he was looking for truth and often felt that certain problems needed better wellness and lifestyle answers and solutions. Capitalism is a recurring theme among the ever-growing list of Democratic presidential candidates. But many Americans of all political stripes have concerns about our free market economy and whether it is working for them, according to Steven Pearlstein, a Pulitzer-Prize winning columnist for The Washington Post and author of “Can American Capitalism Survive: Why Greed Is Not Good, Opportunity Is Not Equal, and Fairness Won’t Make Us Poor.” We talk with Pearlstein about the importance of fairness in economic growth, and consider some ways to reinvent capitalism.
Elise Pearlstein has dedicated her life to non-fiction film, first as award-winning producer, and since 2013, as SVP of Documentary at Participant Media. Her career in docs wasn't always a foregone conclusion--she used to keep lists of other jobs she could do--but now says there's no place she'd rather be. She talks to Matt Holzman about an industry in transition and some of Participant's newest projects.
Jesse Pearlstein graduated from James Madison University in 2006 with a degree in psychology. He spent the next several years as an AmeriCorps volunteer in California and subsequently a Peace Corps Volunteer in the Dominican Republic. Jesse began working as a counselor in 2010 serving at risk youth and their families and graduated with his master's in social work in 2013. He has worked in schools, residential programs, community based settings providing counseling and mental health services to children, youth and adults. Jesse works with both children and adults presenting with a wide range of issues ranging from ADHD, Depression, and Trauma, among others. He utilizes a client centered approach to assist people with cultivating their personal strengths to aid them in their healing process. In his spare time he enjoys being physically active and likes to perform as an amateur stand-up comedian. After his recent meeting with Russian president Vladimir Putin and comments about Russian interference in the 2016 elections, President Donald Trump has sparked a new controversy relating to the constitutional definition of treason, and to what extent actions taken in support of a foreign nation might constitute treason. Jeffrey Rosen leads a discussion about the Treason Clause of the Constitution, what it means, and how it has been interpreted. Paul Crane is an assistant professor of law at the University of Richmond Law School. Previously, he served as a Bigelow Fellow at the University of Chicago Law School. He has also worked as an Assistant United States Attorney in the District of Columbia, as a Bristow Fellow for the Office of the Solicitor General of the United States, and clerked for Chief Justice Roberts on the U.S. Supreme Court. Deborah Pearlstein is a professor of law at the Benjamin N. Cardozo School of Law at Yeshiva University. Previously, she served in the White House from 1993 to 1995 as a Senior Editor and Speechwriter for President Clinton, and served as the founding director of the Law and Security Program at Human Rights First, where she worked on military commission trials at Gitmo.
