Abstract: Environmental insults that activate the maternal immune system are potent primers of developmental neuropathology and maternal immune activation (MIA) has emerged as a risk factor for neurodevelopmental and psychiatric disorders. Animal models of MIA provide an opportunity to identify molecular pathways that initiate disease processes and lead to neuropathology and behavioral deficits in offspring. MIA-induced behaviors are accompanied by anatomical and neurochemical alterations in adult offspring that parallel those seen in affected human populations. We performed transcriptional profiling and neuroanatomical characterization in a time course across mouse embryonic cortical development, following MIA via single injection of the viral mimic polyinosinic:polycytidylic acid (polyI:C) at E12.5. Transcriptional changes identified in the cortex of MIA offspring at E17.5 were validated and mapped to cortical neuroanatomy and cell types via protein analysis and immunohistochemistry. MIA induced strong transcriptomic signatures, including induction of genes associated with hypoxia, immune signaling, and angiogenesis. The acute response identified 6h after the MIA insult was followed by changes in proliferation, neuronal and glial differentiation, and cortical lamination that emerged at E14.5 and peaked at E17.5. Decreased numbers of proliferative cell types in germinal zones and alterations in neuronal and glial cell types across cortical lamina were identified in the MIA-exposed cortex. MIA-induced transcriptomic signatures in fetal offspring overlap significantly with perturbations identified in neurodevelopmental disorders (NDDs), and provide novel insights into alterations in molecular and developmental timing processes linking MIA and neuropathology, potentially revealing new targets for development of novel approaches for earlier diagnosis and treatment of these disorders. We’ve had a bit of a conversation to and fro through LinkedIn. We’ve had a little bit of a look at each other’s leadership aspirations and we are both super-enthusiastic about leadership as a whole and have big visions about what we want to create and what, but I’m not going to spoil that part yet until we get into that. I’ll let Jay share that one. Nathan Simmonds: First of all, let me introduce him completely by some of his accolades and his current celebratory points along his journey. So he’s an award-winning lecturer, consultant and public speaker. He has the vision to enhance the practice of leadership at a global level and he’s already doing this through his training of aspiring managers in Mauritius, Malaysia, Indonesia, Singapore and Morocco which, in itself, is pretty astounding for any leadership trainer. In fact, he’s already clocking up over 7,000 leaders so far that he has supported, guided and mentored. Amongst all this, he’s also achieved five fellowships which, in itself, is a unique accomplishment. And one of the interesting places that he likes to start his conversations is about how he likes to introduce himself as the magician. But we’re going to get into that in a minute. We’re not going to cover that yet. Nathan Simmonds: But first of all, Jay, I just want to say a massive thank you for being here. I first met Susan at her flat in Walthamstow around 10 years ago, when I was asked to help her with her insurance claim for dry rot. I remember at the time Susan telling me she was a comedian—as I’d never met one in the flesh before, so it really stuck with me. Perhaps not the most auspicious start. Since then I’ve tried to keep up with Susan’s thriving career as a much in demand stand up comic. Not only does Susan gig all over the UK, but she also runs and is resident MC at Red Imp in Walthamstow, where she has brought many famous faces to E17, including Alan Carr, Stephen K Amos, Russell Kane, Phil Jupitus, Arthur Smith and many more. I was just leaving to meet Susan for this recording when she asked if her good friend and multi lingual stand up comedian, Maureen Younger, could join us. Well, I thought, why not indeed. Two for the price of one. I love this meandering, somewhat random off the wall chat and I hope it brightens your day as much as it did mine. Susan’s been described as a cross between Frankie Boyle and Julie Walters, so stand by your beds for a bit of a raucous ride. “Obviously the more experienced you are, how good you are, you can work yourself out of situations—but they’ll be one gig at some point in the future where they will hate you—where you will die on your ass, and you just don’t know when that’s going to be.” Dying on stage, every comic has it happen. It always happens right after you feel like you’ve been nailing your material. Susan and Maureen have both experienced this phenomenon—but they’re quick to note that it happens to every comic at some point. In fact, they feel a little better when they see someone big on stage fail to really get the crowd on their side. It goes to show that the struggle to perform live on stage is real, and there are a host of factors that go into a successful bout of stand up. [16:26] Believe it or not – the physicality of a room affects a comic’s material and delivery. The difference in how a comedian performs while in a massive theatre vs an up-close café can make or break their set. Susan and Maureen note that large venues are often more difficult, because the gap between the comedian and audience is physically wider—eating up the comic’s energy if they are depending on their actions to help deliver their jokes. Susan and Maureen share some horror story events, including a charity event with a ten minute speech beforehand about the organizers friend—while people were playing beer ping pong in the back. “As a male comic—you’re given more rope to hang yourself.” [20:07] Comics only have so much time to win a crowd over. Susan and Maureen agree that for a man this is usually around five minutes, but they note it is usually way shorter for female comics—especially if the crowd has never heard of them before. Men just seem to get more benefit of the doubt—however Maureen and Susan agree that there are many factors that go into a booking an act, and many bookings (included ones they run) are very pro female. Red Imp Comedy Club has a whole first season of female headlining comics, showing that women aren’t going anywhere in today’s comedy scene. “I’ve never met a comic, irrelevant of how well their career is going, who is happy with their career. The closely related synaptic vesicle membrane proteins synaptophysin and synaptoporin are abundant in the hippocampal formation of the adult rat. But the prenatal hippocampal formation contains only synaptophysin, which is first detected at embryonic day 17 (E17) in perikarya and axons of the pyramidal neurons. At E21 synaptophysin immunoreactivity extends into the apical dendrites of these cells and in newly formed terminals contacting these dendrites. The transient presence of synaptophysin in axons and dendrites suggests a functional involvement of synaptophysin in fibre outgrowth of developing pyramidal neurons. Synaptoporin expression parallels the formation of dentate granule cell synaptic contacts with pyramidal neurons: the amount of hippocampal synaptoporin, determined in immunoblots and by synaptoporin immunostaining of developing mossy fibre terminals, increases during the first postnatal week. Moreover, in the adult, synaptoporin is found exclusively in the mossy fibre terminals present in the hilar region of the dentate gyrus and the regio inferior of the cornu ammonis. In contrast, synaptophysin is present in all synaptic fields of the hippocampal formation, including the mossy fibre terminals, where it colocalizes with synaptoporin in the same boutons. Our data indicate that granule neuron terminals differ from all other terminals of the hippocampal formation by the presence of both synaptoporin and synaptophysin. This difference, observed in the earliest synaptic contacts in the postnatal hippocampus and persisting into adult life, suggests distinct functions of synaptoporin in these nerve terminals.
