It’s World Mental Health Day and the end of Mental Health Week as I release this episode and I couldn’t pass up this chance to talk about a topic I’ve felt more bold opening up about during the last few months. This podcast was definitely part of that. Chasing a new endeavor, cobbling together a dream and laughing with my best friends along the way has been life-altering, I crap you not, especially during the pandemic. Before we get really going here I bet you were wondering where episode #33 was! Probably not, but I like to think some of the type A’s out there may have been curious. But why 33? Let me tell ya about it and tell you a story close to my heart. So I gritted my teeth, fought through the self-created tension and did E14 Mental Health with ole buddies Mitch & Paul. It centered around me seeking grief counseling after the deaths of my two brothers, Collin and Alex. Collin left us in March of 2015 after a quick trip to the hospital and I didn't get a chance to see him before he passed away. Alex passed away in May of 2019. I'm fortunate to have talked to him one last time before he passed away. Both were 33. Now you see where I’m going here. I listened back to that E14 a couple times and to me it was awkward and cringe-worthy. How’s that different from a regular episode or J.P.-moment you ask (Ba-DOOM Boom CHING)?!? But the cringe-worthy nature of it is what made it good. Did I just unearth a paradox? Embrace the awkward as the mental health saying goes or to you fans of the pod, “It’s only tense if you make it tense,” as Paul and I broke down in his Get to Know episode 20 referring to an epiphany in college at the Paisano’s in Commerce, America. Context is important. During my senior year in college as I was mapping out my future dominance in the internet sports broadcasting world, I took a capstone class in sociology that looked interesting called Drugs & Society. It was probably the easiest class I ever took. It just came naturally. It turns out my curiosity in what I thought would be a blow-off class on the way to graduation was part of a foundation for the rest of my career and more so for the future of The Texas Porch. I don’t want to bury the lead because this is about a newfound passion of mental health awareness. On this episode you'll also hear from one of my compadres on the Friday Night Frenzy on the great KICKS 105.1, Darrell Derbin. Some quick background on “Double D." He's Tik Tok famous. I trust him on that. They say it’s a big deal. Darrell has a history of friends and family that have died of suicide. And he recently released a song relating to that. During this show we'll hear him speak about that. Context is important as to where we take it from here and this is my platform so how about a little background first? I’ve worked in tobacco prevention for about nine years. In 2009 I got a job at the Alcohol and Drug Abuse Council of Deep East Texas. I had grown tired of my radio gig for reasons that will fill a chapter or two in a future book. I also wanted to chase that college play by play dream that was made possible by a call from KETR Program Director Jerrod Knight to broadcast games at my alma mater, A&M-Commerce. The executive director of ADAC, Phyllis Grandgeorge actually walked up to me at church and said “I think I have a job for you,” and I would have had to be pretty dense and/or thick-headed to not realize God was telling me, “TAKE THIS JOB!” I left that gig for the Rice job in 2012 and started back circa 2014. Now I work at another non-profit, The Coalition and get to teach a tobacco prevention curriculum to sixth-graders, serving a few hundred students per year. All of this non-profit work if you bundled it together has opened my eyes to many things, but mainly, and here’s the payoff of the long tease: It has helped me think more about people who can’t defend themselves. It’s also stressed that prevention is the best medicine. About 3.3 million adults in Texas are living with a mental illness according to www.mentalhealthtx.org. 61% of adults in Texas who needed mental health treatment in the last year did not receive any. And sadly 754,000 Texans have had serious thoughts of suicide in the past year. If we prevented the onset of these mental health episodes in young people before they get to adulthood we change their life. My counselor told me a stat that was jarring. One out of every four students has a mental health need. And only one out of those four gets treated. Staggering. So for every 500 students at a Texas school district we’re missing 94 students that cut, overeat, undereat, get bullied, bully, drink, use marijuana and perform other forms of self-harm because they feel like they’re not good enough. Hopefully it goes without saying but school counselors and teachers do a wonderful job trying to fill that gap and I’m just a guy with a dream at the beginning of this journey. But I’m all ears. Where can we help? It’s amazing to see the strides we’ve made in the last two years alone discussing mental health. Bottom line, I just want to encourage anyone to get the help you’ve earned. You’re living this life and you deserve to live it to the full. If you have anxieties, don’t think you’re good enough, abused drugs and alcohol to cope, you’ve bottled up stuff for decades, or if you’ve been abused please ask for help from a licensed counselor. Oh, let me tell you it’s awkward at first. But it’s the first step to a journey of forgiving yourself. There’s still a lot I don’t want to talk about. And that’s ok. “You’re still friends” as it were if you know the vernacular of the show (Get to Know Mitch E23). Collin and Alex’s passing rocked me to the core and I can tell you this. I’ll be comfortable sharing more in the future as my own wounds continue to heal. One last thing I do want to talk about and if you’ve made it this far I’ll leave you here with a big, bold, audacious goal. Back to that E14 I’ve seemingly promoted over and over. I told Mitch and Paul I would like to.... Walk. Across. Texas. So yeah. Most importantly, I’ve saved the best for last. As if walking across the best state there is wasn't a big enough goal, Alex and Collin were both 33 when they passed. I am determined somehow to bring good out of that. I have felt led to opening up a foundation, starting a 501-c3 - whatever the lingo- called Forever 33 that will address mental health awareness and prevention. I don’t know how the best way to form a foundation or anything like that but I like to ask questions so if you’ve got any pointers fire away. I will walk across Texas for Forever 33. Let’s do it. M., Legendre, P., Czarnecki, A. Abstract: A remarkable feature of the developing central nervous system is the generation of spontaneous network activity (SNA) at the onset of synaptogenesis. In the spinal cord (SC) of the mouse embryo, SNA occurs around the 12th embryonic day (E12.5) and involves a recurrent functional loop between motoneurons (MNs) and GABAergic interneurons. We previously showed that Renshaw cells (V1R), which constitute a functionally homogeneous population in the adult, are the first neurons enriched with GABA in the mouse embryonic lumbar SC. V1R make synaptic-like contacts with MNs and likely participate to the genesis of SNA. However, how electrophysiological intrinsic properties of V1R evolve during early development remains poorly understood. Here, using patch-clamp recordings, cluster analysis and biophysical modeling, we analyzed the mechanisms underlying the firing patterns of V1R between E11.5 and E16.5. This developmental period covers the initial phase of development of SC activity (E11.5-E14.5) when SNA is present, and a pivotal period (E14.5-E16.5) when locomotor-like activity emerges. We discovered that V1R can be subdivided into different clusters before E13.5 and that many cells are capable of sustaining repetitive firing or plateau potentials. These V1R then lose transiently this firing ability, which is later recovered at E16.5. This is in contrast to the classical developmental scheme, i.e. an increase of firing capability with time. Combining pharmacology and computational modeling, we showed that the firing patterns of embryonic V1R rely on the synergy between two opposing voltage-dependent currents, namely the slowly inactivating persistent sodium current and the delayed rectifier potassium current. These two currents are responsible for the repetitive firing of action potentials in a vast majority of neurons. Such a synergy is sufficient to explain the clustering of embryonic V1R and determines the developmental trajectories of the electrical phenotype. Taken together our findings reveal a simple mechanism that is at the core of the heterogeneity of firing patterns. Abstract: Environmental insults that activate the maternal immune system are potent primers of developmental neuropathology and maternal immune activation (MIA) has emerged as a risk factor for neurodevelopmental and psychiatric disorders. Animal models of MIA provide an opportunity to identify molecular pathways that initiate disease processes and lead to neuropathology and behavioral deficits in offspring. MIA-induced behaviors are accompanied by anatomical and neurochemical alterations in adult offspring that parallel those seen in affected human populations. We performed transcriptional profiling and neuroanatomical characterization in a time course across mouse embryonic cortical development, following MIA via single injection of the viral mimic polyinosinic:polycytidylic acid (polyI:C) at E12.5. Transcriptional changes identified in the cortex of MIA offspring at E17.5 were validated and mapped to cortical neuroanatomy and cell types via protein analysis and immunohistochemistry. MIA induced strong transcriptomic signatures, including induction of genes associated with hypoxia, immune signaling, and angiogenesis. The acute response identified 6h after the MIA insult was followed by changes in proliferation, neuronal and glial differentiation, and cortical lamination that emerged at E14.5 and peaked at E17.5. Decreased numbers of proliferative cell types in germinal zones and alterations in neuronal and glial cell types across cortical lamina were identified in the MIA-exposed cortex. Müller Licht "Tint" bei Aldi zum Kampfpreis? Sollen das Reste sein oder taugt das was?Unfassbar. Nach Philips, Ikea und vielen weiteren kommt nun auch Aldi Süd um die Ecke, und steigt in das Business mit steuerbaren Heimgeräten ein. Verkauft werden die von Müller Licht produzierten Leuchtmittel mit Namen "Tint" in E27 und GU10 sowie E14 Form. Preislich sind sie ein Hit, wenn man Philips als Benchmark nimmt, glatt die Hälfte. Und von der Qualität und der Leuchtkraft? Ein erster Testbericht nach zwei Wochen Müller Licht in meinem Flur! [caption id="attachment_1690" align="aligncenter" width="409"] Starter-Set "Tint" von Müller Licht bei Aldi Süd (teilweise noch erhältlich!) für 29,95€ / Quelle: aldi-sued.de Die Sets kamen bei Aldi, was wäre es denn auch sonst, zu Preisen raus, die NOCH günstiger waren, als bei Müller Licht selbst zu kaufen. Das nur am Rande, hier die Preise: das Starter Set mit Fernbedienung und einer E14 oder E27 Birne liegt bei knapp 30 Euro, die E14/E27/GU10-LEDs alleine liegen bei 20 Euro für die farbige und bei 14 Euro je Stück für die reinweiße Variante. Ich hab die Preise von Ikea jetzt nicht im Kopf, aber im Vergleich zu Philips sind die Birnen somit minimal 50% günstiger. ABER: obwohl sie ebenfalls den ZigBee-Funkstandard zur Steuerung verwenden, können sie nicht über die Philips-Bridge gekoppelt werden! Das heißt: ihr braucht hierfür eine "freie" (nicht Philips-)Zigbee-Bridge, wie z.B. einen Raspi mit Modul oder einfach ein amazon Echo Plus, mit eingebauter Bridge. Nun wird es kurz haarig: bei einem Starterkit MIT Fernbedienung ist die Lampe an die Remote gebunden und muss, bevor ihr sie mit Eurer Bridge verbinden könnt, erst resettet werden. Das ist allerdings in beiliegender Anleitung beschrieben und einfach machbar... was mir bei den Philips-Leuchtmitteln, auch mit Hilfe des Supports und ungefähr drei verschiedenen und sich widersprechenden Anleitungen bis heute noch nicht gelungen ist. Sonst ist es ganz einfach: Licht anmachen, Suche starten - und das neue Gerät hinzufügen, mit Namen versehen - und schon kann es gesteuert werden. Bei Alexa per Sprachbefehl, bei allen anderen Lösungen über die Hard-/Software/App, die ihr auch sonst benutzt - nehme ich an... Ich kann jetzt nur für die mehrfarbige E27-Birne sprechen, für den Strahler und die kleine E14-Fassung habe ich nur im Netz gelesen, dass sie von der Helligkeit und der Farbmischung zwar mit 16,7 Millionen möglichen Farben weit mehr können, als die vergleichbare Philips-Lampe, aber ein wenig "dunkler" leuchtet. Für die reinweißen Lampen kann ich mich an keinen Test erinnern, aber Google ist hier sicherlich Euer bester Freund! Die Tint E27 reagiert über mein Echo Plus viel schneller als die Philips über deren Bridge, die ich auch über amazon Echo steuere. Im persönlichen Vergleich ist die Helligkeit besser im Weißlicht-Modus als die Philips. Auch die Farben, die sich größtenteils zuverlässiger und freier steuern lassen, als die vorgegebenen Sets der Philips-Birne, haben mehr Brillanz als das von mir zu Vergleichszwecken aus der Schublade gezogene niederländische Pendant. Ich bereue den Kauf für 20 Euro keine Sekunde. Die Lampe war in Sekunden mit dem Echo Plus verbunden, schnell in der zugehörigen App umbenannt und ab dieser Sekunde Sprachbefehl-bereit. Sie reagiert schnell und direkt, spendet helles klares Licht und brillante Farben. Bei meinem Aldi standen letzte Woche immer noch mehrere Lampen herum, vielleicht, wenn ich jetzt Euer Interesse wecken konnte, habt ihr in Eurem auch Glück, und könnt Euch ein Testobjekt besorgen. Aber: wenn ihr kein Starterset bekommt, müsst ihr ZigBee-Steuerung ermöglichen - sonst ist die Lampe nur eine LED-Glühbirne, die mit einem Schalter an der Wand an und aus geht und weißes Licht spendet - und dafür ist sie dann doch zu teuer! Pax6 encodes a specific DNA-binding transcription factor that regulates the development of multiple organs, including the eye, brain and pancreas. Previous studies have shown that Pax6 regulates the entire process of ocular lens development. In the developing forebrain, Pax6 is expressed in ventricular zone precursor cells and in specific populations of neurons; absence of Pax6 results in disrupted cell proliferation and cell fate specification in telencephalon. In the pancreas, Pax6 is essential for the differentiation of α-, β- and δ-islet cells. To elucidate molecular roles of Pax6, chromatin immunoprecipitation experiments combined with high-density oligonucleotide array hybridizations (ChIP-chip) were performed using three distinct sources of chromatin (lens, forebrain and β-cells). ChIP-chip studies, performed as biological triplicates, identified a total of 5,260 promoters occupied by Pax6. 1,001 (133) of these promoter regions were shared between at least two (three) distinct chromatin sources, respectively. In lens chromatin, 2,335 promoters were bound by Pax6. RNA expression profiling from Pax6⁺/⁻ lenses combined with in vivo Pax6-binding data yielded 76 putative Pax6-direct targets, including the Gaa, Isl1, Kif1b, Mtmr2, Pcsk1n, and Snca genes. RNA and ChIP data were validated for all these genes. In lens cells, reporter assays established Kib1b and Snca as Pax6 activated and repressed genes, respectively. In situ hybridization revealed reduced expression of these genes in E14 cerebral cortex. Moreover, we examined differentially expressed transcripts between E9.5 wild type and Pax6⁻/⁻ lens placodes that suggested Efnb2, Fat4, Has2, Nav1, and Trpm3 as novel Pax6-direct targets. Collectively, the present studies, through the identification of Pax6-direct target genes, provide novel insights into the molecular mechanisms of Pax6 gene control during mouse embryonic development. In addition, the present data demonstrate that Pax6 interacts preferentially with promoter regions in a tissue-specific fashion. Nevertheless, nearly 20% of the regions identified are accessible to Pax6 in multiple tissues.

Background: Lack of appropriate tools and techniques to study fate and functional integration of newly generated neurons has so far hindered understanding of neurogenesis' relevance under physiological and pathological conditions. Current analyses are either dependent on mitotic labeling, for example BrdU-incorporation or retroviral infection, or on the detection of transient immature neuronal markers. Here, we report a transgenic mouse model (DCX CreERT2) for time-resolved fate analysis of newly generated neurons. This model is based on the expression of a tamoxifen-inducible Cre recombinase under the control of a doublecortin (DCX) promoter, which is specific for immature neuronal cells in the CNS. Results: In the DCX-CreERT2 transgenic mice, expression of CreERT2 was restricted to DCX+ cells. In the CNS of transgenic embryos and adult DCX-CreERT2 mice, tamoxifen administration caused the transient translocation of CreERT2 to the nucleus, allowing for the recombination of loxP-flanked sequences. In our system, tamoxifen administration at E14.5 resulted in reporter gene activation throughout the developing CNS of transgenic embryos. In the adult CNS, neurogenic regions were the primary sites of tamoxifen-induced reporter gene activation. In addition, reporter expression could also be detected outside of neurogenic regions in cells physiologically expressing DCX (e. g. piriform cortex, corpus callosum, hypothalamus). Four weeks after recombination, the vast majority of reporter-expressing cells were found to co-express NeuN, revealing the neuronal fate of DCX+ cells upon maturation. Conclusions: This first validation demonstrates that our new DCX-CreERT2 transgenic mouse model constitutes a powerful tool to investigate neurogenesis, migration and their long-term fate of neuronal precursors. Moreover, it allows for a targeted activation or deletion of specific genes in neuronal precursors and will thereby contribute to unravel the molecular mechanisms controlling neurogenesis.

Changes in ATP-induced increase in {[}Ca2+], during collecting duct ontogeny were studied in primary monolayer cultures of mouse ureteric bud (UB) and cortical collecting duct (CCD) cells by Fura-PE3 fluorescence ratio imaging. In UB (embryonic day E14 and postnatal day P1) the ATIP-stimulated increase (EC50 approximate to 1 muM) in fluorescence ratio (DeltaR(ATP)) was independent of extracellular Ca2+ and insensitive to the P2 purinoceptor-antagonist suramin (1 mM). From day P7 onward when CCD morphogenesis had been completed DeltaR(ATP) increased and became dependent on extracellular Ca2+. This ATP-stimulated Ca2+ entry into CCD cells was non-capacitative and suramin (11 mM)insensitive, but sensitive to nifedipine (30 muM) and enhanced by Bay K8644 (15 muM), a blocker and an agonist of L-type Ca2+ channels, respectively. Quantitative RT-PCR demonstrated similar mRNA expression of L-type Ca2+ channel alpha1-subunit, P2Y(1), P2Y(2), and P2X(4b) purinoceptors in UB and CCD monolayers while the abundance of P2X(4) mRNA increased with CCD morphogenesis. In conclusion, both embryonic and postnatal cells express probably P2Y(2)-stimulated Ca2+ release from intracellular stores. With development, the CCD epithelium acquires ATP-stimulated Ca2+ entry via L-type Ca2+ channels. This pathway might by mediated by the increasing expression of P2X(4)-receptors resulting in an increasing ATP-dependent membrane depolarization and activation of L-type Ca2+ channels.
