À Los Angeles, les pompiers essayent toujours de contenir les flammes, plus d'une semaine après le début des incendies. Selon le dernier bilan, au moins 24 personnes ont trouvé la mort. Une catastrophe historique, marquée par un flot d'infox d'une rare intensité sur les réseaux. L'origine de ces incendies, le travail des pompiers, les rescapés, rien n'échappe à la désinformation, à tel point que certaines des images les plus partagées en ligne sont irréelles ou sorties de leur contexte. Si vous avez passé du temps sur les réseaux sociaux ces derniers jours, vous avez probablement été confronté à l'une de ces deux images aériennes, montrant une maison au toit rouge, prétendument épargnée par les flammes à Los Angeles. Tout le quartier semble avoir été détruit, sauf ces luxueuses résidences. Si les deux maisons se ressemblent, ce ne sont pas les mêmes. L'une est en bord de mer, l'autre en pleine ville.Les internautes qui partagent ces photos affirment qu'elles montreraient « la maison d'un croyant de Los Angeles, miraculeusement protégée par Dieu ». Un récit sur lequel plusieurs auditeurs nous ont alertés.Image détournée et générée par IAVérification faîte, ces deux images n'ont rien à voir avec les incendies en cours à Los Angeles. La première photo est bien authentique, mais elle n'a pas été prise dans la cité des anges. Une recherche par image inversée permet de retrouver sa trace dans la banque d'images de l'Agence France Presse (AFP). On y apprend en légende qu'elle montre une maison dans le quartier de Lahaina, à la suite des incendies mortels à Hawaï, en août 2023. La résidence avait été épargnée notamment grâce à sa toiture en tôle épaisse et à l'absence de végétation à proximité.Concernant le deuxième cliché, si visuellement tout semble réel, l'image a en réalité été générée par une intelligence artificielle. Le seul moyen de s'en rendre compte consiste à faire une recherche par image inversée sur Google, puis d'aller dans l'onglet « À propos de l'image ». Un message apparaît alors indiquant « image créée avec l'IA de Google ».Comment Google sait-il que quelqu'un a généré cette image avec son intelligence artificielle ? Pour son outil de génération d'image, baptisé Imagen, la firme américaine utilise ce que l'on appelle des watermarks, des filigranes, invisibles à l'œil nu, mais identifiables par un logiciel. Ce tatouage numérique atteste donc de l'utilisation de l'intelligence artificielle, mais sans faire cette démarche de vérification, il est impossible de s'en rendre compte.Instrumentalisation complotiste et religieuseCette image synthétique fait l'objet de nombreuses instrumentalisations. Religieuse d'abord, avec plusieurs messages parlant d'une intervention divine. Certains affirment, à tort, que cette maison appartenait à une famille chrétienne, d'autres à une famille musulmane.Et puis certains complotistes ont aussi diffusé cette image, assurant que l'incendie était volontaire et ne ciblait que certaines habitations. Deux récits trompeurs vus des dizaines de millions de fois sur les réseaux sociaux. Résultat, cette infox figure aujourd'hui parmi les images les plus vues concernant les incendies de Los Angeles. On est donc là face à un énième exemple de détournement de l'IA qui perturbe les perceptions d'un événement mondial et invisibilise les vraies images de la catastrophe.

Ginkgo biloba, known for its distinctive fan-shaped leaves, has been used in traditional medicine for centuries, particularly in Asia. Ginkgo biloba is often touted as an herb for brain health, such as improving memory and cognition. This reputation does a great disservice to the most versatile herb in the world. Ginkgo biloba can be used as a potent antiviral agent for a variety of viruses. Ginkgo biloba has some key bioactive antiviral components: Flavonoids and Terpenoids: Ginkgo leaves contain high levels of flavonoids and terpenoids, compounds known for their antioxidant properties. These substances contribute to the antiviral activity of the plant. Ginkgolides and Bilobalides: These are unique terpene trilactones found in Ginkgo biloba, which have specific antiviral activities. Ginkgo Biloba's Mechanisms of Antiviral Action The antiviral properties of Ginkgo biloba are multi-faceted, involving multiple mechanisms: Inhibition of the fusions and synthesis of proteins in the viruses herpes simplex 1 and 2 (HSV-1 and HSV-2). Inhibition of genome replication in cytomegalovirus (HCMV) and Zika virus (ZIKV). Inhibition of viral fusion proteins in HIV, Ebola virus (EBOV), influenza A virus (IAV), and Epstein-Barr virus (EBV). Inhibition of the targeting protein and DNA of coronoviruses (SARS-CoV-2), varicella zoster virus (VZV), and measles virus. Inhibition of Viral Entry and Replication: Some studies suggest that Ginkgo biloba extracts can interfere with the ability of viruses to enter host cells or replicate. This is a key step in preventing the spread of viral infections. Immune System Modulation: Ginkgo biloba might enhance the body's immune response against viral infections. By modulating immune functions, it could help in controlling viral spread and severity. Anti-inflammatory Effects: The anti-inflammatory properties of Ginkgo biloba can be beneficial in reducing the severity of symptoms associated with viral infections. Research on Ginkgo Biloba's Antiviral Properties Anti-MERS-CoV and Anti-HCoV-229E Properties: A study focused on the antiviral activities of Ginkgo biloba leaf extracts against Middle East Respiratory Syndrome Coronavirus (MERS-CoV) and Human Coronavirus 229E (HCoV-229E). Inhibition of Enveloped Viruses: Research published in Scientific Reports discussed how ginkgolic acid, a component of Ginkgo biloba, inhibits the fusion of enveloped viruses. The study found that ginkgolic acid had a strong inhibitory effect on Human Cytomegalovirus (HCMV) and also tested its effects on Herpes Simplex Virus-1 (HSV-1) and Zika Virus (ZIKV). Researchers also found broad spectrum inhibition by ginkgolic acid of all three classes of fusion proteins including HIV, Ebola virus (EBOV), influenza A virus (IAV) and Epstein Barr virus (EBV). In addition, inhibition was found of a non-enveloped adenovirus. The authors conclude that ginkolic acids may potentially be used to treat acute infections (e.g. Coronavirus, EBOV, ZIKV, IAV and measles), and also topically for the successful treatment of active lesions (e.g. HSV-1, HSV-2 and varicella-zoster virus (VZV)). It was observed that ginkgolic acid could inhibit the entry of these viruses into cells, thereby blocking viral replication. Another study entitled, “Ginkgolic acids inhibit SARS-CoV-2 and its variants by blocking the spike protein/ACE2 interplay” found that ginkgolic acids from ginkgo biloba inhibited the SARS-CoV-2 virus from binding to the ACE2 receptor and thus could potentially be helpful in acute COVID-19 infections. I use VascuSelect from Moss Nutrition which contains 120 mg of ginkgo biloba, 100 mg of grape seed extract, and 100 mg of mango fruit powder per capsule for acute and chronic viruses dosed 1 capsule once or twice a day with or without food. P., Kornum, B. R. Abstract: Infection with Influenza A virus (IAV) causes the well-known symptoms of the flu, including fever, loss of appetite and excessive sleepiness. These responses, mediated by the brain, will normally disappear once the virus is cleared from the system, but a severe respiratory virus infection may cause long-lasting neurological disturbances. These include encephalitis lethargica and narcolepsy. The mechanisms behind such long lasting changes are unknown. The hypothalamus is a central regulator of the homeostatic response during a viral challenge. To gain insight into the neuronal and non-neuronal molecular changes during an IAV infection, we intranasally infected mice with an H1N1 virus and extracted the brain at different time points. Using single-nucleus RNA sequencing (snRNA-seq) of the hypothalamus, we identify transcriptional effects in all identified cell populations. The snRNA-seq data showed the most pronounced transcriptional response at 3 days past infection, with a strong downregulation of genes across all cell types. General immune processes were mainly impacted in microglia, the brain resident immune cells, where we found increased numbers of cells expressing pro-inflammatory gene networks. In addition, we found that most neuronal cell populations downregulated genes contributing to the energy homeostasis in mitochondria and protein translation in the cytosol, indicating potential reduced cellular and neuronal activity. This might be a preventive mechanism in neuronal cells to avoid intracellular viral replication and attack by phagocytosing cells. Abstract: Many viruses that threaten public health establish condensates via phase transitions to complete their lifecycles, and knowledge on such processes is key for the design of new antivirals. In the case of influenza A virus, liquid condensates known as viral inclusions are sites dedicated to the assembly of its 8-partite RNA genome. Liquid viral inclusions emerge near the endoplasmic reticulum (ER) exit sites, but we lack the molecular understanding on how the ER contributes to their biogenesis. We show here that viral inclusions develop at remodeled ER sites and display dynamic interactions using the ER, including fusion and fission events and sliding movements. We also uncover a novel role for the host factor, ATG9A, in mediating the exchange of viral inclusions between the ER and microtubules. Depletion of ATG9A arrests viral inclusions at microtubules and prevents their accumulation at the ER, leading to a significantly reduced production of viral genome complexes and infectious virions. In light of our recent findings, we propose that a remodeled ER supports the dynamics of liquid IAV inclusions, with ATG9A acting locally to facilitate their formation. This work advances our current knowledge regarding influenza genome assembly, but also reveals new roles for ATG9A beyond its classical involvement in autophagy. Eloim, também conhecido como Iavé, Jeová, Deus e outras alcunhas, segundo contam saiu de cena para descansar no sétimo dia. O job durou só seis dias e aquele do qual o homem é a imagem e semelhança, saiu para descansar no sétimo e nunca mais voltou. Mandou o filho, deu ruim pro pupilo, foi julgado, esculachado, crucificado, morreu, ressuscitou no terceiro dia só para constar. Partiu, está sentado ao lado direito do Pai e também nunca mais apareceu para um rolê por esta bocada chamada Terra. Abstract: Single-molecule experiments have been helping us to get deeper inside biological phenomena by illuminating how individual molecules actually work. Digital bioassay, in which analyte molecules are individually confined in small compartments to be analyzed, is an emerging technology in single-molecule biology and applies to various biological entities (e.g., cells and virus particles). However, digital bioassay is not compatible with multi-conditional or multi-parametric assays, hindering understanding of analytes. This is because current digital bioassay lacks a repeatable solution-exchange system that keeps analytes inside compartments. To address this challenge, we developed a new digital bioassay platform with easy solution exchanges, called multi-dimensional (MD) digital bioassay, and tested its quantitativity and utility. We immobilized single analytes in arrayed femtoliter (10-15 L) reactors and sealed them with airflow. The solution in each reactor was stable and showed no cross-talk via solution leakage for more than 2 h, and over 30 rounds of perfect solution exchanges were successfully performed. To show the utility of our system, we investigated neuraminidase inhibitor (NAI) sensitivity on single influenza A virus (IAV) particles in a multi-conditional assay. We proved that IAV particles show a heterogeneous response to the NAI. Further, to demonstrate multi-parametric assays, we examined the sensitivity of individual IAV particles or model enzyme molecules to two different inhibitors. Abstract: In this study, influenza A virus (IAV) and nanoparticle diffusion in human airway mucus was quantified using fluorescent video microscopy and multiple particle tracking. In previous work, it was determined that mucin-associated sialic acid acts as a decoy receptor for IAV hemagglutinin binding, and that virus passage through the mucus gel layer is facilitated through the sialic-acid cleaving enzyme, neuraminidase (NA), also present on the IAV envelope. However, our data suggests the mobility of IAV in mucus is significantly influenced by the mesh structure of the gel, as measured by nanoparticle probes, and NA activity is not required to facilitate virus passage through mucus gels. Using newly developed analyses, the binding affinity of IAV to the 3D mucus meshwork was estimated for individual virions with dissociation constants in the mM range, indicative of weak and reversible IAV-mucus interactions. We also found IAV diffusion significantly increased in mucus when treated with a mucolytic agent to break mucin-mucin disulfide bonds. In addition, IAV diffusion was significantly limited in a synthetic mucus model as crosslink density was systematically increased and network pore size was reduced. The results of this work provide important insights on how the balance of adhesive and physical barrier properties of mucus influence the dissemination of IAV within the lung microenvironment. Abstract: Influenza A virus (IAV) is a highly contagious human pathogen responsible for nearly half a million deaths each year. Non-structural protein 1 (NS1) is a crucial protein expressed by IAV to evade the host immune system. Additionally, NS1 has been proposed to stimulate translation because of its ability to bind poly(A) binding protein 1 (PABP1) and eukaryotic initiation factor 4G (eIF4G). We analyzed the interaction of NS1 with PABP1 using quantitative techniques. Our studies show that NS1 binds as a homodimer to PABP1, and this interaction is conserved across different IAV strains. Unexpectedly, NS1 does not bind to PABP1 that is bound to poly(A) RNA. Instead, NS1 only binds to PABP1 free of RNA, suggesting that translation stimulation does not occur by NS1 interacting with the PABP1 molecule attached to the mRNA 3'-poly(A) tail. We propose that NS1 binds to the eIF4G complex at the 5'-end of the mRNA and recruits the RNA-free PABP1, which may stimulate translation initiation by promoting the association of the ribosomal subunits. Abstract: The antigenic diversity of influenza A virus (IAV) circulating in swine challenges the development of effective vaccines, increasing zoonotic threat and pandemic potential. High throughput sequencing technologies are able to quantify IAV genetic diversity, but there are no accurate approaches to adequately describe antigenic phenotypes. This study evaluated an ensemble of non-linear regression models to estimate virus phenotype from genotype. Regression models were trained with a phenotypic dataset of pairwise hemagglutination inhibition (HI) assays, using genetic sequence identity and pairwise amino acid mutations as predictor features. The model identified amino acid identity, ranked the relative importance of mutations in the hemagglutinin (HA) protein, and demonstrated good prediction accuracy. Four previously untested IAV strains were selected to experimentally validate model predictions by HI assays. Error between predicted and measured distances of uncharacterized strains were 0.34, 0.70, 2.19, and 0.17 antigenic units. These empirically trained regression models can be used to estimate antigenic distances between different strains of IAV in swine using sequence data. By ranking the importance of mutations in the HA, we provide criteria for identifying antigenically advanced IAV strains that may not be controlled by existing vaccines and can inform strain updates to vaccines to better control this pathogen.
