This week we're excited to present a conversation with director Ira Sachs, whose new film, Passages, is currently playing in our theaters. A masterful work of psychosexual intensity, the newest film from Ira Sachs offers one of the director's most cutting variations on desire and intimacy. Co-written by author and longtime collaborator Mauricio Zacharias, Passages follows Tomas (Franz Rogowski), a mercurial German filmmaker living in Paris whose commitment to his husband, Martin (Ben Whishaw), falls short when he pursues a dalliance with a young schoolteacher, Agathe (Adèle Exarchopoulos). Martin begins his own affair soon after, while Tomas swings between both relationships and unleashes a reckless succession of breakups and makeups. With fearless performances from Rogowski, Whishaw, and Exarchopoulos, Sachs crafts a cinematic rarity in which the white-hot pleasures and compulsions of a particularly dysfunctional amour fou are kept on par with ferocious honesty. This conversation was moderated by film critic, Esther Zuckerman.

It's an early morning in May when Lane is pulled from a deep sleep by a concerning phone call from Scotland—her grandfather has had a heart attack. Lane hastily makes plans to fly overseas, and a dejected Inspector Darling prepares himself for a stint of bachelorhood. But before he can begin to dwell on it the Nelson Police learn that Ben Arden, a local cad, has gone out for a late-night boat ride and not returned, which immediately sets the town rumour mill churning.In Scotland Lane finds her grandfather on the mend but her estranged sister Diana caught up in stressful—and mysterious—circumstances of her own. As Lane follows the thread leading from South Africa to Aberdeen to the War Office in London it becomes apparent that her sister is on the run, and that keeping secrets seems to be a Winslow family trait.Back at the Nelson Police station a strange assemblage of clues begins to amass around the Arden case, stretching from the local printshop all the way back to the Great War. But progress is interrupted when the mayor himself issues a demand that Darling travel to England to tie up a decades-old mystery with local connections. Women Talking is a 2022 drama film written and directed by Sarah Polley. It is based on the Canadian 2018 novel of the same name by Miriam Toews, and inspired by the gas-facilitated rapes that occurred at the Manitoba Colony, a remote and isolated Mennonite community in Bolivia. It features an ensemble cast that includes Oscar-Winning actresses, Rooney Mara , Claire Foy (young “Queen Elizabeth” in Seasons 1 & 2 of Netflix's The Crown), Jessie Buckley, Judith Ivey, Ben Whishaw, and Frances McDormand, who also served as a producer on the film. Women Talking premiered at the 49th Telluride Film Festival on September 2, 2022, and was released in the United States via select theaters on December 23, 2022, before a wide release on January 27, 2023, by United Artists Releasing. The film received positive reviews from critics, who lauded Polley's screenplay and direction, the performances of the cast (particularly of Foy, Buckley, and Whishaw) and score. It was named one of the top ten films of 2022 by the National Board of Review and the American Film Institute, won Best Adapted Screenplay at the 28th Critics' Choice Awards, 75th Writers Guild of America Awards, and the 95th Academy Awards, where it was also nominated for Best Picture. Ep. 94: HBO's Looking (MEMBERS ONLY): It's not often that we get media that is unabashedly gay, depicting gay spaces and the gay community in a way that might make heterosexuals uncomfortable. HBO's Looking was pioneer for this on TV, including the way it depicted gay sex and intimacy. Sebastián Silva's Rotting in the Sunalso pushes the envelope, though in a much more confronting (and depressing) way. Abstract: Cooperative forelimb and mouth movements during eating contribute to diet selection among vertebrates including the oromanual manipulatory skills in rodents and primates. Whereas spinal and brainstem circuits implement forelimb and orofacial actions, whether there is a specialized cortical circuit that flexibly assembles these to achieve cross-body and oromanual coordination for skilled manipulation remains unclear. Here we discover a cortical region and its cell-type-specific circuitry that orchestrates body postures and oromanual coordination for food manipulation in mice. An optogenetic screen of cortical areas and projection neuron types identified a rostral forelimb-orofacial area (RFO), wherein activation of pyramidal tract (PTFezf2) and intratelencephalic (ITPlxnD1) neurons induced concurrent posture, forelimb and orofacial eating-like movements. In a pasta-eating behavior, RFO PTFezf2 and ITPlxnD1 activity were closely correlated with picking up the pasta, adopting a sitting posture, oromanual manipulation, and hand-assisted biting. RFO inactivation and inhibition of RFO PTsFezf2 and ITsPlxnD1 impaired posture and oromanual coordination, leading to deficient pasta manipulation and biting. RFO is reciprocally connected to forelimb and orofacial sensorimotor areas as well as insular and visceral areas. Within this network, ITsPlxnD1 project bilaterally to the entire network and the ventrolateral striatum and PTsFezf2 project to multiple subcortical areas associated with forelimb and orofacial control. These results suggest that ITsPlxnD1 select and coordinate the feeding program involving multiple body parts and PTsFezf2 implement the fine details of movements. Abstract: In contrast to most transgenic mouse models of Alzheimer disease (AD), knock-in mice expressing familial AD-linked mutations of the amyloid precursor protein (App) gene exhibit stereotypical age-dependent amyloid beta (A{beta}) pathology and cognitive impairment without physiologically unrealistic App overexpression. This study investigated the effect of familial AD-linked App mutations on hippocampal CA1 neuronal activity and function. To enable calcium imaging of neuronal activity, AppNL-G-F/NL-G-F knock-in (APPki) mice were crossed with Thy1-GCaMP6s+/- (C-TG) mice to generate AppNL-G-F/NL-G-FxThy1-GCaMP6s+/- (A-TG) mice, which were characterized at 12 months of age. A-TG mice exhibited A{beta} pathology in the hippocampus. In several configurations of an air-induced running task, A-TG mice and C-TG mice were equally successful in learning to run or to stay immobile. In the Morris water place test, A-TG mice were impaired, but learned the task. Comparisons of hippocampal CA1 neuronal activity in the air-induced running task showed that A-TG mice displayed neuronal hypoactivity both during movement and immobility. A-TG mice and C-TG CA1 neuronal encoding of distance or time in the air induced running task were not different. These results suggest that knock-in of familial AD-linked mutations in A-TG mice results in A{beta} pathology, neuronal hypoactivity, and cognitive impairment without severely affecting CA1 neuronal encoding. In comparison to APPki mice, A-TG mice had less severe AD-like memory impairments at 12 months of age (Saito et al., 2014; Mehla et al., 2019), suggesting that the disease onset was delayed in A-TG mice. The effect of APP mutations may have been mitigated through genetic mechanisms when APPKi mice were crossed with C-TG mice. Copy rights belong to original authors. It was developed from a story by King and Hamish McColl, which was based on the stories of the character Paddington Bear created by Michael Bond. Produced by David Heyman, Paddington stars Ben Whishaw as the voice of the title character, with Hugh Bonneville, Sally Hawkins, Julie Walters, Jim Broadbent, Peter Capaldi, and Nicole Kidman in live-action roles. The film tells the story of Paddington, an anthropomorphic bear who migrates from the jungles of "Darkest Peru" to the streets of London, where he is adopted by the Brown family. Kidman plays a taxidermist who attempts to add him to her collection. A British and French venture produced by StudioCanal UK, TF1, and Heyday, Paddington's principal photography began in September 2013 and wrapped up in June 2014. Colin Firth was originally set to voice Paddington, but dropped out in post-production and was replaced by Whishaw. Paddington was released in the United Kingdom on 28 November 2014 to critical acclaim for Whishaw's vocal performance, humour, screenplay, visual effects and appeal to children and adults. It grossed $268 million worldwide on a €38.5 (~$55) million budget. It received two nominations at the BAFTAs: Best British Film and Best Adapted Screenplay. A sequel, Paddington 2, was released in 2017, with King and much of the cast returning.

Summary: The cortex is generally thought to be the seat of consciousness in human beings. A body of literature indicates that the cortex is not integral to many intuitively conscious behaviors in rats. This suggests that either rats don't rely conscious experiences to guide their behavior in the way we do, or else that consciousness depends on distinct mechanisms in humans and rats. All told, we should think this body of literature is evidence against the hypothesis that mammals are generally conscious and non-mammalian vertebrates are not. Epistemic status: I'm not an expert in much of the material surveyed here. Particularly the function of brain regions and lesioning studies in animals. These issues are important and my impression is that very few people have the comprehensive expertise to think well about these things. Caveat: I'm wary of the decortication studies because they seem so out of line with what I would expect given the prevailing opinions and they are discussed very little in contemporary neuroscience. That said, I have a hard time seeing how they could be grossly inaccurate and I've never seen anyone challenge them. Main ideas are bolded. Intro All vertebrate brains follow a similar plan in rough outline. At the highest level, they are generally divided into three parts: the fore, the mid, and hindbrain. In humans and other mammals, the forebrain consists in the cerebral cortex and several smaller structures. The mammalian cerebral cortex houses a number of specialized sensory processors and muscle controls along with general association regions of less specific purpose. In humans and other apes, the cerebral cortex is also widely believed to be largely responsible for generating our conscious experiences. While structures in the mid or hindbrain control aspects of body regulation and arousal, our experiences are thought to be produced in the cortex. We have successfully mapped out sensory regions and have some understanding of how their activity correlates with conscious contents. Damage to regions of the cortex radically alters cognition and affects conscious experiences in a variety of ways. Moreover, severe damage to the human cerebral cortex destroys all signs of awareness and intentional activity. A human without a cerebral cortex is, for the most part, a living body with no mind. Primates differ from many other animals in the central role their cortex plays in their cognitive lives. The effects of cortical loss in rats has been studied in depth in a series of neuroscientific experiments (Whishaw 1990)[1], carried out mostly in the 1960s-80s. The results of these studies are striking. The behavior of decorticate rats is remarkably unaffected by extensive damage or removal of their cerebral cortex. The afflicted rats are impaired by the loss of cortical tissue, but not nearly as much as one would expect. The obvious behavioral implications to decortication noted by experimenters are extremely benign relative to what I would have antecedently expected. They include: A laundry list of small idiosyncratic changes, e.g.: Impaired muscle control over the tongue and jaws Postural changes while grooming Increased use of forepaws during swimming Various social changes Notably, inferior maternal care Difficulty with some complex cognitive tasks Inability to reason abstractly about locations Absence of food hoarding behavior I suspect that there are a lot of complicated ways that decortication does change behavior, but which require subtle experimental designs to detect. Surely, the cortex must do a lot of important things to be worth its metabolic cost. However, losing the cortex does not radically aff...

Abstract: We present a deep neural network for data-driven analyses of infant rat behavior in an open field task. The network was applied to study the effect of maternal nicotine exposure prior to conception on offspring motor development. The neural network outperformed human expert designed animal locomotion measures in distinguishing rat pups born of preconception nicotine dams versus control dams. Notably, the network discovered novel movement alterations in posture, movement initiation and a stereotypy in warm-up behavior (repetition of movement along specific dimensions) that were the most predictive of nicotine exposure. This suggests that maternal preconception nicotine exposure delays and alters offspring motor development. In summary, we demonstrated that a deep neural network can automatically assess animal behavior with high accuracy, and that it offers a novel, data-driven approach to investigating brain alterations in development Copy rights belong to original authors. Abstract: A network of acetylcholinergic (ACh) cell groups in the basal forebrain innerve the forebrain and are proposed to contribute to a variety of functions including attention, and cortical plasticity. This study examined the contribution of the ACh-nucleus basalis projection to the sensorimotor cortex on recovery on a skilled reach-to-eat task following photothrombotic stroke in the forelimb region of the somatosensory cortex. Mice were trained to perform a single-pellet skilled reaching task and their pre and poststroke performance, from Day 4 to Day 28 poststroke, was assessed frame-by-frame by video analysis with end point, movement and sensorimotor integration measures. Somatosensory forelimb lesions produced impairments in endpoint and movement component measures of reaching and increased the incidence of fictive eating, a sensory impairment in mistaking a missed reach for a successful reach. Upregulated ACh activity, as measured by electroencephalogram (EEG) recording, elicited via optogenetic stimulation of the nucleus basalis improved recovery of reaching and improved movement scores but did not affect a sensorimotor integration impairment poststroke. The results show that the mouse cortical forelimb somatosensory region contributes to forelimb motor behavior and suggest that basal forebrain ACh upregulation could serve as an adjunct to behavioral therapy for the acute treatment of stroke. Episode 19 of The Words Are Not Enough is a Bond 25 Emergency News Update!In this episode Griffin (@griffschiller) and Brody (@brodyserravalli) discuss the following:1. Bond 25 has found a DP* Earlier in the week, Variety's Kristopher Tapley broke the story that Academy Award-winning Swedish DP, Linus Sandgren (best know for La La Land and First Man) will be joining Bond 25 as Director of Photography, saying "It appears Linus Sandgren is shooting the next Bond". IndieWire has now confirmed this story, citing Sandgren himself as their source. 2. Seydoux to return* The Daily Mail's Baz Bamigboye reported that Cary Fukunaga had confirmed Lea Seydoux's return to the franchise, offering the concise comment "Lea will be returning".* Variety's Justin Kroll then corroborated Bamigboye's reporting, stating that a source had confirmed the story to Variety. * What this return could mean for 25 (continuation of Spectre's story) * What kind of role will Madeleine play in the movie (OHMSS-style death speculated among fans) - we weigh in * Confirms that Monica Bellucci probably won't be returning as rumoured (although does confirm they were looking to bring a Bond girl back for 25 - something Barbara Broccoli has been trying to do since TND)3. Casting call for two women and a villain* Kroll also reported that "EON is looking for two other actresses - one would play an MI6 agent who works with Bond and another, a mystery woman - as well as the Bond villain."* Alleged insiders then added that "Rami Malek was being eyed for the [villain] role, but his schedule on the last season of Mr Robot, which shoots from March through July, makes it unlikely he will be able to take the gig, even if EON offered him the part". * Does this mean Blofelwill not be returning as the villain?4. Harris, Whishaw, and Fiennes to return* In his conversation with Cary Fukunaga, Baz Bamigboye also confirmed that Ralph Fiennes, Ben Whishaw, and Naomi Harris would be returning as M, Q, and Moneypenny respectively. Cary seemed eager to work with the returning crew and the rest of the British talent likely to fill out the cast saying "You have some of the best actors in the world here, why wouldn't I have the best coming back?!" * Interesting that Rory Kinnear as Bill Tanner was not included in that list of MI6 regulars returning as he has been a staple of the Craig era Vauxhall.5. Cary Fukunaga suggested to be writing Bond 25 in addition to directing* In Justin Kroll's Variety piece, Cary Fukunaga is reported to now be writing Bond 25. In the article, Fukunaga is listed as the sole writer two times and no reference is made to any other writers. Bamigboye suggested that Purvis and Wade were on-board to write and that Fukunaga was having "major input" in the direction of the script, but this does not appear to have been based off of new information obtained during his interview with Fukunaga. It is very likely Bamigboye is basing this off of the uncited rumour that Bond 25 would be going back to using the Purvis and Wade treatment following John Hodge's departure. Fukunaga rewriting the film lines up with his earlier comments from last week about the film still being written. * Confirms earlier story that EON were looking for a writer/director for Bond 25 * If Purvis and Wade are in fact out - were EON looking for a writer/director because they were keen to rewrite/replace their original treatment6. Norway as a Bond 25 filming location* Norwegian outlet, Bergens Tidende, has reported that a filmed dubbed "B25" has applied for a 25% subsidy of costs intensive for filming in Noway. It's time to dust off those headphones and crank up the volume for Episode 12 of The Words Are Not EnoughIn this episode Griffin (@griffschiller) and Brody (@brodyserravalli) discuss the following:Tomorrow Never Lies* Ben Whishaw still not confirmed for Bond 25 * In a recent interview with BBC discussing his return to Shakespearean theater in the role of Marcus Brutus in Julius Caesar, Ben Whishaw shed some light on whether or not he will be retuning for Daniel Craig's final outing as James Bond * Whishaw emphasises he knows nothing about the next film and can't even be sure he'll be in it. But he enjoyed stretching his muscles in Spectre in a way Q hadn't before. * "I'm secretly hoping that I'll get to do more action in the next one. I'd like to be a little bit more involved - it would be wonderful. * "But we'll see. Like everything to do with James Bond, it's all a mystery at the moment.” * It would seem odd for Whishaw not to reprise his role in 25, and seeing as the only announcement surrounding the project that has been made is Craig's return and the release date, there's not reason yet to doubt the young Q's return…still…who knows there could be a slight chance Whishaw's days as Q are over. * Christoph Waltz will NOT be returning for Bond 25 and has regrets about Spectre * It would seem that the heresy surrounding Christoph Waltz's return as Blofeld in Bond 25 can be put to rest as the actor has once again stated that he will not be coming back * This comes from an interview with The Times in which Waltz says the following: * "It was an incredible experience to be part of this postwar cultural phenomenon. Yet I can't say that I was happy at every step of the process. I feel that there are some loose ends with my engagement. And I'm not saying that I'm going to be in the next one. I'm not going to be in the next one.” * The actor then went on to discuss some regrets he had with Spectre: * "But let's say, in terms of my feeling that I have sufficiently served a purpose, there are a few white blotches on that map for me, personally. I don't want to be critical of something that doesn't deserve to be criticized, so that's why I'm being a little cryptic. There are just certain things I am unhappy with. I wish I would have gotten the opportunity to iron those glitches out.” * With this lingering question seemingly put to rest, the question remains as to whether another actor will take over the mantle as Blofeld in Bond 25 (a common bond trope) or will Blofeld simply be put to the side?* All Bond actors Definitively, scientifically, stupidly...Ranked based on their attractiveness * A new scientific study has analysed the faces of all the Bonds, according to the Golden Ratio of Beauty Phi, and revealed the best and worst looking of them all. * Dr De Silva, who runs the Centre For Advanced Facial Cosmetic And Plastic Surgery explains how the study works: "The Phi ratio of 1.618 has long been thought to hold the secret of beauty, and now with the computer mapping we can calculate exactly how it applies to men." * The ratio can be applied to faces (or bodies), and the closer the ratios are to 1.618, the more beautiful they supposedly are. * The Ranking: 1.
