In this episode of the Epigenetics Podcast, we talked with Luca Magnani from Institute of Cancer Research and UNIMI in Milan about his work on epigenetic mechanisms of drug resistance and cancer cell dormancy in breast cancer. We start the interview by putting our focus on his significant contributions to the understanding of estrogen receptor-positive breast cancer. In a foundational study from 2013, Professor Magnani and his colleagues illuminated the role of genome-wide reprogramming of the chromatin landscape in conferring resistance to endocrine therapy. This research marked a departure from a purely genetic mutation paradigm, proposing instead that epigenetic modifications play a pivotal role in the development of drug resistance. A fascinating part of our conversation centers on the role of pioneer transcription factors, particularly PBX1, in regulating the estrogen receptor's transcriptional response. Professor Magnani explains how PBX1, typically associated with hematopoietic development, influences estrogen receptor activity, thereby shaping the cancer cell's fate and response to treatment. Continuing our exploration, we discuss the critical distinctions between primary and metastatic breast cancer through the lens of epigenetic reprogramming. By analyzing samples from women with breast cancer, Professor Magnani's work identifies specific enhancer usage that marks the transition to a drug-resistant state which was a breakthrough in linking epigenetic alterations to real-world patient outcomes. He emphasizes that the reliance on genetic mutations alone does not adequately explain the mechanisms of drug resistance, pushing the field to consider the epigenetic landscape more deeply. Our conversation also touches on the evolution of experimental techniques. Professor Magnani shares insights into the transition from traditional ChIP-seq methods to CUT&RUN, demonstrating the need for techniques that cater to the limited material available from clinical samples. This adaptability mirrors the dynamic nature of cancer itself, as cells continuously evolve under therapeutic pressure. As we traverse through the complexities of dormancy and reactivation in cancer cells, Professor Magnani enlightens us on the unpredictable nature of tumor behavior. He describes how cancer cells can enter dormant states and how their awakening is influenced by environmental factors, akin to an evolutionary response to stressors, thus revealing the intricate balance between survival and proliferation. In the latter part of the episode, we explore Professor Magnani's vision for the future of breast cancer research, which includes the need for better animal models that mimic human disease. His pursuit of understanding estrogen receptor behavior both in healthy and cancerous cells reflects a holistic approach to cancer biology, aiming to decipher the transition from normal tissue to malignancy. We didn't manage to get to much of the recent Luchino Visconti retrospective at BFI South Bank but we somehow wanted to mark the moment, and how better than a discussion of BELLISSIMA (1951), particularly through the great Eureka/ Masters of Cinema blu-ray. We discuss its themes of obsession, mother love, fantasy, cinema, the effects of media on private and collective aspirations; how it's a film that announces its fluency from the opening shots; its relation to neo-realism through on-location shooting and the use of non-professional actors; Anna Magnani's tour de force performance, drawing particular attention to the scene where she gets the neighbours involved in the beating by her husband; we note how it's an unusual film for Visconti in that it's central role is a woman's role, a vehicle for Magnani; we discuss the elements of camp, something not usually associated with Visconti; a very entertaining film of great depth; a critique of cinema by one of its greatest exponents; a film one can't imagine bettered; a film worth seeing
https://notesonfilm1.com/2024/11/29/thinking-aloud-about-film-volver-pedro-almodovar-2006/ Richard and I return to discuss VOLVER (Pedro Almodóvar, 2006), interesting to see always but particularly for Richard now having seen WHAT HAVE I DONE TO DESERVE THIS (1984) and his earlier films, which he hadn't seen before, and thus more fully appreciating one of the ‘returns' in VOVER, that of Carmen Maura. We discuss the recurring motifs: Strong women, dreadful men, female solidarity, rape, relationships between mother and child, cemeteries, a star entrance through a tombstone. We note the evidence of Almodóvar's rural background, how his films always reference and often feature the rural as setting. Almodóvar is of the few directors to represent rural customs (the white shirts and dark trousers of the men at the funeral); the rural is also evident in his use of dialogue, which feels so true, and adds a particular texture to his films, phrases that are structures of feeling, modes of understanding. We note how this is in tension with the representation of Madrid, often a character of its own in his films, and in this instance, the particular, working-class Madrid neighbourhoods of this film. We discuss the narrative, and how the film creates a world in which death and violence hover, the work of a filmmaker who doesn't believe in ghosts but is aware of the relevance of the spectral in people's lives and people's understandings. The film looks luminous with a depth and texture to the visuals. Flowers turn up significantly in the film, represented in the poster's design. We can't think of another director who features rape so recurringly, in this case there's a parallell rhyming rape of a father/ daughter with significantly different responses from the mothers; we note too how fathers are often shown as abusive, absent, neglectful. We discuss how Almodóvar often structures his story telling around a history of cinema, a telling through allusions, quotations, references to film, here we note how the film evokes Magnani, Loren, MILDRED PIERCE (Michael Curtiz, 1945), STELLA DALLAS (King Vidor 1937, the Magnani clip in the film is from Visconti's BELLISSIMA (1951). Lastly, we tie all of this to the film's themes: the past, making amends, building bridges, making connections, various kinds of returns.

As a swordsman, he specialises in northern Italian fencing from the early 15th to the 16th centuries, i.e., Fiore, Vadi and into the Bolognese. And he's a successful competitor and sought after instructor. In the late 2010s Dario became a historical martial arts professional, between his teaching activities and the founding of Thokk Personal Armor, a business through which he designs and sells innovative historical martial arts gear, such as the Thokk gloves. The reason I'm chatting to Dario is because I met him in Spain at the Panoplia, and we ended up spending probably five or six hours of the weekend discussing details of Fiore stuff and other things. I'm joined by Peter Magnani - Peter was my very first boss in advertising - and I have to say with all honesty - he helped guide me and mentor me for those key early years of anyones career. We then worked in different and competitive agencies - always in touch but not close enough for me to know what he was experiencing Very candidly Peter shares his mojo story - of great success and peak moments, and of the grind and sacrifice which characterise many careers. You'll hear that when he should have been celebrating his greatest business triumph he was actually in the midst of a profound stress-induced episode. Peter decided to make a profound change his life and founded Beam in 2019, developed on the twin foundational pillars of “open data for good” and ‘helping people to help themselves” by empowering them to better manage everyday micro-stressors and prevent them building up into anxiety or depression. During her career, she received three Outstanding Speaker Awards from the Association for Clinical Chemistry (AACC); and from the College of American Pathologists (CAP)/CAP Foundation, the 2012 Outstanding Communicator Award, the 2014 Distinguished Patient Care Award, and the 2014 Gene and Jean Herbek Humanitarian Award. Dr. Magnani also received the Distinguished Career in Teaching Award from the Tufts University School of Medicine in 2019. In addition, she has been named a Top Doctor in Boston magazine and was included by Castle Connelly in Exceptional Women in Medicine. Dr. Magnani was also named one of the Top 100 Most Influential Laboratory Medicine Professionals in the World by The Pathologist. Dr. Magnani—the former Chair of the CAP Toxicology Committee and member of the CAP Council on Scientific Affairs—has served as the member-at-large of the TDM and Toxicology Division of the AACC and as an editor of Clinical and Forensic Toxicology News.
