A new research paper was published in Oncotarget's Volume 14 on May 26, 2023, entitled, “Deconstructing the role of MALAT1 in MAPK-signaling in melanoma: insights from antisense oligonucleotide treatment.” The long non-coding RNA (lncRNA) MALAT1 is a regulator of oncogenesis and cancer progression. MAPK-pathway upregulation is the main event in the development and progression of human cancer, including melanoma and recent studies have shown that MALAT1 has a significant impact on the regulation of gene and protein expression in the MAPK pathway. However, the role of MALAT1 in regulation of gene and protein expression of the MAPK-pathway kinases RAS, RAF, MEK, and ERK in melanoma is largely unknown. In this study, researchers Valentin Feichtenschlager, Yixuan James Zheng, Wilson Ho, Linan Chen, Ciara Callanan, Christopher Chen, Albert Lee, Jose Ortiz, Klemens Rappersberger, and Susana Ortiz-Urda from the University of California San Francisco and Medical University Vienna demonstrated the impacts of antisense oligonucleotide (ASO)-based MALAT1-inhibition on MAPK-pathway gene regulation in melanoma. “Our results showed that MALAT1-ASO treatment decreased BRAF RNA expression and protein levels, and MALAT1 had increased correlation with MAPK-pathway associated genes in melanoma patient samples compared to healthy skin.” Additionally, drug-induced MAPK inhibition upregulated MALAT1-expression, a finding that resonates with a paradigm of MALAT1-expression presented in this work: MALAT1 is downregulated in melanoma and other cancer types in which MALAT1 seems to be associated with MAPK-signaling, while MALAT1-ASO treatment strongly reduced the growth of melanoma cell lines, even in cases of resistance to MEK inhibition. MALAT1-ASO treatment significantly inhibited colony formation in vitro and reduced tumor growth in an NRAS-mutant melanoma xenograft mouse model in vivo, while showing no aberrant toxic side effects. “Our findings demonstrate new insights into MALAT1-mediated MAPK-pathway gene regulation and a paradigm of MALAT1 expression in MAPK-signaling-dependent cancer types. "I've always prioritized training and running in a way...I've just continued to do it no matter what."Hilary Cairns is a staple in the DC running and triathlon community. She is the founder and leader of Track Pack, a group of hearty runners who meet at 5:45am on Wednesday mornings year round. She sits on the board for Run Hope Work, an organization founded by Jose Ortiz (and modeled after his own experiences) that combines running, mindfulness , and workforce development. She casually "jumps in" many local races. Most recently, she jumped into the Army 10 Miler and won her age group. Hilary has a bit of a different relationship than many others on this shows. Rather than loving the marathon or going after ultras, her favorite distances to race range from the 10K to 10 mile. While her mom was a runner - who ran Grandma's Marathon, a favorite destination race amongst DC runners - Hilary played tennis growing up. She found running in college. While she is an accomplished runner and an organizer in the sport, she approaches training with a bit of lightness. She doesn't overthink the Track Pack workouts she plans, she finds local races on RunWashington, and being a mom of three, she understands that she understands that sometimes you have to adapt to what life throws at you.I really appreciated hearing about how Hilary has found longevity in running and triathlon and managed to prioritize the sport over decades - I think she provides a strong example for those of us who are faced with pressures from all directions and still want to be part of the larger running community and go after big goals. She doesn't overthink running, she just does it and adapts where needed.

Eurico Rosa de Silva is a Brazilian native who is best known for his outstanding career as a jockey in Canada where he won two Queen's Plates and was the leading rider at Woodbine for seven seasons, including five consecutive years until his retirement in 2019. He is now a life coach for several professional athletes. There were many diverse roads and obstacles this talented reinsman took to the eventual successes of his career and they are revealed in his recently published book – “RIDING FOR FREEDOM.” Born on a dairy farm in rural Brazil, Da Silva determined at an early age that he would become a champion jockey. This is the story of Da Silva's battle with his demons. While it describes his ascent to the highest ranks of his athletic profession, it also lays bare his struggle to heal the emotional scars inflicted by an absent and negligent father and his victory over the forces that threatened at time to end his life. Fourth of July weekend will be sparkling with competitive action from sea to shining sea and plenty of reasons to pull down the E-Z Win Forms at Winningponies. The picnic starts on Friday in Altoona, Iowa at Prairie Meadows where they will fire off three rich stakes races – the G3 $225,000 Iowa Oaks; $250,000 Iowa Derby and the continuance of the G3, $300,000 Cornhusker Handicap, a race that has attracted Knicks Go. Participating in the evening fireworks will be the likes of jockeys Florent Geroux, Jose Ortiz, Joel Rosario, Ricardo Santana, Junior Alvarado and trainers Brad Cox, Steve Asmussen, Bill Mott and Mike Maker to name a few. John Hernandez will join our “Regular Guy” to give us the best bets from central Iowa. John Hernandez has been in and around horse racing for almost 40 years now. The late, great West Coast publicist Nat Wess hired him for his first job in the business for the inaugural Fall-Winter meeting at Hollywood Park in 1981. He was the “press box steward” and for $50 bucks a day, he was rubbing elbows with the likes of Gordon Jones, Ernie Mason, Harry Henson, Joe Hirsch, and many others while watching some great races there. He also worked for Trackmaster in the early days of their business in the 1990s, traveling the Eastern seaboard doing on-track seminars for their hand-held telephone modem device that was sort of like “the next gen Mattel Horse Racing Analyzer.” John has been with Prairie Meadows for over a decade, where he serves as the track handicapper and television commentator.

Spark follows the Hijos del Sol crew as they finish up a mural for a group called Healthy Start, which provides social services to immigrant families in East Salinas. The mural is full of vibrant colors and symbolic images. The focus is on the family and the most striking image is of a mother letting her child go so that he may soar over the fields of strawberries. Like many immigrant children, Jose Ortiz had trouble adjusting to living in new country when his family moved the United States from Mexico when he was 11 years old. His awakening happened when a teacher noticed his artistic talents and encouraged him to start working on canvas. However, it wasn't until Ortiz was in college that he took meaningful art classes. Now a successful artist, Ortiz decided that other kids who needed a place to learn about art shouldn't have wait as long as he did. So in 1992 he started Hijos del Sol, an after school program in East Salinas focusing on murals helping keep youth off the streets. All are welcome at Hijos del Sol but most of the participating youth are immigrants due to the fact that Salinas is a farming town, where migrant workers and their families have settled.
