Bisherige Testformate zum Nachweis von Antikörpern gegen enteropathogene Yersinia spp. bei Schweinen konnten lediglich eine Aussage darüber treffen, ob eine Infektion stattgefunden hat, jedoch nicht darüber, welcher der beiden Erreger, Y. enterocolitica bzw. Y. pseudotuberculosis, die Infektion verursachte. Ziel der Arbeit war daher die Entwicklung eines Immunoassays auf Basis der Beadtechnologie, der Serum und Fleischtropfsaft (FTS) von Schlachtschweinen sowohl als positiv bzw. negativ klassifizieren kann, als auch gezielt zwischen Y. enterocolitica und Y. pseudotuberculosis differenziert. 551 Serum- sowie 368 Fleischtropfsaftproben wurden mit dem im Rahmen der Arbeit entwickelten recomBead Yersinia pig Assay (Firma Mikrogen, Neuried, Deutschland), der die rekombinant hergestellten Antigene YopD, YopE, YopH, YopM, YopN, MyfA, PsaA und VAG-A enthält, gegen einen Referenztest, den PIGTYPE® YOPSCREEN ELISA (QIAGEN Leipzig GmbH, Leipzig, Deutschland) auf IgG Antikörper gegen Yersinien getestet. Verglichen mit dem ELISA zeigte der Beadassay im Serum eine Sensitivität von 98,3% und eine Spezifität von 98,4 %. Für FTS lagen Sensitivität und Spezifität bei 93,6 % bzw. 99,0 %. Von 307 im Beadtest positiv getesteten Seren konnten 180 Seren einem Erreger zugeordnet werden; das entspricht einem Prozentsatz von 58,6 %. Dabei handelte es sich um 179 Y. enterocolitica positive Seren sowie ein Y. pseudotuberculosis positives Serum. Beim FTS wurden 164 als positiv getestet, von denen 85 dem Erreger Y. enterocolitica zugeordnet werden konnten, das entspricht einer Differenzierungsquote von 51,8 %. Kein FTS wurde als Y. pseudotuberculosis positiv differenziert. Dies ist die erste Evaluierung eines Testsystems zum Nachweis von Antikörpern gegenüber beiden enteropathogenen Yersinia-Spezies sowie der Fähigkeit einer gleichzeitigen Erregerdifferenzierung. Der entwickelte Beadassay eignet sich zum Nachweis von Antikörpern aus Blut- und Fleischtropfsaftproben von Hausschweinen.

Die Pathogenität von enteropathogenen Yersinien-Spezies wird von einem Virulenzplasmid pYV kodiert. Das pYV kodiert für den Typ-III-Sekretions-/Translokationsapparat und die Yops. Sechs Yop-Effektorproteine sind bekannt, die in die eukaryote Zelle transloziert werden: YopE, YopH, YopM,YopO/ YpkA, YopP/ YopJ und YopT. Die intrazellulären Angriffspunkte und Wirkungen der einzelnen Yops auf die Zielzelle sind Gegenstand aktueller Forschung. In dieser Arbeit wurden erstmals systematisch potentielle Zytoskelettveränderungen von Thrombozyten durch Inkubation mit Yersinia-Monosekretionsmutanten YopH, YopE, YopO, YopP, YopM und den Yersinia-Monodeletionsmutanten DeltaYopH, DeltaYopE, DeltaYopT, DeltaYopO und DeltaYopM untersucht. Im zeitlichen Verlauf adhärieren uninfizierte Thrombozyten durch eine Zunahme der Fläche durch Ausbildung von Filopodien und Lamellipodien. In den durchgeführten Experimenten ähneln die mit der YopP-Monosekretionsmutante inkubierten Thrombozyten uninfizierten Thrombozyten, d.h. YopP zeigt als Einziges der untersuchtes Yops keinen Einfluss auf das Zytoskelett. Mit der YopH, YopM, YopE bzw. YopO-Monosekretionsmutante inkubierte Thrombozyten zeigen kleinere Zellflächen und eine Dominanz von abgerundeten Zellen. Dies deutet auf eine schlechtere Adhäsion der Zellen an der extrazellulären Matrix hin. In mit WA-C(pTRANS,pCJYM-HA) infizierten HeLa-Zellen konnte YopM zunächst im Zytosol, nach 2-4h Infektion zusätzlich im Zellkern lokalisiert werden. Ausserdem zeigten mit WAC(pTRANS,pCJYM-HA) infizierte HeLa-Zellen Zytoskelettveränderungen: ein ungeordnetes Zytoskelett mit Vakuolen, vereinbar mit einer Ablösung der Zelle von der Matrix. Kaya et al. konnten im Affinitätsblot in vitro eine Interaktion von YopM mit der intrazellulären Cystein-Protease Calpain zeigen. Wegen der morphologischen Ähnlichkeit zwischen mit WA-C(pTRANS,pCJYM) inkubierten bzw. mit Calpeptin, einem Calpain-Inhibitor, inkubierten Thrombozyten wurde die von Kaya et al. dargestellte Interaktion zwischen YopM und Calpain in vivo in dieser Arbeit überprüft. Die Präzipitationsversuche aus infizierten HeLa-Zellen über Calpain sowie über YopM ergaben keinen Hinweis auf eine Interaktion von YopM und Calpain. McDonald et al. publizierten RSK 1 und PRK 2 als intrazelluläre Interaktionspartner von YopM. Funktionelle Studien über die Proteine RSK 1 und PRK 2 lassen darauf schließen, dass sie regulierende Einflüsse auf das Zytoskelett, die Translation und das Zellüberleben haben. Diese Funktionen und die beobachteten Einflüsse von YopM auf das Zytoskelett der Zielzelle sind mit einem Modell vereinbar, in dem YopM die Zytoskelettveränderungen durch Interaktion mit RSK 1 und PRK 2 hervorrufen könnte. Diese Hypothese muss durch zukünftige Forschungen geklärt werden.

Salmonellen verfügen über ein sogenanntes Typ III-Sekretionssystem (T3SS), mit dessen Hilfe Effektormoleküle mit zellmodulatorischer Funktion durch die Wirtszellmembran direkt in das Zytosol von Makrophagen, dendritischen Zellen oder Epithelzellen transloziert werden. Das Salmonella-T3SS kann verwendet werden, um heterologe Proteine in den endogenen MHC Klasse I-Antigenpräsentationsweg von eukaryontischen Zellen einzuschleusen. Als T3SS-Trägerprotein hat sich das „Yersinia outer protein E“ (YopE) bewährt. Als Modellantigene dienten die immundominanten Proteine Listeriolysin O (LLO) und p60 des intrazellulären Bakteriums Listeria monocytogenes. Am Beispiel der murinen Listeriose konnte im Tiermodell demonstriert werden, dass die einmalige orale Immunisierung mit einem rekombinanten S. typhimurium-Impfstamm, der entweder chimäres YopE/LLO oder YopE/p60 exprimiert und transloziert, zu einer effektiven und protektiven CD8 T-Zellantwort führt. In der vorliegenden Arbeit wurde erstmals der Einfluss von Booster-Immunisierungen auf die Induktion antigenspezifischer CD8 T-Zellantworten im oralen Mausmodell untersucht. Es konnte zunächst gezeigt werden, dass der rekombinante S. typhimurium-Impfstamm nach homologen Booster-Immunisierungen zeitlich in seiner Fähigkeit beeinträchtigt war, Darm, mesenteriale Lymphknoten und die Milz geimpfter Mäuse zu kolonisieren. Die schnelle Eliminierung der Salmonellen nach wiederholter oraler Gabe beruhte dabei nicht auf p60217-225-spezifischen zytotoxischen CD8 T-Zellen, die durch die primäre Immunisierung induziert worden waren, sondern auf einer gegen den Impfstamm gerichteten Immunantwort, der sogenannten anti-Vektor-Immunität. Die herabgesetzte Kolonisierungsdauer nach Boost-Immunisierungen verhinderte dabei auch eine verstärkte antigenspezifische CD8 T-Zellantwort. In weiteren Experimenten konnte gezeigt werden, dass im Gegensatz zu einer Langzeit-Kolonisierung über einen Zeitraum von 21 Tagen, eine Kurzzeit-Kolonisierung oral immunisierter Mäuse über 6 Tage nicht ausreicht, um eine schützende p60-spezifische CD8 T-Zellantwort gegen Listeriose zu induzieren. Durch die Verwendung von zwei verschiedenen Salmonella-Serovaren (S. dublin und S. typhimurium) im Rahmen einer heterologen „prime-boost“-Immunisierung konnte die Salmonella-spezifische anti-Vektor-Immunität umgangen werden. Diese Strategie führte zu einer verlängerten Kolonisierungsdauer von über 14 Tagen und zu einer verstärkten antigenspezifischen CD8 T-Zellantwort nach Booster-Immunisierungen.

Yersinia enterocolitica produzieren Pathogenitätsfaktoren, die es ihnen ermöglichen, an die Zellen des Wirtes zu adhärieren, diese zu invadieren und protektive Immunantwort des Wirtes zu modifizieren. Dazu gehören die Yersinia outer proteins (YOP), die durch den Typ III Sekretionsweg über die Bakterienmembran in die Wirtszelle transloziert werden. Diese Sekretion wird durch das Chaperon SycE unterstützt. SycE bindet an YopE, verhindert die Degradation, stabilisiert die Faltung des Proteins und leitet es an die Sekretionspore. Mittels Reporterfusionstechnologie mit GFP und BFP sollte durch einen Fluorescence Resonance Energy Transfer (FRET), bei dem die Fluoreszenzenergie von BFP strahlenlos auf GFP übertragen wird wenn beide Proteine in einer Distanz von 1-10 nm liegen, die Interaktion der beiden Proteine intrazellulär dargestellt werden. Es wurden verschieden lange Yop-Fragmente mit dem Reprotergen gfp und SycE mit BFP fusioniert. Ferner sollte mit Hilfe der Reportergene gfp und luc die Produktion von YopE und SycE in vitro verfolgt werden. Zusammenfassend ist die Darstellung der Interaktion mittels FRET schwierig, da mikroskopisch eine erwartete Grünfluoreszenz gezeigt werden konnte, die Negativkontrollen aber durch die Eigenschaften der Reporterproteine nicht eindeutig negativ waren. Desweiteren zeigt sich eine hohe Ausbleichrate von BFP, ferner überlappt sich das Emissionsspektrum von BFP und GFP in einem grossen Bereich, sodass es unabhängig von einem möglichen FRET von BFP zum Auftreten einer entsprechenden Grünfluoreszenz aufgrund der Anregung durch die Lichtquelle kommen kann. Im zweiten Teil zeigt sich, dass die Produktion von YopE und SycE kontinuierlich stattfindet und mit dem Beginn der in vitro Stimulation produziert werden, mit einem Verhältnis deutlich zugunsten von SycE. Diese Ergebnisse sprechen für ein Modell der Chaperon-induzierten, posttranslationalen Sekretion von YopE. Gleichzeitig ist durch die hohe Stabilität von GFP die fluoreszenzmikoskopische Bestimmung der Produktionskinetiken von YopE-GFP und anderen Proteinen begrenzt.

Pathogene Yersinien injizieren während einer Infektion über ihr TypIII-Sekretionssystem bakterielle Moduline, sogenannte Yops, in Immunzellen, um das Immunsystem zu stören. Zu Beginn dieser Arbeit war bekannt, dass die injizierten Yersinia enterocolitica-Moduline YopE und YopT das Aktinzytoskelett angreifen. Mit dem Ziel der näheren Charakterisierung der zugrunde liegenden Mechanismen wurde insbesondere der Effekt von Y. enterocolitica-YopE auf aktinregulierende Signaltransduktionswege in menschlichen Endothelzellen (HUVEC) untersucht. Zu diesem Zweck wurde ein Yersinia-Stamm hergestellt, welcher YopE als einzigen Effektor transloziert. Mit diesem Stamm infizierte ruhende HUVEC zeigten eine Veränderung des Aktinzytoskeletts ähnlich wie nach Mikroinjektion von dominant negativem N17Rac, was auf eine Inaktivierung von Rac hindeutete. Zur weiteren Untersuchung wurden in infizierten Endothelzellen durch extrazelluläre Stimuli einzelne Rho-GTPasen aktiviert und die dabei ausgebildeten Aktinstrukturen beobachtet. Dabei ergab sich keine Beeinträchtigung der Neubildung CDC42- und Rho-vermittelter Aktinstrukturen (Filopodien und Stressfasern) durch YopE, jedoch eine spezifische Hemmung Rac-induzierter Lamellipodien. Frühere Untersuchungen hatten demonstriert, dass es sich bei YopE um ein sogenanntes GAP („GTPase activating protein“) handelt, welches in vitro die Proteine Rho, Rac und CDC42 hemmt. Die Ergebnisse der vorliegenden Arbeit weisen darauf hin, dass in Endothelzellen transloziertes YopE höchst selektiv auf die Ras-ähnliche GTPase Rac wirkt, jedoch keinen Effekt auf CDC42 oder Rho ausübt. Diese Ergebnisse zeigen, dass YopE von Yersinia Rho- GTPase-abhängige Signaltransduktionswege mit einer bemerkenswerten Spezifität in primären Zielzellen beeinflussen kann. Überdies wurde die genannte Spezifität auch in primären Makrophagen nachgewiesen. Weiterhin zeigte sich im HUVEC-Infektionsversuch, dass die Hemmung der typischerweise Rho-vermittelten Aktin-Stressfasern YopT-abhängig ist. Morphologische Veränderungen von Aktinstrukturen, wie sie typischerweise bei der Unterbrechung von CDC42- oder Racvermittelten Signalen vorkommen, wurden nicht beobachtet. In Zusammenhang damit konnte gezeigt werden, dass genannter Effekt auf eine chemische Modifikation und folgliche Inaktivierung von RhoA zurückzuführen ist (Zumbihl et al., 1999). Damit unterscheiden sich YopT und YopE in ihrem Wirkmechanismus und spezifischen Zielmolekül, greifen andererseits jedoch beide direkt an Rho-GTPasen an. Sie könnten deshalb synergistisch bei der Pathogenität von Y. enterocolitica wirken. Darüber hinaus könnten YopE und YopT aufgrund ihrer Spezifität zukünftig als wertvolle Hilfsmittel zur Untersuchung zellulärer Regulationsvorgänge dienen

Podosomen sind aktinreiche Strukturen des Zytoskeletts primärer humaner Makrophagen. Für Adhäsion, Polarisation und Chemotaxis sind diese Strukturen von essentieller Bedeutung. Ihr ständiger Umbau und ihre Regulation unterliegt einer fein abgestimmten Balance der Rho GTPasen Rho, Rac und Cdc42. Pathogene Yersinien spp. haben Aktinzytoskelett von Wirtszellen durch Modulation von Rho GTPasen als Angriffsobjekt gewählt. Mit ihrem plasmidkodierten Typ III Sekretions- und Translokationsapparat werden wichtige Immunfunktionen paralysiert. In dieser Arbeit wurde in primären humanen Makrophagen der Einfluss von Yersinien-Effektoren auf Podosomen untersucht. Konkret interessierte die Frage, welchen Effekt YopE auf diese Strukturen hat. Hierzu wurden in einem standardisierten Verfahren gewonnene und gereinigte Makrophagen gesunder Spender mit unterschiedlichen Mutanten der Spezies Yersinia enterocolitica für verschiedene Zeiten infiziert. Nach Färbung der Zellen mit Rhodamin-Phalloidin wurde die Anzahl der verbliebenen Zellen mit Podosomen im konfokalen Mikroskop ermittelt und statistisch ausgewertet. Es konnte erstens gezeigt werden, daß ein voll virulenter Yersinien Stamm in der Lage ist, nach einer Infektion von bereits 30 min die podosomalen Strukturen der Makrophagen vollkommen zu zerstören. Zweitens sind an diesem Effekt verschiedene Yersinien-Effektoren und zusätzlich der Typ III Sekretions- und Translokationsapparat beteiligt. Drittens reicht YopE für die Zerstörung von Podosomen alleine aus. Viertens ist die GAP-Aktivität von YopE für die Destruktion von Podosomen nicht notwendig und lässt auf GAP-unabhängige Mechanismen von YopE schliessen. Zusammenfassend lassen die Ergebnisse dieser Arbeit vermuten, daß YopE ein wichtiger aber nicht der alleinige Effektor der Yersinien bei der Paralyse von menschlichen Makrophagen und insbesondere der Zerstörung podosomaler Adhäsionsstrukturen ist.
