In this episode of the Epigenetics Podcast, we talked with Dr. Stephan Hamperl from the Helmholtz Zentrum Munich about his work on how conflicts between transcription, replication, and R-loop formation influence genome stability in human cells. During the early stages of his career Stephan studied conflicts between transcription and replication in human cells, particularly focusing on R-loop structures. In our discussion, he explains the formation of R-loops and their impact on genome stability, emphasizing the importance of the orientation of replication forks approaching R-loops in determining DNA damage outcomes. Stephan then delves into his work on the MATAC-Seq method, which analyzes chromatin domains at DNA replication origins to understand replication timing variability. The method involves methylating DNA linkers between nucleosomes and using nanopore sequencing for single-molecule readouts, revealing heterogeneity in chromatin structure at replication origins. Finally, Stephan discusses his automated image analysis pipeline for quantifying transcription and replication activity overlap in mammalian genomes, addressing the challenge of visualizing these processes simultaneously. The conversation concludes with insights into Stefan's future research directions, focusing on understanding transcription-replication conflicts' molecular basis and their potential implications in cancer cell transformation. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2022.11.09.515872v1?rss=1 Authors: Whitebirch, A. C., Barnett, A., Santoro, B., Scharfman, H. E., Siegelbaum, S. A. Abstract: A significant proportion of temporal lobe epilepsy (TLE) patients experience drug-resistant seizures associated with mesial temporal sclerosis, in which there is extensive cell loss in the hippocampal CA1 and CA3 subfields, with a relative sparing of dentate gyrus granule cells and the CA2 pyramidal neurons. A role for CA2 in seizure generation was suggested based on findings of a reduction in synaptic inhibition (Williamson & Spencer, 1994) and the presence of interictal-like spike activity in resected hippocampal tissue from TLE patients (Wittner et al., 2009). We recently found that in the pilocarpine-induced status epilepticus mouse model of TLE there was an increase in CA2 intrinsic excitability associated with a loss of CA2 synaptic inhibition. Furthermore, chemogenetic silencing of CA2 significantly reduced seizure frequency, consistent with a role of CA2 in promoting seizure generation and/or propagation (Whitebirch et al., 2022). In the present study we explored the basis of this inhibitory deficit using immunohistochemical and electrophysiological approaches. We report a widespread decrease in the density of pro-cholecystokinin-immunopositive interneurons and a functional impairment of cholecystokinin-expressing interneuron-mediated inhibition of CA2 pyramidal neurons. We also found a decrease in the density of CA2 parvalbumin-immunopositive interneurons and disruption to the pyramidal neuron-associated perisomatic perineuronal net in the CA2 subfield. These data reveal a set of pathological alterations that may disrupt inhibition of CA2 pyramidal neurons and their downstream targets in epileptic mice. Copy rights belong to original authors. For my lady friends who do not have kids.... what does your friend group look like? Do you also have friends who are child-free? Or do you maybe have a lot of girlfriends you just plain haven't seen in a while, where your friendship has petered out a bit after they decided to have kids? For a lot of single people who are of child-bearing age, especially women, the dropoff of your friend group can be particularly devastating. Comedians Julie Whittner and Katie Von Till are two such women who met in an acting class but inevitably bonded over not just their child-free status but their determination to not ever become mothers. Leaning into this idea, Katie and Julie decided to create a series pilot called "Not Moms" that highlights the trials, tribulations, highs and lows of losing your social group to parenthood. If anyone has ever fed you a line of "hot having anyone to take care of you when you're old" or "not knowing the meaning of life" or "never experiencing true love," or if you've ever wondered if you made the wrong choice, no matter what that choice is, then you'll relate to this conversation I have with the Not Moms about how bullshit the whole gd system is. "Not Moms" is currently on the festival circuit and therefore not available to the public, but we are rooting for its future life so we can improve the visibility and representation of child-free women on tv!
Welcome to the third instalment of our In Her Shoes series, in partnership with the iconic Australian footwear brand Wittner. This exciting new series gives us the rare opportunity to step into the lives of the women who inspire us and discover the strength, courage and ingenuity that has got them to where they are today... So, let’s meet the fabulous Rey Vakili. It’s hard to know where to start with Rey. You could credit her success to the fact that her parents taught her the importance of education and a strong work ethic from a young age. They clearly had an impact on her - at 18 years old Rey went to study Political Science at Yale and then later did an MBA at the Stanford Graduate School of Business. But it was after Yale, that she landed a job as an assistant to the editor-in-chief of American VOGUE Anna Wintour. Her next career move was working for LTK (the largest Influencer Marketing community) after her friend and Bumble founder Whitney Wolfe Herd introduced her to LTK founder Amber Venz Box. She’s now the managing director of LTK Australia. Want to step into her shoes and find out what makes her tick? And also, what shoes she’s coveting this season? Let’s meet the incredible Rey. When Wittner CEO Catherine Williamson set herself the goal of becoming CEO by 35, she wasn’t counting on giving birth to twin girls shortly after her 34th birthday. Despite the vision board she’d created around that particular milestone, she accepted that it wasn’t going to happen. Or not on schedule, at least. But manifestation is powerful, she says. And get there she did – at 35 years old. It had been a long road to CEO, with Williamson starting out at Wittner as a casual employee attracted by the staff discount. She was picked out for head office, and she remembers receiving the call while at a Ben Harper concert. But while Williamson runs a tight ship, responsible for steering one of Australia’s beloved heritage brands and its 500 staff through Covid, and a new chapter in sustainability - never mind the responsibilities of parenting twin pre-schoolers – don’t for a moment think she’s having it all. To kick off our In Her Shoes series, in partnership with Wittner, our founder Georgie Abay has interviewed Williamson. This exciting new series gives us the rare opportunity to briefly step into the lives of the women who inspire us and discover the strength, courage and ingenuity that has got them to where they are today. Williamson tells us about her leadership style and why she thinks women make great CEOs, as well as the traits her best bosses demonstrated, and what she’s learned about the motherhood penalty. It’s safe to say, Wittner’s future is in good hands. And great shoes, of course. Go to wittner.com.auSee omnystudio.com/listener for privacy information.

Currently, 3.2 million Australians live below the poverty line and more than half of those people are women and young girls. Being able to afford sanitary products is a basic necessity – but sadly one that not everyone can afford. Period poverty is something we can’t ignore. Which is why this week, we’re stepping into the shoes of two exceptional women – the Young Australian Of The Year 2021 Isobel Marshall and her best friend and business partner Eloise Hall. Together, they founded TABOO, a brand of menstrual health products that uses 100% of its profits to fund sanitary care and education programs. Did we mention that they founded the company while they were at school? Our new ‘In Her Shoes’ series, in partnership with the iconic Australian footwear brand Wittner, gives us the rare opportunity to briefly step into the lives of the women who inspire us and discover the strength, courage and ingenuity that has got them to where they are today. We want to inspire you, motivate you, and for those in lockdown, help to lift you up. We also hope next time you’re purchasing sanitary products, you’ll consider TABOO – a company driven by passion and purpose. In this episode, we ask the TABOO co-founders about how they’re tackling period poverty, making real change in the world, and building a brand from the ground up. Also don’t miss our photo shoot with Isobel and Eloise, dressed in the joyful new Wittner Spring/Summer collection. Dio pelea. No paraba nunca.

From 2009: We're celebrating the 20th anniversary of the Acme Comedy Theatre. Joining us first will be Dan Kane, the owner of the theatre. He’ll then be joined by the theatre’s founder and director, Travis Oates, who will be followed in succession by performers Julie Wittner, Kevin Small, Jen Parker, Rich Keith and Claudia Dolph.
Brett Wittner Fourteen-year-old high school freshman Brett Wittner of Covington, Louisiana, was brought into the St. Tammany Parish Hospital the morning of Feb. 25, 2012, with a gunshot wound an inch or so behind his right ear. The bullet had fragmented, and bits of it had made their way toward the left side of his head, causing extensive damage to his brain, according to medical records. He died later that day. The police determined that Brett had accidentally shot himself while at a friend’s house. A grand jury subsequently examined the case and declined to indict anyone. However, Brett’s parents don’t agree with that finding and are continuing to look for avenues that will lead to a different outcome. Brett’s mother, Donna Wittner, will discuss the case and where it will go from here. Welcome to the first episode of Stockton to Malone, a podcast from Jacobin magazine. For the first episode, ahead of protests and counterprotests at Planned Parenthood clinics around the country and forthcoming attacks on abortion rights under President Donald Trump, hosts RL Stephens and Micah Uetricht interview Judy Wittner. Before Roe v. Wade in 1973, abortion was illegal throughout most of the country. In 1969, Wittner, who was involved in the feminist movement in Chicago, discovered she was pregnant and wanted an abortion. She sought out assistance from doctors around the Chicago area but was turned away. Eventually, she turned to an illegal feminist abortion service, the Jane Collective, and ended up receiving an abortion on her kitchen table in Evanston, Illinois. We sat down with Judy in that same kitchen to talk with her about that experience and the state of reproductive rights today.

On the thirteenth episode of American History Too! we embark on our very first sequel – picking up where episode six left off in our discussion of Nuclear Fallout. Why did one researcher collect thousands of baby teeth and why are her results quite terrifying? When and where did the US almost nuke its own citizens – and how was disaster averted? Were fallout shelters a genuine attempt to help the population in the event of nuclear warfare or were they merely ‘for show’? Our resident nuclear aficionado has all the answers. Finally, how was nuclear fallout represented in film and literature during the 1950s and 1960s? We explore On the Beach, Dr Stangelove, and why the British government chose to censor Peter Watkins’ The War Game (1965) which depicted the impact of nuclear warfare on Great Britain. And always remember, ye cannae spend a dollar when your deid! We’ll be back in a fortnight with a discussion of the contentious decade that was the 1980s. Lawrence S. Wittner‘s memoir is a retrospective of a life and career that has straddled between academia and social engagement. While many scholars adopt a detached perspective, Wittner has strived to integrate his research with action. He has worked at seemingly every scale, on issues ranging from U.S. foreign policy to local labor relations. He has numerous noteworthy publications to his credit, including a three-volume study of the nuclear disarmament movement, entitled The Struggle Against the Bomb. Wittner’s career as a public intellectual has carried him around the globe, making him a first-hand student of and frequent participant in events of historical importance. His memoir Working for Peace and Justice (University of Tennesee Press, 2012) tells that story, and is available through the University of Tennessee Press. Lawrence S. Wittner is emeritus professor of history at the University at Albany, SUNY. Among other awards and honors, he has received major grants or fellowships from the National Endowment for the Humanities, the American Council of Learned Societies, the Aspen Institute, the United States Institute of Peace, and the John D. and Catherine T. MacArthur Foundation. Learn more about your ad choices. Visit megaphone.fm/adchoices

----more----Pearlsong Press publisher Peggy Elam, Ph.D. chats with the authors of ExtraOrdinary: An End of Life Story Without End in this month's Pearlsong Conversation. Publishers Weekly & others have praised ExtraOrdinary as a true-life tale of transformation: "In this deeply moving and inspiring account, [Michele] Tamaren and [Michael] Wittner share the life story of a man close to their hearts. When Michele’s mother, Betty, married Herman Liss (she was 55 and he was 65), Tamaren finally had a father figure she adored. Always joyful, Herman exuded a love for life that was contagious. His sincere concern for others, his keen intelligence, and his natural grace generated hundreds of meaningful friendship throughout his life. Wittner, who first met Herman as a result of a community service requirement in his high school, became a devoted friend to the older man when Herman nearly lost the resolve to live after Betty’s death. The authors recall, in simple but evocative language, how Herman always encouraged others to follow their dreams and how deep was his commitment to treat others as he would want to be treated. Readers can expect tears to flow as Herman’s life inspires them to be better." (Publishers Weekly) Click on the Podbean player to listen to the recording of today's Conversation. In 1983, when I was in college, I participated in something called a “Die-In.” A group of us set up crosses on the commons and threw ourselves on the ground as if we were dead. The idea, such as it was, was to suggest that nuclear weapons were bad and should be done away with. Quite honestly, I didn’t really think it would work (to put it mildly). But as Larry Wittner shows in his compelling Confronting the Bomb: A Short History of the World Nuclear Disarmament Movement (Stanford, 2009), I was wrong, or at least partially wrong. Larry demonstrates that the nuclear disarmament movement had an impact on government policy. Politicians, not just here in the US but also in unlikely places like the USSR, actually listened to the protesters. But they sensed that a lot of people–like Einstein and me–were very uncomfortable with mutually assured destruction and wanted something done about it. Ronald Reagan listened. And so did Mikhail Gorbachev. After reading Larry’s book, I’m thinking I may organize another “Die-In.” Please become a fan of “New Books in History” on Facebook if you haven’t already.
