After that: Interleukin-1 inhibition in TTP. Researchers explore the potential of recombinant IL-1 receptor antagonist, anakinra, in a murine model of thrombotic thrombocytopenic purpura—an uncommon but potentially fatal disorder with limited therapeutic options. Finally, we'll learn about CAR T-cell therapy outcomes by race and ethnicity in large B-cell lymphoma. This episode's guest is arguably one of the most influential scientists in the human brain mapping community. Dr. Peter Fox, director of the Research Imaging Institute at the University of Texas Health, San Antonio. Early in his career he wrote the seminal paper that showed, using positron emission tomography , that brain-activation related increases in blood flow are accompanied by only small increases in oxidative metabolic -  resulting in the blood locally increasing in oxygenation. This paper set the foundation for understanding all of Blood Oxygen Level Dependent Contrast used in fMRI today. The true purpose of activation-related flow increases is still an open question. The story of the events and details surrounding this are in his review article from the 2012 NeuroImage special issue. It's titled, simply "The coupling controversy." Dr. Fox was also among the first to promote data sharing and pooling with his brainmap database, and early on, established stereotactic coordinates and spatial normalization as a way to put data into a shareable space. He started the annual meeting that pre-dated the Organization for Human Brain Mapping, and also founded one of the major brain mapping journals today, titled: Human Brain Mapping.   Peter had his formative undergraduate education at the extremely unique St. Johns college in Annapolis. He received his MD from Georgetown University, interned at Duke University, then carried out his residency and fellowship at Washington University where he worked closely with Dr. Mark Raichle, who was at the time pioneering PET scanning.   In this discussion, we delve into his contributions in a wide range of topics, from neurovascular coupling to the challenge of spatial normalization - particularly at high resolution - to subject variability, to clinical applications and the ongoing evolution of scientific publishing. Lots of history, content, and insight here. We hope you enjoy it! My name is Rainetta Jones, I was born in Port of Spain Trinidad and Tobago. I came to the United States at the age of 14 with my family. I attended Martin Luther King Jr. HS in Manhattan NYC. I would like to note that I do not have any memories of High School Lunch Break for three years. I graduated at 17 years of age and attended University of Buffalo. My tenure at UB was short lived due to serious neurological anomalies which I later learned was “Brain Scrambling Technologies. While at UB I wanted to pursue a degree in Micro and Molecular Biology; however, that didn't pan out. After Buffalo, I attended Sheffield University in England as a study abroad student. Upon returning to the US, I took time off to Invent, Model, and Dance. In 2000 I had a meeting with a “friend” about my two inventions, the iPod and Kindle and also a novel conceptual hosiery design. All of which were stolen. Later on I learned he had connections to Steve Jobs and was invited by him to the launch of iTunes in Cupertino California. After that meeting I was followed by a Russian Jew who befriended me. He invited me to go to England a couple of days after September 11th, 2001. It wasn't until 2016, I was told that both David and Yuri were Mossad agents and that they had to get me out of the US for the launch of the iPod. I later became a Certified Administrative Professional and worked at the United Nations and several other Government agencies in NYC. In 2019 I graduated from Empire State College in Business Administration. I have no memories of a Secret Space Program however, I do know that my life has been filled with extraterrestrial experiences. After graduation I created the Micro- Business Model using business concepts on a micro-scale for those who would like to start a business with very little money. Today I do spiritual consultations and teach online classes oncea month. My website is

GBP3-STING Interaction in Glioblastoma Coordinates Poor Response to Temozolomide

BUFFALO, NY- October 17, 2023 – A new editorial paper was published in Oncotarget's Volume 14 on May 19, 2023, entitled, “GBP3-STING interaction in glioblastoma coordinates autophagy, anti-oxidative, and DNA repair programs in response to temozolomide.” In their recent editorial, researchers Jun Ma, Ziyu Wang, Clark C. Chen, and Ming Li from the University of Minnesota discussed the methylating agent, Temozolomide (TMZ). TMZ is the standard adjuvant chemotherapeutic drug for glioblastoma, which constitutes 17.7% of overall primary central nervous system tumors. The survival rate of this WHO Grade IV tumor has achieved a clinically significant prolongation of 2.5 months overall and an increase of 16.3% 2-year survival rate. However, one intractable challenge is the diverse reactions to temozolomide treatment. “The acquired resistance to the standard adjutant radiochemotherapy including temozolomide has favored the recurrence of some glioblastoma cases and has kept the milestone from moving forward for more than 15 years.” Guanylate-binding proteins (GBPs) are a group of dynamin-related large (~65 kDa) GTPases expressed in response to interferon and mediate intracellular immunity. Consisting of 7 members in humans, little is known about the function of GBPs beyond their role in innate cellular immunity. After recent years of dedication to the GBP family, its role in glioblastoma's development and recurrence has drawn great attention. “More recently, Li's lab did informatic analysis of clinically annotated glioblastoma datasets, laboratory studies of protein-protein interaction, and functional characterization after depletion or exogenous expression.” GBP family members such as GBP1, GBP2, GBP3, and GBP5 are highly elevated and play pro-tumor roles through multiple mechanisms in glioblastoma. Although other GBP family members did not show a prominent relationship with treatment resistance at the present stage, GBP3 showed significant up-regulation in response to temozolomide. Furthermore, it's revealed that high levels of GBP3 expression in glioblastoma was associated with a worsened survival after temozolomide treatment. Consistent with this observation, exogenous expression of GBP3 induced temozolomide resistance in independent patient-derived glioblastoma neurosphere lines, while GBP3 silencing conferred temozolomide sensitivity, both in vitro and in vivo. “This sensitivity was associated with the accumulation of cytoplasmic DNA fragments, suggesting the involvement of Stimulator of interferon genes (STING).” DOI - https://doi.org/10.18632/oncotarget.28370 Correspondence to - Ming Li - m2li@umn.edu Sign up for free Altmetric alerts about this article - https://oncotarget.altmetric.com/details/email_updates?id=10.18632%2Foncotarget.28370 Subscribe for free publication alerts from Oncotarget - https://www.oncotarget.com/subscribe/ Keywords - cancer, GBP3, MGMT, glioblastoma, temozolomide resistance, STING About Oncotarget Oncotarget (a primarily oncology-focused, peer-reviewed, open access journal) aims to maximize research impact through insightful peer-review; eliminate borders between specialties by linking different fields of oncology, cancer research and biomedical sciences; and foster application of basic and clinical science. Lead Story:   Heterogeneous neuroimaging findings across substance use disorders localize to a common brain network  Nature Mental Health This study used network mapping approaches and a functional connectome from a large cohort of healthy participants (n = 1,000) to test whether neuroimaging abnormalities across substance use disorders map to a common brain network. Starting with coordinates of regional brain atrophy from 45 studies, researchers found that 91% of the neuroimaging findings mapped to a common brain network specific to substance use disorders compared to atrophy associated with normal aging and neurodegenerative disease. Coordinates of functional MRI abnormalities from 99 studies mapped to a similar brain network. Neuroimaging abnormalities across substance use disorders map to a common brain network that is similar across imaging modalities, substances, and lesion locations that cause remission from substance use disorders. Dr. Friedlander underscores the significance of the mitochondria, our cell's powerhouse, attributing improved sleep, reduced disease, and enhanced overall health to its proper function. He highlights the critical influence of sunlight in life's evolution on Earth and its symbiotic relationship with mitochondria. However, the contemporary world isn't without its health pitfalls. Dr. Friedlander cautions against the adverse health implications of prevalent technologies, such as computers, cell phones, and Wi-Fi. Their frequencies, vastly differing from the human body's natural Schumann Hertz at 7.83 Hertz, pose potential risks to our cognitive and physical well-being. Emphasizing the rejuvenating power of natural light, both physicians recommend basking in morning and afternoon sunlight to realign our body's circadian rhythm, boost healing, and potentially thwart specific ailments. The conversation also touches upon collagen's indispensable role in our health, from skin sustenance to muscular and skeletal support. They underscore the merits of collagen supplementation, particularly from superior sources, and its integral role in athletic prowess and recuperation. Dr. Friedlander champions a holistic health approach: prioritizing outdoor activities, indulging in a nutrient-rich diet emphasizing proteins and fats, and minimizing contact with detrimental technologies. The overarching theme? Nature and our interaction with it often house the remedy.   References: ===================================== Click here for Dr. J's links: https://www.justinhealth.com/links ===================================== ***Click below to SUBSCRIBE for more Videos http://www.youtube.com/subscription_center?add_user=justinhealth =====================================

If you work in construction, you know that we have a problem. We need more young people! Someone needs to connect the countless dots of classroom learning, workplace skills, career awareness, and student interests. Lydia Burns is that connector of dots.  More formally, Lydia is known as a senior manager with Allied Construction Industries, a trade group that works on advocacy, networking, and solutions for anyone within the construction industry. Lydia's specific focus is on the Construction Career Center, and even more specifically their K-12 programs, which work to familiarize students with various aspects of construction. The goal of these programs is to increase the number of young people heading into construction.  Here is what the ACI program does: Takes a systemic, big-picture approach to increasing student awareness and working them toward actual jobs, Fosters relationships between companies and groups of students as early as 5th grade, Seeks to reduce the anxiety many students feel regarding the math skills that are foundational to jobs in engineering and construction, Helps schools develop and implement project-based learning curriculum focusing on connecting math lessons to the real-world applications, Engages students in an actual small-scale construction project within the school building, Coordinates sequential training: 7th-grade framing, 8th-grade pipefitting, 9th-grade electrical, and so on. As you can imagine, there are challenges to this kind of work. Changing public school curriculum takes time. Putting a bunch of inexperienced high school kids in the middle of a mock construction project has risks. It's difficult to coordinate the schedules and align the instruction of various educators and contractors.     Here are some of the things ACI does to meet these challenges:  Becomes familiar with the entire public school administrative structure within a state and district; Facilitate communication and policy alignment among OSHA, insurance companies, attorneys, and schools regarding the presence of minors on work sites;  Establishes and enforces GPA requirements and other eligibility requirements for participants; Equips classroom teachers with lesson plans that focus on hands-on-learning .  As a result of their work, ACI in the Cincinnati Public Schools has seen students receive industry-recognized credentials before they leave high school.  Here are some other bits and pieces from this episode: Tyler and Lydia share their own frustrations with math back in the day. We explore how working in residential construction can lead to work in industrial construction.  Lydia explains the message she communicates to school districts: “Hope is not a strategy.” We celebrate that most state departments of education have finally acknowledged that not everyone should be heading to college.  Learn more about Lydia, ACI, and their programs: Lydia's LinkedIn -  ACI's web page - Info about ACI's k-12 Program Find Us Online: BrosPodcast.com - LinkedIn - Instagram - Facebook - Eddie's LinkedIn - Tyler's LinkedIn If you enjoy the podcast, please rate us on Apple Podcasts or wherever you listen to us!
