Les chats tricolores, aussi appelés "écaille de tortue" ou "calico", présentent une combinaison de trois couleurs : noir, roux et blanc. Fait fascinant : ces chats sont presque toujours des femelles. Cette particularité n'est pas liée au hasard, mais à la génétique des chromosomes sexuels, et plus précisément au gène de la couleur du pelage porté par le chromosome X.Chez les mammifères, le sexe est déterminé par deux chromosomes : XX pour les femelles, XY pour les mâles. Or, les gènes responsables des couleurs rousse et noire chez le chat sont situés sur le chromosome X. Le blanc, lui, est déterminé par un autre gène, indépendant, qui agit en masquant certaines zones de couleur.Concrètement, une femelle (XX) peut hériter d'un gène roux sur un chromosome X et d'un gène noir sur l'autre X. Mais les cellules d'un organisme femelle ne peuvent pas exprimer les deux chromosomes X en même temps. Un phénomène appelé inactivation du chromosome X, ou "lyonisation", entre alors en jeu : dans chaque cellule, un des deux chromosomes X est désactivé au hasard. Résultat : certaines zones du corps expriment le gène du pelage noir, d'autres celui du pelage roux. C'est ce mélange aléatoire qui produit l'apparence tricolore. Les taches blanches, elles, sont dues à un gène différent qui bloque la pigmentation à certains endroits.Chez les mâles (XY), les choses sont différentes : ils n'ont qu'un seul chromosome X, donc un seul gène de couleur parmi le noir ou le roux. Ils ne peuvent donc pas naturellement avoir les deux couleurs. Voilà pourquoi les mâles sont en général soit entièrement roux, soit entièrement noirs, parfois avec du blanc, mais rarement tricolores.Cependant, il existe une exception génétique rare : le syndrome de Klinefelter, chez les chats mâles porteurs de deux chromosomes X et un Y (XXY). Ce déséquilibre chromosomique leur donne la capacité d'exprimer à la fois les gènes du noir et du roux, avec une inactivation d'un des X comme chez les femelles. In episode 276 of The Just Checking In Podcast we checked back in with James Roffey. James is a personal trainer and semi-professional footballer. He was previously the business owner of his personal training brand ‘The Mental Health PT' but due to personal and business reasons, has rebranded and relaunched it as ‘Ultimate Body and Mind'. James first came on the pod in JCIP #183 where he discussed his journey into personal training and his struggle and recovery from a long-term eating disorder, which was triggered by three major life events. Since then, a massive amount has happened in James's life, positively and negatively. For the former, he signed for non-league club Sheffield FC, only for the team to sign a professional goalkeeper the day before the transfer window shut and make him the number two goalkeeper at the last minute. As a 36-year-old, James needed to play minutes and had to leave the club and join another team as a result. For the latter, he has separated from his wife, which became messy and caused James severe mental health difficulties. During this separation, James discovered he was infertile because of a genetic condition he had called Klinefelter syndrome which, according to the NHS website, is characterised by boys being born with an extra X chromosome, so for James he has XXY, as opposed to XY for other men without the condition. It's important to state that the X chromosome is not a "female" chromosome and is present in everyone. The presence of a Y chromosome denotes male sex. Symptoms of Klinefelter Syndrome in childhood can include: shyness and low self-confidence, problems with reading, writing, spelling and paying attention, mild dyslexia or dyspraxia, low energy levels, and difficulty socialising or expressing feelings. In teenagers, it is characterised by growing taller than expected. Finally in adulthood, as James tragically discovered, it is characterised by infertility as well as low sex drive, smaller testes and erection problems, which James bravely admits on the podcast but originally attributed it to his eating disorder. Kleinefelter Syndrome is also recognised as a Difference in Sexual Development (DSD). James also began suffering from cluster migraines because of the stress of his marriage breaking down, as well as a myriad of other events happening which we can't discuss on this podcast. James's uncle also tragically took his own life in 2024 after trying to take his own life previous to that three times. James says his uncle was let down by the system; the healthcare system for not providing adequate care, and the family court system as his uncle separated from his wife and claims he was parentally alienated from his step-children by his ex-wife. In Part 3, we discuss James's business rebrand and why he did it, a deep dive into his marriage breakdown, his infertility diagnosis, drama in his football career and how he took himself from rock bottom to where he is now. Infertility is one of the most stigmatised experiences one can feel as a man for obvious reasons and we are very proud of James for the bravery he has shown on this podcast to discuss it so openly and courageously for all of the men who live with Klinefelter Syndrome in the UK, which is currently as high as 1 in 600 in this country. Virginia (Ginnie) Isaacs Cover has worked throughout her career with children and adults with complex medical conditions and developmental disabilities. She is an advocate for those with disabilities and their families and published a widely read guidebook for those affected by X and Y chromosome variations. She explores the impact of a prenatal diagnosis on a young family in her new book, Supplemental Needs: A Novel. Ginnie became a “special needs parent” when her younger son was diagnosed prenatally with 47,XXY, also known as Klinefelter syndrome. Although sex chromosome aneuploidy (SCA) is more common than Down syndrome at an estimated 1 in 400 live births, it is recognized and properly diagnosed in fewer than 25 percent of the affected population. She became a passionate advocate for families affected by disabilities, including the X and Y chromosome variations, and provides educational programs for families and professionals who work with this population. In 2012, she published Living with Klinefelter Syndrome, Trisomy X, and 47,XYY. The book provides clear explanations of the genetics involved, as well as diagnosis, disclosure, development from infancy through potential health and fertility issues, and educational and psycho-social considerations. This guidebook has helped thousands of people grow their understanding of this condition. She believes it is equally important for literature to cover the emotional impact that prenatal testing has on parents while also improving early identification of X and Y chromosome conditions. She felt this could only be done through the sharing of stories. Her goal in writing the novel was to explore the impact of a prenatal genetic diagnosis on a young family who decides to continue a pregnancy despite a medical geneticist's advice. Supplemental Needs: A Novel portrays a young family whose lives change on learning through prenatal genetic diagnosis that their baby will have Klinefelter syndrome, most likely causing some degree of disability, as well as infertility and possible sexuality issues. In this episode, Ginnie speaks about prenatal counseling; termination of pregnancy when chromosomal variations exist; societal acceptance of KS, the neurodiverse, LGBTQ individuals and how it has changed over time; and attitudes in Judaism toward LGBTQ individuals and why these are so different across the branches (Reform, Conservative, Orthodox) Supplemental Needs incorporates accurate information and creates needed conversation around what we each believe about continuing a pregnancy where there may be some level of disability, or when there may be a slightly elevated rate of homosexuality, or of gender dysphoria. Readers gain greater awareness and understanding about the parental experience of when a child is not following a typical developmental trajectory, familiarizing the public with sex chromosome aneuploidy in a non-sensational manner. "I want readers to experience the sadness, fear, and hopefulness felt by [the characters] as they wrestle with the choices that must be made, and the impact they have on their son, and their family," Ginnie says. "Through the characters and events, readers will also confront the research suggesting that sexual orientation and identity may be heavily influenced by DNA." Ginnie lives in the Washington, D.C., metro area and has two adult sons and three grandchildren. She volunteers through her synagogue for the Interfaith Refugee Support group. She also is a board member for L'Arche Long Island, a non-profit agency providing housing for adults with developmental disabilities, vocational and life skills training. Ginnie holds a Master of Social Work degree from the University of Michigan. Ryan is the president and founder of California-based non-profit Living with XXY. Ryan is a 37-year-old with Klinefelter syndrome or 47, XXY. Klinefelter syndrome results from an extra X chromosome, affecting roughly 1 in 500 males. Ryan's parents learned about his XXY diagnosis during his mother's pregnancy. His experiences navigating XXY and those of the over 600 families he's met help guide and inspire Ryan's work today. While Ryan's background includes work as a fine dining chef and action sports photographer, he currently spends most of his time advocating for his community through his non-profit. Living with XXY was officially established in 2019 and focuses on creating a community, spreading awareness, and sharing positive traits for those with an XXY diagnosis. Living with XXY is growing daily, reaching over 40 countries. Ryan is traveling around the US, meeting with clinics, professors/students, other professionals, and families to share the work of Living with XXY and create opportunities for connection for those with XXY.
Brittany is the mother of a 4 1/2-year-old boy named Callan. Their son was experiencing bloody noses at a young age. After a week in the hospital, Callan was diagnosed with Aplastic anemia, which is a rare but serious blood condition that occurs when your bone marrow cannot make enough new blood cells for your body to work. At the same time, he was also diagnosed with Klinefelter syndrome.

The Reverend Harcourt ("Harky") Klinefelter joins Rachel and David to share memories from the time he spent working with the Rev. Dr. Martin Luther King, Jr. Harky, a DA member, talks about serving as Dr. King's press officer, and the Selma and Montgomery marches. He brings the voices of long-ago freedom fighters to life, and about working on Dr. King's funeral. I had planned to go all the way but I wasn't sure they were going to be able to take me because they taken all the names and I didn't take anything but my coat. So I had to sleep (in the) cold, and the heaters (in the tent) went out and the wind and everything was real terrible, and so I stayed woke (awake) that night by the little gas burner they had outside. The next day-- the next day we marched and marched and it rained, and that's the day we really had to walk hard all that rain. I didn't get a cold, the next day it rained which was Tuesday, that was through Lownes country, Yeah, that's the day you had to `pick them up and put them (your feet) down'. Blisters and everything, oh boy I'm telling you, I said I know they (the racists) they're going to try something even if they have to wait until we come back. I said `Lord,I say, I know these people, in Lownes country. All I want you to do is take care of me and give me the strength to walk faster through this stretch. I mean, I prayed that prayer all the way through there. I didn't have any water all day because if we stopped we had to run to catch up, you know, and I wanted to be credited for walking all the way So I just did without water, and my tongue was swole (swelled) up. The next day, ah, the nurses out there they waited on me and she came and she say (said) `Little girl are you going to march today?' I say (said) `Sure I am.? All those blisters?' I say (said) `There not hurting, only when I stop'. And you stayed out all the time? `All the time, every camp we stayed in. I slept in it.' How did you feel when you marched up the street to the Capital? `My feet were so bad! and when I had the flag carrying, one of the biggest ones out there, I felt so happy, I didn't knew what to do Dr King, I think he's the greatest that's happened. Really, it's just like heaven. And I'll go anywhere he tells me to go, really, with blisters, sores, sore head, anything. You know, I couldn't hardly breathe when I was out there, I fainted, I passed out, and everything, but I ended up going on ahead. What do you feel the march was accomplished? `We're 50 years ahead from where we started when this voter registration started. Really in Selma, 50 years, because people can walk around and not be afraid. If someone get killed that doesn't stop them, I didn't walk 50 miles in vain.' You feel that people will be afraid to register vote? `Well, I don't think they should be afraid because after all they have been killing Negroes all the time and noting has be done about it. They do find out who killed'em (them) now. This has been a big improvement? `If you will excuse me for the slang, "It's just a nigger dead."'

Kelly Stine is the mother to Connor, who is 17, a senior in high school. Connor encourages other boys his age to share their stories and help build a community for teenagers with XXY. Connor loves to play Roblox and Brawlstars. He also enjoys his government class because his teacher is super funny. At the end of 10th grade, she noticed his maturity was not developing like others. After some blood work, Kelly noticed his FSH levels were off the charts. She started to learn about Klinefelter syndrome and asked her doctor to do a Karyotype. Once he was diagnosed, they did Micro-Tese which came back negative. Kelly says "starting testosterone has been a night-and-day difference for Connor."
Carson Blake shares with us about her son Louden, who is now four years old and has been in Early Intervention since he was six months old. Carson talks about why EI has been super helpful for her son and his accomplishments. Carson also got her state of Missouri to add Klinefelter syndrome to the First Steps Early Intervention system for infants and toddlers, birth to age three, who have delayed development or diagnosed conditions associated with developmental disabilities.
Anne Price had an NIPT screening due to her age of 41. One week later, while their family was on vacation, her two older kids received a call while waiting for the Finding Nemo ride at Epcot. Her phone rang. Her OBGYN, who had a very somber tone of voice, delivered the news horribly, telling her the results were a sex chromosome abnormality and the mention of Klinefelter syndrome. He said, "The good news is that your child doesn't have Down syndrome." The call got disconnected, and Anne couldn't leave the line, so she started to cry uncontrollably during the ride.
Colleen Sander's son, Jackson, was born at 29 weeks and wasn't speaking at the age of two. Once Jackson started school, he was given speech therapy, and over time, something didn't seem right for Colleen. Jackson, 14, had some routine blood work done a few days before receiving a strange phone call from the NHS on a Sunday evening around 9 p.m. The doctors asked for him to come back and do some follow-up testing. Colleen jumped on Google to research what the results might have meant. She started to cry when she was reading about Klinefelter syndrome, checking all the boxes off that this was most likely what her son could have.

After his two boys were all grown up, Russell was diagnosed with mosaic Klinefelter syndrome in his mid 40's. After extensive research on mosaic XXY, even after his doctors told him there was no way he could have XXY, a karyotype test years later revealed he did. What is Mosaic Klinefelter syndrome? Seth and Erin have a son named Chipper, who was diagnosed with Klinefelter syndrome in utero from non-invasive prenatal testing. They have embraced their son's diagnosis, and Chipper has met all his milestones.
Nathan Johns started trying for kids around 28 years old. After trying for over a year and nothing, he went in for testing. He was told he had a low sperm count and low testosterone, but no further testing was done. After his first marriage failed he met his second wife with her knowing he couldn't have kids. Charlie Plaisance got married when he was 27 and felt old enough to start trying for kids immediately. After trying for some time, he and his wife decided to get testing done. After giving a sperm sample, charlie learned he had three immobile sperm and later found out he would be diagnosed with Klinefelter syndrome (47 XXY). Ryan Turesson is from Brisbane, Australia. In 2007, he was trying for kids with his partner. Nothing was happening, so she went in for fertility testing. Her tests returned, saying nothing was wrong. So Ryan went in for testing and later found out he had Klinefelter syndrome. Learning he was sterile and couldn't have kids was devastating, sending him into a dark depression. Yvette Maano is the mother of a young boy named Sammy. During her pregnancy, she was flagged for her baby boy having Klinefelter syndrome. After her diagnosis, her doctor handed her outdated information and asked if she wanted an abortion. Ariel Ze'evi might be the first man to share his story openly with the world from Israel. Ariel was diagnosed with Klinefelter syndrome while trying to create a family with his wife. He hid his diagnosis from family and friends for many years. After a successful Micro-Tese surgery and two beautiful boys, Ariel speaks out to break the stigma. Jared Pike was diagnosed with Klinefelter syndrome nearly 10 years ago while trying to have kids. After trying for more than five years and multiple sperm tests, a new doctor suggested doing a Karyotype. With his wife's support, Jared decided to take a year off from their fertility journey to process, understand, learn and grieve his new diagnosis. After Sierra Westley Wilson became pregnant at 27, she wanted to know the gender of their baby, so they did NIPT, and they were told they had a 36% chance of their baby having Klinefelter syndrome. Once her son was born, they did a routine blood draw, and two weeks later, they learned their son had 48, XXXY.
At 41 years old, Ryan Brown opens up to spread more awareness about his life with Klinefelter syndrome. When he had been trying to have kids with his ex-wife for over a year, the doctors recommended they both do fertility testing. After a sperm sample and a series of other tests, Ryan discovered he had XXY. Ryan is now on track to compete in bodybuilding and is a head chef at a group home.
