Yet, little is known about how dopamine modulates hippocampal representations of emotionally-salient stimuli to inform appropriate resolution of approach versus avoidance conflicts. We here study dopaminoceptive neurons in mouse ventral hippocampus (vHipp), molecularly distinguished by their expression of dopamine D1 or D2 receptors. We show that these neurons are transcriptionally distinct and topographically organized across vHipp subfields and cell types. In the ventral subiculum where they are enriched, both D1 and D2 neurons are recruited during anxiogenic exploration, yet with distinct profiles related to investigation and behavioral selection. In turn, they mediate opposite approach/avoidance responses, and are differentially modulated by dopaminergic transmission in that region. Together, these results suggest that vHipp dopamine dynamics gate exploratory behaviors under contextual uncertainty, implicating dopaminoception in the complex computation engaged in vHipp to govern emotional states. Abstract: Hyperexcitability in the orbitofrontal cortex (OFC) is a key clinical feature of anhedonic domains of Major Depressive Disorder (MDD). However, the cellular and molecular substrates underlying this dysfunction remain unknown. Here, cell-population-specific chromatin accessibility profiling in human OFC unexpectedly mapped genetic risk for MDD exclusively to non-neuronal cells, and transcriptomic analyses revealed significant glial dysregulation in this region. Characterization of MDD-specific cis-regulatory elements identified ZBTB7A, a transcriptional regulator of astrocyte reactivity, as an important mediator of MDD-specific chromatin accessibility and gene expression. Genetic manipulations in mouse OFC demonstrated that astrocytic Zbtb7a is both necessary and sufficient to promote behavioral deficits, cell-type-specific transcriptional and chromatin profiles, and OFC neuronal hyperexcitability induced by chronic stress, a major risk factor for MDD. These data thus highlight a critical role for OFC astrocytes in stress vulnerability and pinpoint ZBTB7A as a key dysregulated factor in MDD that mediates maladaptive astrocytic functions driving OFC hyperexcitability. Abstract: The cellular complexity of the human brain is established via dynamic changes in gene expression throughout development that is mediated, in part, by the spatiotemporal activity of cis-regulatory elements. We simultaneously profiled gene expression and chromatin accessibility in 45,549 cortical nuclei across 6 broad developmental time-points from fetus to adult. We identified cell-type specific domains in which chromatin accessibility is highly correlated with gene expression. Differentiation pseudotime trajectory analysis indicates that chromatin accessibility at cis-regulatory elements precedes transcription and that dynamic changes in chromatin structure play a critical role in neuronal lineage commitment. In addition, we mapped cell-type and temporally specific genetic loci implicated in neuropsychiatric traits, including schizophrenia and bipolar disorder. Together, our results describe the complex regulation of cell composition at critical stages in lineage determination, serve as a developmental blueprint of the human brain and shed light on the impact of spatiotemporal alterations in gene expression on neuropsychiatric disease. Abstract: Brain region- and cell-specific transcriptomic and epigenomic molecular features are associated with heritability for neuropsychiatric traits, but a systematic view, considering cortical and subcortical regions, is lacking. Here, we provide an atlas of chromatin accessibility and gene expression in neuronal and non-neuronal nuclei across 25 distinct human cortical and subcortical brain regions from 6 neurotypical controls. We identified extensive gene expression and chromatin accessibility differences across brain regions, including variation in alternative promoter-isoform usage and enhancer-promoter interactions. Genes with distinct promoter-isoform usage across brain regions are strongly enriched for neuropsychiatric disease risk variants. Using an integrative approach, we characterized the function of the brain region-specific chromatin co-accessibility and gene-coexpression modules that are robustly associated with genetic risk for neuropsychiatric disorders. In addition, we identified a novel set of genes that is enriched for disease risk variants but is independent of cell-type specific gene expression and known susceptibility pathways. Our results provide a valuable resource for studying molecular regulation across multiple regions of the human brain and suggest a unique contribution of epigenetic modifications from subcortical areas to neuropsychiatric disorders. Abstract: Prior to identifying clusters in single cell gene expression experiments, selecting the top principal components is a critical step for filtering out noise in the data set. Identifying these top principal components typically focuses on the total variance explained, and principal components that explain small clusters from rare populations will not necessarily capture a large percentage of variance in the data. We present a computationally efficient alternative for identifying the optimal principal components based on the tails of the distribution of variance explained for each observation. We then evaluate the efficacy of our approach in three different single cell RNA-sequencing data sets and find that our method matches, or outperforms, other selection criteria that are typically employed in the literature. Resting-state functional magnetic resonance imaging (rsfMRI) measures brain activity at rest, which is closely linked with cognition and clinical outcomes. The role of genetics in human brain function is largely unknown. Here we utilized rsfMRI of 44,190 multi-ethnic individuals (37,339 in the UK Biobank) to discover the common genetic variants influencing intrinsic brain activity. We identified and validated hundreds of novel genetic loci associated with intrinsic functional signatures (P < 2.8 * 10^{-11}), especially for interactions of the central executive, default mode, and salience networks involved in the triple network model of psychopathology. A number of intrinsic brain activity associated loci had been implicated with brain disorders (e.g., Alzheimer's disease, Parkinson's disease, schizophrenia) and cognition, such as 17q21.31, 19q13.32, and 2p16.1. Genetic correlation analysis suggested the shared genetic influences among intrinsic brain function, brain structure, and brain structural connectivity. We also detected significant genetic correlations with 26 other complex traits, such as education, cognitive performance, ADHD, major depressive disorder, schizophrenia, sleep, and neuroticism. Heritability of intrinsic brain activity was enriched in brain tissues. The reported risk genes of Alzheimer's disease typically had stronger associations with intrinsic brain activity than brain structure, and the associated genes of intrinsic brain activity were enriched in multiple biological pathways related to nervous system and neuropathology (P < 1.8 * 10^{-9}). Abstract: Strong phenotype-genotype associations have been reported across brain diseases. However, understanding underlying gene regulatory mechanisms remains challenging, especially at the cellular level. To address this, we integrated the multi-omics data at the cellular resolution of the human brain: cell-type chromatin interactions, epigenomics and single cell transcriptomics, and predicted cell-type gene regulatory networks linking transcription factors, distal regulatory elements and target genes (e.g., excitatory and inhibitory neurons, microglia, oligodendrocyte). Using these cell-type networks and disease risk variants, we further identified the cell-type disease genes and regulatory networks for schizophrenia and Alzheimer's disease. The cell-type regulatory elements (e.g., enhancers) in the networks were also found to be potential pleiotropic regulatory loci for a variety of diseases. Further enrichment analyses including gene ontology and KEGG pathways revealed potential novel cross-disease and disease-specific molecular functions, advancing knowledge on the interplays among genetic, transcriptional and epigenetic risks at the cellular resolution between neurodegenerative and neuropsychiatric diseases. Finally, we summarized our computational analyses as a general-purpose pipeline for predicting gene regulatory networks via multi-omics data. Copy rights belong to original authors. Abstract: The basal ganglia, best known for processing information required for multiple aspects of movement, is also part of a network which regulates reward and cognition. The major output nucleus of the basal ganglia is the striatum, and its functions are dependent on neuronal compartmentation, including striosomes and matrix, which are selectively affected in disease. Striatal projection neurons are GABAergic medium spiny neurons (MSNs), all of which share basic molecular signatures but are subtyped by selective expression of receptors, neuropeptides, and other gene families. Neurogenesis of the striosome and matrix occurs in separate waves, but the factors regulating terminal neuronal differentiation following migration are largely unidentified. We performed RNA- and ATAC-seq on sorted murine striosome and matrix cells at postnatal day 3. Focusing on the striosomal compartment, we validated the localization and role of transcription factors and their regulator(s), previously not known to be associated with striatal development, including Irx1, Foxf2, Olig2 and Stat1/2. In addition, we validated the enhancer function of a striosome-specific open chromatin region located 15Kb downstream of the Olig2 gene. These data and data bases provide novel tools to dissect and manipulate the networks regulating MSN compartmentation and differentiation and thus provide new approaches to establishing MSN subtypes from human iPSCs for disease modeling and drug discovery. Welcome to your Remember your Music program, Nº14. We continue with the decade of the 70s and find Mike Oldfield, Aphrodite's Child and after his separation Vangelis and Demis Rousos Vangelis:  In any case, the diversity and complexity of the work contained in his discography makes it difficult to catalog him as a purely New Age artist, since he is even considered one of the pioneers of the avant-garde of electronic music born in the mid-1970s. Of my: After the dissolution of Aphrodite's Child, Demis continued recording sporadically with his bandmate, the also outstanding musician Vangelis. They recorded together in 1977 the Magic album whose biggest success was "Because", known in Spanish as "Die next to my love" and recorded in seven languages ??in total. The album Demis (1982), which included "Tu libertad" -a vocal adaptation of the song "Race to the end", is part of the soundtrack of the Oscar-winning film Carros de Fuego-, while Roussos also appears on the soundtrack of the film Blade Runner (1982), all successful and, finally, in 1984 the album Reflection
