He shared a strange experience he had in Idaho Falls around 2008, when all radio stations were playing music from the 1920s or 1930s instead of their usual programming. David recalled hearing news broadcasts mentioning an atmospheric event and wondered if anyone else experienced it. Richard speculated about the possibility of a time slip or time dilation as a cause for the strange radio phenomenon David experienced.Shana is in Missouri shared her experiences communicating with orbs and receiving messages telepathically from them. Sarah Ellerby, CEO, Nova Pangaea Technologies (NPT) is the latest guest to sit down with Al on The SAF Podcast. Nova Pangaea Technologies are a UK based company looking whose patented REFNOVA technology can create fantastic added value to almost all waste plant biomass, including the production of Sustainable Aviation Fuel (SAF).  Currently, Nova Pangaea Technologies is currently most notably working on project Speedbird, in the UK, with British Airways and Alcohol-to-Jet refinery technology company, Lanzajet, which is scheduled to come online in 2026. British Airway's parent company, International Airline Group (IAG) also invested in NPT in June 2023. This is a fantastic discussion where Sarah outlines what NPT's expansion strategy could look like beyond project Speedbird, and which global markets they will look to target. Another critical point of discussion is the challenge of procuring project finance and investment when you are based outside of the US and how this slows down projects in the less regulatory mature markets around the world, as well as so many other critical discussion points.  You can also find out what Sarah did when the IAG investment was confirmed. Abstract: Huntington's disease (HD) is a neurodegenerative disorder caused by an expansion of CAG repeats in exon 1 of the HTT gene, ultimately resulting in the generation of a mutant HTT (mHTT) protein. Although mHTT is expressed in various tissues, it significantly affects medium spiny neurons (MSNs) in the striatum, resulting in their loss and subsequent motor function impairment in HD. While HD symptoms typically emerge in midlife, disrupted MSN neurodevelopment has an important role. To explore the effects of mHTT on MSN development, we differentiated HD induced pluripotent stem cells (iPSC) and isogenic controls into neuronal stem cells, and then generated a developing MSN population encompassing early, intermediate progenitors, and mature MSNs. Single-cell RNA sequencing revealed that the developmental trajectory of MSNs in our model closely emulated the trajectory of fetal striatal neurons. However, in the HD MSN cultures, the differentiation process downregulated several crucial genes required for proper MSN maturation, including Achaete-scute homolog 1 and members of the DLX family of transcription factors. Our analysis also uncovered a progressive dysregulation of multiple HD-related pathways as the MSNs matured, including the NRF2-mediated oxidative stress response and mitogen-activated protein kinase signaling. Using the transcriptional profile of developing HD MSNs, we searched the L1000 dataset for small molecules that induce the opposite gene expression pattern. Our analysis pinpointed numerous small molecules with known benefits in HD models, as well as previously untested novel molecules. A top novel candidate, Cerulenin, partially restored the DARPP-32 levels and electrical activity in HD MSNs, and also modulated genes involved in multiple HD-related pathways. This week Salopcast had the chance to go into the club and sit down with Lawrence Ellerby. Lawrence is the Safety Officer at the club and along with Brian Cauldwell and a cast of others had the mad week last week getting arrangements for the 1000 person trial in place. We give Lawrence a brief introduction to the fans and his role and then move onto the prep for the trial game, the way the day went and the fall out following the change in government advice. The club got a 10/10 for the trial and the club are also keen to express that was the fans mark too. We are also joined by Mike Davis the clubs Supporters Lesion Officer with his experience of the process, how they helped get things sorted and some other issues that will need to be addressed. Apologies for the fridge noise for the first couple of minutes, but after that we stopped and turned off Roland's mini fridge ;)

Abstract: Impaired neuronal proteostasis is a salient feature of many neurodegenerative diseases, highlighting alterations in the function of the endoplasmic reticulum (ER). We previously reported that targeting the transcription factor XBP1, a key mediator of the ER stress response, delays disease progression and reduces protein aggregation in various models of neurodegeneration. To identify disease-modifier genes that may explain the neuroprotective effects of XBP1 deficiency, we performed gene expression profiling of brain cortex and striatum of these animals and uncovered insulin-like growth factor 2 (Igf2) as the major upregulated gene. Here we studied the impact of IGF2 signaling on protein aggregation in models of Huntington disease (HD) as proof-of-concept. Cell culture studies revealed that IGF2 treatment decreases the load of intracellular aggregates of mutant huntingtin and a polyglutamine peptide. These results were validated using induced pluripotent stem cells (iPSC)-derived medium spiny neurons from HD patients. The reduction in the levels of mutant huntingtin was associated with a decrease in the half-life of the intracellular protein. The decrease in the levels of abnormal protein aggregation triggered by IGF2 were independent of the activity of autophagy and the proteasome pathways, the two main routes for mutant huntingtin clearance. Conversely, IGF2 signaling enhanced the secretion of soluble mutant huntingtin species through exosomes and microvesicles involving changes in actin dynamics. Administration of IGF2 into the brain of HD mice using gene therapy led to a significant decrease in the levels of mutant huntingtin in three different animal models. Moreover, analysis of human post-mortem brain tissue, and blood samples from HD patients showed a reduction of IGF2 level. This study identifies IGF2 as a relevant factor deregulated in HD, operating as a disease modifier that buffers the accumulation of abnormal protein aggregates. Abstract: The basal ganglia, best known for processing information required for multiple aspects of movement, is also part of a network which regulates reward and cognition. The major output nucleus of the basal ganglia is the striatum, and its functions are dependent on neuronal compartmentation, including striosomes and matrix, which are selectively affected in disease. Striatal projection neurons are GABAergic medium spiny neurons (MSNs), all of which share basic molecular signatures but are subtyped by selective expression of receptors, neuropeptides, and other gene families. Neurogenesis of the striosome and matrix occurs in separate waves, but the factors regulating terminal neuronal differentiation following migration are largely unidentified. We performed RNA- and ATAC-seq on sorted murine striosome and matrix cells at postnatal day 3. Focusing on the striosomal compartment, we validated the localization and role of transcription factors and their regulator(s), previously not known to be associated with striatal development, including Irx1, Foxf2, Olig2 and Stat1/2. In addition, we validated the enhancer function of a striosome-specific open chromatin region located 15Kb downstream of the Olig2 gene. These data and data bases provide novel tools to dissect and manipulate the networks regulating MSN compartmentation and differentiation and thus provide new approaches to establishing MSN subtypes from human iPSCs for disease modeling and drug discovery. It would exclude the area of the second floor entirely, counting it as a missing area, but including the steps that are from the second story floor and up as being in the second story bagua map. But if I'm to include the top area of the staircase in the first story bagua map, then wouldn't I create a Bagua map sectioned out differently for the second floor with the stairway and the missing area above the foyer both excluded? “ That was an uber long question, but let's get down to the basics. Basically what you would like to find out is how do you apply a bagua to the second floor when the second floor does not have rooms in the same areas as the first floor does or there's a two-story foyer. You're still always going to apply that bagua to the second story home in the same orientation and position as you did for the first floor. The first reason is that “crystal-clear”, natural quartz is extremely rare to find in nature, and so that makes them super expensive to purchase. Another reason, I choose to use human-made crystals, is that natural quartz crystals have imperfections in them, and that makes them very difficult to cut into a multifaceted stone. One of my favorite items in my home is my exquisite collection of Himalayan Salt Lamps. Himalayan salt lamps are hand-chiseled solid block of salt. They have a beautiful pink color or kind of orangey due to the high concentration of trace minerals contained within the salt. The salt block is then hollowed out in the center and a light source is inserted as well as a wooden base so that it can stand. Some of the benefits include things like improved breathing, reduced allergy and asthma symptoms, they have less colds and flu, reduced stress, they have fewer headaches and migraines, reduced symptoms of inflammatory diseases like arthritis and skin conditions…
