Join us this week as we sit down with Conroy Copeland and Frank Clayton. Sensei Copeland began his karate training around the age of 15. He has been taught and trained under renowned martial arts instructors like Sensei Richard Kim, Morio Higaonna, and Professor Wally Jay, gaining valuable knowledge that he now shares with his students, some of whom run their own dojos. In 1975, Conroy met Sensei Don Warrener at a tournament and became his student, training at DWMA until the early 1990s when he began learning from Sensei Teruo Chinen, who has been his teacher for the last 20 years. As a competitor, he has represented national teams worldwide for WKO, Karate Ontario, and CKO, earning numerous championships. Sensei Copeland has been teaching since 1978 and has successfully run Copeland's Martial Arts since then. Additionally, he has hosted the Windsor Open Karate Championships for over 20 years, making it one of the region's longest-running tournaments. In 2023, Sensei Conroy Copeland was inducted into the Canadian Black Belt Hall of Fame. Sensei Clayton, born in Halifax in 1959 and raised in Montreal, began his martial arts journey with Judo and Boxing before fully immersing himself in Karate in 1971 after moving to Hamilton. He joined Don Warrener's Delta club in 1977, earning his black belt in 1980. Over the years, he has helped his students open their own dojos, mentoring them to positively impact their communities. With over 10,000 students taught and more than 300 black belts produced, Sensei Clayton has built a lasting legacy in martial arts. Renshi Clayton is a 7th dan in Okinawan Goju Ryu Karate and a 5th dan in Ryu Kyu Kobudo. Sensei Clayton continues to teach and pursue his own training, regularly traveling to Okinawa. In 2024, Sensei Frank Clayton was inducted into the Canadian Black Belt Hall of Fame. #PKCC #martialarts #karate

Das Marienhospital Osnabrück (MHO) und das Christliche Kinderhospital Osnabrück (CKO) liegen im Evakuierungsgebiet. Das MHO wird am 17.11.2024 ab 7 Uhr nicht durch den Rettungsdienst angefahren und ist auch fußläufig nicht zu erreichen. Für verbleibende Patienten und Mitarbeitende greift ein umfangreiches Sicherheitskonzept.

Abstract: Clustered protocadherins (cPcdhs) are candidates for the neural circuit formation; however, the localization of cPcdhs in pre- and post-synaptic compartments has not been well characterized. Here we examined the localization of cPcdh{gamma} proteins in the mouse hippocampal CA1 region using light and electron microscopy. From postnatal day 7 to 21, cPcdh{gamma} immunosignals were detected in approximately 40-60% of spines of pyramidal cells. SDS-digested freeze-fracture replica labelling revealed that cPcdh{gamma} immunolabeling was found in 50% of PSD 95-positive postsynaptic profiles but only in less than 10% of vGluT1-positive pre-synaptic terminals. Interestingly, cPcdh{gamma}-positive pre-synaptic terminal was exclusively accompanied by cPcdh{gamma}-positive postsynaptic counterpart. In addition, electrophysiological investigations revealed that the miniature excitatory postsynaptic current frequency in cPcdh{gamma} cKO mice was significantly higher than that in wild-type mice. These results suggest that cPcdh{gamma} proteins are unequally distributed in the pre- and post-synaptic membrane during neural circuit development and regulate the number of excitatory synapses. Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) published a new research paper in Volume 15, Issue 4, entitled, “Isoform-specific effects of neuronal repression of the AMPK catalytic subunit on cognitive function in aged mice.” AMP-activated protein kinase (AMPK) functions as a molecular sensor that plays a critical role in maintaining cellular energy homeostasis. Dysregulation of the AMPK signaling has been linked to synaptic failure and cognitive impairments. In a recent study, researchers Xueyan Zhou, Wenzhong Yang, Xin Wang, and Tao Ma from Wake Forest University School of Medicine demonstrated abnormally increased AMPK activity in the hippocampus of aged mice. The kinase catalytic subunit of AMPK exists in two isoforms α1 and α2, and their specific roles in aging-related cognitive deficits are unknown. “Taking advantage of the unique transgenic mice (AMPKα1/α2 cKO) recently developed by our group, we investigated how isoform-specific suppression of the neuronal AMPKα may contribute to the regulation of cognitive and synaptic function associated with aging.” The team found that aging-related impairment of long-term object recognition memory was improved with suppression of AMPKα1 but not AMPKα2 isoform. Moreover, aging-related spatial memory deficits were unaltered with suppression of either AMPKα isoform. Biochemical experiments showed that the phosphorylation levels of the eukaryotic initiation factor 2 α subunit (eIF2α) were specifically decreased in the hippocampus of the AMPKα1 cKO mice. They further performed large-scale unbiased proteomics analysis and revealed identities of proteins whose expression is differentially regulated with AMPKα isoform suppression. Abstract: A lack of the mitochondrial tRNA taurine modifications mediated by mitochondrial tRNA translation optimization 1 (Mto1) was recently shown to induce proteostress in embryonic stem cells. Since erythroid precursors actively synthesize the hemoglobin protein, we hypothesized that Mto1 dysfunctions may result in defective erythropoiesis. Hematopoietic-specific Mto1 conditional knockout (cKO) mice were embryonic lethal due to niche-independent defective terminal erythroid differentiation. Mechanistically, mitochondrial oxidative phosphorylation complex-I was severely defective in the Mto1 cKO fetal liver and this was followed by cytoplasmic iron accumulation. Overloaded cytoplasmic iron promoted heme biosynthesis and enhanced the expression of embryonic hemoglobin proteins, which induced an unfolded protein response via the IRE1 -Xbp1 signaling pathway in Mto1 cKO erythroblasts. An iron chelator rescued erythroid terminal differentiation in the Mto1 cKO fetal liver in vitro. Abstract: Proper brain function depends on intact neurovascular coupling (NVC). Through NVC, local cerebral blood flow (CBF) is rapidly increased in activated cerebral regions to meet the corresponding demands of neural activities. NVC has been believed to support advanced cerebral functions such as complex emotional homeostasis. However, few studies provide causal link evidence with specific molecular and cellular identities. Here, using RNAscope, cell-type specific conditional knockout (cKO) perturbation in BL6/C57 mice, laser speckle contrast imaging, and mouse behavior assays, we demonstrated that the N-methyl-D-aspartic acid (NMDA) receptor noncore subunit NR2D is only expressed in cerebral arteriole smooth muscle cells (aSMCs) but not in peripheral aSMCs. Both aSMC-NR2D cKO and aSMC-NR2D+NMDA receptor subunit 1 (NR1) double cKO mice displayed a severe decline in functional hyperemia induced by whisker stimulation. In addition, the NR2D pathway was independent of previously demonstrated COX-2-PGE2-dependent mechanism and high-fat diet (HFD)-induced impairments in NVC. Caldesmon is a potential downstream protein in the NR2D pathway for NVC regulation. Notably, we demonstrated that NVC deficits caused abnormalities in complex emotional behaviors, such as anxiety-like behaviors (exhibiting increased defecation and urine), reduced curiosity and decreased fear instinct (displaying less fear of aggressive CD1 mice). Furthermore, fear-response gene c-FOS expression in the amygdala neurons was attenuated in aSMC-NR2D knockout mice after response to acute stresses. In contrast, its expression level remained unchanged in the non-fear-related cerebral regions such as the piriform cortex and hippocampus. These findings demonstrated that inadequate functional hyperemia impaired cerebral complex emotional functions and suggested a co-existence of anxiety and fearlessness in single individuals. Abstract: Background and Purpose: Chemotherapy-Induced Peripheral Neuropathy (CIPN) is a common, dose-limiting side effect of cancer therapy. Protease-activated receptor 2 (PAR2) is implicated in a variety of pathologies, including CIPN. In this study, we demonstrate the role of PAR2 expressed in sensory neurons in a paclitaxel (PTX)-induced model of CIPN in mice. Experimental Approach: CIPN was induced in both PAR2 knockout/WT mice and mice with PAR2 ablated in sensory neurons via the intraperitoneal injection of paclitaxel. In vivo behavioral studies were done in mice using von Frey filaments and the Mouse Grimace Scale. We then examined immunohistochemical staining of dorsal root ganglion (DRG) and hind paw skin samples from CIPN mice to measure satellite cell gliosis and intra-epidermal nerve fiber (IENF) density. Pharmacological reversal of CIPN pain was tested with the PAR2 antagonist C781 Key Results: Mechanical allodynia caused by paclitaxel treatment was alleviated in PAR2 knockout mice of both sexes. In the PAR2 sensory neuronal conditional knockout (cKO) mice, both mechanical allodynia and facial grimacing were attenuated in mice of both sexes. In the dorsal root ganglion of the paclitaxel-treated PAR2 cKO mice, satellite glial cell activation was reduced compared to control mice. IENF density analysis of the skin showed that the paclitaxel-treated control mice have a reduction in nerve fiber density while the PAR2 cKO mice had a comparable skin innervation as the vehicle-treated animals. Similar results were seen with satellite cell gliosis in the DRG where gliosis induced by PTX was absent in PAR cKO mice. Finally, C781 was able to transiently reverse established PTX-evoked mechanical allodynia. Conclusions and Implications: Our work demonstrates that PAR2 expressed in sensory neurons plays a key role in paclitaxel-induced mechanical allodynia, spontaneous pain and signs of neuropathy, suggesting PAR2 as a possible therapeutic target in multiple aspects of paclitaxel CIPN. E., Barros, L. F., Nave, K.-A., Weber, B., Saab, A. S. Abstract: The integrity of myelinated axons relies on homeostatic support from oligodendrocytes (OLs), which is essential for brain function. However, the mechanisms by which OLs detect axonal spiking and rapidly control axon-OL metabolic coupling are largely unknown. Here, we combine optic nerve electrophysiology and two-photon imaging to study activity-dependent calcium (Ca2+) dynamics in OLs and metabolite fluxes in myelinated axons. Both high-frequency axonal firing and extracellular potassium (K+) elevations trigger a fast Ca2+ response in OLs that is facilitated by barium-sensitive, inwardly rectifying K+ channels. Using OL-specific Kir4.1 knockout mice (Kir4.1 cKO) we now demonstrate that, in addition to being crucial for K+ clearance, oligodendroglial Kir4.1 regulates axonal energy metabolism and long-term axonal integrity. Before the manifestation of axonal damage, we observed reduced glucose transporter GLUT1 and monocarboxylate transporter MCT1 expression in myelin of young Kir4.1 cKO mice, suggesting early deficits in metabolite supply to axons. Strikingly, we found lower resting lactate levels and activity-induced lactate surges in optic nerve axons of young Kir4.1 cKO mice. Moreover, both axonal glucose uptake and consumption were hampered in the absence of oligodendroglial Kir4.1, uncovering a new role of OLs in regulating axonal glucose metabolism. Our findings reveal a novel model of axon-OL signaling and metabolic coupling in which OLs detect high-frequency axonal activity through K+ signaling, which is critical in adjusting the axon-OL metabolic unit and in preserving long-term axonal health. Using human data as well as the Plasmodium berghei ANKA (PbA) model of experimental cerebral malaria (ECM), we sought to define contributions of brain endothelial cell MHC class I antigen (Ag) presentation to CD8 T cells in this pathology. We crossed novel H-2Kb and H-2Db LoxP mice to Cdh5-Cre mice to achieve targeted deletion of discrete class I molecules on endothelium. RNA transcriptomes of brains during PbA infection are differential between H-2Kb and H-2Db expressing mice, highlighting differential regulation of neuropathogenesis. Distinct patterns of disease onset, CD8 T cell infiltration, targeted cell death, and regional BBB disruption and vascular leakage are observed. Strikingly, ablation of H-2Kb or H-2Db from brain endothelial cells resulted in diminished interactions of T cells with brain vasculature and lessened infiltration of activated CD8 T cells, resulting in prevention of blood-brain barrier (BBB) disruption, CNS vascular leakage and death. Overall, endothelium-specific conditional knockout (cKO) mice were protected from development of ECM resulting in survival of the mice. These data show the first in vivo evidence that interactions of CD8 T cells and cognate peptide:MHC on brain endothelium regulate development of ECM neuropathology. Therefore, targeting MHC class I interactions therapeutically may hold potential for treatment of cases of severe malaria. Copy rights belong to original authors. You need a buffer, and that's the legal operations buffer. With that in place, you can express the problem. The legal ops professional can go share it as the buffer: "I'm generally hearing from these people, And here's what I suggest we start to do to help facilitate."Often, I believe people in the profession think that legal operations are putting technology in, right? Or you're putting a process in. And I feel like the conduit that legal offspring is legal ops is not all those things. Legal ops is a mindset that needs to happen in the culture of the org, for the department to get better. We also believe that without sharing, without creating true standards in the market, it will take ages to go through this digital transformation.Struggling with digital transformationWe see that people are really struggling with digital transformation. They first don't understand why all this is happening. They're missing the digital mindset.The idea that digital transformation is something positive. They do not get the link back to legal because, they are thinking that what we do is something that's handcrafted; it's something that relies on very special knowledge.We do a perfect job of creating the perfect, beautiful contract that nobody else understands. We see beauty. And we see our role in protecting our company and defending our companies, defending our client's interests. It's just that the other side doesn't understand it.So the digital aspect is that we need to get more out of this dormant contract that's archived in the file cabinet somewhere. To make it valuable information, accessible for the digital company that we are working in.The whole idea of digital transformation is to make information that sits somewhere in an unstructured format, deep in contracts, more accessibleWe miss a huge opportunity for our profession to sit at the C-suite table and contribute to the overall goals of the enterprise. Second, everything moves so fast. We are overwhelmed with the speed of change. And we need to understand that change is constant and it will never go back to a slow motion mode. It will not happen. It will be fast. So we need to learn how to become adaptable, call it agile, call it whatever, but we need to adapt fast to a changing world. That bill is high. Maybe you bought a company, in an M&A scenario, that had subsidiaries or locations in highly regulated, highly risky territories - where there's a ton more work to be done. It's not an apples-to-apples kind of company. So you don't look at what I got for the price I paid? You're just saying the bills are higher now.Price discriminationAb: How do you demonstrate that we're going to charge you X, and it's worth more than X to you as an outcome?Keith: Price discrimination is the short answer to that. Not everything is worth the same amount. By nature, and this is not any judgment or me trying to take a position on the actual value or the value of the practitioners doing this kind of work. But by nature, there's a spectrum of complexity, and the stuff that's really complex on the highly complex side tends to be more scarce. There is a more scarce supply of people who can do it.On the opposite side, on a more commoditized side or the more routine side - it's not as complex, and there are more options to avail yourself of to do that work.Client sophistication - apply the scalpel, not the hatchetMy particular role is to ensure that the firm is as profitable as possible and that we're making our clients as happy as possible in terms of service delivery models and getting the outcomes they want.The conversation with the client depends on the level of sophistication:I'd say architecting and implementing some of those solutions depends on the sophistication that the client or the person on the buy-side has. There are only so many levers in this business in terms of what I can do. It's all about service delivery models and the underlying economics. This isn't astrophysics. There are not huge, massively complex data jobs in most cases (there are some benefits to that in different scenarios).There are a couple of levers on who's going to do the work and how much they have to do, what's the cheapest way to do it, and what the outcome will be. How do those things correlate (the price to the product)? Having those conversations with people that understand that better is important.I've worked with several legal operations groups where they have the knowledge, the understanding, and the sophistication. Still, there's a gap in the level of influence they have over the in-house attorneys...Different sides of the table, same challenges:We have very similar challenges - in-house and the law firm side - as it relates to getting the lawyers to understand the economics, the process, and to engage attorneys with it - to really adopt it. As opposed to, 'Yeah, but I'm skeptical of that because I don't understand it. So let's just do it the old way.'Next service delivery modelWhere can I get efficiencies out of technology? Which is a hugely growing market, that's going to be the new paradigm shift. It's going to be a much more dynamic service delivery model...Given that we've got a shortage of supply of people with the requisite skills to do a lot of this work and that's why we have to charge more. They [the attorneys] also have been working ridiculous man hours, and that's why we have to pay more for that. On top of that, there are obviously the mental health and quality of life concerns which is why a lot of people leave the workforce.I've been trying to make the case, and we work with our innovation group a lot, too, with several different teams internally to say: 'if you don't want people burnt out, you want them to have a better level of work-life balance and job satisfaction. Don't force them to brute force their way through everything all the time.'If you can get 30% more productivity out of the same people and let them sign off at seven or eight at night and use some tool that scales their productivity and delivers efficiencies then do it. Joseph's story is one of determination, hard work and taking chances. CKO Kickboxing was originally formed in Hoboken, NJ as “Take It To The Max” in November, 1997 by Joseph Andreula. Joseph designed and created the intense workout, which was the first of its kind. CKO became so popular in Hoboken, that by 2004, nine additional locations opened throughout NJ and NY. In the summer of 2005, CKO opened the doors to its second Hoboken gym. CKO has a massive army of devoted followers and has been recognized in countless national fitness magazines and television programs such as NBC's The Today Show. We have one of our low back pain patients, Juliana Magnifico shared with us her story.Juliana is a CKO kickboxing coach as well as a nutrition coach. She truly understands the field of fitness and has been part of it for the past 8 years.
