Jason and Kaitlyn began their journeys in different ways, from a family that was open to following an artistic path, to another that was more resistant to it. However, via routes from lighting design to training as an architect, their paths collided in the world of projection design, and they eventually founded PXT Studio, building bespoke designs and making the seemingly impossible possible. When Jason first met the opera director Yuval Sharon, it was the start a unique relationship that would draw Jason and Kaitlyn into the world of opera, where they have been delighting and astonishing audiences ever since.  They have created work from The Industry in LA, to the Vienna State Opera, to Oper Frankfurt, Carnegie Hall, and on Broadway among many other venues. Collectively they have won two Los Angeles Ovation Awards, three LADCC awards, as well as having their work showcased at the Prague Quadrennial on three separate occasions... Abstract: Motivation: Drug re-positioning allows expedited discovery of new applications for existing compounds, but re-screening vast compound libraries is often prohibitively expensive. "Connectivity mapping" is a process that links drugs to diseases by identifying drugs whose impact on expression in a collection of cells most closely reverses the disease's impact on expression in disease-relevant tissues. The high throughput LINCS project has expanded the universe of compounds, cellular perturbations, and cell types for which data are available, but even with this effort, many potentially clinically useful combinations are missing. To evaluate the possibility of finding disease-relevant drug connectivity despite missing data, we compared methods using cross-validation on a complete subset of the LINCS data. Results: Modified recommender systems with either neighborhood-based or SVD imputation methods were compared to autoencoders and two naive methods. All were evaluated for accuracy in prediction of both expression signatures and connectivity query responses. We demonstrate that cellular context is important, and that it is possible to predict cell-specific drug responses with improved accuracy over naive approaches. Neighborhood-based collaborative filtering was the most successful, improving prediction accuracy in all tested cells. We conclude that even for cells in which drug responses have not been fully characterized, it is possible to identify drugs that reverse the expression signatures observed in disease. You've more than likely heard the saying, "A bird in the hand is worth two in the bush." In the third installment of the Inside JABA Series, Dr. Iser DeLeon, along with Inside JABA regulars Drs. Tiger and St. Peter we do a deep-dive into the topic of accumulated versus distributed reinforcement arrangements to see if this adage is true in the context of skill acquisition and problem behavior. I'm excited to have Dr. DeLeon, better knowns as Willie to friends and colleagues, on the show. Willie's contributions to the field are too lengthy to list here but to summarize, he is a Professor of Psychology at The University of Florida, and he has done great work in the area of translating concepts derived from the Experimental Analysis of Behavior for use in the applied realm. I'll read you a bit from his bio: "[Willie's] work is notable for its use of concepts derived from behavioral economics, behavioral momentum, and other conceptual models as they apply to behavior analytic interventions. In addition to his impressive research and scholarly contributions, Dr. DeLeon has served on the editorial boards of a number of behavior analysis journals, including as associate editor of the Journal of Applied Behavior Analysis and editor of translational research for the Journal of the Experimental Analysis of Behavior." So as you can see, Willie, along with my other Inside JABA co-hosts, are eminently qualified to talk about this subject. In this episode we reference three different papers on distributed versus accumulated reinforcement. For the purposes of short-hand, I referred to them in the conversation as "Willie's paper," "Claire's paper," and so on. Allow me a minute to give full and proper recognition to all of the authors. The papers we discussed include the following: DeLeon, Chase, Frank-Crawford, Carreau-Webster, Triggs, Bullock, and Jennett (2014). Distributed and accumulated reinforcement arrangements: evaluations of efficacy and preference. Fulton, Tiger, Meitzen, and Effertz (2019). A comparison of accumulated and distributed reinforcement periods with children exhibiting escape-maintained problem behavior. Robinson and St-Peter (2019). Accumulated reinforcers increase academic responding and suppress problem behavior for students with Attention-Deficit Hyperactivity Disorder. An alternative to this distributed delivery of reinforcement is to arrange conditions in which reinforcement can accumulate following multiple target responses before then being consumed. This episode’s discussion highlights three studies that compared task engagement among individuals with intellectual and developmental disabilities (DeLeon et al., 2014) including those identified with escape-maintained problem behavior (Fulton et al.,  2020) and adolescents with attention deficit/hyperactivity disorder (Robinson & St. Peter, 2019) under accumulating and distributed reinforcement arrangements. Across each study, engagement was increased and problem behavior reduced during accumulated reinforcement conditions. Further, these studies each included preference assessments in which participants were provided with the opportunity to choose between accumulated and distributed reinforcement arrangements. There was some variability across participants, but many individuals preferred to engage in extended work periods for the opportunity to accumulate reinforcement prior to consumption. Learning Objectives: Describe the difference between distributed and accumulated reinforcement arrangements. Describe why reinforcement efficacy may be increased through accumulation of reinforcers? A few years ago, my DREAM THINK DO guest, Tom Pietras, asked himself a simple but profound question: “What would I regret if I got run over by a beer truck on the way home from work?“ (Hey… he lives in Madison, Wisconsin… so it's not a crazy question!)  His answer (and his next steps) changed everything for the better. Listen To The Podcast:                         By all outward appearances, Tom had everything going for him. He was a successful executive doing challenging work he enjoyed. Plus, he had a happy marriage and was surrounded by some great friends. What more could a guy want? Here's the thing. When he answered his “beer truck” question, he got hit with a surprise. The surprise… was that he wanted to dig back into a former passion. Music. Drumming to be more specific. He had drummed as a kid.  His dad had taught him.  He was good too.  But then a disagreement made him put down the sticks. They stayed down… for 29 years! But the “beer truck” question and a chance encounter at a dinner party changed everything… for the better. During our convo, we dig into Tom's story but we also talk through some of the specific strategies he used to set a new course for his life and career. The latest advanced technology in breast cancer screening and diagnosis, called DBT (digital breast tomography), 3D or Tomosynthesis, creates a layered, 3-dimensional image. While the patient experience is very similar to traditional digital mammography, the new technology provides a series of thin images that a radiologist can view almost like turning the pages in a book. The results include better detection of invasive breast cancers and fewer call-backs for false positives. Elizabeth S.
