S., Ravuri, S., Huard, J. Abstract: Mesenchymal stem cells (MSCs) have long been viewed as a promising therapeutic for musculoskeletal repair. However, regulatory concerns including tumorgenicity, inconsistencies in preparation techniques, donor-to-donor variability, and the accumulation of senescence during culture expansion have hindered the clinical application of MSCs. Senescence, in particular, is a driving mechanism for MSC dysfunction with advancing age. Often characterized by increased reactive oxygen species, senescence-associated heterochromatin foci, inflammatory cytokine and chemokine secretion, and reduced proliferative capacity, senescence directly inhibits MSCs efficacy as a therapeutic for musculoskeletal regeneration and repair. Furthermore, autologous delivery of senescent MSCs can further induce disease and aging progression through the secretion of the senescence-associated secretory phenotype (SASP) and mitigate the regenerative potentetial of MSCs. To combat these issues, the use of senolytic agents to selectively clear senescent cell populations has become popular. However, their benefits to human MSCs during the culture expansion process have not yet been elucidated. To address this, analyzed markers of senescence during culturing of human primary adipose-derived stem cells (ADSCs), a population of fat-resident MSCs commonly used in regenerative medicine applications. Next, we used the senolytic agent fisetin to determine if we can reduce these markers of senescence within our culture-expanded ADSC populations. Our results indicate that ADSCs acquire common markers of cellular senescence including increased reactive oxygen species, senescence-associated {beta}-galactosidase, and senescence-associated heterochromatin foci. Furthermore, we found that the senolytic agent fisetin works in a dose-dependent manner and selectively attenuates these markers of senescence while maintaining the differentiation potential of the expanded ADSCs. Seeing his neighbors and acquaintances play a board game in which the object was to buy and sell property, he decided to publish his own version of the game, with the help of his first son, William, and his wife Esther. Darrow marketed his version of the game under the name Monopoly. In truth, Darrow was just one of many people in the American Midwest and East Coast who had been playing a game of buying and trading property. The game's direct ancestor was The Landlord's Game, created by Elizabeth Magie. The Darrow family initially made their game sets on flexible, round pieces of oilcloth instead of rigid, square carton. Charles drew the designs of the properties with drafting pens, and his son and wife filled in the spaces with colors and made the title deed cards and Chance and Community Chest cards. In 1970, three years after Darrow's death, Atlantic City placed a commemorative plaque in his honor on The Boardwalk, near the corner of Park Place. In 1973 Ralph Anspach, an economics professor at San Francisco State University, produced Anti-Monopoly, a game similar to Monopoly, and for this was sued by Parker Brothers. The leading role was intended for Steve McQueen, who starred in The Blob two years earlier, also produced by Harris and directed by Yeaworth. McQueen passed on the film to make The Magnificent Seven instead. The dinosaurs were filmed using the technique of stop-motion animation as well as puppets for close-ups. The film promulgates the naïve idea that herbivorous animals (such as the brontosaurus) are not dangerous (a similar claim was made in Spielberg's 'Jurassic Park', 1993). The cape buffalo is one of the most aggressive and dangerous animals in Africa (and only weighs about 5% of what a brontosaurus is estimated to have weighed). Marcel Delgado was given less than half the time originally agreed upon to create the dinosaur models used in the film. The studio initially agreed to give him five to six weeks, as he requested, but two weeks later he was told that they would begin production on Tuesday. It began the orbital insertion maneuver which placed the spacecraft into a three-hour, nine minute, elliptical orbit that brought the spacecraft 295-kilometers from the surface at about 10 degrees North during the periapsis and out to 7762-kilometers during apoapsis. During each orbit, the space probe captured radar data while the spacecraft was closest to the surface, and then transmit it back to Earth as it moved away from Venus. This maneuver required extensive use of the reaction wheels to rotate the spacecraft as it imaged the surface for 37-minutes and as it pointed toward Earth for two hours. The primary mission intended for the spacecraft to return images of at least 70 percent of the surface during one Venusian day, which lasts 243 Earth days as the planet slowly spins. To avoid overly-redundant data at the highest and lowest latitudes, the Magellan probe alternated between a Northern-swath, a region designated as 90 degrees north latitude to 54 degrees south latitude, and a Southern-swath, designated as 76 degrees north latitude to 68 degrees south latitude. However, due to periapsis being 10 degrees north of the equatorial line, imaging the South Pole region was unlikely. The resulting maps were comparable to visible-light photographs of other planets, and are still the most detailed in existence. In 1988, Mike was crowned Mountain Bike Cross Country World Champion and throughout his cycling career stacked up an impressive resume of world cup wins and podiums as well as many other titles, including a silver medal in the Mountain Bike Downhill World Championships. Upon retiring from professional cycling, Mike went on to adventure racing. He won the Eco-Challenge 3 times, Primal Quest 5 times and won the World Championships 4 times. and he is still going, this past summer he added the Go Pro Games Ultimate challenge win to him palmeres. This week's guest is adventure racing legend Mike Kloser, who is one of the top adventure and endurance athletes of all time. Mike is a former mountain bike world champion and multi-time adventure racing world champ as well. Even though he is retired now, he still competes regularly in a number of events and even came back to take part in the new Eco-Challenge race that took place last fall. In the interview, Mike shares stories from throughout his career, tells us how he got started in adventure racing, and provides some insights on his friend Lance Armstrong too. We even discuss his latest venture, Out There Adventure Travel, which organizes cycling, mountain biking, and wellness trips to parts of Europe. This is just a small part of what we chat about, with a guy that both Dave and Kraig know very well. Before we jump into the interview, our intrepid co-hosts get us up to speed on the latest happenings from the world of mountaineering, where it looks like things have mostly come to a standstill thanks to the virus. The vast majority of us have talked about writing a book. Problem is, the vast majority of us never make time to sit down and get the words on the page. Christine Kloser got immersed in publishing when her husband decided to write a book involving interviews with Major League Baseball players. She pored over Dan Poynter's Self-Publishing Manual, learning the nuts and bolts of putting a book together to assist her husband in becoming a published author. At the same time, Kloser was involved in the leadership of the Network for Empowering Women Entrepreneurs, and she often heard members talk about becoming authors—yet fail to take action. She decided to apply her knowledge of the writing process and help these women get published: Kloser offered to compile the book if each woman would agree to write one chapter. That book, entitled Inspiration to Realization, made the 2005 Entrepreneur Magazine's list of top reads for women, and soon other women were begging Kloser for help in writing a book of their own. Inspired to create a company that would help people write transformational books, Kloser designed the Get Your Book Done process. Ten years later, she has trained 70,000 writers in more than 125 countries, serving as a master facilitator and trainer in the area of excavating your message and writing books that positively impact the world. Austrian musician, film composer, producer, and screenwriter Harald Kloser began his career in Europe playing in a band and collaborating with pop artists such as Falco. As a musician he appeared on recording projects by Tom Waits, Jose Feliciano, Elton John, Al Jarreau, and others. After a career scoring for television, Harald eventually broke through to national prominence with his work on Alien vs. Predator and the sci-fi disaster film The Day After Tomorrow, starring Dennis Quaid and Jake Gyllenhaal. Both films were directed by Roland Emmerich, with whom Kloser went on to work on 10,000 B.C. and 2012, starring John Cusack. Though the two films were scored by Harald, he also co-wrote the scripts with Emmerich. In addition to creating the score for the movie Anonymous, Harald served as both composer and film producer on the recent hits White House Down, starring Jamie Foxx and Channing Tatum, and Independence Day: Resurgence, starring Liam Hemsworth and Jeff Goldblum. He has won three BMI Film & TV Awards for Alien vs. Predator, The Day After Tomorrow, and 2012.
