Meet Shaik Salauddin, a 38-year-old cab driver from Hyderabad, who is fighting for the rights of eight million gig workers from across the country. While India's gig economy is burgeoning, the workers on whose backs it is built barely enjoy any rights or legal protections. Salauddin realised this early on and in 2019, after five years of relentless pursuit, the Indian Federation of App-based Transport Workers (IFAT) was born. With over 25,000 members working for aggregators like Uber, Amazon, and Zomato, through IFAT, Salauddin is redefining the way we look at trade unions. To begin with, the union has no political affiliations. Instead, Salauddin encourages all of its members to understand power structures and approach the right people to drive change.Thanks to his efforts, two states, Karnataka and Rajasthan, have introduced legislations to protect the rights of gig workers. When Salil's accidental death attracted the media glare, Zomato CEO Deepinder Goyal took to Twitter and assured help in processing an insurance payout of Rs 10 lakh. Zomato employees contributed Rs 12 lakh to the family and a fundraising page started by his wife raised about Rs 9 lakh. While Zomato's move came as a welcome step, this instance has emphasised the need to provide social security benefits to gig and platform workers. Gig workers currently depend on the generosity of companies. In the absence of a legislation that grants protections to gig workers, the companies employing them  don't have a uniform policy on the kind of insurance cover they should provide to their drivers or delivery partners in cases of accidents or medical emergencies. Zomato covers its delivery partners with accident and life insurance along with an OPD allowance, whereas Swiggy offers Rs 6 lakh worth of medical and accident insurance cover. However, Shaik Salauddin, the national-general secretary of the Indian Federation of App-Based Transport Workers (IFAT) told Business Standard that there have been several instances where Zomato and Swiggy haven't done enough to compensate their delivery partners for ‘loss of pay' after they met with an accident while on the job. The IFAT represents gig workers employed by food delivery and taxi apps.  The recent Fairwork India report 2021 ranked Indian startups based on how they treat their gig workers. It said that most Indian startups don't score well when judged against the principle of ‘Fair Conditions'. To address this issue and many more, the Central government has come up with the Code on Social Security which recognises gig and platform workers. But according to reports, the Centre is unlikely to implement it before state elections in Uttar Pradesh and Punjab this year, as it is worried about the possibility of protests by labour unions, after having had a similar experience with the three contentious farm laws that had to be withdrawn.  The Supreme Court too has admitted a petition by the Indian Federation of App-Based Transport Workers (IFAT) that seeks classification of gig and platform workers as ‘unorganised workers' under the Unorganised Workers Act, 2008. This would entitle them to benefits such as provident fund, health and maternity benefits and old age protection. “This is obviously an attempt to distance itself from any language that would make the aggregator responsible for providing the drivers with social security or any form of protection or acknowledging any form of ‘employer-employee' relationship,”  reads the IFAT petition. The IFAT's petition points out that recently, two large cab aggregators ‘updated' the service agreements for their riders and drivers to “essentially absolve the ride sharing/ hailing company (the aggregator) of all liabilities and/or responsibilities towards the drivers or riders”. One of the aggregators, for instance, has stopped using the word ‘partner' in the agreement and now defines individuals utilising its app service for commercial gains as ‘customers'. I Was There: Stories of Production Incidents II

Corey Martin leads the discussion with two developers about production incidents they were personally involved in. Their goal is to inform listeners on how they discovered these issues, how they resolved them, and what they learned along the way. Ifat Ribon is a Senior Developer at LaunchPad Lab, a web and mobile application development agency headquartered in Chicago. For one of their clients, they developed an application to assist with the scheduling of janitorial services. It was built with a fairly simple Ruby on Rails backend, leveraging Sidekiq to process background jobs. As part of its feature set, the app would send text messages to let employees know their schedule for the week; these schedules were assembled by querying the database several times, fetching frequencies and availabilities of workers. Unfortunately, a client noticed a discrepancy between how many notices were being sent out, versus how many jobs they knew they had: of the 400 jobs total, only 150 had notifications. It turned out that all of the available database connections were being exhausted--but that was only half of the issue. Sidekiq was attempting to process far too many jobs at once, and each of these jobs were responsible for connecting to the database, exhausting the available pool. The solution Ifat settled on was to reduce the number of parallel jobs processed while increasing the number of connections to the database. From this experience, she also learned the importance of understanding how all these different systems interconnect. Christopher Ostrowski is Chief Technology Officer at Dutchie an e-commerce platform for the cannabis industry. One Christmas Eve, while celebrating with his family, Chris began receiving pager notifications warning him about some sluggish API response times. Since it didn't really have any significant end user impact, he ignored it and went back to the festivities. As the night went on, the warnings became significant alerts, and he pulled together a response team with colleagues to figure out what was going on. By all accounts, the website was functioning, but curiously, the rate of orders began to drop off. Through some investigation, they realized what was going on. Customers' order numbers were assigned a random, non-sequential six digit numbers. Dutchie was about to track its one-millionth order, a huge milestone. Before any orders are created, though, the app generates a six digit number, and tries to create one that doesn't already exist. The database was constantly being hit, as less and less six digit numbers were available for use. The solution ended up being rather simple: the order number limit was increased to nine digits. Although they had monitoring in place, the data was set up as an aggregate reporting; even though the "create order" API was slow, all of the others were low, keeping the average within tolerable levels. Christopher's solution to avoid this in the future was to set up more groupings for "essential" API endpoints, to alert the team sooner for latency issues on core business functionality. In der vorliegenden Arbeit wurden Real-time PCR-basierte Nachweisverfahren für E. canis und A. phagocytophilum entwickelt, validiert und im Anschluss für die Untersuchung von Patientenproben eingesetzt. Für E. canis wurden für zwei Tests Primer und Sonden des Typs „Molecular Beacon“ konstruiert, die auf unterschiedliche Zielgene gerichtet waren, die Reaktionsbedingungen optimiert und die Leistungsfähigkeit beider Tests verglichen. Die PCR EC-16S hatte hierbei die 16S rDNA als Zielgen, während die PCR ECP-p30 auf das p30-10-Gen von E. canis gerichtet war. Bei der Ermittlung der analytischen Sensitivität und analytischen Spezifität ergab sich, dass beide Tests in ihren Leistungen sehr ähnlich waren. Beide PCRs waren spezifisch und lieferten nur für DNA von E. canis ein positives Ergebnis, während die übrigen getesteten Erreger A. platys, N. risticii, A. phagocytophilum, Babesia canis, B. gibsoni und H. canis in der PCR negativ reagierten. Da die PCR ECP-p30 bei der Sensitivitätsprüfung geringfügig besser beurteilt wurde, wurde entschieden, die weiteren Untersuchungen mit diesem PCR-Protokoll durchzuführen. Zur Bestimmung der diagnostischen Sensitivität und Spezifität dieses Tests wurde eine extern kontrollierte Validierung mit geblindeten Proben durchgeführt. Hierbei ergab sich eine diagnostische Spezifität von 100 %. Die diagnostische Sensitivität der Real-time PCR betrug 82 %. Der prädiktive Wert des positiven Testergebnisses lag für die PCR ECP-p30 bei 100 %, während der prädiktive Wert des negativen Testergebnisses 87,5 % erreichte Als Nachweisgrenze wurden 14,5 Moleküle des Zielgens pro 50 µl Ansatz ermittelt. Im Anschluss wurde die Eignung des Tests an Blutproben von Hunden aus einem für E. canis endemischen Gebiet untersucht. Dabei wurden 244 Blutproben einbezogen und die Ergebnisse der PCR mit denen eines IFATs verglichen. Die Blutproben stammten aus Kampanien, Italien und wurden dort durch Tierärzte von Hunden gewonnen, die in einer Tierarztpraxis mit angeschlossenem Tierheim vorgestellt wurden. Die Tiere waren drei Gruppen zuzuordnen: Ein Teil der Hunde, und zwar 19 Tiere, wurde von privaten Besitzern in der Praxis vorgestellt, 52 Tiere waren unmittelbar zuvor von der Strasse aufgelesen worden und die dritte Gruppe, die 173 Hunde umfasste, hielt sich zum Zeitpunkt der Probennahme schon längere Zeit im Tierheim auf. Innerhalb des Tierheims wird ein hoher diagnostischer und medikamenteller Aufwand zur Erkennung und Bekämpfung von E. canis mittels antibiotischer Therapie und Zecken¬prophylaxe betrieben. In die Untersuchung einbezogen wurden jedoch nur Hunde, die mindestens drei Monate lang nicht mehr mit einem gegen E. canis wirksamen Medikament behandelt worden waren. Bei der serologischen Untersuchung der Hunde mittels IFAT ergab sich ein Anteil seropositiver Tiere von insgesamt 41,8 %, der auch bei Betrachtung der drei verschiedenen Gruppen nur wenig variierte. So betrug der Anteil seropositiver Tiere innerhalb der Gruppe der Hunde aus dem Tierheim 43,4 %, während 40,4 % der Straßenhunde und 31,6 % der Tiere in privatem Besitz seropositiv waren. Diese Unterschiede waren nicht signifikant. Ein direkter Erregernachweis mittels Real-time PCR erfolgte bei 13,9 % der untersuchten Tiere. Beim Vergleich der Untersuchungsergebnisse von PCR und IFAT wurde eine Überein¬stimmung bei 61,9 % der untersuchten Proben ermittelt. Bei Betrachtung der einzelnen Hundegruppen lag der Anteil der in der PCR positiven Tiere bei den Straßenhunden mit 23,1 % ungefähr doppelt so hoch wie bei den Tieren in Privatbesitz (10,5 %) oder den Tierheim¬hunden im Tierheim (11,6 %). Der Unterschied zwischen den Straßenhunden und den Tierheimhunden war somit signifikant. Diese Ergebnisse weisen auf ein häufiges Vorkommen von E. canis im Untersuchungsgebiet hin und stützen die Auffassung, dass eine Erreger¬elimination mittels Antibiotikatherapie nur schwer zu erreichen ist. Für A. phagocytophilum wurde in der vorliegenden Studie ebenfalls eine Real-time PCR entwickelt und das Testverfahren unter Einbeziehung zweier bereits publizierter Real-time PCR-Protokolle und zwar von Pusterla et al. (1999a) und von Courtney et al. (2004), validiert. Bei der Entwicklung der Real-time PCR für A. phagocytophilum wurde als Zielgen die 16S rDNA herangezogen, da nur hierfür vergleichbare Sequenzen nahe verwandter Ehrlichienspezies verfügbar waren. Alle drei vorliegenden Testverfahren wurden auf ihre analytische Spezifität und ihre analytische Sensitivität überprüft und zusätzlich im Rahmen einer extern kontrollierten Validierung mittels geblindeter Proben verglichen. Hierbei zeigte sich, dass nur die PCR nach Courtney et al (2004), hier als PCR AP-MSP2 bezeichnet, eine sehr gute Spezifität für A. phagocytophilum besaß. Die anderen Tests lieferten auch für N. risticii und A. platys positive Ergebnisse. Die analytische Sensitivität war bei diesem Test ebenfalls um mindestens eine Zehnerpotenz höher als bei den anderen beiden PCRs. Im Rahmen der Validierung wurde für die PCR AP-MSP2 eine diagnostische Spezifität von 96 % ermittelt, während die im Rahmen dieser Studie entwickelte PCR AP-16S eine Spezifität von 64 % und die PCR nach Pusterla et al. (1999a) einen Wert von 36 % erreichten. Der prädiktive Wert des positiven Testergebnisses betrug für die drei PCRs somit 96 %, 74 % bzw. 61 %. Für die Untersuchung von Patientenproben auf Befall mit A. phagocytophilum wurde deshalb die PCR AP-MSP2 ausgewählt. In die Studie wurden Hunde aus Deutschland einbezogen, und zwar sowohl 72 Blutproben, die eigens für diese Studie auf Anforderung von Tierärzten eingesandt worden waren, als auch 133 Proben, die aus verschiedensten Gründen in das Routinelabor des Institutes eingesandt worden waren. Die 72 eigens für die Studie gewonnenen Proben wurden mittels Buffy-coat-Ausstrich, PCR und IFAT auf A. phagocytophilum untersucht. Lichtmikroskopisch konnten in keinem Fall Einschluss¬körperchen des Erregers in den Granulozyten nachgewiesen werden. Mittels PCR wurde jedoch bei einem Hund (1,4 %) der Nachweis von A. phagocytophilum erbracht. Im IFAT konnten bei 16,7 % der 72 untersuchten Hundeseren spezifische Antikörper gegen den Erreger nachgewiesen werden. Eine Übereinstimmung der Ergebnisse von PCR und Buffy-coat-Ausstrichen lag bei 98,6 % der Proben vor. Beim Vergleich der Ergebnisse der Buffy-coat-Ausstriche mit den Ergebnissen des IFAT wurde eine prozentuale Übereinstimmung von 65,3 % errechnet. Identische Ergebnisse bei PCR und IFAT wurden bei 66,7 % der untersuchten Hunde erzielt. Die 133 Proben, die zufällig aus allen Einsendungen in das Routinelabor des Instituts für vergleichende Tropenmedizin und Parasitologie ausgewählt worden waren, wurden mittels PCR und IFAT untersucht, wobei zwei Tiere (1,5 %) in der PCR und 34,6 % im IFAT ein positives Ergebnis lieferten. Die Identität des PCR-Produktes eines der positiven Tiere wurde durch Klonierung und anschließende Sequenzierung bestätigt. Eine Übereinstimmung der Testergebnisse von IFAT und PCR bestand bei 52,6 % der untersuchten Proben. Diese Ergebnisse stützen die Auffassung, dass Hunde in Deutschland häufig mit A. phagocytophilum in Kontakt kommen, dass es sich dabei aber meist um eine selbstlimitierende, klinisch inapparente Infektion handelt.

During the year 2002 319 cattle from 31 farms (6 districts) of southern Bavaria were investigated for the presence of ticks during the grazing period and 287 serum samples were tested for the presence of antibodies against Borrelia burgdorferi and Babesia divergens. Ticks were detected in all 31 farms and the mean prevalence was 69 %. 3218 out of 3453 collected ticks were Ixodes ricinus. 139 nymphs, 19 larvae and 77 damaged adult specimens could only be determined to the Genus level (Ixodes). The seasonal pattern revealed the highest frequencies of ticks in May/June and September. The intensity of the infestation of the 221 positive animals was generally low. 76,5 % of parasitized individuals had 1-6 ticks/cattle and day of investigation. By contrast, individuals on pastures with appropriate tick-habitats showed up to 250 ticks/cattle and day of investigation. The percentage of infested animals in each herd varied within the period between 0 100 %. The investigation of 287 serum samples for the presence of antibodies against Borrelia burgdorferi by immunfluorescence techniques (IFAT), revealed antibody-titers ≥ 1:64 for 45,6 % of the animals. 27 of 31 farms showed prevalences of Borrelia burgdorferi ranging from 20 to 100 %. There were no significant differences of positive cattle between the various areas. However, a significant correlation could be detected between the number of ticks per cattle and the antibody concentration of anti-Borrelia IgG. By contrast, there was no significant correlation between the age of the animals and the serum antibody-titers. For comparative reasons, 64 IFAT-positive serum samples were tested by Westernblot techniques for the presence of antibodies cross-reacting with Borrelia garinii antigens. These analyses revealed that 69 % of the samples reacted positively, 28 % were unclear and 3 % were negative. The investigation of the 287 cattle sera for Babesia divergens by means of IFAT revealed one positive animal with an antibody-titer of 1:16. Finally, in an additional enzyme linked immunosorbent assay (ELISA) 105 serum samples from 18 farms and 13 pooled sera including 45 cattle from the other 13 farms were tested for the presence of antibodies against Fasciola hepatica. In 96,8 % of the farms antibodies against Fasciola hepatica could be detected.

Investigations on piroplasmosis in domestic cats in South Africa Domestic cats originating from an area in South Africa known to be endemic for babesiosis were included in an investigation on feline piroplasmosis. In order to receive some information about the range of species involved, molecular biological methods were used to characterize the causal agents genotypically. Two segments of the rDNA gene, the regions of the 18S and of the first and second internal transcribed spacers (ITS) were sequenced and phylogenetic analyses were performed. Based on these sequence data a modern and sensitive diagnostic test for a specific detection of B. felis, a real-time PCR, was developed and validated. Using this test, 206 blood samples of cats belonging to the patients of a small animal surgery in Port Elizabeth were screened for infections with B. felis and the test results were compared with results of examinations of stained blood smears and an indirect immunofluorescent antibody test (IFAT). Sequence analysis of the 18S rDNA and comparison with available sequence data from the Genbank NCBI demonstrated that at least 2, possibly 3 species are responsible for piroplasm infections in domestic cats in Port Elizabeth. The species B. felis was diagnosed in 11 of the 13 isolates. In one isolate, K 8, the parasites were identified as B. leo, a piroplasm species which had been documented to occur in lions before. This represents the first detection of B. leo in a naturally infected domestic cat. The sequence of the isolate K 68 could not be assigned to any known species, but it was closely related to the other feline piroplasms. The genetic relationship was slightly closer to B. leo with an identity of 98,3 % than to B. felis where an identity of 97,8 % was found. Compared to two other feline piroplasms isolated from a caracal, Babesia sp. Caracal Strain A and B, the isolate K 68 shared identities of 96,8 % and 96,7 % respectively. Phylogenetic trees clearly separated the isolate K 68 from the other feline piroplasms indicating a separate species status. Furthermore the results of the IFAT support a separation at least from B. felis. The isolate did not show any cross reactions with B. felis antigen. However, the parasites could not be differentiated from B. felis or B. leo in stained blood smears. In phylogenetic analyses including several Babesia and Theileria species all the feline piroplasm species of the present study were grouped together with B. rodhaini, B. microti and T. annae and seperated from the group of the `typical´ babesia and the classical theileria. When sequence analyses of the ITS rDNA of the 13 selected isolates were performed, the 11 isolates diagnosed as B. felis showed a polymorphism rate of 2,6 % and high pairwise identities between 98,6 % and 99,9 %. The sequences of the isolate K 8, already identified as B. leo, and of the isolate K 68 were less homologous with identities of 77,0 % and 76,0 % respectively compared with B. felis. In the phylogenetic tree the isolates K 8 and K 68 were clearly separated from each other and from the B. felis – isolates. This supports the existence of three distinct species. For one isolate, the isolate K 60, the intraisolate variation was determined in more detail. 7 different genotypes were found. 6 of them were very similar to B. felis and showed sequence identities between 97,2 % and 99,6 %. The sequence of the genotype 7, clone K 60 E.2, clearly differed from the other genotypes. With an identity of 76,9 % to B. felis the sequence could not be assigned to any hitherto described species. In phylogenetic analyses this clone K 60 E.2 was more closely related to B. leo and K 68 than to the B. felis isolates. Based on the sequence data of the ITS rDNA, a real-time PCR method was developed for the specific detection of B. felis. This test was proven to be sensitive and very specific in a blinded, externally controlled evaluation. The specifity was shown to be 100 %. The sensitivity of the test was 75 %. The positive prediction value reached 100 % and the negative prediction value was 82,1 %. A detection limit of 7 to 77 parasites/µl blood was determined. With this real-time PCR a modern, sensitive and highly specific method for the detection of B. felis is now available. The performance of the real-time PCR were compared with those of stained blood smears and the IFAT by investigating 206 blood samples of domestic cats of a small animal surgery in Port Elizabeth.When the results of the blood smears were compared with the results of the real-time PCR an agreement was found in 94,6 % (192/203) of the results. Identical results of the blood smears and IFAT were demonstrated in 91,2 % (187/205) of the cases. The results of the PCR and the IFAT corresponded in 96,1 % (196/204) of the investigations. The isolates K 8 and K 68, which had been diagnosed as B. leo and a closely related species by 18S rDNA sequence analyses and which had revealed babesia in stained blood smears, showed a negative test result in the PCR as well as in the IFAT. 9 isolates that were found to be babesia negative in blood smears gave positive results in the PCR as well as in the IFAT. 8 isolates were positive in the IFAT but negative in the PCR. 7 out of these isolates were also proven negative in the blood smears whereas in the remainig one case the blood smear was not examined. Infections with piroplasms were diagnosed in a high portion of the cats examined. The prevalence of babesia determined by stained blood smear was 32,2 % (66/205). DNA of B. felis was detected in 35,3 % (72/204) of the cats by real-time PCR. B. felis specific antibodies were found in 39,3 % (81/206) of the examined cats in the IFAT. No specific breed predilection was evident. The results of blood smears and PCR did not correlate with the sex or age of the cats. However B. felis specific antibodies were detected more frequently in male than in female animals and in a higher portion of the cats older than 2 years. Concerning the housing conditions, significantly more “indoor/outdoor” cats were affected. DNA of B. felis and specific antibodies were found in a portion above average of the cats coming from the “Walmer” area. Typical clinical signs attributed to feline babesiosis were observed in 28,8 % (19/66) of the cats showing babesia in the blood smear, in 25 % (18/72) of the cats with DNA of B. felis detected by PCR and in 22,2 % (18/81) of the cats that showed a positive test result in the IFAT. The part of latently infected was thus very high. No specific breed or sex predilection for clinical affection was evident. However a correlation of the age of the cats with the manifestation of a clinical babesiosis is assumed. In the age group between 0,5 and 2 years the percentage of infected cats showing clinical signs was above average.

Evaluation of antigens for the serodiagnosis of kala-azar and oriental sores by means of the indirect immunofluorescence antibody test (IFAT)

Antigens and corresponding sera were collected from travellers with leishmaniasis returning to Germany from different endemic areas of the old world. The antigenicity of these Leishmania strains, which were maintained in Syrian hamsters, was compared by indirect immunofluorescence (IFAT). Antigenicity was demonstrated by antibody titres in 18 sera from 11 patients. The amastigotic stages of nine strains of Leishmania donovani and four strains of Leishmania tropica were compared with each other and with the culture forms of insect flagellates (Strigomonas oncopelti and Leptomonas ctenocephali). Eighteen sera from 11 patients were available for antibody determination with these antigens. The maximal antibody titres in a single serum varied considerably depending on which antigen was used for the test. High antibody levels could only be maintained when Leishmania donovani was employed as the antigen, but considerable differences also occurred between the different strains of this species. The other antigens were weaker. No differences in antigenicity between amastigotes and promastigotes of the same strain were observed. It is important to select suitable antigens. Low titres may be of doubtful specificity and are a poor baseline for the fall in titre which is an essential index of effective treatment.

Five cases of Kala-Azar carried in from the Mediterranean area between May 1977 and March 1979 suggest that in this age of mass tourism these diseases, considered by us as rarities until now, may become more common. In all cases considerable difficulties in diagnosis were first encountered. Serology was always indicative, the indirect fluorescence antibody test (IFAT) being found very reliable. In 4 of the 5 cases the identity of the pathogen was previously established by different methods.
