I wrote an essay on John and Barbara Ehrenreich's concept of the PMC, and how it relates to Mark Fisher's concept of the Vampire Castle, which I read in this episode. In my essay I argue Mark Fisher makes the same mistake many Marxists have made. They conflate the petite bourgeoisie with the professional managerial class. This is an understandable mistake, but it is time for Marxists to begin using the Ehrenreich's definition of the professional managerial class in their class analysis. At another time I will quibble further with the Ehrenreichs. I think they make a few mistakes, but on the whole I think they have brought a tremendous improvement to the discipline of class analysis. Here, we provide encyclopedic transcriptional hippocampal profiling of mice treated with rhEPO. Based on ~108,000 single nuclei, we unmask multiple pyramidal lineages with their comprehensive molecular signatures. By temporal profiling and gene regulatory analysis, we build a developmental trajectory of CA1 pyramidal neurons derived from multiple predecessor lineages and elucidate gene regulatory networks underlying their fate determination. With EPO as tool, we discover novel populations of newly differentiating pyramidal neurons, overpopulating to ~200% upon rhEPO with upregulation of genes crucial for neurodifferentiation, dendrite growth, synaptogenesis, memory formation, and cognition. Using a Cre-based approach to visually distinguish pre-existing from newly formed pyramidal neurons for patch-clamp recordings, we learn that rhEPO treatment differentially affects excitatory and inhibitory inputs. There are good reasons to hesitate using the term "PMC." There are also a lot of people who need to think more about the designation of this term. The subject-matter is ideology at a level deeper than most people tend to go. The assumptions, habits, and general mode of the PMC are distinguishable from the rest of the working class. The "Professional Managerial Class" is not just a cluster of professionals and managers, but specifically an analysis of the way that class society is reproduced. It is essential for understanding capitalism today, but simultaneously serves as a critique for a lot of purported "alternatives" to capitalism. Socialism, fascism, and New Deal liberalism, are all three different flavors of managerial control correcting for certain contradictions that Marx had thought would destine it to ruin. It is hardly surprising that Marx left the PMC, its existence and role, out of his analysis; he was writing at a time when most of its current roles did not exist or were being carried out by either the capitalists or the workers themselves. But Marxists aren't the only ones who need to take the PMC critique seriously. Liberals and conservatives do as well. Revolutionary or reformist, left or right, insofar as you want to think seriously about what constitutes society, much less positive ways of changing it, PMC critique is something you will need to think through. I am being joined by my friend and colleague Elton L.K. of Working Class Intelligentsia podcast in leading a series of lectures and discussions on the topic of the PMC, its critique and consciousness. Abstract: Heterozygous mutation of Ambra1, known as a positive autophagy regulator, produces autism-like behavior in mice and autistic phenotypes in humans in a female-specific manner. However, the substantial roles of the Ambra1 mutation in neurons are still unknown. We find that Ambra1 heterozygotes display a moderate decrease in excitatory synaptic release in-vitro and ex-vivo exclusively in females without autophagy activity, resulting in significant alterations in {gamma}-oscillation power and seizure susceptibility by excitatory/inhibitory (E/I) imbalance. Specifically, Ambra1 deficiency has no effect on neurogenesis and morphogenesis, but selectively decreases excitatory synaptic activity without changes in synapse number, quantal size, synaptic release probability, and synaptic plasticity. Therefore, the limited excitatory synaptopathy by Ambra1 expression levels ultimately determines E/I imbalance in global neural networks leading to the female-specific ASD. Abstract: A key feature of advanced brain aging includes structural defects of intracortical myelin that are associated with secondary neuroinflammation. A similar pathology is seen in specific myelin mutant mice that model 'advanced brain aging' and exhibit a range of behavioral abnormalities. However, the cognitive assessment of these mutants is problematic because myelin-dependent motor-sensory functions are required for quantitative behavioral readouts. To better understand the role of cortical myelin integrity for higher brain functions, we generated mice lacking Plp1, encoding the major integral myelin membrane protein, selectively in ventricular zone stem cells of the mouse forebrain. In contrast to conventional Plp1 null mutants, subtle myelin defects were restricted to the cortex, hippocampus and underlying callosal tracts. Moreover, forebrain-specific Plp1 mutants exhibited no defect of basic motor-sensory performance at any age tested. Surprisingly, several behavioral alterations reported for conventional Plp1 null mice (Gould et al., 2018) were absent and even social interactions appeared normal. However, with novel behavioral paradigms, we determined catatonia-like symptoms and isolated executive dysfunction in both genders. This suggests that loss of myelin integrity has an impact on cortical connectivity and underlies specific defects of executive function. These observations are likewise relevant for human neuropsychiatric conditions and other myelin-related diseases. Copy rights belong to original authors. This episode was unplanned, but when Barbara Ehrenreich died on September 1, 2022, we felt an urge to honor her memory and the profound influence she has had on the American left, socialism, feminism, and our collective thinking about class struggle. From her work in the women's health movement of the 1960s, to her theorizing (with ex-husband John Ehrenreich) of the "professional-managerial class" in the 1970s, to her explorations of Reagan-era yuppie pathologies, and her renowned exposé of low-wage work in 2001's Nickel and Dimed, Barbara Ehrenreich has been an essential and nuanced guide to the inner-life of American class conflict in the latter half of the 20th century and the beginning of the 21st. To undertake this journey through an extraordinary body of work, we're joined by two brilliant writers who have both — in their own way — taken up Ehrenreich's profound ethical and intellectual challenge: Alex Press, staff writer at Jacobin magazine (and KYE's favorite labor journalist); and returning guest Gabe Winant, University of Chicago historian and author of The Next Shift: The Fall of Industry and the Rise of Health Care.As Gabe writes in his stunning obituary last week, "Ehrenreich's specialty was to reveal her readers to themselves by showing them the other. Her humor and projection of personal vulnerability were particularly deft techniques for asking the reader to see their own position, often through identification with Ehrenreich: she invites this, beckoning you to follow her into her subject, and then suddenly wheels around on you—and you are caught out." Tras la biopsia, cirugía, radioterapia y quimioterapia, con unos efectos secundarios a veces difíciles de soportar, Bárbara, como muchas personas en su misma situación, busca en sus redes de apoyo un espacio donde poder compartir su experiencia, su malestar y su incertidumbre. Pero pronto se encuentra con la imposibilidad de poner en común los sentimientos que experimenta acerca del proceso, porque se da cuenta que en las personas a las que recurre prevalece un discurso de forzado optimismo basado en la convicción de que sólo una actitud positiva permitirá enfrentar y superar la enfermedad, mientras que los sentimientos asociados a la tristeza o la ira son considerados obstáculos, incluso posibles favorecedores de un curso negativo para la recuperación. Y hablamos de convicciones porque, a día de hoy, diversos estudios científicos discrepan en resultados, siendo imposible avalar que un estado de ánimo positivo fortalezca nuestro sistema inmunológico.
