We discuss a post-hoc study comparing the efficacy of Syfovre (pegcetacoplan) and Izervay (avacincaptad pegol) with Paul Hahn, MD, PhD, of NJ Retina.
As noted on Episode 144, there has been a steady decline in certified DRE officers over the past decade. As a result, in some jurisdictions there has been an increased use of DRE officers to provide testimony in drug impaired DWI cases where no DRE was utilized at the time of the initial DWI investigation. In this episode, Jake reviews two recent unpublished cases from the Court of Appeals on DRE Testimony, State v. Lewis and State v. Williams. Highlights: · Discover how State v. Lewis and State v. Williams, when read together, make it difficult for any officer to provide opinion testimony in a drug impaired driving case at trial. · Understand the collective importance of defense attorneys statewide to object to post hoc DRE testimony. · Learn attack points to post hoc DRE testimony if it is allowed to come into evidence over defense counsel's objection.

H., Cai, D. Abstract: Over the last decade, the advances in Brainbow labeling allowed labeling hundreds of neurons with distinct colors in the same field of view of a brain [1, 2]. Reconstruction (or "tracing") of the 3D structures of these images has been enabled by a growing set of software tools for automatic and manual annotation. It is common, however, to have errors introduced by heuristics used by tracing software, namely that they assume the "best" path is the highest intensity one, a more pertinent issue when dealing with multicolor microscope images. Here, we report nCorrect, an algorithm for correcting this error by reanalyzing previously created neuron traces to produce more physiologically-relevant ones. Specifically, we use a four dimensional minimization algorithm to identify a more-optimal reconstruction of the image, allowing us to better take advantage of existing manual tracing results. We define a new metric (hyperspectral cosine similarity) for describing the similarity of different neuron colors to each other. Our code is available in an open source license and forms the basis for future improved neuron tracing software. Copy rights belong to original authors. In this episode, Dave and Jamison answer these questions: Listener Sara asks, How can I deal with favoritism towards informal leaders in a group? The group is losing group intelligence because the informal leader's reasoning and direction is favored. Example: when member A propose an argument is dismissed, but when the informal leader proposes the same argument it is cherished. How do I react to the question “why didn't you do it this way” for features already in production? I am frustrated by being asked that. I got scolded for an idea that turned out to be bad after I implemented it (in production), although I asked the Lead for his opinion ahead of time. As soon as trouble came up a.k.a performance issue in production, he pointed the finger at me. Lost all kinds of respect for him.
When your intuitive mind senses a pattern and begins to search for the completion of that pattern, we call this, “curiosity”. But sometimes our searching for the completion of a pattern goes sideways, takes a shortcut, gets it wrong. The false logic that springs to mind as a result of this wrong turn is so common that it has a Latin name, “Post Hoc, ergo Propter Hoc.” Blame Isaac Newton. Newton taught us to think of cause and effect as sequential: a pool cue strikes a ball, which strikes another ball. As a result of our trust in Newtonian physics, the often-wrong logic of Post Hoc ergo Propter Hoc is almost irresistibly seductive because it begins with the observation that two events occurred in sequence. Remember a TV show called The West Wing? Jed Bartlet: C.J., on your tombstone it's going to say Post Hoc ergo Propter Hoc. C.J.: Okay, but none of my visitors are going to be able to understand my tombstone. Jed Bartlet: It means, “One thing follows the other, therefore it was caused by the other.” But it's not always true. In fact, it's hardly ever true. We did not lose Texas because of the hat joke. Do you know when we lost Texas? C.J.: When you learned to speak Latin? Do you remember The Big Bang Theory? In a 2009 episode, Sheldon Cooper was speaking to his mother on the phone: “The Arctic expedition was a remarkable success, I'm all but certain there's a Nobel prize in my future. Actually, I shouldn't say that. I'm entirely certain… (audience laughter) No, Mother, I could not feel your church group praying for my safety… (audience laughter) The fact that I'm home safe does not prove that it worked. That logic is Post Hoc ergo Propter Hoc… (audience laughter) No, I'm not sassing you in Eskimo talk.” Similar to “Post Hoc” is the broken logic of simultaneous occurrences, Cum Hoc ergo Propter Hoc, “With this, because of this.” “The bigger a child's shoe size, the better the child's handwriting. Therefore, having big feet makes it easier to write.” The mental sleight-of-hand of “Post Hoc” and “Cum Hoc” are what make advertising – and conspiracy theories – so easy to believe. @WardQNormal writes, “The trouble with conspiracy theories is that a lack of evidence is taken as proof that the conspiracy is everywhere. This is like thinking that the reason you never see elephants hiding up in treetops is because they're good at it.” Seeing patterns where they don't exist can be costly and dangerous. But still, I am wildly in favor of curiosity.Zora Neale Hurston wrote, “Research is formalized curiosity. It is poking and prying with a purpose.” Albert Einstein said, “Curiosity has its own reason for existing. One cannot help but be in awe when he contemplates the mysteries of eternity, of life, of the marvelous structure of reality. It is enough if one tries merely to comprehend a little of this mystery every day. Never lose a holy curiosity.” Gemma Stone, “When we enter a conversation with curiosity, we allow ourselves to see things differently and to be surprised by what we discover.” Tom Robbins, “Curiosity, especially intellectual inquisitiveness, is what separates the truly alive from those who are merely going through the motions.” Dorothy Parker, “The cure for boredom is curiosity. There is no cure for curiosity.” And none other than Daniel Boone – yes, THAT Daniel Boone – said, “Curiosity is natural to the soul of man and interesting objects have a powerful influence on our affections.” Like I said, I am wildly in favor of curiosity. This episode follows S2 E43 in discussing the possible role of NASH cirrhosis clinical trials in the transition from biopsy as gold standard to a post-biopsy world. Stephen Harrison starts this conversation by reviewing a post hoc analysis of Phase 2 belapectin data from Galectin Therapeutics, followed by a review of the FALCON 2 stud of pegbelfermin in NASH cirrhosis, sponsored by Bristol Myers Squibb. Jörn Schattenberg, Mazen Noureddin and Roger Green add their perspectives to the conversation.Stephen's primary point from the belapectin post hoc analysis is a practical one: the high correlation between an AST:ALT ratio>1 and HVPG levels can help clinicians suspect which patients have cirrhosis and possibly define candidates for NASH cirrhosis clinical trials. Mazen states that given the lack of trial data in this area, any secondary data that can be interpreted or integrated into a broader analysis is exceptionally helpful. Stephen asks what the ideal design would be for a NASH cirrhosis trial starting today. While answers vary, they all rely heavily on non-invasive tests to provide richness to the data and possibly set a foundation to become primary endpoints over time. This leads Roger to observe how much more complex and disease-focused trial design discussions have become over just the past two or three years.

Dr. Streiner’s advice for researchers: Pick a small number of primary outcomes and develop a narrow hypothesis. Then use post hoc analysis as a means of assessing future questions that can be investigated in a subsequent study. Dr. Streiner’s advice for clinicians: Treat a post hoc analysis as a hypothesis that requires further study. It should be viewed with some degree of suspicion because it may have been significant only by chance. Phil & Matt talk about cooking. Matt tells us his secret ingredient to making a killer bechamel sauce and Phil gives us his take on sweet n’spicy BBQ chicken and homemade turkey stock. Warning: this intro will make you hungry! Admittedly, it was a meat heavy conversation. Phil’s pasta sauce can be made vegan, which is how he normally likes it. Logical Fallacies & Fantasy Politics (15:09) Mel settles in to talk about logical fallacies, truth tables & syllogisms. Matt & Phil find out that they could either be a podcast, be on the internet or are the internet. It got a bit confusing to the non-initiated, but Mel does a good job at walking us through the details of logically sound v.s. empirically false arguments. Our baseball team fandom differences are put aside as we go off the rocker and talk fantasy fiction and politics in the era of Trump. Mel brings up The West Wing episode titled Post Hoc, Ergo Propter Hoc as we go down the rabbit hole on this one. Aye yai yai! Mel’s favorite fallacy is Ad hominem (Tu quoque), which is the “you too” argument. Matt goes meta on us, Phil wants to see more civility in arguments and Mel simmers us down, bringing the conversation back on point. We make connections around emotions, oratory and politics, revealing the shortcomings and limits of truth tables and logical expressions. Mel makes a strong argument that all undergraduates should take a course in philosophy to hone critical reasoning and argumentation skills. Stronger writing will result. Matt & Phil couldn’t agree more, especially in the current political climate. We end on a connection between works of fiction and logic. Phil asks how logic can operate in a fantasy world. Mel sheds light on how the world building that is done in fantasy (and high fantasy) needs to create the conditions for logical arguments, while not being factually or empirically true according to our earthly world. Sarah J. Maas’s ‘Throne of Glass’ series is an excellent example of this, according to Mel. Phil realizes that Plato and Aristotle had creativity and the arts in mind the whole time. Mel’s tips: find a passion, discuss a counter point, debate openly and follow a sound structure. This, especially in writing and arguing, is a good thing. Trust us. You’ll hear these in politics, advertising, etc. Only 2 groups of people in the world… Upcoming stuff! Karl Smith arrives in April to teach the Kinetic Shift! Chris and Mike will be students in the class, so come and join them. Architecture of Hypnosis is nearly sold-out for May 2017. Get in now, or wait until June…Then there’s only November left… Mike does his really stupid old British guy voice, purely for his own amusement. Bandwagon is the first logical fallacy in the podcast. Mike admits that he used to be a telephone operator. Bell Canada sent a salesman to Mike’s house to sell him an upgrade. Note to Bell: It didn’t work. We all want to be part of a Tribe! Groups are safer than lone wolves. Chris invokes Robert Cialdini, who’s an awesome author! Mike’s wife decides by committee… Being a Brit, Mike manages to bring the Germans into the discussion. Let’s invade France! Strawman! When losing an argument, redefine the terms of the argument and then attack the new definition. We must respect the elderly, because Mike’s getting close… A Red Herring is often used if you’re winning the argument. It’s pure distraction. Chris brings up reciprocity at this point, which is a good addition. Mike tells the tale of the patient NLP Trainer who kept spatially anchoring the point of interruption… Post Hoc Ergo Propter Hoc! (The Great Courses are awesome! Google them.) I got my car washed and it made it rain! Mike invokes Magical Thinking! Mike’s friend of 53 years, Brian McDowell thinks carpet fluff can change the course of a hockey game that’s happening elsewhere. Check cause and effect, and listen for Post Hoc! The No True Scotsman fallacy. Just keep redefining things until it works for you. Mike mentions Occam’s Razor. Friction doesn’t exist! It’s all about demons… The Gambler’s Fallacy or the Law of Independent Events. The dice and cards have no memory! There is no equilibrium to be restored. Chris talks about the stock market…he knows what he’s talking about. The taxi driver Webb, and the lottery tickets. Sound is getting weird now! Keep listening though, and just pretend someone’s tapping his fingers on the table in a senseless rhythm, and you’ll be okay! Being hit by lightning is more likely than winning the lottery! Would you pick the numbers that won last week? Human Givens says that strong emotions make us stupider! Even really good emotions do it. When you’re in love, you’re stupid. Seriously. Stupid. Mike says that good hypnotherapy gets the emotions involved. He invokes the example of a stupid inlaw he had to deal with. You can’t use logic to say that you can’t use logic! Thanks Aristotle! Circular Reasoning A proves B because B proves A… Chris makes a tedious point about spreadsheets, and Mike pretends to be interested. Ad Hominem is to attack the person, rather than refuting what they’re saying. “Your hat looks stupid, so I don’t have to listen to you!” Mike says that white dudes lie all the time! They don’t. Neither do other races, although Martians might… Get somebody angry and flustered and then point it out to win the argument! (It’s a cheap shot. Don’t do it.) False Dilemma is related to the double-bind. Sometimes there are gray areas…Politicians are great at this one.
