Notes: Seg 1 All of life sciences and biotech is healthcare. Diagnostics is the central organizing portion in healthcare, and consists of testing if needed. There are five (5) levels to diagnostics. Level One is Screening – where a patient may or may not be sick, check the health status, and also could be regular check ups for something pre-diagnosed. Level Two is the Diagnosis - where tests are taken and then you see the results of the test. Level Three is the Prognosis – what symptoms does the patient have, and where you need to go from here, what will happen with these symptoms. Level Four is a newer part called Interacting - This is where you can do personalized medicine and what's called theranostics or drug treatment. You combine drugs with the treatment. You have to analyze what type of drug is needed. What someone's body is like, how they metabolize drugs. What dose central levels are needed. People's reactions to drugs is not just body size or weight as suggested previously, but could be very much based on the genes. Level Five also new is called Monitoring - If disease treatment is over how do you monitor over the long term. What test do you take to confirm that there's been no return of the disease. Examples would be an MRD test which stands for a minimal residue disease. You also may be doing things like CT scans or x-ray scans in the past, but sometimes these do not detect disease properly. Some cancers could be very small and you need a blood test. People may check in through cycles of a disease treatment also. Seg 2 Regarding monitoring and Level Five using cancer as an example. People may go through 6 rounds of chemotherapy treatment. The doctor should monitor by round 3 and check is the tumor reduced or how is the chemo working. Do you want to have benchmarks to see if you're reaching the health goals and if the medicine is effective. Future treatment might be biopsy of a tumor and testing glass to see what drug may work on it. Revisiting Level Four in personalized medicine you may see in the future Chino metric analysis. Check the risk level for certain diseases so you can have a preemptive process to prevent potential predisposed diseases. This is level A. Level B would be when diagnosed with a disease, you check the tumor, mutations and genes checking both the DNA and RNA. Mara is on the board of AZ Bio (.org). The goal of AZ Bio is bringing educated people about biotech together to affect the state, the universities, and companies. Move research along with the responsibility to patients in the medical field. Mara is involved in venture capital which is critical, because it funds entrepreneurs in good companies to research and create Biotech product processes and products. There are other levels of investment, which typically start with friends and angel investing then move on to venture capital, and then private equity for really big rounds. She was the founder of Blue Stone and they invested in Southwest companies. In the fall of 2023 she became a partner at Illumina Ventures. They deal with 35 different companies. They've invested in all levels, biotech and genomics. You're looking for a VC to be specialized and knowledgeable about an industry so they can help the business grow provide it with money and add expertise. Seg 3 Mara and Illumina Ventures recently put out a report - Diagnostics Year in Review. This report covers how did the industry is doing, what stocks earn money, what IPOs there were, financials, clinical, innovation, and mergers and acquisitions. How does venture-capital work? Typically you have individual funds one through five. You raise money from investors and invest in new emerging companies. Companies may be doing research, drug development, or working other areas of life sciences. Examples of companies in their portfolio: Delphi deals in cancer, diagnosis test and early screening for cancer and detection test. Level one screen checking for lung cancer and what early symptoms there are. Biopsy can be dangerous so you need other tests. Let's Get Checked - a telehealth online company that brings testing to the patient either at home or in an urgent care or ER. This is faster and easier. Examples of test are Covid test and STD test. Serimmune human immunity research company Genome Medical a genetic counseling company, working with patients to discuss what diseases they may be predisposed to, and ways to not pass it to the Seg 4 Future of Biotech We saw with the pandemic, the rise of telemedicine with faster online diagnosis. We will also see in the future that AI changes drug development and reading tests The creation of better and faster equipment. The rise of nanotech, and early detection of diseases with blood samples - for example, at home blood draw. A major leak in Waipukurau's drinking water network has sent the town into immediate level four water restrictions. Water levels in the Pukeora Reservoir are critically low, at 26 percent. Central Hawke's Bay Mayor Alex Walker spoke to Corin Dann.
Wellingtonians are being urged to store emergency water supplies as the capital faces possible severe restrictions. Wellington Water is preparing for level four restrictions, which would ban all outdoor water use and cut indoor use by up to half. The region's Emergency Management Office recommends storing enough for at least 20 litres per person, per day, for seven days. Regional manager Jeremy Holmes spoke to Charlotte Cook.

This level includes VO2max testing efforts (so a 10 second flying 200 on the track to a 5 minute VO2Max effort), — you are using everything you got. All out for a short time. Pacing is hard without practice but what is notable is how, even with proper pacing, you fail to maintain the power output after a very short time. This is similar to weight lifting…the bigger the weight, the fewer the reps. The fast twitch muscles fatigue quickly from using up ATP and accumulation of waste products that make the muscle environment more acidic and interfere with the muscle function. At this level, oxygen is the limiter. After a rest, the muscle recovers and can go again but not quite as well. This process can be repeated a few times but the fast twitch muscle will get fatigued (maybe damaged?) and need to heal and adapt before the next effort (hopefully a couple days hence if you went to failure). At this level, you really need a power meter or smart trainer to measure your power output for the training interval. HR won't help because of the delay in HR response. LEVEL FOUR (4): Lactate threshold / FTP/ steady state (around an hour) — it's a concept as much as it's a power output that can be sustained as long as glucose is available to burn and lactate can be cleared and used by the mitochondria. Similar to this is the Functional Threshold Power….It's called functional because an hour is a nice round number and is approximately the time needed to complete a 40k TT which is a standard TT distance in cycling. This might also be about the time it takes to burn through available glucose. You are breathing heavily but steadily. You are not talking. At this level, if you paced it correctly, glucose is the limiter, not oxygen. A power meter is helpful but you could also do an FTP test based on distance covered in 1 hour or time to compete 25 miles. LEVEL THREE (3) Tempo / Sweetspot (also called “no man's land”) is in-between lactate threshold and endurance pace. This is where racing lives unless it is a very short or very long race. This is advertised as being more time-efficient than polarized training. Sweetspot is the high end, just below Threshold, while Tempo is the lower end, just above Endurance/FatMax level. LEVEL TWO (2): Endurance/ fat max (a long time). This is the famous Zone 2

"Today is the eight day of the month. Tomorrow is the 13th." ~Zen Koan Skills vs. habits. Habits vs. skills. What's the difference? Bound by language, experts in behavioral science tend to struggle as they attempt to delineate the two. MG suggests these scientists are missing the most important aspect of their relationship: How similar habits and skills are! There are many favorable reasons to regard habits and skills as nearly identical. First, it helps to remove the predominately negative connotation that habits carry. Skills are rarely, if ever, thought of as "bad" or negative. Second, it reinforces how POSITIVE HABIT BUILDING IS THE SAME AS SKILL BUILDING. This is precisely what The Habit Factor's P.A.R.R. method has taught since 2010. When it comes to developing good habits, the same sort of desire, effort, and practice is required that skills demand. And, knowing they are nearly the same helps to highlight how ineffective "The Habit Loop" is when it comes to positive habit building. By "The four levels of competence" model, developed by Martin M. Broadwell, we can see how both habits and skill capability increases. LEVEL ONE: Unconscious Incompetence: I don't even know that tying my shoes will be helpful. LEVEL TWO: Conscious Incompetence: I've just started to learn, and I'm really struggling. LEVE THREE: Conscious Compentence: I'm much better at tying my shoes now, and it's getting easier. LEVEL FOUR: Unconscious Compentence: The highest level: I tie my shoes without even looking or much thought! Level four is where both habits and skills share the greatest similarity and, interestingly enough, they reside in the same place in the brain!. Today is going to be a little different - some adjustments. In Episode 100 - you'll get to deconstruct how I listen to the guests. I've interviewed over the past 100 episodes. If time allows after the interview has formally concluded, I have a simple and consistent habit where I ask the guests, just one question - What did you notice about my listening? Now, this is a Level Four listening technique. It's designed as a way for me to make incremental improvements in each conversation. When I hear what people notice in the way I listen, I am making some very simple notes in my mind, that's a very important listening signal, make sure I continue to do it the next time. Occasionally people will highlight things that surprise me. They highlight things that wow, I didn't realize that was a listening signal for the person speaking. It's critical to understand that when you listen deeply, gently, thoroughly, carefully, you will change the way the speaker communicates. Not just what they say, not just what they think, but also what they make of the conversation, what it means for them. What can you expect today? You'll hear reflections of 11 people and their perspectives on how I was listening to them. You'll notice some very, very consistent themes. And yet you'll notice some subtle variations as well. You'll hear from six females, five males from deaf and blind people you'll hear from people whose first language is English and you'll hear from people whose home language isn't English. You'll hear from authors, musicians, professors, former military leaders, researchers, psychotherapists, and a range of many others. As you listen to them, deconstruct my listening, please keep these points in mind. This is just the way I listen. My listening context is very specific. Listening is situational. It's relational and contextual. The way I listen during an interview is with a listening orientation for the audience, for you. There are many questions I would love to ask the people that I interview yet, they're only appropriate for me. They're not going to help you and I play with this duality while I'm listening. How do I stay in the moment long enough - not to listen, but to listen on behalf of you. In chapter one of the upcoming book - how to listen and at the end of every chapter in the book, we have a series of three invitations, they're practices that we invite the reader or the audiobook listener to explore, we invite them to explore something to practice because we recommend that you read the book one chapter per week while practicing a technique during that week. So at the end of chapter one, we pose these three invitations and. Who's the best listener, you know, and what's one thing they do well? When was the last time somebody fully and deeply listened to you? and what did they do well during that conversation? When you think about that conversation where you were deeply listen to, how did you think speak and feel differently as a result? The only answer, in this election year, is politics. Instead of hurting businesses at one of the busiest times of the year, as well as earning the ire of the anti-lockdown crowd, he chose to hurt millions of other people. The CDC elevated its travel recommendation to "Level Four: Very High" for Canada, telling Americans they should avoid travel to its northern neighbour. This is the first show I've broadcast in more than a month in which Auckland has been out of the dreaded Alert Level 4.And honestly, I thought this time, things *might* be different. I thought after more a month of sitting at home, more of us in the big city might have taken stock, kicked the habit, and set our aspirations a little higher for the move to Level 3. Aucklanders might have learnt from every one else around the country. A new start. A new dawn. A new opportunity to refresh our routines.But no. We all went to McDonalds and Burger King and Wendy's and Carl's Junior, and KFC, instead.Well, not all of us. I'm not trying to be all high and mighty but I haven't had any of that Worldwide-franchisey-plastic-additivesy-supersize-me shit in almost fifteen years and I'll be damned if a move to Level 3 was going to convince me to spend half an hour with an idling engine waiting for some chicken nuggets in a drive-through queue.What's wrong with people? On Wednesday I could see queues down the street for my closest franchise. Addicts waiting for a fix! Honestly, big brand fast food franchise marketing has to be one of the greatest media swindles of all time. Because in case you haven't noticed, the food's really not that good.It's ok I guess. A bit of sugar, salt, and saturated fat plus however many hundred additives. Call me old fashioned but I prefer my food to be prepared with ingredients, not random numbers.And before you accuse me of elitism, know this: I'm not a vegan. I'm not a health nut. I eat sugar and salt and saturated fat. But if I'm going to enjoy some takeaways as a bit of a treat, I'd much rather spend my money on a roti canai at the Malaysian hole-in-the-wall down the road, or on a scoop and a piece of cod at the local fish and chip shop. Independent small businesses need all the help they can get right now.I know we love to joke about it. Haha! Two guys got caught trying to smuggle buckets of K-Fry over the Auckland border. Hilarious. I just think sometimes the fixation is a bit... lame. Apparently health officials have been considering setting up a vaccination system at KFCs, Piza Huts, and Taco Bells. Part of me thinks it's smart. Part of me thinks it's funny. Part of me thinks it's pretty sad state that there are apparently people in our society who would rather get a Cheesy Gordita Crunch than a vaccination for a deadly virus.I saw Population Nutrition Professor Boyd Swinburn on Breakfast this week, lamenting the fast food ‘culture of craving' that has dominated Covid-19. I totally agree with him. It's a conversation our society don't really want to have and you shouldn't need me to make this point: If you think Covid-19 puts pressure on our health system, just wait until you hear about bad nutrition!There are plenty of reasons I hope we never go back to Level Four. Plenty. Top of the list: I'm sick of people treating a quarter pack or a McChicken combo like a sweet sip of water after a long march through the desert.

Auckland's Level Four lockdown could be making the grass even greener on the other side. Real estate agents in regions such as Tauranga are reporting record levels of interest from Auckland home buyers. Some are even making lockdown sales where the buyers haven't been able to see the property. Valocity valuation director James Wilson told Kate Hawkesby it was already happening pre-lockdown, but now it's coming to a head. "We've now seeing and hearing those stories around interest levels spiking, you look at agencies reporting massive interest for listings that do come up." LISTEN ABOVE
Building supplies like plumbing goods and insulation have reportedly run out across the country.The Government is set to sign an order in coming days, to allow Auckland factories to produce building materials in lockdown.The Building Industry Federation says it will work with the Ministry of Business and Innovation so the right products are greenlit.Chief Executive Julien Leys says delays have been worsen by high demand before Level Four. Wellingtonians are already dreaming up their first food orders as the capital prepares for an overnight alert level drop. While alert level 3 has infamously become known as Level Four with takeaways, not everyone is choosing to open up this time around. Those on one side of a Waikato road will wake up tomorrow in level three, but the other will still be in level four. Everywhere south of Auckland will move to level three at midnight but the boundary cuts across Koheroa Road at Mercer. Waikato District Mayor Allan Sanson told Kate Hawkesby he's spoken to the Prime Minister's office about it but they're sticking with their decision. Two weeks is a long time when it comes to Covid. There are 562 community cases in the current Delta outbreak -- with just 53 new cases reported yesterday. That's 30 fewer than the day before. Most of the country comes out of Level Four at midnight and Northland is likely to follow on Thursday night, while Auckland faces another two weeks under Level Four restrictions. Otago University infectious diseases physician Kurt Krause says told Kate Hawkesby while yesterday's drop in case numbers is encouraging, only time will tell whether we have the outbreak under control. "There's new variants brewing around the world, different things are happening. It really has been hard to settle down and be confident about what's going to happen from one week to the next." The focus of her work is on tackling some of the toughest and most demanding behavioural challenges. She works with CEOs and senior leaders from a range of different cultures and countries. She designs & delivers accredited training programs on structural dynamics, generative dialogue and childhood story. Sarah is the Author of “Where did you learn to behave like that? A coaching guide for working with Leaders. She has also co-authored the book The Tao of Dialogue. Sarah holds a PhD and has published articles on collaborative research working with invisible realities & issues of integrity through the lens of Structural Dynamics. Sarah's ambition as a thought leader is to help Leaders recognise that their unresolved childhood stories not only adversely impact themselves but the teams and whole systems of which they are a part. Exposed to the work of David Kantor and her PhD “Separate Lives silenced voices” is a theme through all of her work Sarah referenced the podcast I had with Dr. Paul Lawrence, episode 13 where he explored Structural Dynamics with me Sarah illuminated the 4 different levels of Structural Dynamics Level One: speaks to our action propensities in conversation through four different acts, the move, the follow, oppose and the bystander Level Two: These are the operating rules governing our behaviour, open, closed or random Level Three: describes the communication domain we use or attend to and they include, Power which is about action or task, Affect is about intimacy or a focus on relationship, Meaning is about sense making, thought or knowledge Level Four: relates to your childhood story that is often invisible If I were to self-assess using this model, I might consider that I am naturally inclined towards Move/Random/Meaning/ Sarah shared her story about being uncomfortable with oppose having been in a family dynamic where oppose was used and where for Sarah as a child it was dangerous to oppose or be opposed. That was her internal narrative In high stakes we can react in ways that are embarrassing & shaming. Often something else is playing out behind the shadow behaviour, a fear or old narrative that is now not helpful The work involves understanding what happens in high stakes, what are the triggers that induce shadow behaviour? what are the themes or fears that lie behind our behaviour in high stakes? what are some of our formative experiences? The work is not to dwell on the story but to recraft a new narrative, knowing that the story does not change Structural Dynamics helps to make the invisible visible. It provides a way to understand what might be appearing in conversation and why often there exist model clashes between individuals on teams or in organisations Structural Dynamics is very proactive. It equips teams to change the nature of their discourses by having a common language to explore together. Sarah is of the mind that the work of Structural Dynamics and the associated behavioural change work necessary is still not central enough on teams Sarah appreciates that to do the work teams need to slow down to speed up.
