Usually I just grab a prepackaged 80/20 that's a little over 1 lb)1/2 lb-ish thick-cut bacon, chopped (about 6 strips but fuck it man go wild if you want)1 can (15 oz) of each: light red kidney, black, pinto, beans drained. 1 can (15 oz) fire roasted diced tomatoes with juice1 small can (6 oz) tomato paste2 big ass shallots, chopped (you can use 1 regular onion, sweet or red, instead if you're lame—just don't use white onion)1 red bell pepper, chopped (orange is fine too I guess. Just don't use green)1 big jalapeño, chopped. (Use a serrano if you need more heat and want people to think you're interesting)2 and 1/2 cups beef stock (maybe 3. Keep an eye on your liquid levels and add more if you need it for your desired consistency)1 T dried oregano2 T chili powder2 t cumin2 t cinnamon 2 t kosher salt1 t ground black pepper1 t smoked paprika1 t crushed red pepper2 T Worcestershire sauce (only use Lea & Perrins unless you don't respect yourself)1 T minced garlic (you can do more if you want. it's garlic. Go crazy. No one will question you.)1 big squirt Siracusa (this is for a little extra flavor, not heat)brown ground beef. Drain. Dump into chili receptacle. Brown bacon in same pan. Give the chopped veg a quick sauté in the fatty bacon pan. Scrape everything (yes, all those good brown bits too) into your main Chili cooking receptacle. Add everything else and give it a good stir to combine.How to cook it all: Stock pot - low heat for a long ass time, cover, stir occasionallySlow cooker - low for 6 hrsInstant pot/Pressure cooker - chili setting, 15-20 mins. 19 seems to work well. Vent after and you're good.Serve with sour cream, chopped scallions, cilantro, shredded cheddar and cornbread on the side

E. Abstract: The Locus Coeruleus (LC) is the primary noradrenergic nucleus in the brain with widespread projections driving changes in cognitive state and animal behaviour. The LC is composed of multiple 'modules' with specific efferent target domains enabling discretional neuromodulation. LC neuronal activity releases noradrenaline within the nucleus as a local feedback mechanism, but it is not known how this influences modular output. We address this question using whole-cell recordings and calcium imaging in rat pontine slices in combination with LC neuronal ensemble modelling to assess the influence of local noradrenaline release on cross-modular interactions. Electrophysiological recordings of LC neurons from rats transduced with the optogenetic actuator ChR2 showed auto-inhibition and lateral inhibition (of surrounding non transduced neurons). This inhibition was strongly frequency dependent and was mediated by noradrenaline acting on alpha2-adrenceptors (alpha;2R). To allow calcium-imaging of LC neuronal ensembles a Canine-Adenoviral vector strategy was developed using the PRS promoter to drive selective expression GCaMP6s. Calcium imaging allowed resolution of both increases and decreases in LC activity (to TTX / clonidine or high potassium). Selective chemogenetic activation of subsets of LC neurons (expressing the ionotropic actuator PSAM) revealed both a direct excitation (after application of PSEM308, 3-PSEM308) and an alpha2R-mediated inhibition of neighbouring LC cells (non-transduced). Differential retrograde targeting of PSAM or GCaMP6s to specific LC modules showed the presence of strong, reciprocal cross-modular inhibition (shown for the LC-olfactory bulb vs LC bulbospinal modules) and a subsequent rebound activity inversion. This represents a preferential, targeted, cross-modular, lateral inhibition within the LC rather than a non-specific surround inhibition. Computational modelling showed the emergence of lateral inhibition and biphasic responses to modular activation when alpha2R signalling and noradrenergic reuptake saturation were included. This interaction may facilitate recruitment of neuronal ensembles by coherent inputs and represents a bottom-up differential contrast-enhancement mechanism within the LC to produce a modality specific focus. Dr Perrins research since 1989 into CFS/ME has expanded our knowledge of the disease and how to diagnose and treat it. In July 2005 he was awarded a doctorate by the university of Salford, UK for his thesis on the involvement of cerebrospinal fluid and lymphatic drainage in CFS/ME. Since 2007 he has also held the academic post of Honorary Senior lecturer at the Allied Health Professions Research Unit , University of Central Lancashire, in Preston, UK. He was appointed member of the scientific committee for the 2nd 3rd and 4th World Congress on Neurobiology and Psychopharmacology affiliated with the European Association of Psychiatrists Annual Conference, Greece. In 2015 he joined the international faculty of the German School of Osteopathy, Hamburg; and teaches osteopathic theory and practice to students in colleges all over Europe. He has lectured internationally and in the UK to CFS/ME patient groups and the medical profession on the manual diagnosis and treatment of CFS/ME. He has published papers in major medical journals including the BMJ and is the author of the bestselling book The Perrin Technique: How to beat CFS/ME published by Hammersmith Press, London in 2007. Dr Perrin is a vice-patron of The University College of Osteopathy and in 2015 was the recipient of the inaugural Research and Practice Award from the Institute of Osteopathy. He is a founder member of the newly formed International Osteopathic Research Group and still continues to run clinics in Manchester and London specialising in the treatment of CFS/ME and Fibromyalgia. In a lengthy and beautiful intro, Robert Jordan brings us back to The Stone of Tear, where the Dragon's Banner whips in the wind on the top of the mountain fortress. Perrin and Faile are in their rooms, arguing, believe it or not. We discuss their relationship, and debate a bit about just how intimate it has become. Faile wants Perrin to accept her as a long-term partner, but he doesn't feel right about keeping her near him, considering the imminent existential threat that surrounds him and everyone he knows. Death could come at any time, and Perrin very badly wants to protect her, but refuses to turn his back on his childhood friend, Rand Al'Thor. Shaitan stirs, and like bubbles rising from rotten detritus at the bottom of a pond, a dark force rises to the surface of the Pattern itself. Those bubbles are what gives life, or some kind of sentience, to Perrins weapon, and it tries to kill him. After a harrowing fight, Perrin is able to misdirect it, and the ax drives itself into a wooden door. This essentially “grounds” the energy of the Dark One, dispersing it.

Barry Perrins sailed from England accross the Atlantic and beyond in his sailboat. He talks about his cruise of Portugal, Spain, the Canary Islands, Cape Verde, and Curacao. He was anchored next to the creators of RAN Sailing in Curacao when he was interviewed. Linus also talks about his second longest offshore passage (900 nm) from Aitutaki, Cook Islands to Nuku'alofa, Tongatapu, Tonga. Linus passed up on Beveridge Reef and sailed into a cold front that stoped the Slow Boat's forward progress.The Raymarine and CPT Autopilots failed and the diesel engine got air in the lines and a broken bleed bolt knocked out the Yanmar diesel engine. That forced Linus to sail into anchor at Big Mama's Yacht Club in Tonga where Janna and Sophie will meet Linus in a few days. For this episode I went for a chat with my friend Dave Perrins at his house in Brighton. I first met Dave a couple of years ago, at an event run by our mutual friend, Adam. Over the past few years we’ve had many a chat over cups of tea about mental wellbeing, productivity and getting stuff done, and also about the importance of sharing the lows and the struggles and not just the high points. Dave is the brains behind, Share Your Story, a popular event in Brighton which as the name suggests creates a platform for people to share their experiences, around themes such as immigration, death, building happy businesses, mental health and creativity. He also runs The Dad Course which helps fathers to be learn the ropes as they enter into life as a first time dad. Dave also writes a blog called Do What Matters Most, about living simply and making the world better.At the beginning of the podcast, we mention how we are not going to just talk about our projects - and then proceed to talk about our respective projects - but weaved around that we cover topics like: -how to ask yourself difficult questions, -being honest about your comfort zone, -what is happiness? -Loneliness - the useful side of experiencing pain. -The importance of sharing experiences This is a much more conversational episode than some of the others, and I feel despite the microphone it captures the essence of some of our past chats, and hopefully you will find it fun, entertaining and maybe even find a few nuggets of wisdom buried within it. She is traumatised by her experiences in the Arches and her mentor Liandrin offers her insights on how to cope with becoming an Accepted. Liandrin tells Nynaeve of Perrins's capture by the Seanchan close to Falme, which leads to Nynaeve, Egwene, and Elayne sneaking out of the tower to mount a rescue. But Nyneave is taken off guard by the appearance of Liandrin who ambushes them, using the One Power to subdue them. Meanwhile, Perrin and Elyas look for the Shienarans and Loial, where Perrin is told that he is Wolfbrother by Elyas. As Perrin tries to understand this he bonds with the wolf pack and in particular with one wolf called Hopper befriending Perrin. Elsewhere, as Min and Matt settle down for the night at a Tavern on the road, she discovers that Liandrin's contact she is meant to meet is Ishamael, one of the Forsaken. He offers to rid her of her visions if she brings Mat to Cairhien. Moiraine Damodred arrives in Cairhien, where she heads to her family home to meet her little sister, Anvaere. Their relationship is strained and Moiraine focuses on her task at hand as she sets about her investigations to find Lan. She also meets with Logain, and offers to end his life if he first helps her train Rand to use the One Power. Back at her home her sister forces Moiraine to realise that she no longer holds any power over her, and if she wants to know where her ginger headed boy is she will have to ask politely. Elsewhere, Lan stays with Alanna and her warders, who attempt to help him through losing his bond with Moiraine. Alanna finds a prophecy that reveals that the Forsaken Lanfear has returned to the world.Meanwhile, Rand and Selene leave Cairhien to travel to Selene's mountain lodge, where a Fade attacks them. Rand kills it with the One Power, and confesses his love for Selene, who chooses to stay with him despite his ability to channel, and reveals a secret of her own, as she begins to channel. Before she can do anything, Moiraine arrives and stabs Selene through the heart before cutting her throat. Pleading for Rand to run, she reveals Selene is Lanfear.
