In vivo, mice on a high saturated fatty acid diet had significantly lower platelet counts, which was prevented by eating a PUFA-enriched diet. Fatty acid uptake was largely dependent on CD36, and its deletion in mice resulted in thrombocytopenia. Moreover, patients with a CD36 loss-of-function mutation exhibited thrombocytopenia and increased bleeding. Our results suggest that fatty acid uptake and regulation is essential for megakaryocyte maturation and platelet production, and that changes in dietary fatty acids may be a novel and viable target to modulate platelet counts. We zijn vandaag op Herengracht 380-382 in Amsterdam. Hier staat het "pronkkasteel" dat tussen 1888 en 1890 werd gebouwd voor de nouveau-riche tabakshandelaar Jacob Nienhuys. Het werd ontworpen door architect Abraham Salm die zich liet inspireren door Amerikaanse huizen van superrijke families, zoals de Vanderbilts. Sinds 1997 is het NIOD, Nederlands Instituut voor Oorlogs- Holocaust- en Genocidestudies in het pand gehuisvest. architect Rogier Groeneveld kent alle hoeken en gaten van het imposante gebouw. Het was meer dan een woonhuis, het was een ontvangstmachine. Je kon met je koets het huis binnenrijden, zodat je in geval van regen droog de vestibule bereikte. Daar kon je, terwijl je wachtte tot je belet kreeg, een wijle verpozen in de Cosy Corner. En dat alles in het schijnsel van elektrisch licht, dat al in 1890 werd geïnstalleerd. Het huis zat sowieso vol met de modernste techniek. Koud was het er niet, dankzij de stoomverwarming. En wanneer kregen de Nienhuysjes eigenlijk telefoon? Ook daar weet Groeneveld alles van. Historicus René van Heijningen, zelf werkzaam bij het NIOD, werkt dag aan dag in een Rijksmonument. Went dat ooit? En wat zijn zijn favoriete ruimtes? Dit pronkkasteel is een verbazend staaltje van wat mensen eind Negentiende Eeuw zoal kon den aanschaffen als ze steenrijk waren.
