One of the most important aspects of walking the spiritual path - which is really just a path of freedom through inner refinement - is finding a good teacher. A good teacher enables us to stay connected to our intention by reminding us, again and again, should we forget that intention…whilst getting lost in the wild jungle of the untrained monkey mind. We are all together in this grand classroom, for our soul to play out the karmas that it needs to play out, to learn what it needs to learn on its way home, to the place beyond form, and beyond time. On the heels of yet another holiday season, we close out 2024, and interestingly, this year we are doing it on a New Moon. What better time to take stock of our lives, as we enter this new moon and new year. What better time to realize the abundant nature of the Universe, the many gifts we have been given, and how many of those gifts are immaterial, and actually, in the form of teachers, healers, and friends. People we have learned from, people who have been our guiding light in times of darkness. Today's guest, is one such beloved teacher, healer, and friend to many. We could introduce Ramkumar as a Vaidya, a Vedic Scholar, and one of the founding doctors of Vaidyagrama, an Ayurvedic Healing Village in Coimbatore, India that has helped thousands of people heal from around the world through the ancient practice of Panchakarma. We could make a list of his keynote speeches at various international conferences, share personal testimonies of how his satsangs and teachings have impacted me and fellow students, patients, colleagues, and friends of his. But, none of these descriptions seem to encapsulate him. Because this would be describing someone who identifies with their ego, and Ramkumar simply does not. What he is, or who he is, is an actual embodiment of Ayurveda; a living, breathing, flowing, channel of the Vedas, and we are so thankful that today, we get to share him with all of you. Abstract: Human-derived cortical organoids (hCOs) recapitulate cell diversity and 3D structure found in the human brain and offer a promising model for discovery of new gene therapies targeting neurological disorders. Adeno-associated viruses (AAVs) are the most promising vehicles for non-invasive gene delivery to the central nervous system (CNS), but reliable and reproducible in vitro models to assess their clinical potential are lacking. hCOs can take on these issues as they are a physiologically relevant model to assess AAV transduction efficiency, cellular tropism, and biodistribution within the tissue parenchyma, all of which could significantly modulate therapeutic efficacy. Here, we examine a variety of naturally occurring AAV serotypes and measure their ability to transduce neurons and glia in hCOs from multiple donors. We demonstrate cell tropism driven by AAV serotype and hCO donor and quantify fractions of neurons and astrocytes transduced with GFP as well as overall hCO health. They are able to capture business opportunities that otherwise would not be possible for them. They won't be as strongly affected by shocks e.g if member of family dies or storm destroys your home. Micro finance creates Increased possibilities and opportunities. From survey, 90% of end borrowers say they believe quality of life has improved with access to micro finance. Blended finance works, there are a lot more innovation happening and there is huge interest in it. In this episode, we touch on the SPAC craze that has hit the financial markets in 2020 and continued into 2021. Amrith discusses the history of SPACs from a niche product used by a group of wealthy individuals to it now utilized by most of Wall St. and Silicon Valley. He talks about seasoned investors like Bill Foley and Besty Cohen doing SPACs and how this is an alternative to raising money via a VC or Traditional IPO. He provides examples of SPACs such as the SE Asian “Super App” GRAB going public in a SPAC merger that values the business at nearly $40 Billion, even though they aren't profitable. Lastly, Amirth talks about key investment rounds called (PIPE deals). A Private Investment in Public Equity where investors like Blackrock, Fidelity, Franklin Templeton, T Rowe Price put in vast amounts of money to get exposure to these companies of the future, which has helped make SPAC's a mainstream alternative.

However, variability induced by current culture methodologies precludes using organoids as robust disease models. To address this, we developed an automated Organoid Culture and Assay (OrCA) system to support longitudinal unbiased phenotyping of organoids at scale across multiple patient lines. We then characterized organoid variability using novel machine learning methods and found that the contribution of donor, clone, and batch is significant and remarkably consistent over gene expression, morphology, and cell-type composition. Next, we performed multi-factorial protocol optimization, producing a directed forebrain protocol compatible with 96-well culture that exhibits low variability while preserving tissue complexity. Finally, we used OrCA to study tuberous sclerosis, a disease with known genetics but poorly representative animal models. For the first time, we report highly reproducible morphological and molecular signatures of disease in heterozygous TSC+/- forebrain organoids, demonstrating the benefit of a scaled organoid system for phenotype discovery in human disease models.
