Abstract: Human induced pluripotent stem cells (iPSCs) have already been used in transplantation therapies. Currently, cells from healthy people are transplanted into patients with diseases. With the rapid evolution of genome editing technology, genetic modification could be applied to enhance the therapeutic effects of iPSCs, such as the introduction of secreted molecules to make the cells a drug delivery system. Here, we addressed this possibility by utilizing a Fabry disease mouse model, as a proof of concept. Fabry disease is caused by the lack of -Galactosidase A (GLA). We previously developed an immunotolerant therapeutic molecule, modified -N-acetylgalactosaminidase (mNAGA). We confirmed that secreted mNAGA from genome-edited iPSCs compensated for the GLA activity in GLA-deficient cells using an in vitro co-culture system. Moreover, iPSCs transplanted into Fabry model mice secreted mNAGA and supplied GLA activity to the liver. This study demonstrates the great potential of genome-edited iPSCs secreting therapeutic molecules. Copy rights belong to original authors. This time around we've chosen a particularly grim chapter in MMA history to focus on, the later-era of MMA legend Kazushi Sakuraba's career. Known as the ‘Gracie Hunter' Sakuraba rocketed to fame for his victories over MMA's first family, defeating Royler Gracie, Royce Gracie, Renzo Gracie, Ryan Gracie, and Gracie proteges Vitor Belfort, Ebenezer Fontes Braga, and Nino Schembri. Despite all that success, the Gracies eventually got some of their own back, starting with Gracie disciple Ricardo Arona's victory over Sakuraba at Pride Critical Countdown 2005. Royce Gracie got his chance to even the score two years later, at Dynamite!! USA. And Ralek Gracie was there to pick up the pieces with a 2010 win over Sakuraba at Dream 14. Tony began his journey to the UFC by walking into a gym in Chicago well over 225lbs and meeting Coach Dino Spencer. At the time Tony was a blue belt in Jiu Jitsu but had plans of becoming a UFC fighter. Dino believed in Tony's dream and had him set a goal to lose enough weight so he could fight at 145lbs. Pat asks Tony about his journey in weight loss and his experience in extreme weight cuts prior to fights. Ultimately Tony achieved his dream of fighting in the UFC and now focuses on training and coaching. In the news section of the show we commented on the interesting news that apparently everyone is training at DDS and Renzo’s this week. We saw multiple main card stand outs and world champs including Buchecha, Gordon, Craig, Romulo, Pena, in the blue basement. This is for sure an ADCC camp, but interesting non the less. We also talk about some potential matchup’s for Ben Askren in wrestling. Maine talks about his interview with BJJ strength Podcast. In the recap section of Kasai Pro 5 in New York. We go deep into every single match of the 205 pound light heavyweight tournament featuring Aaron tex Jhonson, Felipe Pena, John Thor Blank, Kaynan Duarte, Jackson Souza, Craig Jones, and more. We talk about how absolutely crazy it was that Tex was able to Heel Hook Pena; and espouse about how amazing Duarte is. As well as talk about some interesting technical pieces of Craig Jones game with respect to back control. We talk about the super fights on the card as well. Marcos Tinoco impressive takedown game versus Romulo Barral. Dante Leon and him impressive rear naked choke of a top grappler in Edwin Najmi in back to back Kasai events; and Mayssa Bastos impressive performance and submission of Fiona Watson. After that we move into the recap of Quintet Fight Night 3 the ladies. This is the promotions first time showcasing an all women’s event. We talk about all the matchups on this event. In the first round team matchups with Team 10th Planet vs. Team Deep Jewels we saw impressive submissions from Elvira Karppinen that we’re instrumental in progressing team 10th planet into the second team stage. On the other side of the team stage we saw Team BJJ Kunoichi vs. Team Sun Chlorella Rikako Yuasa and Yuki Sugichi submit their teams way into the final team stage against team 10th planet. In the final team stage we talk about Liz Carmouche, Grace Gundrum, Fabina Jorge and the rest of 10th Planet vs. BJJ Kunoichi in a series of draws where 10th planet was able to secure the victory. We talk about the superfghts on the card as well, looking at the rebroadcast of Sakuraba and his crazy Japanese theatrics matchup and more. Then we get into our recap of Fight to Win 107, outside of Boston. Recapping Ana Carolina Viera vs Luisa Montiero. Nick the Show Pony Rodriguez in his Fight To Win debut; and one of the best judo matches the team has ever seen under the old school F2W rules. After all that we move into the preview section where we take a look at Fight to Win 108 in Philadelphia Pennsylvania headlined by Eddie Cummings return and tile defense. Maine is going to be at this card covering it live and is very excited about t given the stack of huge names on the card. This event features DJ Jackson, Murillo Santana, Josh Palmer, Dylan Royce, Mike Padilla vs Andrew Kochel in a match sure to produce fireworks. Plus a rematch the we all want to see Amanda Leve vs Maggie Grindatti for the brown belt Gi title at welterweight. Plus John Battle defending his belt, and Nick Rodriguez back to back weeks, but fighting for a title this week. This is not one to miss. Recorded 4-7-2019

IMMAF Moves Asian Open - Abu Dhabi Warriors 5 - ONE Heros Ascent and Saku Returns - Asian MMA Podcast Episode 14   I’ve got to start out with the big news on the amateur scene. IMMAF The International Mixed Martial Arts Federation has been forced to pull their 2019 Asian Open Championships out of China. The good news is that it will be landing here in Bangkok, they will announce the new date soon. I am going to try and set up an interview with Plai the president of the Thai Mixed Martial Arts Federation to get his take on it, but I am sure he is super excited for this. IMMAF has had bureaucratic issues with MMA in China in the past and I am sure this is no different. Of course, it’s a massive market that everyone wants to tap, but there are barriers to entry there, even ONE Championship and the UFC have struggled with China at various times. Personally, I think IMMAF under Kerith Brown's leadership has done some fantastic things for the sport, and to the same effect TMMAF here in Bangkok under the leadership of Plai has been fantastic so I think Bangkok is the absolute best options for the Asian Open Championships. I’ll definitely have more info on this as it develops. Over the weekend we saw Abu Dhabi Warriors 5 take place in, well Abu Dhabi. The biggest outcome there was UFC vet Oli Thompson get a second round stoppage against Cage Rage vet Roman Webe via TKO. Another notable fight on that card was UFC vet Rolando Dy out of the Philippines who also pulled a second round stoppage via TKO. This is the 5th outing for Abu Dhabi Warriors, but this was also their first event since 2016. unfortunately, this type of promotional inactivity is a common thread across Asia as the economics of the fight game are clearly difficult to figure out at a smaller scale. With any luck, we will see some more shows from Abu Dhabi Warriors in the near future. On to ONE Championships Hero’s Ascent in Manila over the weekend. The main event saw Adriano Moraes face off against the champ Geje Eustaquio, this was an amazing fight that went back and forth, at the end, I fully expected this to go in the Champs favor, but I was surprised, as I think a lot ware that the decision went to Moraes. I’ve taken a lot of issues with ONEs judging over the years, I am not a fan of judging a fight as a whole, It’s not a street fight, it’s a sport and they fight in rounds for a reason. I don’t think that the whole 10 point must system is the best judging criteria out there either, but judging by round is a much better fit for the format of the sport. I’ve seen a lot of judging decisions in ONE that were very questionable over the years. The most recent Angela Lee vs Mei Yamaguchi fight from 2018 comes to mind, after 5 rounds everyone I was sitting with in the press section thought for sure Mei had just easily handed Angela her first defeate but when the decision was announced Angela won via Unanimous Decision. It’s all a conversation for another day, if you guys want to see me do a whole episode on Judging Criteria let me know. Lastly, in super fanboy news, the legend, the Gracie Killer, Sakuraba will hit the mats for Quintet Fight NIght 2 for a submission grappling match on February 3rd. We get an immediate reaction to UFC 212 from Rio De Janeiro, Brazil. The rest of the card was a rollercoaster of highs and lows, but overall very entertaining.
