Meet Our Guest: Joyce Murphy brings a wealth of health care experience and a deep understanding of the New England health care landscape to our board. She retired from Commonwealth Medicine and the UMass Medical School in 2018, after a dozen years focused on developing solutions to the most challenging health care policy and public health populations. From 1991 through 2005, she held various positions at Caritas Christi Health Care System, including president of Caritas Carney Hospital, president of St. Mary's Women and Children's Center, and vice president of St. Margaret's Hospital for Women. She was a 2019 Advanced Leadership Fellow at Harvard Business School and has been repeatedly honored as an outstanding leader. In Drosophila melanogaster, the widespread inversion polymorphism In(3R)P is involved in climate adaptation, exhibiting non-neutral latitudinal clines on multiple continents. Here, I summarize new results suggesting that this chromosomal rearrangement represents a long-term (equilibrium) balanced polymorphism of ancestral African origin and that it harbors alleles that are maintained by balancing selection on several continents. Our findings indicate that In(3R)P spread out of its ancestral subtropical/tropical range and then become latitudinally along similar but independent climatic gradients, always being frequent in subtropical/tropical areas but rare or absent in temperate climates.Thomas Flatt is Full Professor of Evolutionary Biology and Head of the Department of Biology at the University of Fribourg, Switzerland. Thomas' research interests are the genomic basis of adaptation, population genetics, and the evolution life histories and aging, mainly using Drosophila as a model system. He received his M.Sc. from the University of Basel in 1999 (supervisor: Prof. Stephen Stearns), for work done at the University of Sydney with Prof. Richard Shine, and his Ph.D. from Fribourg in 2004 (supervisor: Prof. Tadeusz Kawecki). Between 2004 and 2008, he was a postdoctoral researcher at Brown University with Prof. Marc Tatar and a visiting postdoc with Prof. Neal Silverman at UMass Medical School, funded by fellowships from the Swiss National Science Foundation (SNSF) and the Roche Research Foundation. Prior to taking up his position in Fribourg in 2017, he was a SNSF Professor in the Department of Ecology and Evolution at Lausanne (2012-17), a Fellow at the Wissenschaftskolleg Berlin (2012), a faculty member of the Vienna Graduate School of Population Genetics and a tenured group leader at the Institute of Population Genetics in Vienna (2009-12). Between 2018 and 2021 he held a DFG Mercator Fellowship and Visiting Professorship at the University of Münster. Synopsis: Joel Schneider, Ph.D. and Robert Kotin, Ph.D. are the President & CEO and Founder & Chief Technology Advisor, respectively, of Carbon Biosciences, an emerging leader in the development of novel parvovirus-derived gene therapies. Joel and Robert sit down with host Rahul Chaturvedi to discuss the arc of their careers, how the AAV field has changed over the last decade, and how they approach building out their team at Carbon. They also talk about Carbon's focus on cystic fibrosis and the implications of The Cystic Fibrosis Foundation investing in their Series A. Finally, they both weigh in on what excites them when thinking about the future of gene therapy. Biographies: Dr. Joel Schneider joined Carbon as CEO in 2022, after serving as Chief Operating Officer at Solid Biosciences. As Solid's first employee, he played an instrumental role in building the company's unique disease-focused business model. Dr. Schneider is an accomplished biosciences executive with a track record of achievement in identifying, developing, and financing high potential therapeutic modalities and has diverse leadership experiences across R&D, technical and corporate operations, and corporate development organizations. Dr. Schneider holds a Ph.D. from Rutgers University and an undergraduate degree from Brandeis University and is the author of numerous peer-reviewed articles related to Duchenne and stem cell biology. He completed a postdoctoral fellowship at Harvard University in the Department of Stem Cell and Regenerative Biology, where he characterized and developed the small molecules that enhance skeletal muscle regeneration. Dr. Robert Kotin has been a leader in adeno-associated virus (AAV) research for 35 years, focusing on the molecular biology of the virus's non-structural proteins and then leveraging this understanding to develop novel AAV vectors for somatic cell gene therapy. Beginning as a postdoctoral fellow at Cornell University Medical Center, Dr. Kotin discovered a common integration site for AAV DNA in human chromosome 19, which he designated AAVS1 locus. He spent most of his career in the Intramural Research Program at the National Heart, Lung, and Blood Institute (NHLBI) of the National Institutes of Health (NIH), first as a tenure-track investigator and then as a tenured senior investigator and Head of the Laboratory of Molecular Biology and Gene Therapy. While at the NIH, Dr. Kotin's laboratory invented and developed a scalable recombinant adeno-associated virus (rAAV) production process in Sf9 cells which was licensed by UniQure, ThermoFisher, Voyager, Biomarin, and others and was used to produce Glybera™, the first rAAV product granted regulatory agency approval for sale. Additional research from the Kotin lab resulted in the discovery of an AAV replicative product that has been described as closed-ended linear duplex DNA (ceDNA) and became the basis of the non-viral gene therapy company Generation Bio (NASDAQ: GBIO). Dr. Kotin served as vice president of virology and gene therapy at Voyager Therapeutics from 2014 to 2016. Since 2016, he has served as an adjunct professor at UMass Medical School, where his research interests include vectorizing and characterizing ancestral parvoviruses based on inferred sequences from endogenous virus elements (EVEs) as novel gene therapy vectors. Dr. Kotin earned his B.A. in biology from the University of California, Santa Cruz, and his doctorate in microbiology from Rutgers University and the University of Medicine and Dentistry of New Jersey (now Robert Wood Johnson Medical School).

Nutritionist Judy Palken discusses healthy food choices that can help us maintain our cardiovascular and cognitive health, keep our bones strong, and preserve muscle strength as we get older.  She provides tips for making healthy food choices every day as well as during the holiday season.  Judy Palken is a registered dietitian with a master's degree in Clinical Nutrition from Cornell University. She has spent many years working in Boston hospitals, with a specialty in cardiac nutrition. She worked in nutrition research in the Division of Preventive and Behavioral Medicine at UMass Medical School in Worcester. She  teaches classes in community settings, promoting healthy eating and activity for good health.  Judy was a guest speaker on the panel at eternalHealth's “Big Papi Intentional Walk” at the Natick Mall in Natick, Massachusetts along with eternalHealth's CEO Pooja Ika, and Baseball Hall of Famer David Ortiz. Born in Camaguey, Cuba, and raised in California, she completed her medical training at the University of California, San Francisco, after earning a bachelor's in Cellular and Molecular Biology from California State University, Northridge. She completed her residency at UMass Memorial and has worked as a board-certified physician in Internal Medicine in the Worcester community for more than 28 years, including as an associate professor of Internal Medicine, Family Medicine and Psychiatry at UMass Medical School. Along with serving as medical director with LAHA, Dr. Castiel has served on the boards of many of Worcester's nonprofit organizations. She has been honored with many awards, including the Public Citizen of the Year Award from the National Association of Social Workers, Massachusetts Chapter. — You already know what it means to live a fulfilling life. You may have had glimpses of this inner knowing and clarity after meditation or other activities like skiing or playing music that help us to return to ourselves. In these moments we are present, connected, creative, and open to possibilities. However, once we step back into the fast-paced world, our default habits - to rush and react - kick back in. We feel tempted to return to the comfort of what's familiar and easy, even though the outcomes are not ultimately healthy or fulfilling. We know that we're not leading a fulfilling life when we don't have enough energy, time, or whatever it takes to make meaningful choices—be it at home, work, school, or in relationships. Instead, we can train ourselves to return to ourselves – to the field of non-judging awareness within us. In returning to ourselves, we reconnect with what's present, what's important, and our potential. The possibilities that exist in this vast, open field of awareness are endless. Audrey is the Director of Child and Family Policy at the UMass Medical School. She is responsible for identifying, developing, overseeing the management of new programs, research, and evidence-based policy initiatives all related to children and families. Her current focus is on trauma, particularly racial trauma, and the long term negative impacts on the trauma of youth.  Also joining me is Thula Sibanda. She is the coordinator of the Juvenile Detention Alternatives Initiative (JDAI) in Massachusetts. Thula's focus is on the oversight and implementation of the JDAI initiative in Massachusetts. In Episode 021 of A Medicinal Mind: Wisdom and Wellbeing, we get into the world of mindfulness, meditation and the capacity of these practices, to relieve suffering and promote flourishing with founder of the Long Island Center for Mindfulness, Cory Muscara.   To put it simply, Cory is one of the most fascinating people I have ever met. While we are all unique, yet paradoxically all of one collective whole, Cory's background and experiences are quite astonishing.   Cory Muscara is the founder of the Long Island Center for Mindfulness, where he utilizes his extensive professional training in Mindfulness, Positive Psychology, and Integrative Health Coaching to facilitate a client's creation of, and movement toward, their optimal vision of health and wellbeing.   Cory has undergone professional training to integrate mindfulness within healthcare, schools, and businesses, through teacher training programs in Mindfulness-Based Stress Reduction (MBSR) at UMass Medical School with Jon Kabat-Zinn, Mindful Schools, Search Inside Yourself, and Breathworks Chronic Pain. He has completed his Integrative Health Coach training at Duke Integrative Medicine, is a 200-Hour Certified Yoga Instructor, and holds a Masters degree in Applied Positive Psychology (MAPP) from the University of Pennsylvania. In 2012, Cory spent six months in silence practicing mindfulness meditation 14+ hours a day as a Buddhist Monk in Southeast Asia. The experience taught him how powerful mindfulness can be as a transformative practice for cultivating greater peace, clarity, wisdom, and wellbeing in one's life, and became the inspiring force behind his mission to share this work with others. Cory currently serves as faculty at Columbia Teachers College where he teaches mindfulness to school leaders and principals, and is an assistant instructor for the Master of Applied Positive Psychology (MAPP) at the University of Pennsylvania, where he completed his graduate work. Cory has presented mindfulness to organizations such as Johnson & Johnson, Travelers, Prudential Insurance; universities such as NYU, Wharton Business School, and Dartmouth; a number of hospitals and healthcare systems in the New York area; and he regularly appears on the Dr. Oz show as a guest expert in the topic of mindfulness meditation. Cory's teachings and work with students stem from a deep passion for helping others, and personal experience with the transformative power of mindfulness, coaching, and positive psychology. What in the world do we mean when we say mindfulness anyway?   Cory reflects on his background and rather remarkable course to get to his current work with the Long Island Center for Mindfulness.   As part of the discussion, Cory lays out in exquisite detail, some of his approaches to cultivating a meditative practice with others, providing one of the most insightful practical examples I have ever heard when it comes to describing the meditative experience.   Digging deeper, I push Cory to explore the word spirituality and ask him to draw upon his unique experiences to share his views of the world “spirituality” and perhaps what we should adopt as a more unifying collective perspective.   Not wanting to forget the medical applications, Cory shares some of his knowledge having trained as a health coach incorporating principles of mindfulness and positive psychology into his work.
