Understanding How Nitric Oxide Affects Our Health

For quite some time now, ever since entering the world of red light therapy  — but particularly when getting into the realm of methylene blue  — I've wondered what nitric oxide's true impact on our body and wellness is. For the vast majority of my life, I was under the belief that all nitric oxide = good. That was ingrained at an early age with the countless pre-workout supplements I was taking during my heavy lifting days during high school through college. But it didn't stop there. The message of nitric oxide = good and that "more is better" has been perpetuated in the wellness, and even longevity, industries by countless experts and influencers. But how is that possible if one of methylene blue's main actions is nitric oxide inhibition? How is that possible if nitric oxide is a free radical and an environmental pollutant and, thus, is effectively an "electron stealer"?For whatever reason, I have never been able to get a straight answer from anyone on this topic. But serendipitously, I was recently reading a book that provided the most clearcut and sensible explanation that would allow for nitric oxide to be both "good" and "bad". The book is Methylene Blue: Magic Bullet: The Ultimate Mitochondrial Support, by Dr. John Lieurance. As I learned from the book, it turns out that nitric oxide is quite similar to how inflammation can be both a positive and a negative for our physiology; but with some additional nuances through in there. Today's episode is about nitric oxide and the roles it plays in our wellness and what the best strategies are for best harnessing the "good" nitric oxide while downregulating the "bad" nitric oxide.  If you found the information in today's episode particularly interesting and/or compelling, please share it with a family member, friend, colleague and/or anyone that you think could benefit and be illuminated by this knowledge. After that: Interleukin-1 inhibition in TTP. Researchers explore the potential of recombinant IL-1 receptor antagonist, anakinra, in a murine model of thrombotic thrombocytopenic purpura—an uncommon but potentially fatal disorder with limited therapeutic options. Finally, we'll learn about CAR T-cell therapy outcomes by race and ethnicity in large B-cell lymphoma. Blood is a symbol of life, which makes sense given that it plays such an important role in so many body functions, including our immune system. Blood makes up approximately 8% of your normal body weight and unfortunately, cancers of the blood, including lymphoma and leukemia, account for ~10% of all diagnosed cancers in the U.S. each year.  CAR-T cell therapy has emerged as a promising method to engineer a subject's own immune cells to fight bloodborne cancer. Our guest for this episode, Raquel Munoz from the Hospital Universitario Virgen del Rocío in Seville Spain, is doing research in this exciting CAR-T cell therapy space. Specifically, she is working to develop methods to help better quantify and understand the expansion of CAR-T cells in the body to help monitor treatment and predict outcomes.  We learn about why digital PCR was selected for her work and how it's helped raise confidence in the results they're getting. We even hear about how she believes this treatment will find success in treating solid tumor cancers. In Cassie's career corner, we learn how Raquel found her career path and love of immunology and working in a hospital setting. Podcast Intro: Immunity refers to an organism's ability to withstand a specific virus or poison through the use of specialized antibodies or sensitized white blood cells. We prefer to sacrifice healthy selections for the sake of saving time in this fast-paced atmosphere where fast foods are abundant. On the other hand, what are the negative consequences of these actions? With many businesses competing with one another, and with the high demand for food relative to the continuous growth in population, companies are now churning out processed and cheap food rapidly. The lack of education and the rise of daily expenses on necessities, such as food, allowed people to compromise. Now, the pandemic accelerated and made things worse. What are the factors and effects that unhealthy food consumption leads to? Tune in as Dr. Emeran Meyer discusses the gut-immune relationship in this episode. What you'll get out of tuning in: What is the “One Health Concept” What is the importance of the soil microbiome for plant and human health What are the mind-gut microbiome system and its role in health and disease What is the key to longevity and good health How the microbiome determines the strength of your immune system and your immune resistance. How to increase the abundance of microbes in your gut. How lifestyle determines your chances of surviving pandemics that are about to come in the future. Many people think it has a lot to do with the altered interaction we have, as infants in a much more sterile environment. Where we have banned microbes from the time of deliveries, or C sections and sterile hospital environments, to antibiotics early on, prematurely born babies ending up in intensive care units being loaded with antibiotics for four weeks sometimes.” “Early in life in terms of the natural exposure, Homo sapiens have had to their environment, including going through the birth canal, and picking up some of the good bacteria in the birth canal, which helps start to build the baby immune system is even like upon injection, right from the mom's body to then what we do. And if we look back, historically, there was a lot of skin-to-skin contact with newborns. There's a lot of mixing of the environment with the infant. And that helped create a baseline immune system, an innate immune system that was stronger.” “When we look in any indigenous culture, usually there's some relationship with a specific type of traditional fermented foods. And so in ayurvedic medicine, it was always considered to pacify Vata, which is the wind force, which creates the issues primarily in the nervous system first, and then the digestive system second, and it's like the nerves that upset the digestive system, like just being nervous, being anxious, being worried more of the people that are on the lighter side have less consistency with digestive capacity.” “The immune activation happens in the gut. So, these microbes are only microns away from immune cells. And it's the only separation not even from the real physical barrier, but from this mucous layer produced by some cells in the gut in the colon. And so, the margin for error is pretty small, and if you change that mucous layer just a little bit, you will already get contact between microbes and these immune sensors.” “People always talk about how we need to boost our immune system. With COVID-19, it's the opposite. You want to prevent this excessive response. An excessive response happens because the immune cells are programmed in a non-adaptive way in their passage through the gut.” Guest BIO: Emeran Mayer Dr. Mayer is a Distinguished Research Professor in the Department of Medicine at the David Geffen School of Medicine at UCLA, and Executive Director of the G Oppenheimer Center for Neurobiology of Stress & Resilience. He has been one of the pioneers in the science and practice of brain gut microbiome interactions with applications in a wide range of diseases in gastrointestinal, psychiatric, and neurological disorders. He has published close to 400 scientific papers and several books and has received multiple awards. In addition to his academic interests, Mayer has a longstanding interest in ancient healing traditions and has been involved in documentary film productions about the Yanomami people in the Orinoco region of Venezuela, and the Asmat people in Irian Jaya. He has recently co-produced the award-winning documentary “In Search of Balance” and is working on a new documentary “Interconnected Planet”. He is a strong believer in Buddhist philosophy and got married in a Tibetan monastery by Choekyi Nyima Rinpoche in Kathmandu. He regularly pursues meditative practices. He has spoken at UCLA TEDx on the Mysterious Origins of Gut Feelings in 2015 and has been interviewed on National Public Radio, PBS, and by many national and international media outlets including the Los Angeles and New York Times, the Atlantic, and Time magazine. He has appeared on numerous podcasts, including Lewis Howes' The School of Greatness, Tom Bilyeu's Impact Theory, and Mark Hyman's The Doctor's Pharmacy. He is the author of the 2016 bestselling book The Mind Gut Connection published by Harper&Collins and translated into 16 languages. In his new book, The Gut Immune Connection, Mayer proposes a radical, unifying concept about the chronic non-communicable disease epidemic we are finding ourselves in. He discusses how changes in our diet, lifestyle, and the way we interact with the world during the last 75 years have led to a profound dysregulation of the community of trillions of microbes living in our gut, resulting in a progressive chronic activation of our immune system. This aberrant immune system activation is emerging as the root cause of our current epidemic of interrelated chronic diseases affecting every part of our body. In addition, it makes us more vulnerable to viral pandemics. He uses the One Health concept to explain the intricate interconnectedness between the microbes living in our gut, in the soil, the health of our plants, and our own health.

Last year, while most shuls had limited and restricted minyanim from Rosh Hashana through Simchas Torah, this year, most shuls were open for business, albeit with some cautionary guidelines. A year ago, we were still in the “mask” and social distancing zone, this year, we sat near each other. Yet, as the doctors will tell you, we are from being out of the woods. Today's guest, Rabbi Dr. Aaron Glatt,  is one of the most sought-after experts on Covid and other infectious diseases. Rabbi Dr. Glatt is the Associate Rabbi at the Young Israel of Woodmere, and is the Director of the Medical Halacha Commission of the Rabbinical Alliance of America. Heis Board certified in Internal Medicine and Infectious Diseases, and is Chairman of Medicine and Chief of Infectious Diseases / Hospital  at Mount Sinai South Nassau, and a full Professor of Medicine at the Icahn School of Medicine at Mount Sinai. A spokesperson for the Infectious Diseases Society of America, he previously, was the President & CEO of St. Joseph Hospital; Executive Vice President at Mercy Medical Center; and Associate Dean at NY Medical College. Whenthe promise of a vaccine was first rolled out under the Trump Administration, many in the media, and his many political critics said, “it's not trustworthy.” Then under the Biden Administration, the messaging became, “follow the science….it saves lives,” and while millions of people across the globe have been vaccinated, there are still many who are reluctant. Leaving out those who believe in world conspiracies, there are others who are very reasonable and intelligent people who have legitimate questions based on sound and documented research. There have been many reports of people becoming violently ill or suffering serious side effects after being vaccinated, possibly from overstimulation of the immune system, known as the Cytokine Storm. There is no one more qualified to answer these difficult questions and concerns, than Rabbi Dr. Glatt.

Jim Joyce, co-founder, chairman, and CEO of Sigyn Therapeutics discusses the development of Sigyn Therapy, the company's blood purification technology to overcome the limitations of drugs and other devices to treat Cytokine Storm induced inflammatory conditions that lead to sepsis. He explains the relationship between Cytokine Storm and COVID and how this blood purification technology is an approach that is getting alot of attention right now. He talks about how this technology differs from current blood purification technologies and what the current treatment landscape looks like. #SigynTherapeutics #BloodPurificationTechnology #CytokineStorm James “Jim” Joyce has 30+ years of diverse public market experience, which includes two decades of public company CEO and Corporate Board leadership roles. He is also an inventor or co-inventor underlying 18 pending or issued patents. Prior to establishing Sigyn Therapeutics, Mr. Joyce was the founder and former Chairman and CEO of Aethlon Medical, a therapeutic technology company that he navigated from single shareholder start-up to Nasdaq-traded Company with 8000+ shareholders. During his tenure at Aethlon, Mr. Joyce oversaw the development of the Hemopurifier®, a first-in-class blood purification technology to address life-threatening viruses and cancer-promoting exosomes. Under his leadership, the Hemopurifier® became the first therapeutic candidate to be awarded two FDA “Breakthrough Device” designations and was the first and only device to receive “Emergency Use Authorization” (EAU) approval from both the FDA and Health Canada to treat Ebola virus. Time Magazine named the Hemopurifier® one of the “11 Most Remarkable Advances in Healthcare” and designated the device to its “Top 25 Best Inventions” award list. The Hemopurifier® has since been cleared by the FDA to treat severe COVID-19 infections in a clinical setting. Under Mr. Joyce's leadership, the Hemopurifier® was the subject of two Department of Defense (DOD) contract awards and a National Cancer Institute (NCI) contract. Mr. Joyce led the completion of approximately $100 million of equity financings on behalf of Aethlon Medical and established preclinical and clinical collaborations with more than twenty government and non-government research institutes. Based on the use of the Hemopurifier® to treat HIV and Hepatitis-C infected individuals in India, Mr. Joyce was the recipient of the “Spirit of India Award” sponsored by the Bill & Melinda Gates Foundation and awarded each year by the American India Foundation to the American business leader who has demonstrated a commitment to accelerate social and economic change in India. Mr. Joyce testified before Congress and lobbied Capitol Hill to promote the Hemopurifier® as a broad-spectrum countermeasure against bioterror and pandemic threats, which contributed to expanding the government-wide definition of treatment countermeasure to be inclusive of medical devices under U.S. law.

With the prospect of a universally effective vaccine in doubt, many of us have turned to diet and lifestyle to “shore up our Covid-19 defenses.” While many of the supplement regimens marketed as cure alls for Covid-19 are lacking scientific validation, natural compounds like vitamin D are showing some promise in serious research settings. What other nutrients can battle Covid-19? Are there pharmaceutical drugs that can be repurposed to assist in treating the coronavirus? Our guest today, Dr. Theoharis Theoharides has some answers. Dr. Theoharides is an MD, and member of the faculty at Tufts Medical School, who also holds a PhD in pharmacology, and a master's in immunology, from Yale. We discuss the latest research on how flavonoids and other natural compounds impact the immune system and why this research matters for our approach to treating Covid-19 in the coming months.
