Abstract: The ER Ca2+ channel ryanodine receptor 2 (RyR2) is required for maintenance of insulin content and glucose-stimulated insulin secretion, in part, via regulation of the protein IRBIT in the insulinoma cell line INS-1. Here, we examined store-operated and depolarization-dependent Ca2+entry using INS-1 cells in which either RyR2 or IRBIT were deleted. Store-operated Ca2+ entry (SOCE) stimulated with thapsigargin was reduced in RyR2KO cells compared to controls, but was unchanged in IRBITKO cells. STIM1 protein levels were not different between the three cell lines. Basal and stimulated (500 M carbachol) phospholipase C (PLC) activity was also reduced specifically in RyR2KO cells. Insulin secretion stimulated by tolbutamide was reduced in RyR2KO and IRBITKO cells compared to controls, but was potentiated by an EPAC-selective cAMP analog in all three cell lines. Cellular PIP2 levels were increased and cortical f-actin levels were reduced in RyR2KO cells compared to controls. Whole-cell Cav channel current density was increased by 65% in RyR2KO cells compared to controls, and barium current was reduced by acute activation of the lipid phosphatase pseudojanin preferentially in RyR2KO cells over control INS-1 cells. Action potentials stimulated by 18 mM glucose were more frequent in RyR2KO cells compared to controls, and insensitive to the SK channel inhibitor apamin. Taken together, these results suggest that RyR2 plays a critical role in regulating PLC activity and PIP2 levels via regulation of SOCE. RyR2 also regulates {beta}-cell electrical activity by controlling Cav current density, via regulation of PIP2 levels, and SK channel activation. Copy rights belong to original authors. No es algo generalizado, pero ocurre. A veces incluso nos llega algún mensaje preguntando ¿cómo puedo deslimitar mi ebike? En este vídeo os vamos contar lo que les respondemos. Te vamos a explicar las consecuencias reales de deslimitar o trucar tu ebike: 1.- La multa La multa… Sí. Deslimitar una bike te puede ocasionar una multa de la Guardia Civil o de la Policia Local de tu municipio. Pero no por el hecho de deslimitarla a nivel técnico. Sino porque al deslimitar, conviertes tu ebike en un ciclomotor. Una ebike estaría dentro de la categoría EPAC. Eso supone tener: • Asistencia al pedaleo por un motor eléctrico auxiliar hasta una potencia nominal máxima continua de 250W, sólo y exclusivamente siempre que se pedalee. • Asistencia al pedaleo hasta 25km/h. Esto está regulado por reglamento europeo e incluido en el Reglamento General de Vehículos y en el Reglamenteo General de Circulación. Si deslimitas la ebike pasa a ser el equivalente a un ciclomotor. Y necesita por tanto, matriculación, seguro e ITV… Y si no llevas todo eso: - Por ir sin seguro: Multa de entre 600 y 3000€ y el depósito o precinto del vehículo. El dinero depende del vehículo, si está circulando, reiteración… - Por ir sin ITV: 200€ Y si encima se da el caso de que no tienes el carnet de conducir… pues podrías hasta ser detenido. En fin, que puedes acabar con infracciones administrativas e incluso penales. En fin, no es fácil que un agente te ponga todo el pack… Pero algo te llevas seguro. 2.- Adiós garantía. He visto pisapapeles de 5.000€. Como os lo digo. Bicis trucadas que llegan al fabricante, éste detecta la trampa y… no hay garantía. La marca de la bici y del motor no van a mirar más. Si detectan que la bici ha sido deslimitada en algún momento, se acabó. Y no vale con volver a limitarla. Hemos consultado con varias marcas esta semana y nos confirman que la deslimitación queda en el software y se detecta. Y, además, también pueden hacerlo mediante el análisis de otros parámetros. Si el problema afecta a motor y batería, el coste total de la jugada puede ser de más de entre 2000 y 3000€. 3.- ¿Y quién la truca? En 2020 AMBE y CONEBI avisaron sobre lo obvio. Que tiendas y talleres que deslimitan o trucan ebikes también pueden tener responsabilidad. Porque, a fin de cuentas, si alguien tiene un accidente grave con una ebike, su familia, su seguro o incluso el mismo podrían buscar “responsables” y “culpables”. Y ahí la tienda o el taller podría estar incurriendo y cito “en la incitación a una infracción administrativa”. Es decir, que pueden acabar comiéndose un marrón interesante. Preparando este guión, he hablado con un par de servicios técnicos del sector y me han asegurado que ya han recibido peticiones de informes por parte de aseguradoras en casos de accidente. 4.- Una vida más corta… No la tuya. Pero sí la de tu bici. En una bici deslimitada el motor sufre más y la batería dura mucho menos. Y no nos referimos a autonomía. Solo en cargas tendrá más y, por tanto, tendrá menos vida útil. Y además, sufre más por el tipo de uso. 5.- El fin de las ebikes Es uno de los mayores riesgos para el fin de las ebikes. Es la excusa perfecta que están buscando muchos gestores u otros colectivos para quitarnos de en medio: no son bicis, son motos; son unos piratas, son un peligro…

Kate, Ryan, and Joel shared their perspective on what's happening in their respective region of the state and also weren't shy boasting about how great the climbing is in Pennsylvania. Pennsylvania is rich with a wide variety of climbing opportunities. Perhaps it's mostly known for its world-class bouldering but opportunities abound to sport and trad climb as well. Each organization does a great job addressing needs and any issues in their area and have been incredibly supportive of each other. Join us with your lively host Ray Lowe, better known as the luckiest guy in the world.Ray Loewe00:17Well, this is the lively host, Ray Lowe, and welcome to our brand new studio in Willow Street, Pennsylvania, wherever that might be. And we've got a great guest today. But before we get into our guest, I want to remind everybody that changing the rules is about the fact that all through our lives we're given rules, we're given them by our parents, and then we went to school in the school gave us rules in the church gave us rules, and our jobs gave us rules. And I think it was Steve Jobs, the Apple guy who came back and said, You know, when you're living your life under somebody else's rules, you're not living your life, you're living somebody else's life. And we're lucky enough that every week we interview one of the luckiest people in the world. Now we have a definition for that. The luckiest people in the world are those people who take control of their own lives and live them under their own terms. And we certainly have one of the luckiest people in the world with us today. And I want to start out with a statement and you're gonna see why it's so important as we go through. You know, just because you reach a certain age in life doesn't mean that you have to retire and that you're washed up. And in fact, many people when they reach a certain age are useful. And sometimes they're outstanding, and sometimes they're even become the best there is regardless of their age. So I want to do is introduce today, Candice O'Donnell. You prefer Candice or Candy?Candace O'Donnell 01:45Candice? Candice. I think Candy sounds like a retired stripper at my age.Ray Loewe01:53Oh, well,Candace O'Donnell 01:55I go with Candace,Ray Loewe01:56you know, you'd probably do that well, too. But we'll get into that one. Okay, so So Candice has a really interesting career. And her background is she's raised four children. Okay, not a small feat. While she was doing that she taught English at Elizabethtown University, she has always been active in the theater. And then she got to a point where she had a chance to create some projects that were of interest to her. Okay, and a let's and that started later in life. So So let's, let's tell everybody how young you are.Candace O'Donnell 02:31I'll be at in about a month 27th two months 27 of JuneRay Loewe02:37And you know, many people, when they reach these certain ages, say it's time to shut down? Well, not Candice. Okay, so tell us a little bit about these projects that you created. And tell us about them in general. And then let's get specific about the three specific ones that you chose to put into life.Candace O'Donnell 02:56Well, as you said, I've been doing theater here in Lancaster for maybe 25 years. I've done the Fulton I've done EPAC, my favorite role until I started doing this. This one-woman show was Driving Miss Daisy. That's a wonderful play with a fabulous message. But I guess it was about six, seven years ago. I started doing these one-woman shows I had done small skits for the anniversary of the Fulton 200 and 50th anniversary of people who had appeared at the phone, one of them being Sarah Bernhardt. And so I started I had done a little bit on Carrie Nation, the Temperance leader I had done Abigail Adams, but I started going in earnest into these one-woman shows. I had always wanted to do Mary Lincoln. And I hesitated on Mary Lincoln because it was such a tragic life. She was mentally ill, and she lost every single person that she loved. Every single person that she loved was taken away from her. And I couldn't figure out a way to get into humor in it. And so I kept hesitating, because I thought can I put in audiences through 70 minutes, 75 minutes of hell, her life was hell. And then I remembered one of her funny lines. When she first met Lincoln. She was the belle of the ball and he was a country bumpkin. And he came up to her and he said, Ms. Todd, I want to dance with you in the worst kind of way. And then she said, and then he proceeded to do exactly that. So that's where I got a little humor and I developed that. And then I decided to undertake Sarah Bernhardt an entirely different person. I go for a through-line with each of my characters. The through-line for Mary Lincoln was much madness is divinest sense, which is Emily Dickinson. And was it the track it was a fact that she was mentally ill. Sarah Bernhardt entirely different story, my throughline for her was Edsp ofs. Riojan was not ago not at all. And Sarah Bernhardt lived life on her own terms. She was a survivor. She invented the casting couch. She invented the PR agent. And she invented the cougar. She was amoral, rather than immoral. She was a tremendous survivor. She continued to perform 10 years after her leg was amputated. And incidentally, she did perform at the Fulton Theatre in Lancaster. And finally, I worked my way to Queen Victoria. I had had a strong interest in her for years. And the subtitle there is he was my all in all, Victoria is about her obsession, obsession with her husband, Albert. And particularly funny because they had nine children, she hated babies. You do the math, you put it together? Why did they have nine children? She hated them. So that's how I got into these. And I've really enjoyed them.Ray Loewe06:03Okay, well, I'm sorry, you don't have any passion for any of these at all. But you know, I think what does it take to do this? So let's go back to the first one to Mary Todd Lincoln. First of all, you had to make the decision that this was a character that you were going to bring to life. Okay. And so what did you have to do? I mean, because you wrote the script, right?Candace O'Donnell 06:29You I, it takes me about two to three years to research each person. And, but it's, it's amazing. Ray, the, the through-line comes to you almost instantly, at least it did to me when you see what the glue of this character is what you're going to emphasize. Now, another writer might not emphasize it. But then your research all falls into place. AndRay Loewe06:56okay, so you write the script, you're starting two years ahead of before you're going to deliver this Right, correct. And you got to go where do you find the background data on these people?Candace O'Donnell 07:07There, you're gonna really be surprised at this. It shows you what a low-tech dinosaur I am. I get it out of books. You've heard of books, B. O. O. K. S. I do not get online. Most people today would do their research online.Ray Loewe07:23Yeah. You know, we have our engineer here, who is college age, you know, and I think he's a digital book guy. Oh, is he? okay. Well, maybe not. Maybe he knows what a book is. Okay, You read books in college? Yeah, he did. Okay, so you dig in, and you've got two years of finding a character? Have you ever started on any and then found out halfway through that you couldn't get enough material and you killed the character?Candace O'Donnell 07:48No, I'm a little bit too cautious a person for that? I wouldn't. I'm usually interested in the character and know something about the character. And also I use films and plays as my sources too. I know enough about the character that I have yet to launch into one and thought, oh, no, this is actually a boring character. In fact, the more I researched them, the more fascinating they become.Ray Loewe08:13Okay, so So you start digging into this and you got this two-year process and you're writing your own script? Yeah. Okay. Which probably helps you memorize the script. Okay, and now you're going to deliver this. Okay, so how do you deliver this do you need to get sponsors for this as something that you go to somebody and do a trial.Candace O'Donnell 08:38I'm really glad you asked me that question, because it gives me a chance to pay tribute to Betsy Hurley of the Lancaster Literary Guild, and I haven't been asked that question before. She's the person who got me into the Ware Center with Mary Lincoln. Okay, and once those were very successful, and then I didn't have trouble getting into the Ware center after that. Most of them the more sellouts. My difficulty was COVID. You know, I had a delay of several well, all told this production was delayed four years because of COVID.Ray Loewe09:16Okay, so this is why Candice is one of the luckiest people in the world. I want you to think about this as our listeners here. Okay, so she took on a project several years ago, she knew it was going to take several years to do this. She ran into the COVID barrier most other people use as an excuse to quit, but not here. We were going to deliver this and we're gonna get into a couple of other things later as we go. So all of a sudden, Mary Todd Lincoln appeared on the stage, and you have a script. And do you have any plans to do anything with that script? Now that you've given the character life?Candace O'Donnell 09:55You mean Mary Todd Lincoln? Yeah. I've been asked to do a program here at Willow Valley and what I sometimes do with my programs, I'll do 20 minutes of Mary Lincoln. I'll do 20 minutes of Sarah Bernhardt. I'll do 20 minutes of Queen Victoria. I'm developing that now.Ray Loewe10:14Okay, so you've finished, Mary Todd. She's now alive. Okay. Yes. And now you sat there and you said, Okay, what's next? You didn't stop. Right? So how did you get the drive to go on to the next one? Candace O'Donnell 10:34I, because I'm an incorrigible ham. That's what my husband would tell you. Okay, that's where I get the drive. Okay, I have to admit it.Ray Loewe10:42Well, this is where the passion meets the excellence, though, so go ahead.Candace O'Donnell 10:47Well, that's what motivates me. But also, right, I really get passionate about these women. That's why I don't choose anybody that I don't admire. I see their foibles. We all have our foibles. But I couldn't do it fair, if I were doing man, I couldn't do Trump because I wouldn't, I couldn't admire him enough to do him, okay, I admire all these women. And the more I know about them, the more I see the hell they went through in various ways, and they triumphed over it. So it's not at all hard to motivate myself to do this. It was hard to keep the faith during COVID. With all the delays, like um, and of course, as you and I discussed, I'm getting older. So I'm wondering if I'm gonna go into dementia. Oh, and by the way, I'm losing my balance. I take the balance classes here at Willow Valley. So I won't fall down on stage. Okay. So you're wondering, you are wondering, is the body is the mind going to fail me. And you just sort of leap out in faith,Ray Loewe12:00but you didn't give up? And it worked. So let's talk about being queen. Okay. So I met you when you were going into this role of Queen of the empire Victoria. Okay. And, to tell you the truth, when I met you, I went to your performance with some trepidation. I mean, I'm sitting there saying, you know, can I sit through an hour plus of this? And I'll tell you, I was wrapped for 75 minutes, I don't think I moved in my seat, and to your little heart and to get me to do that. This is not me. I you know, so you know, you're an athlete. So you did something special here. And, it was a wonderful performance, and you brought this character to life. And I could just see in your eyes and your, the way you moved on stage that you are not you that don't you are Queen, Victoria. Okay. So let's talk a little bit about putting this one together. Because you had to start two years ago, you'd already done a couple of these. So you knew you could do it. Yeah. But now you started asking these questions in one of the things that you told me was about two weeks before you were gonna give this guess what, what happened?Candace O'Donnell 13:12Really, really nasty cough? And, of course, immediately tested for COVID. No, it wasn't COVID chest X-ray, is it pneumonia. And that was frightening I, people, I don't get frightened by performing because as I already confessed, I'm a ham I love to perform. But this cough frightened me. Because I was really terrified that I would not be able to deliver the performance. I was thinking of some other actresses I've worked with, but that was too late for them to memorize a 70-minute script. And I remember my daughter, saying, Mom, well, you may just have to give up on this. And she said I said, Well, I'm, you know me. I'm not giving up at this point. Don't you know my personality? And she said, Well, would you rather die mom? And I said yes. Yes. I would rather die than have to call Keegan my granddaughter was in the show introducing it. She's a temple, a student at Temple called Keegan and say, Keegan, we're not doing it. I would. So that was our big family joke. Mom would rather die than not do it so. As you know, you were there. Well, I was coughing right before I went out, I had to sucrets, I had tea. But now this, you said I'm the luckiest person in the world. And you are and I am and we are but that this was also a blessing. Because I absolutely believe this was God. I mean, I go out there and I'm not coughing. It's unbelievable to me, nor did I fall down on stage. Ray Loewe14:39And the show must go on. The show must go on. So I think this is a message that I want our listeners to get across. Most of us during our lives, put off projects that we want to do because life gets in the way. You know, here you were. You're raising four kids. You're teaching English. You know you're doing all of these things and then somewhere along the line, I think this germ woke up in your head and said, this has been there for a long time I have to do this.Candace O'Donnell 15:09Yeah. It's, it's, I think, if you have a particular passion, you almost have it from the womb.Ray Loewe15:17And it's never too late to do that. And even at your stage of the game, when you are worried about health issues and things like that, guess what? You know everything falls in place, it was no problem. You got it done.Candice O'Donnell 15:32I was flabbergasted by it myself. Oh, I want to say one other thing, because there were so many Willow Valley people in the audience, I had two very sharp audiences, you being one of the members of the audience, who were completely with me, and you can tell that when you step out on stage, you can feel the button. You know, Bruce Springsteen, performing as an exchange of energy between the audience and the performer. You can tell when they're with you, they were laughing ahead of my jokes. That before I got to my punch line, they were laughing. I thought, Oh, boy.Ray Loewe16:10Well, you know, what was the gift? Well, when we had to stand up and sing God, save the queen, and do the royal wave to greet you in there. I mean, you had us at the beginning. But I think this is a really good lesson for people because here you are. And I'm going to predict you're going to do another one. I have no idea what it might be.Candace O'Donnell 16:30My husband will kill me but yeah, we can all see I'm incorrigible.Ray Loewe16:33And the other thing that you're doing here is you're creating scripts, that maybe somebody else will do not as well as you do, but they'll do it at some point in time. And, and the research that you've done is just phenomenal projects. And I think you're to be congratulated for doing that. And I think it just makes you younger and younger and younger. So there all right, it keeps you going forward. Okay, so, unfortunately, we're near the end of our time here. So it's flies. Do you have any, any parting comments, any words of wisdom to anybody who wants to do these things? Or anything for the good of mankind?Candace O'Donnell 17:12Well, I just want to say I am hoping to eventually sell the scripts so that they will live on after me. Again, you may think I sound like a religious fanatic here. If you can get the guts to get out there and do it. Something in my case, I believe it was God, but something will see you through. Lo que tiene que cumplir los patinetes eléctricos llega con mucho retraso y demasiados años para entrar en vigor realmente. ¿Qué es un vehículo de movilidad personal? Sección 1. Definición de Vehículo de movilidad personal (VMP) Vehículo de una o más ruedas dotado de una única plaza y propulsado exclusivamente por motores eléctricos(1) que pueden proporcionar al vehículo una velocidad máxima por diseño comprendida entre 6 y 25 km/h. Solo pueden estar equipados con un asiento o sillín si están dotados de sistema de autoequilibrado(2). – VMP para transporte personal (alto máximo 140 cm, ancho máximo 75 cm y largo máximo 200 cm) – VMP para transporte de mercancías u otros servicios (alto máximo 180 cm, ancho máximo 100 cm y largo máximo 200 cm. Con una masa máxima admisible de hasta 400 kg) ¿Qué NO es VMP? – vehículos diseñados específicamente para circular fuera de las vías públicas o vehículos concebidos para competición, – los vehículos para personas con movilidad reducida, – los vehículos con una tensión de trabajo mayor a 100 VCC o 240 VCA, – los vehículos considerados juguetes, siendo tales los que su velocidad máxima no sobrepasa los 6 km/h, – vehículos diseñados y fabricados para ser utilizados exclusivamente por las Fuerzas Armadas, – los ciclos de pedales con pedaleo asistido (EPAC), – aquellos vehículos incluidos dentro del ámbito del Reglamento (UE) n.o 168/2013. ¿Quién tiene que solicitar este certificado o manual técncico? El fabricante del VMP o representante autorizado por el mismo será responsable de obtener un certificado de circulación para cada modelo y versión de VMP Van a tener los VMP una placa interna, ¿qué debe reflejar? Tendrán algo parecido a un número de bastidor en el que tiene que poner: – Velocidad máxima. – Número de serie o identificación. – Número de certificado. – Año de construcción. – Marca y modelo. ¿Tendrán que tener retrovisores? Abstract: Cyclic AMP is a ubiquitous second messenger used to transduce intracellular signals from a variety of Gs-coupled receptors. Compartmentalisation of protein intermediates within the cAMP signaling pathway underpins receptor-specific responses. The cAMP effector proteins protein-kinase A and EPAC are found in complexes that also contain phosphodiesterases whose presence ensures a coordinated cellular response to receptor activation events. Popeye proteins are the most recent class of cAMP effectors to be identified and have crucial roles in cardiac pacemaking and conduction. We report the first observation that Popeye proteins exist in complexes with members of the PDE4 family in cardiac myocytes thus restricting cAMP signaling. We show that POPDC1 preferentially binds the PDE4A sub-family via a specificity motif in the PDE4 UCR1 region and that PDE4s bind to the Popeye domain of POPDC1 in a region known to be susceptible to a mutation that causes human disease. Using a cell-permeable disruptor peptide that displaces the POPDC1-PDE4 complex we show that PDE4 activity localized to POPDC1 is essential to maintain action potential duration in beating cardiac myocytes. Now there’s a lot to it before you can actually get your product on a retailer’s shelf. In this episode we’re going to talk about packaging. While this is a topic I’ve covered in great detail on other episodes about compostable packaging, recyclable packaging, what the difference is, what you need to know, et cetera, the focus on this episode is on answering those questions. What do you need to know when you think about putting your product in a package? Who do you go to? Do you go to a packaging broker, or do you go directly to the converter? A converter is an industry term. These are the people that work the magic to create the packaging that your product goes in, the packaging that you see on retailer shelves. What questions do you need to ask them? What are the options that are available to you as a brand, and then how does that packaging supplier work with your co-packer, your marketing company, and anyone else who’s involved in designing your packaging and your product? Essentially everything that has to go on behind the scenes before you can get your product on more store shelves and into the hands of more shoppers. But before we can go down that path we’ve got to talk about some of the other very important elements that you need to have in place before you even develop your packaging. For example, how can your packaging to tell a compelling story about your brand? What are some of the ways that you can leverage your packaging to help grow incremental sales to make it easier for a retailer to say yes to your product? What can you do to catch the consumer’s eye so that they buy more product and then evangelize your product to their friends? Once you come up with your packaging design, what’s next? What format does it need to be in and how does that work? What are the limitations around your packaging? How customized can you make your packaging? What are some of the different strategies and technologies that you can take advantage of today to gain a significant competitive advantage in your category? Then more importantly, what are the different options available to you? This can vary greatly depending on what kind of product you have, whether it’s hot, cold, how much liquid is in it. There’s a lot to know here. Anyhow, don’t fret. This no longer needs to keep you up at night. Förster (Fluorescence) Resonance Energy Transfer (FRET) has become a powerful tool to study protein-protein interactions and signal transduction in living cells. FRET is commonly read out either by detecting the ratio of the donor and acceptor intensities (sensitized emission) or by detecting the excited state lifetime of the donor, which decreases with increasing FRET (Fluorescence Lifetime IMaging or FLIM). FLIM is robust, immune to bleaching and inherently quantitative. On confocal microscopes, FLIM is typically read out by Time-Correlated Single Photon Counting (TCSPC). This requires expensive add-on hardware and is inherently very slow, necessitating accumulation of many consecutive scans to arrive at low-noise lifetime images. For this reason fast lifetime changes, such as those encountered when reading out live-cell signaling events with FRET sensors, cannot be detected. Accumulation of images and/or very slow scanning can also cause morphological artifacts in the acquired images. We present the Leica SP8 FALCON, a new high-end confocal instrument with built-in very fast FLIM capabilities. The instrument applies a novel technology to records photon arrival times at up to 80 MHz (160 MHz if two detectors are used) using the built-in spectral HyD detectors. To arrive at such high counts rates, the instrument uses real-time 10 GHz sampling and analysis algorithms that render it largely immune to pulse pile-up. Falcon FLIM has been fully integrated in the LAS-X software, allowing convenient acquisition of FLIM time-lapses, stitched FLIM overview images, FLIM XYZ-stacks and spectral FLIM stacks, among others, as well as combinations thereof. We scrutinized the performance of the SP8 FALCON and applied it to record agonist-induced changes in concentration of second messengers such as cAMP and Ca2+ with sub-micrometer precision at high speed- on the order of several (512×512) frames per second (FPS). To detect cAMP by FLIM, we used our newest generation of EPAC-based dedicated FLIM sensors, which use mTurquoise2 as a donor and a tandem of two monomeric dark Venus proteins as acceptors. Reducing the image format to 128×128 pixels, we acquired good quality FLIM time-lapses at 25 FPS (83 FPS using resonant scanning), allowing detection of even very fast signaling events, including e.g. the activation of G-proteins and Ca2+ sparks. The large numbers of detected photons reduce pixel-to-pixel variability in the calculated lifetimes. Finally we demonstrate that the SP8-FALCON is ideal for fast FLIM screening applications in both fixed- and live-cell formats.

-TUMORE, AL VIA LA CARTA EUROPEA DEI DIRITTI DEL MALATO Nasce la Carta Europea dei Diritti del Malato di Cancro. È presentata oggi in occasione della Giornata mondiale contro il Cancro, a Strasburgo, con la partecipazione del Commissario Europeo per la Salute, Tonio Borg. Il documento è realizzato grazie ad una forte alleanza tra associazioni di pazienti (European Cancer Patients Coalition - ECPC) ed esperti in ricerca, innovazione e advocacy del “pianeta cancro” (European Cancer Concord – ECC), con il sostegno condiviso dell’Associazione degli Europarlamentari contro il cancro (MAC). La Carta è una vera e propria chiamata alle armi delle istituzioni europee e nazionali per un impegno concreto nell’affrontare l’epidemia di cancro che colpisce l’Europa e che porterà alla morte di 1 malato ogni 10 secondi nei prossimi 20-25 anni. In Italia nel 2013 sono state registrate 366mila nuove diagnosi e 173mila decessi. L’Italia ha contribuito concretamente alla redazione della Carta Europea dei Diritti del Malato di Cancro grazie al lavoro della Federazione italiana delle Associazioni di Volontariato in Oncologia (FAVO) e dell’Associazione Italiana Malati di Cancro, parenti e amici (AIMaC). La Carta Europea dei Diritti del Malato di Cancro è uno Statuto del Paziente che punta a risolvere le diseguaglianze che le persone affette da neoplasie affrontano ogni giorno in Europa, principalmente legate a status socioeconomico, età anagrafica, accesso a cure di qualità e mancanza di un Piano Oncologico Nazionale. Il cancro pesa sull’economia di tutto il mondo per un valore complessivo che sfiora i 1.000 miliardi di Euro, cosa che ne fa la malattia numero 1 in termini di prosciugamento delle risorse finanziare globali. -EPATITE C: SI PUO’ GUARIRE. CAMPAGNA EPAC E' partita oggi la campagna nazionale: “Epatite C – Le novità ci sono. Informati adesso”, promossa da EpaC Onlus, con l’obiettivo di sensibilizzare i pazienti con Epatite C affinché si informino sulle attuali opportunità di cura della patologia presso la rete dei Centri specialistici autorizzati dall’AIFA. L’Italia è il Paese europeo con il maggior numero di persone positive al virus dell’Epatite C: circa il 3% della popolazione italiana è entrata in contatto con l’HCV. Si stima che i portatori cronici siano almeno 1,6 milioni, di cui 330.000 con cirrosi epatica. Di questi, ogni anno, circa 20.000 muoiono per malattie croniche del fegato causate dall’Epatite C. Si tratta di una malattia subdola perché nella maggior parte dei casi rimane asintomatica per anni o addirittura per decenni, ma silenziosamente il virus lavora, provocando gravi danni al fegato. Quasi la metà dei pazienti può sviluppare cirrosi nell’arco di 25-40 anni, altri possono sviluppare manifestazioni extraepatiche, come vasculiti e danni renali fino ad arrivare all’epatocarcinoma e al trapianto. È importante eradicare il virus prima che si sviluppino questi elementi di gravità clinica. -DIABETE, LA DISINFORMAZIONE UCCIDE. APPELLO SID - AMD Le Società Scientifiche e Professionali di Diabetologia e le Associazioni delle persone con diabete denunciano come, con sempre maggiore insistenza, vengano presentate al pubblico e alle persone con diabete sulla rete e sui giornali ‘soluzioni’ miracolistiche, in larga parte basate su diete drastiche (cibi crudi, diete alcaline, vegetariane o altro), con la promessa di ‘guarire’ in pochi giorni il diabete e di abbandonare le terapie farmacologiche, inclusa l’insulina. La comunità scientifica nazionale e internazionale, l’Organizzazione Mondiale della Sanità e lo stesso Ministero della Salute da decenni sostengono a chiare lettere che la migliore prevenzione del diabete mellito è e rimane una sana igiene di vita (regolare attività fisica, dieta equilibrata e sana, moderato consumo di alcool e abolizione del fumo). Il fatto che la dieta sia un pilastro fondamentale del trattamento del diabete è un dato indiscutibile e irrinunciabile. Il problema nasce quando la terapia dietetica viene proposta come unica alternativa ai farmaci. Questo rischia, infatti, di creare situazioni estremamente pericolose, soprattutto per le persone con diabete di tipo 1 che, senza insulina possono andare incontro a condizioni a rischio di vita. Il diabete tipo 1 è una malattia che deriva dalla perdita delle cellule che producono l’insulina, per distruzione autoimmunitaria, per cui l’organismo ne diventa totalmente privo. -ANMCO: “SETTIMANA DEL CUORE” DAL 10 AL 16 FEBBRAIO Nella “Settimana del Cuore” in oltre 700 ospedali italiani i cardiologi ANMCO saranno a disposizione dei cittadini per incontri formativi, visite gratuìte ed iniziative educative. La Campagna sarà sostenuta anche dalle squadre della Serie A del Campionato di Calcio, che il 16 febbraio dedicheranno alla “salute del cuore” la 24a giornata di campionato. A sostegno dell'iniziativa anche la Federazione Italiana Gioco Calcio attraverso il Commissario Tecnico della Nazionale Cesare Prandelli, che parteciperà alla realizzazione di uno spot come testimonial della Campagna. La “settimana del cuore” sarà inoltre l'occasione per una raccolta fondi da dedicare alla ricerca e alla prevenzione cardiovascolare.
Background: Prostate smooth muscle tone is regulated by alpha 1-adrenoceptor-induced contraction and cAMP-mediated relaxation. EPAC is an effector of cAMP, being involved in smooth muscle relaxation and cell cycle control outside the lower urinary tract. Here, we investigated the expression and function of EPAC in human prostate tissues from patients undergoing radical prostatectomy. Results: mRNA and protein expression of EPAC was detected in all prostate tissues by RT-PCR and Western blot analysis. Immunoreactivity was observed in stromal cells, and colocalized with immunofluorescence for a-smooth muscle actin and calponin. Under normal conditions, noradrenaline-or phenylephrine-induced contraction of prostate strips in the organ bath was not affected by the EPAC activator pCPT (SP-8-pCPT-2'-O-Me-cAMPS.NA) (30 mu M). However, when the cyclooxygenase inhibitor indomethacin (50 mu M) was added, EPAC activators pCPT and OME (8-CPT-2'-O-Me-cAMP.Na) (30 mu M) significantly reduced contractions by low concentrations of phenylephrine. These effects were not observed on noradrenaline-induced contraction. OME and pCPT caused phosphorylation of the transcription factor Elk1 in prostate tissues. Elk1 activation was confirmed by EMSA (electrophoretic mobility shift assay), where OME and pCPT incresed Elk1 binding to a specific DNA probe. Conclusions: EPAC activation may reduce alpha 1-adrenergic prostate contraction in the human prostate, although this effect is masked by cyclooxygenases and beta-adrenoceptors. A main EPAC function in the human prostate may be the regulation of the transcription factor Elk1.
Eny2, the mammalian ortholog of yeast Sus1 and drosophila E(y)2, is a nuclear factor that participates in several steps of gene transcription and in mRNA export. We had previously found that Eny2 expression changes in mouse pancreatic islets during the metabolic adaptation to pregnancy. We therefore hypothesized that the protein contributes to the regulation of islet endocrine cell function and tested this hypothesis in rat INS-1E insulinoma cells. Overexpression of Eny2 had no effect but siRNA-mediated knockdown of Eny2 resulted in markedly increased glucose and exendin-4-induced insulin secretion from otherwise poorly glucose-responsive INS-1E cells. Insulin content, cellular viability, and the expression levels of several key components of glucose sensing remained unchanged; however glucose-dependent cellular metabolism was higher after Eny2 knockdown. Suppression of Eny2 enhanced the intracellular incretin signal downstream of cAMP. The use of specific cAMP analogues and pathway inhibitors primarily implicated the PKA and to a lesser extent the EPAC pathway. In summary, we identified a potential link between the nuclear protein Eny2 and insulin secretion. Suppression of Eny2 resulted in increased glucose and incretin-induced insulin release from a poorly glucose-responsive INS-1E subline. Whether these findings extend to other experimental conditions or to in vivo physiology needs to be determined in further studies.
