The reliability of testing epigenetic DNA methylation using Illumina beadchips is of paramount importance due to the specific intricacies of this technology. Illumina beadchips are widely used platforms for high-throughput epigenetic analysis, employing thousands of probes to measure DNA methylation levels at specific genomic loci. In this week's Everything Epigenetics podcast, Dr. Karen Sugden and I talk about how the reliability of these probes directly impacts the accuracy and validity of the results obtained.Keep in mind that in the context of Illumina beadchips, reliability refers to the consistent and accurate performance of each individual probe across multiple samples and experimental replicates. Each probe is designed to target a specific CpG site, and the methylation signal it generates must be dependable and reproducible.We discuss how reliable probes ensure the accuracy of DNA methylation measurements and how the reliability of probes becomes crucial for reproducibility when conducting large-scale studies using Illumina beadchips, such as epigenome-wide association studies (EWAS).Dr. Sugden and I also discuss how the reliability of probes on Illumina beadchips has implications for cross-study comparisons. Abstract: Migraine headache is thought to involve the activation and sensitization of trigeminal sensory afferents that innervate the cranial meninges. However, this notion is based mainly on preclinical studies in anesthetized animals with surgically exposed meninges. Here, we used two photon calcium imaging via a closed cranial window in awake, behaving mice to investigate the responses of meningeal afferents that might lead to migraine pain using a preclinical model of migraine involving cortical spreading depolarization (CSD). A single CSD episode caused a second-slong wave of calcium activation that propagated across afferents and along the length of individual afferents. Surprisingly, few afferents exhibited prolonged activation or suppression following CSD. However, a specific subset of afferents - those sensitive to acute meningeal deformation - exhibited stronger activation during locomotion bouts. Moreover, following CSD, a subset of previously silent afferents developed sensitivity to locomotion-related meningeal deformation. Our data point to acute calcium signaling in meningeal afferents as a potentially critical nociceptive factor contributing to migraine pain and support the notion that enhanced responsiveness of meningeal afferents to local mechanical deformation of the meninges is the neural substrate underlying the worsening of migraine headache pain during physical activity. Listen to a conversation that Frank van Laerhoven had with Valentina Fonseca Cepeda . Together with Daniel Castillo-Brieva, Luis Baquero-Bernal, Luz Angela Rodríguez, Eliane Steiner, and John Garcia-Ulloa, Valentina co-authored a recent IJC publication entitled Magical Realism for Water Governance Under Power Asymmetries in the Aracataca River Basin, Colombia, an article on wetlands, water and conflict in the Ciénaga Grande de Santa Marta wetland complex in Colombia. As many wetlands worldwide, this system is characterized by its high biodiversity and various livelihoods that are closely linked to water. Unfortunately, also as many wetlands worldwide, this system is vulnerable to and affected by many threats and conflict. Valentina and her colleagues unravel and examine this case employing a mix of methods that include a hydrological analysis, semi-structured interviews, and role-playing. With a background in ecology, Valentina's work focuses on local communities, more in particular the use of traditional ecological knowledge for the governance of natural resources. Valentina is affiliated with The Pontificia Universidad Javeriana in Colombia, teaching on the governance of socio-ecological systems, among other thing. At the same time, she works with the NGO ONF Andina where she helps designing and implementing participatory methods with institutional actors and community members to analyze ecosystem services and value chains with the purpose of co-creating alternative scenarios. Dr. Sugden first called Jackson Hole home back in the 40s when his family owned a dude ranch in Kelly. Dr. Sugden helped establish the first air ambulance service in Jackson Hole back in 1980. He has been inducted into the Wyoming Aviation Hall of Fame and is the recipient of the Harrison Ford Aviation Legacy Award. Currently, Dr. Sugden is a family medicine doctor based in Jackson Hole. In this episode, Dr. Sugden shares what it was like to grow up in a Navy family. He talks about spending part of his childhood in Kelly, WY, Rome and San Francisco. Dr. Sugden goes into what brought him back out to Jackson and how he helped create Jackson Hole Air Ambulance, the first of its kind in the greater Teton area. We'll be hearing a lot more from Laurel Sugden in the years to come. She has been studying Huachuma, - aka, the San Pedro cactus, for years, both on the ground in Peru and in the academy. Laurel has now become a remarkable source of knowledge on most subjects related to the sacred cactus.
