N., Jain, M. K., Qu, C.-K. Abstract: While mitochondria in different tissues have distinct preferences for energy sources, they are flexible in utilizing competing substrates for metabolism according to physiological and nutritional circumstances. However, the regulatory mechanisms and significance of metabolic flexibility are not completely understood. Here we report that the deletion of PTPMT1, a mitochondria-based phosphatase, critically alters mitochondrial fuel selection -- the utilization of pyruvate, a key mitochondrial substrate derived from glucose (the major simple carbohydrate), is inhibited, whereas the fatty acid utilization is enhanced. PTPMT1 knockout does not impact the development of the skeletal muscle or heart. However, the metabolic inflexibility ultimately leads to muscular atrophy, heart failure, and sudden death. Mechanistic analyses reveal that the prolonged substrate shift from carbohydrates to lipids causes oxidative stress and mitochondrial destruction, which in turn results in marked accumulation of lipids and profound damage in the knockout muscle cells and cardiomyocytes. Interestingly, PTPMT1 deletion from the liver or adipose tissue does not generate any local or systemic defects. These findings suggest that PTPMT1 plays an important role in maintaining mitochondrial flexibility and that their balanced utilization of carbohydrates and lipids is essential for maintaining both skeletal muscle and heart despite the two tissues having different preferred energy sources. It's been scientifically proven that consistent mindfulness practice can actually improve your sex life (and most other areas of your life as well) – isn't that just amazing! But the issue is that most people don't really have a clear understanding of what mindfulness is, often get frustrated with the practice and tell themselves that they “aren't good at it” or they “don't have time.” To explain to you just how powerful this practice can be, I chatted to Dr Lori Brotto, a clinical psychologist, author and researcher in Canada, who is considered the world's leading expert on the subject. Lori and I take a deep dive into just how useful mindfulness can be for your sexual experience. Mindfulness, the act of paying attention to the present moment, moment-by-moment, without judgement is not supposed to be easy. It can help us manage our emotions and improve our ability to connect with our bodies. We also delve into the difference between compassion and complacency – a concern so many people express when I talk about using mindfulness to change one's relationship with sex. Dr Brotto practices in British Columbia, Canada and is a faculty member of the University of British Columbia. Her best selling book, “Better sex through mindfulness” is in my top 10 books I've ever read!

In this episode, I speak with Dr. Lori Brotto (@ubcshr), Registered Psychologist, Professor in the Department of Obstetrics and Gynaecology at the University of British Columbia, Executive Director of the Women's Health Research Institute, leading international expert on the topic of sexuality, & author of the powerful book "Better Sex Through Mindfulness" (https://greystonebooks.com/products/better-sex-through-mindfulness). Dr. Brotto conducts research on women's sexual health & difficulties, develops & tests mindfulness-based & psychoeducational interventions for women with sexual desire & arousal complaints, & studies many aspects of sexual health including: culture & sexuality, hormones & sexual desire, cancer & sexuality, concerns about HPV & sexuality, asexuality, & more. In this episode, Dr. Brotto highlights how essential satisfying & embodied sexual lives are to our overall health & general quality of life, not just our sexual health & well-being. She explains what mindfulness means as it applies to sexual well-being & why it is such an optimal tool for promoting "magnificent sex" that involves full aliveness, presence, empowerment, attunement & connection. She highlights important research regarding the multitude of influences that contribute to sexual desire, arousal & response, debunking common myths & emphasizing the importance of understanding what turns us on sexually. She also shares tips about how to compassionately embark upon this exploration when we find it challenging, especially when it comes to the messaging we've internalized related to sex, memories of unpleasant sexual experiences, & high emotional charge around sex due to trauma or other life experiences. We also discuss the benefits of mindfulness-based tools compared to other interventions for low sexual desire (e.g., medicines & cognitive behavioral strategies). We conclude with Dr. Brotto describing two powerful evidence-based mindfulness practices from her new workbook: 1) a "sexual sensations awareness" practice that promotes alignment of physical & mental arousal, and 2) "mindfulness of difficulty" which helps us bring a spirit of compassion & openness to the range of experiences both sexually & beyond. Did you know your bones and muscles are intrinsically connected and in uninterrupted communication to make your body properly function? How about the research on 3D bioprinting (and we'd like to emphasize on bio) to incorporate it into the medical field sooner than you may be imagining? Small changes can lead to a healthier life, and Jello could be the tissue of the future. As weird as the last part of that statement may sound, it is not even that far from the truth. Today's episode is both educational and mind-blowing, as Dr. Rosie Sendher and Dr. Erica Fisk sit down with Dr. Marco Brotto and Dr. Venu Varanasi to chat about the fascinating concept of bone-muscle crosstalk and the no longer futuristic term of bioprinting. Dr. Marco Brotto is a George & Mary Hazel Jay Endowed Professor, Director of the Bone-Muscle Collaborative Sciences, and the Director of the Bone-Muscle Research Center. Dr. Venu Varanasi received his Ph.D. at the University of Florida in the Department of Chemical Engineering and is currently an assistant professor of Biomedical Sciences at Texas A&M University. These are unprecedented times. Much of the world is currently living under lockdown conditions and practicing social distancing in order to curb the spread of the COVID-19 coronavirus. So how is this affecting people's intimate lives? How are our sex lives and relationships holding up, and is there anything we can do to more effectively navigate this complex and challenging situation? For the answers to these questions, I spoke to Dr. Lori Brotto, a Professor at the University of British Columbia, a practicing psychologist, and author of the book Better Sex Through Mindfulness. I asked Dr. Brotto what she has been seeing in her clinical practice since the pandemic began and how it's different from usual. According to Brotto, some of her clients are reporting more avoidance-related reasons for sex at the moment, meaning they're more likely to report having sex in order to avoid a negative outcome rather than doing it because it's what they truly want. Listen to the podcast to find out why that is. We also address the following questions: What are some effective coping strategies for relieving stress and anxiety in order to pave the way for a more active and fulfilling intimate life during this challenging time? How do you successfully navigate singlehood in the era of social distancing? Some people under lockdown are in abusive or unhealthy relationships. How can they stay safe and find the help they need? What are the potential silver linings of this situation? How can we utilize this time to explore our sexuality and develop a new intimacy with our partners? Enjoy, and be sure to check out Dr. Brotto's latest book Better Sex Through Mindfulness! *** Want to learn more about Sex and Psychology?
