A pact to crowdsurf, the offer to sell a bootleg Rage Against The Machine T-Shirt for $20 at a food court in Long Island, and Greek tanning oil. Brian Schleyer (The Babies, First Bloom) "In 2008, Kevin Morby and Cassie Ramone shared a small apartment in Clinton Hill, Brooklyn for a few months and wrote some songs. After meeting again in a house party, the two decided to start a band. The idea was to recapture the early, stress-free days of playing music before they were both in groups that had gained some success and created accompanying pressures. Despite the intention, nothing much came of the idea until the next winter when both bands again had a break. Justin Sullivan, who had played with Ramone in Bossy, began practicing with them on drums, and The Babies debuted as a three-piece in March 2009 at Dead Herring, their friends' loft in Brooklyn. They played a few times before asking Nathanial Stark, formerly of Bent Outta Shape, to join on bass, and the full lineup's first shows were in that summer. The band released self-made tapes and singles for the small Wild World and Make a Mess labels before releasing their self-titled debut album for Shrimper in February 2011.They toured for much of the year, eventually replacing Stark with new bassist, Brian Schleyer. In early 2012, they left their Brooklyn home to record their second album in Los Angeles. Our House on the Hill was released on Woodsist late in the year."
Dan Jönsson, författare och essäistLitteraturRoberto Garvía: Esperanto and its Rivals. Abstract: Adhesion G protein-coupled receptors (aGPCR) function as metabotropic mechanosensors in the nervous system and other organs. aGPCR are heavily spliced forecasting an extraordinary molecular structural diversity. Many predicted isoforms lack the transmembrane (7TM) signaling subunit, but to what extent these non-GPCR isoforms are produced and what physiological purpose they serve is unknown. Alternative splicing through intron retention of ADGRL/Latrophilin/Cirl mRNA in Drosophila generates transcripts encoding unconventional proteins with an extracellular domain anchored by a single transmembrane helix (Cirl1TM). Here, we show that Cirl1TM transcripts are translated in vivo and that Cirl1TM binds Cirl7TM N-terminal fragment-dependently. This interaction enables mechanosensory neurons to distinguish input intensities through G(alpha)o-dependent signaling. Similarly, a direct interaction was found for mammalian GPR126/ADGRG6 isoforms. Together, our findings define intron retention and isoform-specific heteromerization as extraordinary molecular strategies to adjust Cirl-dependent mechanosensation and demonstrate physiological relevance of versatile aGPCR isoform repertoire to tune cellular responsiveness. Abstract: Predicting positive or negative reinforcement from environmental clues is essential to guide decision making and goal-directed behavior. In insect brains the mushroom body is a central structure for learning such valuable associations between sensory signals and reinforcement. We propose a biologically realistic spiking network model of the Drosophila larval olfactory pathway for the association of odors and reinforcement to bias behavior towards either approach or avoidance. We demonstrate that prediction error coding through integration of present and expected reinforcement in dopaminergic neurons can serve as a driving force in learning that can, combined with synaptic homeostasis, account for the experimentally observed features of acquisition and extinction of associations that depend on the intensity of odor and reward, as well as temporal features of the odor/reward pairing. To allow for a direct comparison of our simulation results with behavioral data we model learning-induced plasticity over the full time course of behavioral experiments and simulate locomotion of individual larvae towards or away from odor sources in a virtual environment. S., Rohwedder, A., Schleyer, M., Gerber, B. Abstract: Larvae of the fruit fly Drosophila melanogaster are a powerful study case for understanding the neural circuits underlying behavior. Indeed, the numerical simplicity of the larval brain has permitted the reconstruction of its synaptic connectome, and genetic tools for manipulating single, identified neurons allow neural circuit function to be investigated with relative ease and precision. We focus on one of the most complex neurons in the brain of the larva (of either sex), the GABAergic anterior paired lateral neuron (APL). Using behavioral and connectomic analyses, optogenetics, Ca2+ imaging and pharmacology, we study how APL affects associative olfactory memory. We first provide a detailed account of the structure, regional polarity, connectivity, and metamorphic development of APL, and further confirm that optogenetic activation of APL has an inhibiting effect on its main targets, the mushroom body Kenyon cells. All these findings are consistent with the previously identified function of APL in the sparsening of sensory representations. To our surprise, however, we found that optogenetically activating APL can also have a strong rewarding effect. Specifically, APL activation together with odor presentation establishes an odor-specific, appetitive, associative short-term memory, whereas naive olfactory behavior remains unaffected. An acute, systemic inhibition of dopamine synthesis as well as an ablation of the dopaminergic pPAM neurons impair reward learning through APL activation. Our findings provide a study case of complex circuit function in a numerically simple brain, and suggest a previously unrecognized capacity of central-brain GABAergic neurons to engage in dopaminergic reinforcement. J., Schleyer, M. Abstract: Neuronally orchestrated muscular movement and locomotion are defining faculties of multicellular animals. Due to its numerically simple brain and neuromuscular system and its genetic accessibility, the larva of the fruit fly Drosophila melanogaster is an established model to study these processes at tractable levels of complexity. However, although the faculty of locomotion clearly pertains to the individual animal, present studies of locomotion in larval Drosophila mostly use group assays and measurements aggregated across individual animals. The alternative is to measure animals one at a time, an extravagance for larger-scale analyses. In principle or in practice, this in particular rules out grasping the inter- and intra-individual variability in locomotion and its genetic and neuronal determinants. Here we present the IMBA (Individual Maggot Behaviour Analyser) for tracking and analysing the behaviour of individual larvae within groups. Using a combination of computational modelling and statistical approaches, the IMBA reliably resolves individual identity across collisions. It does not require specific hardware and can therefore be used in non-expert labs. We take advantage of the IMBA first to systematically describe the inter- and intra-individual variability in free, unconstrained locomotion in wild-type animals. We then report the discovery of a novel, complex locomotion phenotype of a mutant lacking an adhesion-type GPCR. The IMBA further allows us to determine, at the level of individual animals, the modulation of locomotion across repeated activations of dopamine neurons. Strikingly, IMBA can also be used to analyse 'silly walks', that is patterns of locomotion it was not originally designed to investigate. This is shown for the transient backward locomotion induced by brief optogenetic activation of the brain-descending 'mooncrawler' neurons, and the variability in this behaviour. Thus, the IMBA is an easy-to-use toolbox allowing an unprecedentedly rich view of the behaviour and behavioural variability of individual Drosophila larvae, with utility in multiple biomedical research contexts. In this episode, Divya speaks with Sharachchandra or Sharad Lele, Distinguished Fellow in Environmental Policy and Governance at ATREE (Ashoka Trust for Research in Ecology and the Environment) in Bangalore, India. They discuss two of Sharad's books, “Democratizing Forest Governance in India” which he co-edited with Ajit Menon, and his other book titled “Rethinking Environmentalism: Linking Justice, Sustainability and Diversity,” which he co-edited with Eduardo Brondizio, John Bryne, Georgina Mace, and Joan Martinez-Alier. Divya and Sharad use the discussion on the books as a way to unpack some of the fundamental questions like why are rights necessary, what is the meaning of democracy, and what is the role of state in the current context. They also talk about colonialism and its imprints on forest governance in postcolonial India. Sharad argues that, in the context of India, a country that has been traditionally hierarchical and where the state or the federal agency has all the powers, there are still colonial structures that are embedded in the system that reinforces those hierarchies. He suggests that recognizing citizens' rights and their ability to exercise them fully is one way to break away from colonial and hierarchical mindsets. Towards the end, Sharad shares that the commons is an arena with enormous scope for interdisciplinary work, yet academia and its various disciplines tend to fragment those spaces. He suggests that people working on the commons need to be alert to such tendencies and break away from the confines of a discipline by continuously asking how and in what ways their work can produce knowledge for meaningful change. Abstract: Across the animal kingdom, dopamine plays a crucial role in conferring reinforcement signals that teach animals about the causal structure of the world. In the fruit fly Drosophila melanogaster, the dopamine system has largely been studied using a rich genetic toolbox. Here, we suggest a complementary pharmacological approach applying the dopamine-synthesis inhibitor 3-Iodo-L-tyrosine (3IY), which causes acute systemic inhibition of dopamine signaling. Using Pavlovian conditioning, across developmental stages (3rd instar larva versus adult), valence domains (reward versus punishment), and types of reinforcement (natural versus optogenetically induced), we find that 3IY feeding specifically impairs associative learning, whereas additional feeding of L-3,4-dihydroxyphenylalanine (L-DOPA), a precursor of dopamine, rescues this impairment. This study establishes a simple, quick, and comparably low-cost approach that can be combined with the available genetic tools to manipulate and clarify the functions of the dopaminergic system - in D. melanogaster and other animals. When it comes to our apps. What's Apple doing what's Google doing and how did Apple mess this is up so badly, frankly. Hey, you're listening to Craig Peterson. Thanks for joining me today. We've had a problem for a very long time when it comes to any sort of apps. I saw a funny meme about yeah about Bill Gates this week. It said Bill Gates couldn't even stop viruses and Windows. It's a really good point that Windows was never designed to be secure at all. When they came out with NT, that was their first attempt to design it like a real operating system. They took the design of VMS, basically of Dave Cutler and company that had been designed over a DEC - digital equipment. And they said, Hey, we'll use this as a framework. This really works. It's not a hack. Let's just make this work. Then, of course, Microsoft had his fingers on it. So they really messed it up. They wanted to be compatible with everything that they could possibly be compatible with in the past. In the past, Microsoft did not have the barriers walls. If you will between the apps between the operating system, between the hardware and the apps. They didn't have the appropriate protections in place. Programmers used some lazy mechanisms to get around the operating system, going directly to things like graphics cards, because the operating system just slowed it down and they couldn't do the graphics they wanted to do that way. So it's been a real problem, frankly. It's been a real problem for a long time in the windows world, and NT was supposed to fix some of that. It did initially, and then it didn't. Now they're trying to tighten up this whole thing. how do you, if you're Microsoft or Google or Apple, how do you protect the people who are using your products from some of this malicious software? What's the way to do it? What Apple has come up with is a mechanism and Google as well that signs the software. You might have noticed that if you are trying to install software on your computer or your Google device from a third party website, it will come up and say, can't be opened, It can't be installed. There are a few different messages that come up. In the case of Apple now, with MacOS Catalina, which is the latest Operating system. Although, there's another one that is about to hit and it looks like they're going to change the whole nomenclature to, with the next release, but in Catalina, Apple now requires what they call notarization for all apps. Any apps that you are installing on that computer need to be signed digitally by Apple. So the developer, when they write the software, they compile it, they sign it themselves. It's a whole public key cryptography thing. Apple takes that software and checks it and then signs it. Both Google and Apple are using automated systems that try to verify whether or not the software is malicious. So it'll go through this automated system and will try and figure out well, is there any malicious content? Are they doing things they shouldn't be doing? Are they making suspicious calls to the operating system? Is it trying to get into files that it shouldn't be getting into? Now, there are ways to hide things from these automated systems. In fact, obfuscation seems to be the norm when it comes to any program or programming anything. It's just absolutely amazing. It does look for code signing issues and then it's designed to return the results to the developer very quickly and say, okay, it's all set. It is signed. You are ready to go. Then they can put it up for sale on the app store or available for free, et cetera. The same trick over on the Google side. In this case, in the Google side, they've got this alphabet owned malware scanning service called virus total that looks at data from over 60 different antivirus providers to figure out, is this software malicious? Is it using any sort of malicious libraries or routines? Now we have seen that happen, unfortunately, and it's scary because we have now found that even in the Apple app store, there have been many apps that included this library that was designed to basically steal personal information from you. Developers would use this library and it wasn't flagged by this notarization process. Now you install it and it's not the main feature of the app, but the app is spying on you. Now, there is a new piece of software out there that they're actually not all that new. It's been around for quite a while. It's called S H L E Y E R Schleyer and it is a Trojan that has been one of the most prolific pieces of Mac malware now for the last couple of years and it was notarized by Apple. Now, this is interesting, right? This whole notarization thing it's been in the last couple of major releases, but it snuck by. There is if you're an Apple user, there is a piece of software there's you can put on your Mac called brew and it uses open-source software to install all kinds of features. I use many of these pieces of open-source software all of the time. And they provide functionality that does not come with the base Mac operating system. And so in the case of brew, It is verified and validated by the brew people, right? Apple has nothing to do with this. There is another site out there called a homebrew.sh, which is a knockoff of the brew site, which is brew.sh. And the number of people were tricked into using that site, this homebrew.sh, and it apparently had fake flash updates. So it pops up and it says, Hey, you need to update. And we've all seen that before if we have flash on our computer. you click on it and open it and install it. In fact, this Schleyer was slash is so smart. It gives you instructions on how to get around Apple's notarization checks. So in case you didn't know if you install an app on your Mac and you, first of all, you probably can't get it to install, but if you do get it to install or if you download it and you try and run it out of your downloads. If you right-click on it, you then have the option to just open it and get around the signatures from Apple. Not good, not a good thing. So this, bottom line means that you cannot 100% trust these signed apps from Apple or from Google. Just, this is just to remember. Okay. This isn't something that any of these companies messed up recently. It's just normal. So be very careful. About what you install and it goes right back to something. I said a little earlier today, which is, do not install any software you do not absolutely need and make sure that you keep it all up to date. Some of this stuff does clickJacking. It tricks users into installing these cryptographic certificates. It decrypts and reads all of your HTTPS traffic. So if you're going to a secure server that is using SSL, it's decrypting it. It's harvesting your user IDs, everything. Okay. Apples goof, in this case, and they fixed it very quickly. It was reported to Apple. Apple did not figure this particular one out by themselves. So that goes right back to how important it is to have third parties out there that are looking at security, not just for Apple, but for many other pieces of software. All right, stick around. When we come back, we're going to talk about a new little study that was done about the internet of things hardware. Make sure you get all of this and more. ★ Support this podcast ★

In anticipation of Beer Boom, the upcoming documentary on New York City brewing, Jimmy talks to the people who made the movie possible. Filmmakers Eric Schleyer and Andrew Coury, and producer Aaron Bell of Canataro Park Avenue Financial join Jimmy in the studio alongside one of the documentary's main characters, Rich Castagna of Bridge and Tunnel Brewery. The five years Schleyer and Coury spent filming Beer Boom gave them a firsthand look at the city's rapidly changing beer scene. They talk about how Castagna grew a brewery out of his garage, and the ten thousand pounds of rocks he had to move before he could open his current location in Ridgewood, Queens.Beer List: Bridge and Tunnel Brewery's Mango New England IPA, Bridge and Tunnel Brewery's "Lobstah" Lobster SaisonBeer Sessions Radio is powered by Simplecast.

In this episode I talk about Volapük, an international auxilliary language created in the late 19th century by Johann Martin Schleyer, a German priest. I look at the history of the language and its structure and vocabulary, and also talk a bit about Schleyer himself. Volapük was the first international auxillary language, or indeed constructed […]

Carl Schleyer is a reknowned leader in the field of people/talent analytics. He's had accomplished stints at Sears, Home Depot, and now 3D Results. As an HR data scientist, his work has focused on better understanding employee behavior: what drives it, what it affects, and how best to predict and influence it. He's spent the last decade committed to improving the lives of employees, strengthening HR brands, and increasing the profitability of organizations. Finally, the alumni he's developed over the years are now, in addition to Carl himself, some of the most influencial and accomplished people in the talent analytics field.
In West Germany in September and October of 1977, a group of self-described urban guerrillas of the Red Army Faction (RAF) kidnapped industrialist Hanns Martin Schleyer. In exchange for Schleyer, the RAF demanded the release of its imprisoned leaders, Andreas Baader and Gudrun Ensslin. Those months in 1977 following the abduction of Schleyer are often referred to as the German Autumn, and they represent a crescendo of leftist political violence that had its origins, in some ways, almost a decade before. Terror prompted a crisis in the 70s for the West German government and German democracy. Of course, 1977 was not the first time in history that a German republic had been tested by a group of radicals intending to bring it down. That had already happened in the 1930s. But 1977 turned out very differently than 1933–when the Nazis “captured” power in a profoundly embattled and dysfunctional democracy. In fact, as Karrin Hanshew argues in her fascinating book, “West Germany's terrorist crisis helped to usher in the relatively stable civil society that still defines Germany today. Karrin Hanshew‘s new book Terror and Democracy in West Germany (Cambridge University Press, 2012) is, at once, a political history of the FRG, a history of democracy, a history of political theory in the abstract and in action, and a history of social movements, among many other things. I learned so much from it and I think that you will too.

Import of proteins into the mitochondrial matrix requires translocation across two membranes. Translocational intermediates of mitochondrial proteins, which span the outer and inner membrane simultaneously and thus suggest that translocation occurs in one step, have recently been described (Schleyer, M., and W. Neupert, 1985, Cell, 43:339-350). In this study we present evidence that distinct membrane areas are involved in the translocation process. Mitochondria that had lost most of their outer membrane by digitonin treatment (mitoplasts) still had the ability to import proteins. Import depended on proteinaceous structures of the residual outer membrane and on a factor that is located between the outer and inner membranes and that could be extracted with detergent plus salt. Translocational intermediates, which had been preformed before fractionation, remained with the mitoplasts under conditions where most of the outer membrane was subsequently removed. Submitochondrial vesicles were isolated in which translocational intermediates were enriched. Immunocytochemical studies also suggested that the translocational intermediates are located in areas where outer and inner membranes are in close proximity. We conclude that the membrane-potential-dependent import of precursor proteins involves translocation contact sites where the two membranes are closely apposed and are linked in a stable manner.
