The idea of the impintome is still foreign to many people. So, let's start with a simple explanation.For the majority of genes, we inherit two functional copies—one from our mother and one from our father. However, imprinted genes follow a different pattern, as we inherit only one functional copy. Depending on the specific gene, either the copy from our mother or our father undergoes epigenetic silencing. This silencing process typically involves the addition of methyl groups during the formation of eggs or sperm.The epigenetic modifications on imprinted genes typically stay put  throughout the organism's lifespan but undergo a reset during the formation of eggs and sperm. Regardless of their origin, certain genes are consistently silenced in eggs, while others are consistently silenced in sperm.Soon after egg and sperm meet, most of the epigenetic tags that activate and silence genes are stripped from the DNA. However, in mammals, imprinted genes keep their epigenetic tags. Imprinted genes begin the process of development with epigenetic tags in place.Imprinted genes are not the only genes that bypass epigenetic reprogramming in the early embryo. Studying imprinting may help researchers understand how other genes make it through reprogramming without losing their epigenetic tags.–The field of epigenetics and the imprintome has grown exponentially in the past decade, largely fueled by Randy Jirtle's groundbreaking research. Picture this: his 2003 study on how nutrition impacts gene regulation is the single most talked-about paper in the history of science. We had the pleasure of interviewing Gabríel Ólafs over Zoom video!Lullabies soundtrack the most formative season of our development. For centuries, parents have hummed them to children as either a salve to ease stress, a natural aid to trigger sleep, or a simple reminder that all will be well in the world once morning comes. Imprinted upon our memories and carried within our hearts for lifetimes, we pass them from one generation to the next. Icelandic composer and artist Gabríel Ólafs continues this tradition on his 2023 album, Lullabies for Piano and Cello [Decca Records US]. After generating over 100 million streams for his original instrumental compositions and earning widespread international acclaim, he summons the spirit of timeless Icelandic lullabies with a twist of his own magic.Gabríel's uncanny melodic sensibility has elevated him to the forefront of contemporary classical music. Following years of writing and performing, he gained international renown with Absent Minded in 2019 followed by Piano Works a year later, hailed by PopMatters as “a testament to the power of melody and mood.” 2022's Solon Islandus marked his debut for Decca Records US, Atwood Magazine described standout track “The Drifter” as “beautifully tender and achingly visceral.”On Solon Islandus, he effectively traced a creative roadmap through Iceland's artistic and cultural history in order to gain a deeper understanding of its conscious and subconscious impact on his own persona and vision. In particular, he leaned on the writing of renowned Icelandic poet Davíð Stefánsson, drawing inspiration from his body work and absorbing its spirit in the album.Beyond his own family experiences, he recalled hearing of a famous book entitled Icelandic Folk Songs. Originally published in 1906, this out-of-print tome collated various folk numbers and lullabies with rare sheet music. At the suggestion of a friend, Gabríel found a copy. He decided to not only reimagine five of the book's tunes, but to also pen five of his own lullabies as an homage and response to tradition.Accompanied by cellist Steiney Sigurðardóttir, the duo tracked the album in Reykjavik's Eldborg Concert Hall. Following the implementation of choir, orchestra, and cinematic heft on Solon Islandus, Gabríel made a conscious decision to strip the sound down to just these two instruments. As opposed to electronics, multi-tracking provided expansive flourishes. Together, Gabríel and Steiney unlocked a striking sound reliant on their quiet chemistry and a rich connection.We want to hear from you! We had the pleasure of interviewing Malavika over Zoom video!International R&B singer-songwriter, Malavika, has released her empowering single, “Overdrive”. A full-package entertainer, Malavika draws on her globe-spanning influences to craft a mélange of r&b, hip-hop and pop from a unique perspective. "Overdrive" is truly a global collaboration, created across three continents - Berlin, Dubai, and LA; including an Indian vocal sample sung by Malavika herself. She says “"Overdrive” is a song that showcases a different side of my personality. It's confident, sexual and empowering. It's about taking control, reversing gender stereotypes and expressing yourself unabashedly. But at the same time it's a sexy R&B love song that is in an ode to your person.” Malavika's intoxicating vocals are on full display, as she initiates romance with her lover over a pulsing beat, with natural confidence and sultry playfulness.Following the organic success of the only other two singles to her name, “Don't Let Go” and “Sugarcoat,” “Overdrive” was created with all the elements that Malavika is most inspired by. She expands “Vocally I wanted to go in on the song in terms of the harmonies, layers and adlibs. It's stacked and is reminiscent of early 2000s R&B, one of many eras where the ad libs really mattered. I have this moment in the second verse where I wanted to have a Queen inspired harmony moment.” With West Coast legend DJ Quik, as her mentor, “Overdrive” showcases a combination of Malavika's confident writing ability, commanding vocals, and musical influences. Alongside songwriting, Malavika has also found herself an advocate for infiltrating her culture into everything she creates. She expands; “The influences in my music are unique to my experience and upbringing, including the exposure to multilingual music. I try to incorporate sonic and visual elements of my culture whenever I can in a way that's natural and organic, because it's a part of my identity.”Born and raised in India, Malavika began to display a profound love for the performing arts from her earliest years, and began performing at just 3 years old. Being exposed to various contrasting worlds and cultures throughout her childhood has had a transformative role in Malavika's development as an artist. Imprinted with her parents' love for music, Malavika grew up idolizing Whitney Houston, Michael Jackson, Stevie Wonder and Tina Turner, alongside Indian masters like A.R. Rahman - not to mention the harder-hitting hip-hop of Jay-Z, Missy Elliott, Kanye West and Drake. Abstract: Imprinted genes are subject to germline epigenetic modification resulting in parental-specific allelic silencing. Although genomic imprinting is thought to be important for maternal behaviour, this idea is based on serendipitous findings from a small number of imprinted genes. Here, we undertook an unbiased systems biology approach, taking advantage of the recent delineation of specific neuronal populations responsible for controlling parental care, to test whether imprinted genes significantly converge to regulate parenting behaviour. Using single-cell RNA sequencing datasets, we identified a specific enrichment of imprinted gene expression in a recognised 'parenting hub', the galanin-expressing neurons of the preoptic area. We tested the validity of linking enriched expression in these neurons to function by focusing on MAGE family member L2 (Magel2), an imprinted gene not previously linked to parenting behaviour. We confirmed expression of Magel2 in the preoptic area galanin expressing neurons. We then examined the parenting behaviour of Magel2+/- null mice. Magel2-null mothers, fathers and virgin females demonstrated deficits in pup retrieval, nest building and pup-directed motivation, identifying a central role for this gene in parenting. Finally, we show that Magel2-null mothers and fathers have a significant reduction in POA galanin expressing cells, which in turn contributes to a reduced c-Fos response in the POA upon exposure to pups. Our findings identify a novel imprinted gene that impacts parenting behaviour and, moreover, demonstrates the utility of using single-cell RNA sequencing data to predict gene function from expression and in doing so here, have identified a purposeful role for genomic imprinting in mediating parental behaviour. Now that same day two of them were going to a village called Emmaus, about seven miles from Jerusalem. They were talking with each other about everything that had happened. As they talked and discussed these things with each other, Jesus himself came up and walked along with them; but they were kept from recognizing him (Luke 24:13-16). Apart for some of Jesus' parables, this story is one of the more beloved episodes of our Lord's life. Treasured, not because of its suspense, but its space for meditation. It's a slow story, simple, profound, and rather poignant. Two of Jesus disciples (many suspect that the second was Cleopas' wife) have left Jerusalem for home. They may have entered the city a week ago along with Jesus on that donkey, with high hopes of a turning point in Israel's fortune. Israel's long-awaited Messiah had arrived. But everything has disintegrated. If Jesus had been the one to redeem Israel, he should have been defeating the pagans, not dying at their hands! Its likely that they are on their way home, wondering how to start life over. They had pinned everything on Jesus. Now he was dead. He last words, “It is finished” still rang in their ears. Imprinted on their memories, that last breath, along with the image of his lifeless body hanging limp on the cross. That was Friday. Now its late Sunday. A story began circulating early in the day that he was alive. Some of their friends had seen him. But that is crazy. Dead people just don't come back to life. Yes, of course, Lazarus had, but that was Jesus' doing. Now he is dead. Who is there to bring him back to life? It is all too much. They left the city and, on the way, went over events again and again, trying to make sense of it all. Why had their expectations of the Messiah come to such a tragic end? And what were they to do with this story that he was alive again? They could not piece it together. Too many questions and too few answers. As they talk, Jesus joins them though they do not realize it is him. Their eyes are “kept from recognizing him.” And here we pause and ask ‘why'? Why don't they recognize him? Some have postulated that Jesus was somehow different from before, already a little glorious. Maybe it's a case of “We see what we expect to see”. Their conversation has been heavy with sorrow, disappointment, and bewilderment; their hearts are so full with pain, they don't really see the stranger who has joined them. They don't expect him to be alive, so they can't see him when he joins them. Or maybe God just closed their eyes for a while. Luke doesn't tell us. An experience with the living God was what they wanted; but though he came to them, and walked alongside them, their hearts were too full of brokenness to notice him right there with them. Let's pause the story here for today. As disciples of Jesus, we may find ourselves in the very same place. We live with dashed hopes and dreams. Life is full of disappointments. We may even be disappointed with God. And yet we long for God. If you're in such a space, take time to sit there today. Now notice that Jesus joins them in their walk home. He is there with them, even though they do not recognize him. It is Psalm 23 in action. We can be sure that Jesus walks with us too. Like these two disciples, we may not notice at first. We assume God will show up in certain ways, so when he appears differently, we don't see him, at least, not at first. Often, God is not like we expect him to be, he is like himself. And that is good.

When I clean my house, I only use all-natural cleaners. Now, before you start thinking how earth-conscious I am (although that is a great reason to use natural cleaners), there's a deeper reason. When I was about four years old, my baby sister got into a cabinet and put a cap of the skull-and-crossbones kind of cleaner into her mouth. Even though I was young, I clearly remember riding in my dad's car behind the ambulance and praying, “God, please don't let my sissy die!” Thankfully, my sister is just fine today. But this experience left an imprint on my soul: a fear of cleaning chemicals. For years, I wondered why I had anxiety when I helped someone clean their house with different products. When my kids were babies, I was super-vigilant and called the poison control number several times, none of which resulted in anything dangerous. Now, it makes sense. We all have habits that we default to, even ones that may not seem rational. Sometimes, we can look back in our past and find the “trigger” where these behaviors started to form, while others may be buried deep in our souls and take some digging to uncover. Why does our younger sibling's whining set us on edge? Why do we have a compulsion to make sure our hair is just perfect? Why do we have extreme anxiety when we are running late? Whatever your “default” is, there may be an underlying reason lurking in your past. This can start to feel overwhelming, but there is so much hope. If you are God's child, you are imprinted in a different way. You have His imprint on your heart and soul, reminding you that you are His forever (John 10:28-29; Ephesians 1:13; 1 John 3:1). God promises that in Christ, you are a new creation (2 Corinthians 5:17). He will complete the work He began in you until the day He comes again (Philippians 1:6). You have the power of the Holy Spirit, who comforts you, teaches you, and reminds you of everything Jesus has said (John 14:26). You can tell your heart that you are not just a product of your past. You are imprinted by the God of the universe who made you in His image (Genesis 1:27). • Savannah Coleman • How can we extend patience to others when they get stressed by things that don't make sense to us? • Have you ever had a knee-jerk reaction of anxiety, anger, fear, etc.? It feels so frustrating when you don't know the cause behind your reaction. And sometimes, it's even more frustrating when you do know the reason for your “trigger,” but you can't seem to change your reaction. The scars from our past or the pain we have buried deep within us can leave us feeling helpless. But in Jesus, we have hope that He can heal our brokenness. And He can do that through His Spirit living in us, the truth found in His Word, and the love of His people. He has also equipped counselors and therapists to help people heal from past hurts. A Mermaid is a mythical aquatic creature with the upper body of a female human and the tail of a fish.  Legends of these creatures have been reported since the beginning of humanity. There is no evidence that mermaids live anywhere on earth. However, there are a lot of scientific reasons why they physically cannot exist. Seriously, fish and human's last common ancestor was over 170 million years ago. If you're going to say mermaids are mammals, they'd have to breathe air, which means going to the surface often. (So lots of chances for humans to spot them).  AND even then, a human/water mammal would not have had time to evolve. Whales returned to the ocean 50 million years ago, and humans evolved 300,000 years ago…. So there is just not enough time for evolution to take place.But, there is a lot of interesting folklore surrounding these creatures. Welcome to Showcasing House Music DJ Mixes by dattrax!!                                                  Just love the 1st instrumental track by Silicone Soul. The feel reminds me of Fingers Inc.'s Mystery of Love Instrumental. Just stomp your feet SICK! I found it in the 'electronica top 100 downloads' at Beatport.com. You have to look everywhere, don't let other people's labeling limit or confuse you. When you hear house, you'll recognise it.The first track is the most important in the mix. It sets the mood. There are few house tracks with nice intros and even if there are, you mix through them, so beginning of mixes are fun to highlight a favourite. You don't want to pick one with a 3min or more intro because this will turn people off from relistening to the mix.Next you EQ it the way that you like tracks to generally sound. Too bad there is no EQ, Volume or Compression Standard. Every song is made so differently, which is a good thing to have such a variety of house music to choose from. I prefer less bass and more hi's. When the bass is farting/ distorting and not a tight punchy kick, then it needs to be lowered. Jim's the opposite, he loves bass! The track sounds flat (not vibrant) if there is too much bass and not enough hi's.Some tracks have too much compression so the hi's stab your ear in a bad way, so you have to lower it. Some tracks with vocals, congos, guitars, or washes that may have the mid's too high and that produces a result as annoying as nails on a chalkboard, so you have to lower the mid's.Jim: Some tracks are produced and mastered too well relative to most house tracks and the volume is much greater and you have to know/ remember to compensate so that you have even levels.Next you pick the speed. We prefer mixes at 126bpms, not more than 128 (sometimes you get out of control when not paying attention). Can't believe that we grew up listening and dancing to house at 123bpms or less. Everything in electronic music keeps going faster and more aggressive over the years. Most of the 'deep house' that is coming out (and for the last year 2009) has elements of electro, tech & minimal house. Welcome to Showcasing House Music DJ Mixes by dattrax!!                                                 For me house music is an escape vehicle from the things that don't excite me about my life or stresses me out, so in those pockets of time that I spend related to house/ DJing, I enjoy it that much more. House Music is therapeutic in that it relaxes me. Other guys love sports, or are zoned out to TV. I don't care for that stuff.Imprinted, MixOne and Two are filled with tracks bought from Beatport.com. Techy, minimal, but still funky with fat house basslines and crisp high hats. Very little vocals on these two mixes aside from the dubby vocal washes and tracks with repetitive one liners.Or I could be wrong, less than usual, I should say. I love vocals laced into cool house instrumentals, because I don't believe you remember an hour of pounding tech and minimal house by itself. I am not criticizing, don't misunderstand. I love listening to top techhouse DJs like Alland Byallo and have a ton of tech and minimal house.But if they were layered with hot vocals, then the tech and minimal house will invigorate the vocals, making them that much better and exciting, and the vocals will imprint a positive memory of the mix into the listener's minds. That's why you can play one bar of a classic house track and the HouseHeadz will start singing. HOUSE KARAOKE.No one remembers a instrumental. People don't ask DJs: "What's that song that goes BOOM, BOOM, BOOM, WAHHH, DUCKP, UMPH???" They don't ask because they don't care.I love the feel of this mix. Charging and aggressive without being annoying/ irritating. And they stripped him and put a scarlet robe on him, and twisting together a crown of thorns, they put it on his head and put a reed in his right hand. And kneeling before him, they mocked him, saying, "Hail, King of the Jews!" And they spit on him and took the reed and struck him on the head. And when they had mocked him, they stripped him of the robe and put his own clothes on him and led him away to crucify him. (Matthew 27:27-31)'' No one likes to be mocked. The shame that comes with the mockery also causes us not to want to mock anyone. However, that thinking goes out the window when it comes to mocking God. The soldiers unknowingly mocked God with their mockery of Jesus. However, they were fully aware that their religious habits and temple worship were mockeries of the worship that happened in Jerusalem. In fact, all religion is a mockery of the Christian faith. Every religion in the world mocks God by making Him in human image. Since God is in man's image, He does basically whatever we want. Whatever we think is right and good is what He considers right and good. This is why the Greek and Roman gods and goddesses were so ... ungodly. But the true God created man in His image. Imprinted on us the true values of what is good and right. Many of these good and right are counter-cultural. Just like salvation through God's death. God the Son had to sacrifice Himself on the cross so that you might be saved. Mocking God is no small crime. But God offers forgiveness to those who submit to His image. Revealed to us in His Word. Clothing us with salvation just as He was stripped for our salvation. Amen.

Abstract: Over 200 murine imprinted genes have currently been identified, many are expressed in the brain, and a subset have been associated with neurological and behavioural phenotypes. These studies have led to the suggestion that imprinted genes show enriched expression in certain key brain functions; however, this has never been formally tested. Here we use fifteen available single-cell RNA sequencing datasets to investigate imprinted gene over-representation in the brain, with the aim of identifying key hotspots of expression and, by extension, function. We establish that imprinted genes are indeed over-represented in the adult brain compared to other adult tissues, and in neurons specifically compared to other cell-types. Brain wide datasets allowed us to demonstrate enrichment of imprinted genes in the hypothalamus, ventral midbrain, pons and medulla. Finally, using scRNA-seq datasets focusing on these regions of enrichment, we were able to identify hypothalamic neuroendocrine populations and the monoaminergic hindbrain neurons as specific hotspots of imprinted gene expression. These analyses provide a robust assessment of the neural systems on which imprinted genes converge and in turn suggest the neuronal regulation of motivated behaviours such as feeding, parental behaviour and sleep as functional hotspots for imprinting. Abstract: Mammalian genomes include many maternally or paternally imprinted genes. Most of these are also expressed in the brain and several were previously implicated in regulating specific behavioral traits. We have used here a knockout approach to study the function of Peg13. Peg13 codes for a fast evolving lncRNA and is part of a complex of imprinted genes on chromosome 15 in the mouse and chromosome 8 in humans. Two knockout constructs were analyzed, one with a full deletion of the gene, the other with a deletion of the three prime-half. The full deletion is semi lethal, while the partial deletion is fully viable, but the mice show distinctive behavioral differences. They lose interest in the opposite sex and show instead a preference for wildtype animals of the same sex. Further, they show a higher level of anxiety, as well as lowered activity and curiosity and females have a deficiency in pup retrieval behavior. Brain RNA expression analysis reveals that genes involved in the serotonergic system, formation of glutamergic synapses, olfactory processing and estrogen signaling, as well as more than half of the other known imprinted genes are affected in Peg13 deficient mice. Intriguingly, the pathways are differentially affected in the sexes, with the result that the male and female brains of Peg13 deficient mice differ more from each other than those of wildtype mice. Guests :  Othembele Dyantyi What are you doing for the earth in this time of crisis? Our planet is facing an environmental emergency which authorities are doing little to halt. Many of these disasters, such as deforestation and the burning of fossil fuels, are the result of processes generated by major corporations. They’re issues that can be dealt with. Yet the situation gets worse every day. Eleven-year-old Othembele Dyantyi is raising her voice to encourage a union of action against this unprecedented climate injustice. Dyantyi is part of Extinction Rebellion, a global socio-political movement to drive government action to prevent the planet’s breakdown. At the forefront of the South African resistance, Dyantyi and her army of youth activists are spreading awareness about the causes and severity of the climate crisis. Together, they’re calling for the protection of endangered ecological systems and challenging the government to invest in renewable energy. Julie Kynaston Julie Kynaston’s children didn’t grow in her belly. She calls herself a ‘heart mama’ – a reference to where her love for them burgeoned. When Kynaston and her husband, Ryan, decided to start a family, they were determined to adopt. But during the process, they encountered a distinct lack of information online. It didn’t deter Kynaston from making the fulfilling choice. Now, she’s using her words to close the gaps and help other South Africans. With Heart Mama Blog, Kynaston speaks openly about the joys and difficulties of having an adoptive family and being a parent. Here, people can share their stories of what it’s like to adopt, or to have been adopted. Adoption, especially multi-racial adoption, comes with a slew of challenges. Her site includes useful resources for those interested in adopting within South Africa. As Kynaston creates a safe space to educate, she’s changing perceptions. For many children, adoption can be rooted in trauma or loss. Her own family is testament to the love that comes with adoption. With her kids Ilan, Kira and Judah, they show there’s no one way of being a family. Moerieda Joubert Moerieda Joubert’s mother understood the importance of connecting with people. And what better way to do that than by sharing a meal? Even though she had Parkinson’s, she would prepare food for the local old-age home out of her own pocket. After her passing, Joubert has proved the apple doesn’t fall far from the tree. Imprinted with a sense of goodwill, Joubert founded Statice Square Senior in Cape Town to build a community from companionship and love. For the past 15 years, she’s been providing scrumptious five-course meals and organising entertainment for the elderly. A pensioner herself, Joubert uses her limited funds to continue her mother’s legacy. Going beyond her means, Joubert is nourishing people with kindness and creating priceless opportunities to break bread with others. Patrick Seruwu Streaks of colour drip down the canvas as the image of a woman begins to take shape. Patrick Seruwu alternates between slathers and dabs of acrylic paint. Despite the delicate details, his portraits are emotionally-charged and rooted in a heavy reality. Representative of women who’ve been abused and fear speaking out, the artworks convey their trauma. Before the Ugandan-born, Johannesburg-based contemporary artist began painting, he used to braid women’s hair for a living. This, combined with his upbringing alongside three sisters and a single mother, influenced Seruwu thematically. To fairly depict their experiences, he bases his paintings on conversations where women dictate their own stories. One of the questions Seruwu often asks, “Are you safe at home?” reveals an unfortunate reality – many women aren’t. His multimedia works serve as a powerful statement. Though he’s only been painting for two years, Seruwu’s pieces have exhibited at the Turbine Art Fair and the ABSA Gallery in Johannesburg. With his art, he’s furthering the conversation on gender-based violence, reminding people of women’s struggles, and celebrating their resilience. Fanie Viljoen Your body is yours. This is an extreme violation, the effects of which can last a lifetime. In an attempt to prevent this exploitation, Fanie Viljoen has written a book that’s educating children and adults in the face of violence. In 2019, Viljoen published The Day the Dragon Came, a children’s story about sexual abuse in the home committed by a ‘dragon’. He has written different versions of the book for boys and girls, which include illustrations by Tumi K. Steyn. Viljoen explains in simple to understand, child-friendly ways the difference between safe and unsafe touch, and what to do if you’re being harmed. The books also contain information for parents, guardians, and teachers on how to recognise abuse and take appropriate measures. “Some things are uncomfortable to speak about but that doesn’t mean they shouldn’t be addressed,” Viljoen says. Teaching children about consent can protect them from dragons that come their way, even in adulthood. But it’s listening to kids and taking action that ends the cycle of abuse.

Artists Imani Sims, Tani Ikeda, and Jordan Alam share a passion in creating work relating to the ways personal experiences are held in the body. We’re thrilled to welcome them to present their narratives through film, prose, and poetry explorations of the events of their lives that have impacted their bodies, and how those changes have affected their encounters with the world. First we hear from local poet and author Imani Sims, who has been hailed as a “cultural ambassador, leader and powerhouse in our community” (Priya Frank, Seattle Art Museum). Then Jordan Alam, Town Hall’s Inside/Out Resident representing Hillman City and Columbia City, takes the stage to share her prose centered on the topics that inspired her to curate this event. And Emmy-winning director Tami Ikeda presents a screening of her film highlighting the resilience of survivors. Join these three innovative artists as they engage different mediums to tell us the stories of their life experiences that are imprinted in their bodies. The insulin-like growth factor 2 receptor (IGF2R) is essential for prenatal growth regulation and shows gene dosage effects on fetal weight that can be affected by in-vitro embryo culture. Imprinted maternal expression of murine Igf2r is well documented for all fetal tissues excluding brain, but polymorphic imprinting and biallelic expression were reported for IGF2R in human. These differences have been attributed to evolutionary changes correlated with specific reproductive strategies. However, data from species suitable for testing this hypothesis are lacking. The domestic cow (Bos taurus) carries a single conceptus with a similar gestation length as human. We identified 12 heterozygous concepti informative for imprinting studies among 68 Bos taurus fetuses at Day 80 of gestation (28% term) and found predominantly maternal IGF2R expression in all fetal tissues but brain, which escapes imprinting. Inter-individual variation in allelic expression bias, i.e. expression of the repressed paternal allele relative to the maternal allele, ranged from 4.6-8.9% in heart, 4.3-10.2% in kidney, 6.1-11.2% in liver, 4.6-15.8% in lung and 3.2-12.2% in skeletal muscle. Stochastic, environmentally and/or genetically induced disturbances in the genome-wide epigenetic reprogramming processes during male germ-cell development may contribute to male infertility. To test this hypothesis, we have studied the methylation levels of 2 paternally (H19 and GTL2) and 5 maternally methylated (LIT1, MEST, NESPAS, PEG3, and SNRPN) imprinted genes, as well as of ALU and LINE1 repetitive elements in 141 sperm samples, which were used for assisted reproductive technologies (ART), including 106 couples with strictly male-factor or combined male and female infertility and 28 couples with strictly female-factor infertility. Aberrant methylation imprints showed a significant association with abnormal semen parameters, but did not seem to influence ART outcome. Repeat methylation also differed significantly between sperm samples from infertile and presumably fertile males. However, in contrast to imprinted genes, ALU methylation had a significant impact on pregnancy and live-birth rate in couples with male-factor or combined infertility. ALU methylation was significantly high-er in sperm samples leading to pregnancy and live-birth than in those that did not. dattrax: Just love the 1st instrumental track by Silicone Soul. The feel reminds me of Fingers Inc.'s Mystery of Love Instrumental. Just stomp your feet SICK! I found it in the 'electronica top 100 downloads' at Beatport.com. You have to look everywhere, don't let other people's labeling limit or confuse you. When you hear house, you'll recognise it. The first track is the most important in the mix. It sets the mood. There are few house tracks with nice intros and even if there are, you mix through them, so beginning of mixes are fun to highlight a favourite. You don't want to pick one with a 3min or more intro because this will turn people off from relistening to the mix. Next you EQ it the way that you like tracks to generally sound. Too bad there is no EQ, Volume or Compression Standard. Every song is made so differently, which is a good thing to have such a variety of house music to choose from. I prefer less bass and more hi's. When the bass is farting/ distorting and not a tight punchy kick, then it needs to be lowered. Jim's the opposite, he loves bass! The track sounds flat (not vibrant) if there is too much bass and not enough hi's. Some tracks have too much compression so the hi's stab your ear in a bad way, so you have to lower it. Some tracks with vocals, congos, guitars, or washes that may have the mid's too high and that produces a result as annoying as nails on a chalkboard, so you have to lower the mid's. Jim: Some tracks are produced and mastered too well relative to most house tracks and the volume is much greater and you have to know/ remember to compensate so that you have even levels. Next you pick the speed. We prefer mixes at 126bpms, not more than 128 (sometimes you get out of control when not paying attention). Can't believe that we grew up listening and dancing to house at 123bpms or less. Everything in electronic music keeps going faster and more aggressive over the years. Most of the 'deep house' that is coming out (and for the last year 2009) has elements of electro, tech & minimal house. A lot less organic sounding. dattrax: For me house music is an escape vehicle from the things that don't excite me about my life or stresses me out, so in those pockets of time that I spend related to house/ DJing, I enjoy it that much more. House Music is therapeutic in that it relaxes me. Other guys love sports, or are zoned out to TV. I don't care for that stuff. Imprinted, MixOne and Two are filled with tracks bought from Beatport.com. Techy, minimal, but still funky with fat house basslines and crisp high hats. Very little vocals on these two mixes aside from the dubby vocal washes and tracks with repetitive one liners. Or I could be wrong, less than usual, I should say. I love vocals laced into cool house instrumentals, because I don't believe you remember an hour of pounding tech and minimal house by itself. I am not criticizing, don't misunderstand. I love listening to top techhouse DJs like Alland Byallo and have a ton of tech and minimal house. But if they were layered with hot vocals, then the tech and minimal house will invigorate the vocals, making them that much better and exciting, and the vocals will imprint a positive memory of the mix into the listener's minds. That's why you can play one bar of a classic house track and the HouseHeadz will start singing. HOUSE KARAOKE. No one remembers a instrumental. People don't ask DJs: "What's that song that goes BOOM, BOOM, BOOM, WAHHH, DUCKP, UMPH???" They don't ask because they don't care. I love the feel of this mix.
