Repurposing Disulfiram to Overcome Chemotherapy Resistance in Relapsed Sarcomas

Matteo Trucco, MD and Kurt Weiss, MD joins us on OsteoBites to describe the collaboration that led to the current Phase 1 clinical trial repurposing the alcoholism drug disulfiram to see if it can overcome the chemotherapy resistance seen in relapsed sarcomas, including osteosarcoma. Matteo Trucco is a Pediatric Oncologist and the Clinical Director of Pediatric Hematology/Oncology at Cleveland Clinic Children's, specializing in the care of children, teenagers and young adults battling bone and soft tissue cancers. He also directs the Children's Cancer Innovative Therapy Program where he and colleagues design, develop and conduct clinical trials seeking more effective and less toxic treatments for childhood cancers. Dr. Trucco earned his undergraduate degree from the University of Pennsylvania and Medical Degree from Temple University School of Medicine. He completed his Pediatrics Residency at Rainbow Babies and Children's Hospital and his Pediatric Hematology/Oncology Fellowship from Johns Hopkins and the National Cancer Institute. In addition to his roles at the Cleveland Clinic, he is a Co-chair of the National Pediatric Cancer Foundation's Sunshine Project consortium, partnering with other top pediatric cancer centers to develop clinical trials. He is honored to be on the Board of MIB Agents, chairs its Scientific Advisory Board and co-chairs the organizing committee for the FACTOR Conference. He also has the privilege of moderating the MIB Agents TURBO Tumor Review Board for Osteosarcoma. Dr. Weiss directs the Department's Musculoskeletal Oncology Laboratory, a basic science laboratory dedicated to the study of sarcomas. His mission is to help develop a world-class translational sarcoma research program at the University of Pittsburgh. As a bone cancer survivor himself, Dr. Weiss brings passion and enthusiasm to the laboratory, clinic, and operating room. Through the UPMC Hillman Cancer Center, he is also a proud collaborator with scientists who are trying to understand how other forms of cancer spread to and destroy the bone. Dr. Weiss is a Founding Member of the Musculoskeletal Oncology Research Initiative (MORI), Pittsburgh Cure Sarcoma (PCS), the Pittsburgh Sarcoma Research Collaborative (PSaRC), and the Pittsburgh Center for Bone and Mineral Research (PCBMR). He is a peer reviewer for multiple journals including the Journal of the American Academy of Orthopaedic Surgeons, BioMed Central Cancer, Sarcoma, Cancer Research, International Journal of Cancer, and others. He is a former member of the NIH's Center for Scientific Review Early Career Reviewer program. He has served on multiple National Cancer Institute Study Sections. Keelan attended the University of Pittsburgh completing a Master of Physical Therapy and a Master of Science Degree in Health and Rehabilitation Science. He completed the Sports Physical Therapy Residency Program through the University of Pittsburgh and Centers for Rehab Services/University of Pittsburgh Center for Sports Medicine. Keelan is also a certified athletic trainer (ATC), certified orthopaedic (OCS) & sports (SCS) physical therapy specialist. He is currently serving as the Director of Clinical Practice Innovation, Administrative Director of Physical Therapy Residency Programs, and Orthopaedic Physical Therapy Residency Program Director at the Centers for Rehab Services/UPMC Center for Sports Medicine Clinic as well as an adjunct professor for the University of Pittsburgh Departments of Physical Therapy and Sports Medicine & Nutrition. Keelan's time is divided time between clinical practice, residency administration, project consultation, & professional development responsibilities. His areas of clinical interest include determinants of outcomes and treatment options available for active individuals with pathological conditions of the hip joint. He regularly presents nationally & internationally on these topics. Keelan has been an author including peer-reviewed articles for the Journal of Orthopaedic and Sports Physical Therapy, Clinics in Sports Medicine, Archives of Orthopedic Trauma Surgery, Current Reviews in Musculoskeletal Medicine, Operative Techniques in Orthopaedics, The Physician and Sportsmedicine, Journal of Bodywork and Movement, and Sports Health. He has served as a content reviewer for the American Journal of Sports Medicine, the Journal of Orthopaedic and Sports Physical Therapy, Journal of Sport Rehabilitation, Journal of Athletic Training, Journal of Manipulative and Physiological Therapeutics, Journal of Hip Preservation, PM&R and Physical Therapy in Sport, Journal of Sports Rehabilitation, Sports Health, guest editor for Topics in Geriatric Rehabilitation, and is an associate editor for the International Journal of Sports Physical Therapy. Additionally, he has co-authored book chapters & continuing education manuscripts covering the topics of rehabilitation of hip and knee injuries. APCOR was founded in 1956 and establishes cork production and manufacturing standards, promotes Research & Development, and educates both professionals and consumers. Cork is a sustainable and regenerative material for bottle closures, for example a Life Cycle Analysis by PwC found that a plastic closure emits 10 times more CO2 than a cork stopper and aluminum cap closures emit 24 times more CO2 than a cork stopper. APCOR has approximately 270 members who represent more than 85% of cork exports from Portugal. Griffith Foods has been bringing food science into the food industry for over 100 years to improve safety and nutrition. As of 2020, they held an Ecovadis Silver certification with just over 60% of their suppliers being Ecovadis bronze certified or better. Griffith reduced manufacturing waste to landfill by 41% from 2014 to 2020 with 13 facilities sending zero waste to landfill. She earned her MFA in Ceramics at the State University of New York at New Paltz. Grants and fellowships include the Haystack Mountain School of Crafts Technical Assistant, Andrah Scholarship, Research and Creative Projects Award, Bolton Scholarship, and the Virginia Fuller Prize.  She has exhibited at the Dorsky Gallery Curatorial Programs, The Samuel Dorsky Museum of Art, Williamsburg Arts and Historical Center, and Pittsburgh Center for the Arts.   Raised by oceanographers across the street from an intertidal region of Long Island Sound, the marine ecosystem played a powerful role throughout her childhood and continues to inform her practice through visual inspiration and systems of organization. Her work focuses on the physicality of the landscape. In her practice, the ceramic process represents a transformation of natural materials into objects of culture and records of time. When we hear the word research, most of us think of innovative breakthrough treatments and technologies. But applied clinical research could not be possible without another type of research called basic science research in which scientists study the fundamental processes of the human body. In this episode, we'll learn what basic science research is and how it applies to the area of kidneys and kidney disease.   In this episode, we spoke with: Hannah Wesselman Hannah Wesselman is a PhD candidate at the University of Notre Dame studying the genetic mechanisms underlying kidney development in Dr. Rebecca Wingert's lab. Prior to her dissertation work, Hannah worked with Dr. Larissa Myaskovsky at the University of Pittsburgh Center for Research on Health Care, and their collaboration continued as Dr. Myaskovsky moved to the University of New Mexico Center for Healthcare Equity in Kidney Disease. During this time, Hannah worked directly with kidney transplant patients, and later published in CJASN on social determinants and race disparities in kidney transplant. Inspired by the strength of the kidney community, Hannah has returned to bench science in the hopes of identifying new treatment targets and has continued to connect with kidney patients through NKF advocacy. Holly Kramer, M.D., MPH Holly Kramer, M.D., MPH, is a practicing nephrologist who conducts research connecting nutrition and kidney health. Her connection to the National Kidney Foundation was inspired by her mom, who was a dialysis nurse and helped create some of the first dialysis units in Northwest Indiana. Dr. Kramer finds being on the NKF Board important, because it is the largest, patient-centered organization focusing on kidney disease. Her long-term goal is to increase national funding for kidney disease research and to heighten awareness about chronic kidney disease. Joseph V. Bonventre, M.D., PhD Dr. Bonventre is the Samuel A. Levine Distinguished Professor of Medicine and Constantine L. Hampers Distinguished Chair at Harvard Medical School and Professor of Health Sciences and Technology at the Massachusetts Institute of Technology. He is Chief of the Renal Division and Founding Chief of the Engineering in Medicine Division of the Brigham and Women's Hospital. In addition to his B.S. with distinction in Engineering Physics from Cornell, Dr. Bonventre holds M.D. and Ph.D. degrees in Biophysics from Harvard University.  He has honorary doctorate degrees from Mt. Saint Mary's College and from the Norwegian Institute of Science and Technology in Norway. In this episode, Alicia host an interview with figurative artist, scenic painter, and independent curator Heather Heitzenrater. They discuss the artist's ongoing series of work centered around reflective surfaces and the human form, her experiences working as a scenic painter, and other highlights in her career thus far.  Heather Heitzenrater was born in 1992 and grew up in Punxsutawney, PA.  She now lives with her husband and their two cats in Pittsburgh, PA. Heather received her BFA in Painting and Drawing from Edinboro University of Pennsylvania in 2015. Her work has been shown Nationally including Erie Art Museum, Westmoreland Museum of American Art, and Baton Rouge Gallery. She has been published in Poet Artist Magazine, Pikchur Magazine and Minus 37. Heather has won awards, given artist talks, curated and juried local art shows.  In addition, she is as an active member of The Associated Artists of Pittsburgh. Along with showing her work, Heather instructs figurative workshops at Pittsburgh Center of the Arts and is a Scenic Artist for ScareHouse.  Heather and her husband, Christopher Boring, share a space at the Radiant Hall Susquehanna location.

Njaimeh Njie is a multimedia storyteller. From her desires to make a featured documentary to learning how to make a delicious Sunday dinner, Njaimeh shows us no goal is too unreachable or too small. Her photography, video, oral history, writing, and public artwork explores contemporary Black experiences, with a particular focus on how the past shapes the present. Njie's work has been featured in outlets including CityLab and Belt Magazine, exhibited in spaces including the Carnegie Museum of Art and The Mattress Factory Museum, and she has presented at venues including TEDxPittsburghWomen and Harvard University. Among several awards and grants, Njie was named the 2019 Visual Artist of the Year by the Pittsburgh City Paper, and the 2018 Emerging Artist of the Year by the Pittsburgh Center for the Arts. Newly elected PA State Representative Jessica Benham joins Montel on this episode of Free Thinking. Rep Benham is a disability rights activist serving as a member-elect of the Pennsylvania House of Representatives for the 36th District, located in Western Pennsylvania. She is the first openly LGBTQ+ woman and first openly autistic person elected to the Pennsylvania General Assembly. She is a union organizer, community leader, advocate, and role model. She co-founded the Pittsburgh Center for Autistic Advocacy.
