In recent days, the OS team has been reaching out to people on both sides who have lost loved ones in the war; inviting them to tell the stories of those killed. Next week's programme will feature Israeli families. This edition, hosted by James Reynolds, is a conversation with two Palestinians who now live in Scotland and Turkey. Yousef Almqayyad in Istanbul, had to have a heartbreaking discussion with his seven-year-old daughter about the deaths in his family. “Your grandfather, your grandmother, your uncles and aunts and your cousins, right now are in heaven, in a good place,” he said. “Better than Gaza, better than Turkey, better than any place in this world. I told her they are waiting for us.” Dr Ibrahim Khadra also shares his final conversation with a member of his family in Gaza, who said: “If we'll survive, we'll pray to God and if I'll die just pray for me.” “It was our last call,” said Dr Khadra. You can find remarkable people in every corner of this planet. Many of these people have something in common. There is a spirituality about them. They seem to be motivated by the call of something greater. They seem to live their lives in the context of a bigger story. A story that precedes them, and will go on after them. They are on a path of awakening to wisdom. I met Khadra Ali in 2014. I've always been struck by the light that radiates from her face. I have also been intrigued by how she keeps traditional, religious, ways of Islam and remains so open to so much that fits outside of her tradition. I have to brag and say that Khadra was part of the original, FIRST, cohort of the Evolutionary Leadership Workshop. It is an honor and a blessing to count her among our accomplished alumni. Khadra is doing amazing things. She works with the Youth and Innovation Team at the United Nations Population Fund, and she is based in Mogadishu. She is also dreaming up an organization called “Toos,” which means: Wake Up! Khadra is part of the passionate generation of migrants, who came West as children and returned to their homelands as adults. They share the privileges they've been afforded just by living in the rich world. Khadra shares the story of leaving her motherland, and of the advice her grandmother gave her: stay true to your religion. It is a powerful charge from one's elder. And Khadra has stayed true to her grandmother's direction. While also finding ways to dive beneath what is traditional. Her authentic relationship with God keeps her tracking for the pathways to awakening. Religions of each and every ilk have managed to corrupt themselves into systems of myopia, self-righteousness, and oppression. But they still come from wisdom traditions. You can find wisdom in ancient stories. You can find healing in time-tested practices. In prayer, contemplation, meditation, acts of collective worship, and adoration. These are patterns that we find through traditions that have stood the test of time. Getting wise is not an option. It is a necessity. It is how we become good ancestors. It is how we learn to contend with life itself. It feels to me that this is what Khadra is aiming for.  Get to know her. She is a soul worth knowing. Get a sense of her light.

Abstract: Parvalbumin-expressing interneurons (PVINs) play a crucial role in within the dorsal horn of the spinal cord in by preventing touch inputs from activating pain circuits. After nerve injury, their output is decreased via mechanisms that are not fully understood. In this tudy, Wwe show that PVINs from nerve-injured mice change their firing pattern from tonic to adaptive. To examine the ionic mechanisms responsible for this decreased output, we employed a reparametrized Hodgkin-Huxley (HH) type model of PVINs, which predicted (1) the firing pattern transition is due to an increased contribution of small conductance calcium-activated potassium (SK) channels, enabled by (2) impairment in intracellular calcium buffering systems. Analyzing the dynamics of the HH-type model further demonstrated that a generalized Hopf bifurcation differentiates the two types of state transitions observed in the transient firing of PVINs. Importantly, this predicted mechanism holds true when we embed the PVINs model within the neuronal circuit model of the spinal dorsal horn. To experimentally validate this hypothesized mechanism, we used pharmacological modulators of SK channels and demonstrated that (1) tonic firing PVINs from naive mice become adaptive when exposed to an SK channel activator, and (2) adapting PVINs from nerve-injured mice return to tonic firing upon SK channel blockade. Our work provides important insights into the cellular mechanism underlying the decreased output of PVINs in the spinal dorsal horn after nerve injury and highlights potential pharmacological targets for new and effective treatment approaches to neuropathic pain. Today the boys discuss Blues' improved performance in the fourth round of the FA Cup away to Blackburn Rovers. You could argue we were the better team. We certainly were in the opening 10/15 minutes after former Rovers man, Reda Khadra, struck home an early lead, but we had to fight our way back after we conceded two fairly sloppy goals, in fairness. Jubilation was to be left late on though after sub, Jordan James, got the vital equaliser to keep Blues in the cup. The boys talk about whether Eustace should be giving him more game time, as well as why Khadra came off instead of Chong earlier in the second half, and the boys also read more into Eustace's interaction with the fans after the game too. Decoding the relative contributions of extrinsic and intrinsic mechanisms in mediating heterogeneous spiking activities of sensory neurons in vivo using computational modeling

Abstract: Neurons ubiquitously display heterogeneities in spiking activity even within a given cell type. To date, the relative contributions of extrinsic mechanisms (e.g., synaptic bombardment) and intrinsic mechanisms (e.g., conductances, cell morphology) towards determining spiking activity remain poorly understood. Here we address this important question using a novel approach that combines biophysical techniques, in which extracellular in vivo recordings of electrosensory pyramidal cells within weakly electric fish, are combined with computational modeling. Specifically, by varying parameters, a conductance-based computational model successfully reproduced the highly heterogeneous spiking activities seen experimentally. Model parameters that varied the most were then used to gauge the relative contributions of extrinsic vs. intrinsic mechanisms. Overall, extrinsic synaptic input was predicted to be the main factor accounting for spiking heterogeneities. We tested this prediction experimentally by performing two different manipulations: i) pharmacologically inactivating feedback; ii) applying the neuromodulator serotonin. Our model predicted that feedback inactivation should reduce while serotonin application should increase spiking heterogeneities. Experiments corroborated these predictions. Importantly, for serotonin application, increased heterogeneity occurred despite a strong reduction in intrinsic membrane conductance, further demonstrating that extrinsic synaptic input is the primary determinant of spiking heterogeneities in vivo. Taken together, our results demonstrate that devising a computational model to capture spiking heterogeneities in vivo and assessing which parameters are responsible can successfully determine the relative contributions of extrinsic vs. intrinsic inputs. Abstract: In mesenchymal cell motility, several migration patterns have been observed, including directional, exploratory and stationary. Two key members of the Rho-family of GTPases, Rac and Rho, along with an adaptor protein called paxillin, have been particularly implicated in the formation of such migration patterns and in regulating adhesion dynamics. Together, they form a key regulatory network that involves the mutual inhibition exerted by Rac and Rho on each other and the promotion of Rac activation by phosphorylated paxillin. Although this interaction is sufficient to generating wave-pinning that underscores cellular polarization comprised of cellular front (high active Rac) and back (high active Rho), it remains unclear how they interact collectively to induce other modes of migration detected in Chinese hamster Ovary (CHO-K1) cells. We previously developed a 6D reaction-diffusion model describing the interactions of these three proteins (in their active/phosphorylated and inactive/unphosphorylated forms) along with other auxiliary proteins, to decipher their role in generating wave-pinning. In this study, we explored, through computational modeling and image analysis, how differences in timescales within this molecular network can potentially produce the migration patterns in CHO-K1 cells and how switching between them could occur. To do so, the 6D model was reduced to an excitable 4D spatiotemporal model possessing three different timescales. The model produced not only wave-pinning in the presence of diffusion, but also mixed-mode oscillations (MMOs) and relaxation oscillations (ROs). Implementing the model using the Cellular Potts Model (CPM) produced outcomes in which protrusions in cell membrane changed Rac-Rho localization, resulting in membrane oscillations and fast directionality variations similar to those seen in CHO-K1 cells. The latter was assessed by comparing the migration patterns of CHO-K1 cells with CPM cells using four metrics: instantaneous cell speed, exponent of mean square-displacement (called -value), directionality ratio and protrusion rate. Variations in migration patterns induced by mutating paxillin's serine 273 residue was also captured by the model and detected by a machine classifier, revealing that this mutation alters the dynamics of the system from MMOs to ROs or nonoscillatory behaviour through variation in the concentration of an active form of an adhesion protein called p21-Activated Kinase 1 (PAK). These results thus suggest that MMOs and adhesion dynamics are the key ingredients underlying CHO-K1 cell motility. Copy rights belong to original authors. The mutual inhibition exerted by these two proteins on each other's activation and the promotion of Rac activation by an adaptor protein called paxillin have been implicated in driving cellular polarization comprised of front (high active Rac) and back (high active Rho) during cell migration. Mathematical modeling of this regulatory network has previously shown that bistability is responsible for generating a spatiotemporal pattern underscoring cellular polarity called wave-pinning when diffusion is included. We previously developed a 6D reaction-diffusion model of this network to decipher the role of Rac, Rho and paxillin (along with other auxiliary proteins) in generating wave-pinning. In this study, we simplify this model through a series of steps into an excitable 3D ODE model comprised of one fast variable (the scaled concentration of active Rac), one slow variable (the maximum paxillin phosphorylation rate - turned into a variable) and a very slow variable (a recovery rate - also turned into a variable). We then explore, through slow-fast analysis, how excitability is manifested by showing that the model can exhibit relaxation oscillations (ROs) as well as mixed-mode oscillations (MMOs) whose underlying dynamics are consistent with a delayed Hopf bifurcation. By reintroducing diffusion and the scaled concentration of inactive Rac into the model, we obtain a 4D PDE model that generates several unique spatiotemporal patterns that are relevant to cell motility. These patterns are then characterized and their impact on cell motility are explored by employing the cellular potts model (CPM). Our results reveal that wave pinning produces purely very directed motion in CPM, while MMOs allow for meandering and non-motile behaviours to occur. Abstract: Visual attention is a fundamental cognitive operation that allows the brain to evoke behaviors based on the most important stimulus features. Although mouse models offer immense potential to gain a circuit-level understanding of this phenomenon, links between visual attention and behavioral decisions in mice are not well understood. Here, we describe a new behavioral task for mice that addresses this limitation. We trained mice to detect weak vertical bars in a background of checkerboard noise while audiovisual cues manipulated their spatial attention. We then modified a reverse correlation method from human studies to link behavioral decisions to stimulus locations and features. We show that mice attended to stimulus locations just rostral of their optical axis, which was highly sensitive for vertically oriented stimulus energy whose spatial frequency matched those of the weak vertical bars. We found that the tuning of sensitivity to orientation and spatial frequency grew stronger during training, was multiplicatively scaled with attention, and approached that of an ideal observer. These results provide a new task to measure spatial- and feature-based attention in mice which can be leveraged with new recording methods to uncover attentional circuits. The first time that I became aware of the LOA was when I realized that I was experiencing lack in many areas of my life.  I had no idea that I was causing myself to be in this circumstance to be this way.I found myself in multiple side hustles, but still not financially independent.  I was still struggling to pay the bills and feeling exhausted! My quality of life was very poor, I had no time for myself and my kids.  I felt like I was running all the time but never reaching anywhere.One day I was having a casual discussion with my daughters and we were discussing past events that happened, and my daughter Khadra, said to me …”But you know mom, you allowed that to happen”At first, it was difficult to hear that, as no one wants believe that they allowed undesirable - even painful circumstances to exist in their lives.  But once I got over myself and the hesitation to accept the truth in that statement, I realized that YES in fact, I did cause that situation to happen.  Without me allowing it, it would not exist! That was my wake-up call.  What is the LOA?   The LOA is one of the universal laws such as the LOG.  Even though you can't see it, it's there working all the time!  The LOA is based on:1.    Everything is in vibration2.    Vibrations of similar frequencies are drawn together 3.    You have the ability to control your vibration and the condition of your lifeWhat's Manifestation? Dictionary Definition - To show something clearly, through signs or actions“Manifesting is making everything you want to feel and experience a reality... via your thoughts, actions, beliefs, and emotions.”- Angelina Lombardo, the author of A Spiritual Entrepreneur What you think about all day is what you will manifest into your life. This is why you hear people say – Same shit different day! This episode is part two of our conversation with returning guest Khadra Mohamed about what it means to be a leader. In the first part we talked with Khadra about different leadership experiences and opportunities we've had throughout our lives so far, what kinds of leadership styles come naturally to us, and some things that can keep us from showing up as ourselves. In this part we'll go deeper, talking about how we became leaders and what helped us see ourselves that way. We'll talk about women in leadership positions who inspire us and how important it is to see women who look like us taking the reins. Representation matters! Khadra talks about not realizing how important it was to see women who looked like her in leadership roles until it happened, how good it felt to see possibilities for herself, and how connected she felt to those women. This can have such a big impact on our self-image and what we believe we're capable of. We'll also get into how we were encouraged (or if we were encouraged) to take on leadership roles when we were growing up, some differences we've experienced across cultures and families, and what kinds of differences we saw for boys when it came to leadership opportunities. We'll share stories about our first big leadership challenges and how we've grown since then— what was hard, what came naturally, and what we learned about ourselves. Our takeaways: Leading can be challenging, and there can be barriers along the way, but it can also be so rewarding! You might not always see someone who looks like you in a leadership position, but you could be the one who inspires someone else. We hope you find some inspiration in our experiences that can help you take some risks, put yourself out there, and see yourself as a leader. This is part one of a two-part episode about what it means to be a leader with our friend and returning guest Khadra Mohamed. We talked with Khadra about different leadership experiences and opportunities we've had throughout our lives so far like working at camp, taking care of siblings, and stepping up at school or work. Leadership styles and how we approach leading can be very different. For example, Khadra shared that what leadership looks like for her is showing up, being consistent for the girls she works with, and building trust. She's found that quiet leadership is the best fit for her. This means being herself, and not trying to be too outgoing when it doesn't come naturally. Being a leader doesn't necessarily mean being vocal, charismatic, or outgoing - there are lots of different kinds of leaders. There's strength in quiet leadership, and introverts can be leaders just as effectively as extroverts. But that doesn't mean there's anything wrong with being outgoing and extroverted! Leaders can be quiet, loud, really hands-on, or more hands-off. Do what comes naturally to you! Showing up as yourself is important. You'll be more effective and authentic, and build stronger, more trusting relationships. We talked about some things that are hard for us that maybe some girls out there can relate to, like always wanting to seem likable, approachable, and nice. We don't want to come off as mean, aggressive, condescending, or rude, but we know we need to be assertive to get things done in a leadership role. As women and girls, we can stretch ourselves to be comfortable with direct communication and not worry about coming across as being "bossy." Our takeaways: Being a leader doesn't mean you have to know everything, and it's ok to make mistakes. It's easy to think of leadership as having power or being in command, but really the best part is seeing growth happen in the people we lead. There's no one right way to be a leader. Everyone can lead, and it can look different for different people. In this episode we talk with Khadra Mohamed about her experience growing up Somali in America. Khadra is a friend of ours and a young professional woman who works as a program coordinator for Girl Scouts ConnectZ bringing programming to girls in underrepresented communities. She has a degree in child psychology from the University of Minnesota and has worked with youth in lots of different ways. Outside of her work with girls Khadra loves going to concerts, sewing, journaling, and hanging out with her siblings. We had a great conversation about her experience growing up in the Somali-Muslim community in central New York, Ohio, and Minnesota, and some of the challenges she and her family have faced. Khadra and her parents and siblings moved to Minnesota to be closer to extended family and found a thriving community where she was able to connect with other Somalis and learn more about her culture and religion. But while the Somali community in the Twin Cities is strong, there are still lots of barriers and misconceptions. Our conversation got deep, and we learned so much from Khadra, but we still had our fun and funny moments :-) We learned from Khadra that on top of all the other hard things girls go through in middle and high school, Somali girls are navigating other identities that can make things tough and cause them to be targeted— like being black, Muslim, women, and wearing the hijab, a headscarf many Muslim women wear to connect to their religion. Khadra talks about how Somalis are storytellers and poets, how important it is to have the support of family and community and always have each other's backs, and the significance and deep love of good food! Our takeaways— Somali girls are smart, cool, and creative, and are a thriving, incredible force. We can support Somali girls in our own communities by reaching out with resources and opportunities. Girls can reach across cultures and learn about their peers and neighbors, breaking down the barriers that can keep us from making friends and supporting other girls. Not sure where to start? Khadra is actually a man, and took a pseudonym (his wife's!) during his career in the Algerian Army during the civil war. His book follows a group of people struggling to hold on to their humanity in a world where pleasure is a sin and death awaits anyone who breaks the rules.
