The NIEHS Superfund Research Program (SRP) is sponsoring a Risk e-Learning webinar series, hosted by CLU-IN, focused on research efforts to develop tools for sampling, monitoring, detecting, and characterizing per- and polyfluoroalkyl substances (PFAS) contamination. The three-part series will feature SRP-funded researchers and collaborators whose research focuses, in part, on understanding the distribution and fate of PFAS in the environment. The first session will feature SRP-funded investigators working on innovative methods to classify and/or quantify PFAS compounds. To learn about and register for the other sessions in this webinar series, please see the SRP website. Diana Aga, Ph.D., of the State University of New York at Buffalo (University at Buffalo) will discuss work affiliated with her R01 grant, Model-aided Design and Integration of Functionalized Hybrid Nanomaterials for Enhanced Bioremediation of Per-and Polyfluoroalkyl Substances. Discussed will be various complementary techniques used to identify and quantify known and unknown PFAS in complex environmental samples, such as liquid chromatography (LC) with high-resolution mass spectrometry (HRMS), supercritical fluid chromatography (SFC), combustion ion chromatography (CIC), and fluorine nuclear magnetic resonance spectroscopy (19F-NMR). Diana will examine the advantages and limitations of these techniques in terms of their applications in PFAS analysis in wastewater and biosolids samples. Erin S. Baker, Ph.D., of the University of North Carolina at Chapel Hill will illustrate how combining liquid chromatography, ion mobility spectrometry and mass spectrometry enables the evaluation of a broader range of chemical exposure, while uncovering novel species yet to be reported. Throughout life, people are frequently exposed to both naturally occurring and human-made chemicals such as PFAS. Associating these exposures to health risks can however be quite difficult since, for example, there are over 14,000 PFAS listed in the U.S. Environmental Protection Agency's PFAS Master List but only a few hundred standards available for targeted analytical tests and highlights how non-targeted approaches are of great importance. Lee Ferguson, Ph.D., of Duke University will focus on the development of novel non-targeted, high-resolution mass spectrometry methods and cheminformatics approaches for characterizing PFAS in the environment. He will discuss applications of these methods to assessment of PFAS contamination in drinking water and wastewater in North Carolina. The NIEHS Superfund Research Program (SRP) is sponsoring a Risk e-Learning webinar series, hosted by CLU-IN, focused on research efforts to develop tools for sampling, monitoring, detecting, and characterizing per- and polyfluoroalkyl substances (PFAS) contamination. The three-part series will feature SRP-funded researchers and collaborators whose research focuses, in part, on understanding the distribution and fate of PFAS in the environment. The first session will feature SRP-funded investigators working on innovative methods to classify and/or quantify PFAS compounds. Diana Aga, Ph.D., of the State University of New York at Buffalo (University at Buffalo) will discuss work affiliated with her R01 grant, Model-aided Design and Integration of Functionalized Hybrid Nanomaterials for Enhanced Bioremediation of Per-and Polyfluoroalkyl Substances. Discussed will be various complementary techniques used to identify and quantify known and unknown PFAS in complex environmental samples, such as liquid chromatography (LC) with high-resolution mass spectrometry (HRMS), supercritical fluid chromatography (SFC), combustion ion chromatography (CIC), and fluorine nuclear magnetic resonance spectroscopy (19F-NMR). Diana will examine the advantages and limitations of these techniques in terms of their applications in PFAS analysis in wastewater and biosolids samples. Erin S. Baker, Ph.D., of the University of North Carolina at Chapel Hill will illustrate how combining liquid chromatography, ion mobility spectrometry and mass spectrometry enables the evaluation of a broader range of chemical exposure, while uncovering novel species yet to be reported. Throughout life, people are frequently exposed to both naturally occurring and human-made chemicals such as PFAS. Associating these exposures to health risks can however be quite difficult since, for example, there are over 14,000 PFAS listed in the U.S. Environmental Protection Agency's PFAS Master List but only a few hundred standards available for targeted analytical tests and highlights how non-targeted approaches are of great importance. Lee Ferguson, Ph.D., of Duke University will focus on the development of novel non-targeted, high-resolution mass spectrometry methods and cheminformatics approaches for characterizing PFAS in the environment. He will discuss applications of these methods to assessment of PFAS contamination in drinking water and wastewater in North Carolina. Vahue begins by explaining how Brite has grown and transformed over the years (2:17) before explaining how she got started with the company (6:09) and the advantages of both HR and finance teams rolling up into one person (7:59). She then recalls some of the major financial challenges the business faced running on spreadsheets and four different systems, like reporting due to a lack of visibility and access to real-time data (9:26). Brite decided to implement NetSuite in 2015 because they wanted a true cloud system that could integrate with other applications they needed to run the business (11:50). Vahue says the company immediately saw benefits across reporting, revenue recognition and compliance (14:18). She then dives into the HR side of her role, noting her No.1 priority was giving power back to employees to take responsibility for their roles, careers and future at the company (18:27). However, it was difficult to meet that goal while running HR processes–like performance reviews and time off–in word docs and spreadsheets (21:52). Thus, Brite implemented SuitePeople, NetSuite's HRMS offering, to put the power back in employees' and managers' hands (22:26). Vahue says SuitePeople has helped Brite automate and improve time off and expense processes as well as goal and performance management (23:38). Brite even customized SuitePeople to track metrics that are specific to the business and its sales team (24:35). Finally, Vahue says Brite is growing quickly and looks forward to automating more processes within NetSuite, like payroll and project management (28:04). E., Schoeny, H., Baumgartinger, C., Koecher, T., Koellensperger, G. Abstract: Non-targeted analysis by high-resolution mass spectrometry (HRMS) is the essential discovery tool in metabolomics. Up to date, standardization and validation remain a challenge. Community wide accepted, cost-effective benchmark materials are lacking. In this work, we propose yeast (Pichia pastoris) extracts, derived from fully controlled fermentations for this purpose. We established an open-source metabolite library of > 200 metabolites, reproducibly recovered in ethanolic extracts by orthogonal LCHRMS methods, different fermentations (over three years) and different laboratories. More specifically, compound identification was based on accurate mass, matching retention times, and MS/MS spectra as compared to authentic standards and internal databases. The library includes metabolites from the classes of 1) organic acids and derivatives (2) nucleosides, nucleotides and analogues, (3) lipids and lipid-like molecules, (4) organic oxygen compounds, (5) organoheterocyclic compounds, (6) organic nitrogen compounds and (7) benzoids at expected concentrations ranges of sub-nano to micromolar. As yeast is a eukaryotic organism, key regulatory elements are highly conserved between yeast and all annotated metabolites were also reported in the Human metabolome data base (HMDB). A large fraction of metabolites was found to be stable for several years when stored at -80 degree. Thus, the yeast benchmark material enabled not only to test for the chemical space and coverage upon method implementation and developments, but enabled in-house routines for instrumental performance tests. Abstract: The proper storage and release of monoamines contributes to a wide range of neuronal activity. Here, we examine the effects of altered vesicular monoamine transport in the nematode C. elegans. The gene cat-1 is responsible for the encoding of the vesicular monoamine transporter (VMAT) in C. elegans and is analogous to the mammalian vesicular monoamine transporter 2 (VMAT2). Our laboratory has previously shown that reduced VMAT2 activity confers vulnerability on catecholamine neurons in mice. The purpose of this paper was to determine whether this function is conserved and to determine the impact of reduced VMAT activity in C. elegans. Here we show that deletion of cat-1/VMAT increases sensitivity to the neurotoxicant 1-methyl-4-phenylpyridinium (MPP+) as measured by enhanced degeneration of dopamine neurons. Reduced cat-1/VMAT also induces changes in dopamine-mediated behaviors. High-resolution mass spectrometry-based metabolomics in the whole organism reveals changes in amino acid metabolism, including tyrosine metabolism in the cat-1/VMAT mutants. Treatment with MPP+ disrupted tryptophan metabolism. Both conditions altered glycerophospholipid metabolism, suggesting a convergent pathway of neuronal dysfunction. Our results demonstrate the evolutionarily conserved nature of monoamine function in C. elegans and further suggest that HRMS-based metabolomics can be used in this model to study environmental and genetic contributors to complex human disease Copy rights belong to original authors.
