In this episode of the Epigenetics Podcast, we talked with Kyle Eagen from Baylor College of Medicine about his work on BET Proteins and their role in chromosome folding and compartmentalization. In the early days of his research career Dr. Eagen made use of genomics and microscopy to study chromosomes, particularly polytene chromosomes in Drosophila. The correlation between the folding patterns detected by Hi-C and polytene bands highlights the similarities between the two, bridging traditional cytology with modern NGS methods. This work formed the basis of Kyle's thesis and sparked his interest in nuclear organization and chromosome 3D structure. In his independent lab Kyle then studied compartments in chromatin structure and focused on the relationship between histone modifications and the 3D structure of chromosomes. The discovery of BRD4-NUT, a fusion oncoprotein that reprograms chromosome 3D structure, is highlighted as a significant step forward in understanding chromatin structure. The conversation then shifts to the use of a tool to test hypotheses about the involvement of BRD4 in a specific process, leading to consistent results and considerations for manipulating chromosome organization for therapeutic purposes. The role of BET proteins in genome folding and the need for further research on other factors involved in 3D genome structure are discussed. References Rosencrance, C. D., Ammouri, H. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2020.08.05.238519v1?rss=1 Authors: Chereda, H., Bleckmann, A., Menck, K., Perera-Bel, J., Stegmaier, P., Auer, F., Kramer, F., Leha, A., Beissbarth, T. Abstract: Motivation: Contemporary deep learning approaches show cutting-edge performance in a variety of complex prediction tasks. Nonetheless, the application of deep learning in healthcare remains limited since deep learning methods are often considered as non-interpretable black-box models. Layer-wise Relevance Propagation (LRP) is a technique to explain decisions of deep learning methods. It is widely used to interpret Convolutional Neural Networks (CNNs) applied on image data. Recently, CNNs started to extend towards non-euclidean domains like graphs. Molecular networks are commonly represented as graphs detailing interactions between molecules. Gene expression data can be assigned to the vertices of these graphs. In other words, gene expression data can be structured by utilizing molecular network information as prior knowledge. Graph-CNNs can be applied to structured gene expression data, for example, to predict metastatic events in breast cancer. Therefore, there is a need for explanations showing which part of a molecular network is relevant for predicting an event, e.g. distant metastasis in cancer, for each individual patient. Results: We extended the procedure of LRP to make it available for Graph-CNN and tested its applicability on a large breast cancer dataset. We present Graph Layer-wise Relevance Propagation (GLRP) as a new method to explain the decisions made by Graph-CNNs. We demonstrate a sanity check of the developed GLRP on a hand-written digits dataset, and then applied the method on gene expression data. We show that GLRP provides patient-specific molecular subnetworks that largely agree with clinical knowledge and identify common as well as novel, and potentially druggable, drivers of tumor progression. As a result this method could be potentially highly useful on interpreting classification results on the individual patient level, as for example in precision medicine approaches or a molecular tumor board. She is an Army Veteran, entrepreneur, businesswoman, educator, holistic physical therapist, life coach and soon to be published author with over 38 years experience using integrative holistic therapies and energy medicine helping to heal mind, body, spirit of thousands of clients. She is passionate about changing lives to make a difference in this world. Deborah Stegmaier- Passion for Helping Soldiers Deborah started her journey at the University of Connecticut in ROTC and studied Physical Therapy. Deborah was commissioned as a 2nd Lieutenant and served as a Physical Therapist for the Army and absolutely loved it. She was stationed at Fort Knox and Madigan Army Medical Center in Tacoma after doing work in Texas. “ We had the opportunity to work with doctors and do all kinds of testing, evaluations and screenings. We were the TMCs, we saw the military guys before the docs even saw them. So I had freedom to learn and to help these soldiers.” – Deborah Stegmaier Transitioning When Deborah got out of the Army she had a rude awakening transitioning into Physical Therapy in the civilian world. She didn’t have the freedom she had in the Army. Deborah had to learn another way since there were more rules and structure. She stayed in the Army Reserves and started studying holistic therapy, manual therapy using her hands. “I learned early on in my career that what I learned in school wasn't enough. I was working with people with acute and chronic injuries and pain illnesses, disease processes. And I just saw that there was so much more to healing that needed to be learned and used to help these people truly heal. ” – Deborah Stegmaier Well Being for Warriors Deborah used her passion for helping people and holistic medicine to start wellbeing for Warriors, a nonprofit helping veterans with PTSD. Her heart and soul is committed to helping veterans with PTSD, mild traumatic brain injury, chronic pain and military sexual trauma. “We offer free, absolutely free holistic services to veterans, active duty and veterans who have a diagnosis of PTSD, mild traumatic brain injury, chronic pain or military sexual trauma. And we offer a variety of things because everything we do is about mind, body, spirit, and energy because of who we are as human beings. We need to look at the whole person.” – Deborah Stegmaier To hear more from Deborah Stegmaier and how she serves our military community download this episode. Grace & Grit Podcast: Helping Women Everywhere Live Happier, Healthier and More Fit Lives
Hormones are a topic we have talked a lot about on the Grace & Grit Podcast because let’s face it – hormones govern your body – and if they aren’t in their happy place…well…neither are you! As I have crossed over the threshold of 40, I have become a lot more curious about the state of my own hormones. I feel amazing, but there are months when my PMS is simply out of control. My breasts hurt, I am reactive and I just feel like my body is not my own. I have been hearing about the DUTCH Test for a few years now (DUTCH stands for Dried Urine Test for Comprehensive Hormones), which you can do in the comfort of your own home. The Dutch Test is being used by more and more physicians to remedy hormonal challenges in patients and has been endorsed by well known medical experts like Kris Kessler, Dr. Joesph Mercola, Dr. Ben Lynch, Christiane Northrup, and Sara Gottfriend. When I asked my awesome Naturopathic physician, Elizabeth Stegmaier, about it she encouraged me to have the testing done not just for my benefit but to be able to educate you the listeners about the value of this type of testing. Not only has Dr. Stegmaier personally been using the Dutch test for years with her own patients, but she just happens to be friends with the top clinical advisors to the product, one of which agreed to join me on the podcast today, Dr. Debbie Rice. Also included: Frank does a cuss twice and he's real sorry. Josh has inherited his mother's coffee order. Jamey gases up the Boys' egos. Cam does a successful time travel goof involving cheese.

Oooooooh, can you feel it? The nip in the air, the spice of the pumpkin latte, the fur of the Ugg caressing your calf? This can mean only one thing, it's the most wonderful time of the year, the time when the freaks can walk the streets free of judgment hidden behind masks of celebrated serial killers. That's right it's time for the obligatory Halloween episode! Bwaahahaaaaaaa! Now quit peeing your pants in fright we've got a lot of show here for ya! Leeandra joins in the first half before going to trick or treat in China (I guess they have the best candy corn?) She, Lilly and I preview the Kickstarter game Epic Monster Tea Party before she departs. Then I enlist the help of one of the best game designers out there, Jamey Stegmaier and one of the "funniest" pod-casters, Steven Spang, for our big Halloween spooky skeptic segment. Bare with me as I tortuously try to sound intelligent... If only there weren't hours of previous audio to refute this. So join us won't you? As we take you on a fright of fancy across an episode with more bite than Drac, more Friday's than the 13th and more creep than Harvey Weinstein!
In our second interview during First Timers' Day at The Players, Brett Stegmaier joins us to talk Sawgrass, his season struggles and trying to escape them, as well the difficulties for players who try to make a schedule and a career after graduating from the Web.com Tour and Web.com Tour Finals.

And what you all have been waiting for! We dive head first into one of the biggest board game discussion topics and that's Board Game Immersion... what better way to do that then to have the man himself Jamey Stegmaier of Stonemaier Games co-host with us to really take a hard look at what keeps us entrenched in this hobby!
