E., Ahamed, H., Ringstad, N., Nagel, K. I., Schoppik, D. Abstract: Balance and movement are impaired in a wide variety of neurological disorders. Recent advances in behavioral monitoring provide unprecedented access to posture and locomotor kinematics, but without the throughput and scalability necessary to screen candidate genes / potential therapeutics. We present a powerful solution: a Scalable Apparatus to Measure Posture and Locomotion (SAMPL). SAMPL includes extensible imaging hardware and low-cost open-source acquisition software with real-time processing. We first demonstrate that SAMPL's hardware and acquisition software can acquire data from three species (flies, worms, and zebrafish) as they move vertically. Next, we leverage SAMPL's throughput to rapidly (two weeks) gather a new zebrafish dataset. We use SAMPL's analysis and visualization tools to replicate and extend our current understanding of how zebrafish balance as they navigate through a vertical environment. Next, we discover (1) that key kinematic parameters vary systematically with genetic background, and (2) that such background variation is small relative to the changes that accompany early development. Finally, we simulate SAMPL's ability to resolve differences in posture or vertical navigation as a function of affect size and data gathered -- key data for screens. Taken together, our apparatus, data, and analysis provide a powerful solution for labs using small animals to investigate balance and locomotor disorders at scale. More broadly, SAMPL is both an adaptable resource for labs looking process videographic measures of behavior in real-time, and an exemplar of how to scale hardware to enable the throughput necessary for screening. Abstract: Grid cells are neurons in the entorhinal cortex that play a key role in spatial navigation. When subjects navigate through spatial environments, grid cells exhibit firing fields that are arranged in a triangular grid pattern. As direct recordings of grid cells from the human brain are only rarely possible, functional magnetic resonance imaging (fMRI) studies proposed and described an indirect measure of entorhinal grid-cell activity, which is quantified as a hexadirectional modulation of fMRI activity as a function of the subject's movement direction. However, it still remains unclear how the activity of single grid cells is related to the sum activity of a population of grid cells, which may exhibit hexadirectional modulation and thus provide the basis for the hexadirectional modulation of entorhinal cortex activity measured with fMRI. Here, we thus performed numerical simulations and analytical calculations to better understand how the aggregated activity of many grid cells may be hexadirectionally modulated. Our simulations implemented three different hypotheses proposing that the hexadirectional modulation occurs because grid cells show head-direction tuning aligned with the grid axes; are subjected to repetition suppression; or exhibit a bias towards a particular grid phase offset. Our simulations demonstrate that all three hypotheses can, in principle, lead to a hexadirectional modulation of sum grid-cell activity. However, the magnitude of the hexadirectional modulation appears to depend considerably on the subject's navigation pattern and the exact biological properties of grid cells. Our results thus indicate that future fMRI studies could be designed to test which of the three hypotheses most likely accounts for the fMRI measure of grid cells. These findings also underline the importance of quantifying the biological properties of single grid cells in humans to further elucidate how hexadirectional modulations of fMRI activity may emerge. Dieses Kauderwelsch haben deutsche Auswanderer vor Jahrhunderten nach ihrer Ankunft gesprochen. Es hat bis heute überlebt und dabei viele Eigenarten entwickelt - mit Wörtern, die es eigentlich weder im Deutschen noch im Englischen gibt. Doch dieser Dialekt stirbt allmählich aus, zusammen mit den Alten, die ihn noch sprechen. Und die Jungen interessiert dieses Erbe nicht mehr besonders. Die Historikerin eines Museums in Fredericksburg, eine ehemalige Lehrerin, Kulturschaffende aus New Braunfels und der Veranstalter eines Radioprogramms mit deutscher Musik versuchen, den Bedeutungsverlust des Texas German in den vergangenen Jahrzehnten zu erklären. Dieses Mal im Gespräch mit Thomas Auer, Auditor bei American Institute of Baking (AIB) über die aktuell neu erschienene IFS 7 Richtlinie. Thomas Auer findet klare Worte und diskutiert die neue Guideline mit Daniel Schröer, von Futura, der als Guideline Consultant bei der IFS 7 Richtlinie mitarbeiten durfte. Dutzende Experten und Verbände arbeiteten seit knapp 3 Jahren am neuen „Standard“ und nun ist er da. Wie auch der AIB, geht der IFS extrem auf IPM ein. Also Prävention, Monitoring sowie physikalische und biologische Alternativen, die in den letzten Jahren noch nicht in der breiten Masse aufgenommen wurden, aber nun gefordert werden. Natürlich geht es auch um eine nachhaltigere Art und Weise Schädlinge zu managen, da typischerweise viele Rodentizide eingesetzt werden innerhalt der „Dauerbeköderung oder Permanent Beköderung“. Der EU Green Deal und führende Kunden und Pest Control Betriebe haben klar kommuniziert: es müssen weniger Rodentizide werden. Der IFS wie auch der AIB fordern dies schon lange, aber nun noch mit mehr Ausdruck und Verbindlichkeit. Der IFS hat alle Daten der vergangenen Audits analysiert und erkannt, dass Prävention sowie nachhaltige Alternativen auch einen guten Effekt bzgl. schneller und nachhaltiger Bekämpfung haben. Zudem wurden IoT (Internet of Things) Lösungen wie Digitale Fallen und Sensoren empfohlen, mit denen IFS, AIB Kunden schon lange arbeiten. Diese Lösungen erlauben die Früherkennung und 24-7 Überwachung – große Vorteile, die Thomas Auer auch schon seit Jahren kennt und zu schätzen weiß. Es ist klar in welche Richtung die Zukunft geht und diese Themen werden im Detail im Podcast besprochen. Zudem gibt es immer mehr Gerüchte im DACH Raum bzgl. neuer Regularien – Thomas und Daniel sprechen diese an und diskutieren potenzielle Szenarien. Here is a double paradox: The European Union's (EU) set of founding principles—its telos, so to speak—are undergoing a two-track inversion. The block was initially designed to slide gently towards federalization whilst remaining a largely toothless actor on the world stage. And yet the opposite has happened: the EU has since grown into a powerful geopolitical player of its own that is internally at peace with the present deadlock of integration. Sometime between the eurozone crisis of the early 2010s and Russia's ongoing invasion of Ukraine, the EU's entire architecture has been turned inside out. Scholars, journalists and analysts in Brussels and European capitals are still at pains to gauge the depth of this compete revolution. Stefan Auer of Hong Kong University may have lost some sleep over it. A former—and likely future—recipient of the prestigious Jean Monnet Chair for EU studies, he's as astute an observer as any of the block's institutional dynamics. He argues in his most recent book that, instead of seeking to transcend the laws of politics, the EU would be well advised to heed them. In this episode he sits down with us and Glyn Morgan, associate professor at Syracuse University's politics department. "With the eurozone crisis going back to 2010, the refugee crisis that culminated in 2015, the crisis of the EU-Russia relationship going back to the Ukrainian Maidan revolution of 2013-14, to the Covid-19 crisis in 2020, the EU has struggled to live up to the expectations it raised both in relation to its own people and neighbouring countries. This is not an accident". Could this really be by design? In European Disunion: Democracy, Sovereignty and the Politics of Emergency (Hurst in the UK, OUP in the US, 2022), Stefan Auer argues that the EU's hybrid form – falling somewhere between a multinational state and a multilateral organisation – comes closest to the ideals of Germany, its most powerful member. This attempt to bypass politics has weakened the EU in the many emergencies it has faced over the last 15 years. Today, he says, "Europeans do not have the luxury of living in a politics-less world". Stefan Auer is an Associate Professor at the University of Hong Kong, having previously taught in Melbourne and Dublin and twice held Jean Monnet chairs.
