Episode Synopsis: The doctor that Seward had placed in charge of the asylum in his absence, Dr. Patrick Hennessy, writes to Seward describing an unfortunate incident regarding Renfield. Renfield had been witnessed shouting obscenities at some workmen who were retrieving some heavy wooden boxes from the neighboring Carfax Estate. When the men were preparing to leave, Renfield escaped from his room and attacked the men, almost killing one. He struggled against the attendants who managed to restrain him. The workmen were angry about the incident, but calmed down after Hennessy bought them a round of drinks and paid them a sovereign a piece to forget about the incident. Their names were recorded for future reference. Van Helsing, Seward, and Arthur trade turns watching over Lucy as she nears the end. Seward finds it perplexing that when asleep, Lucy pulls the garlic away, and when she awakens, she draws it close. Her teeth continue to look longer and sharper than usual. Van Helsing removes the scarf around her neck and discovers that the wounds on her throat have completely disappeared. He calls for Arthur to be brought in, as he believes Lucy is nearing the end. Arthur tries to comfort Lucy, but Van Helsing will not let him kiss Lucy on the lips. Lucy undergoes a change, actively seducing Arthur to kiss her, but Van Helsing forcefully keeps Arthur back. Lucy regains herself, thanking Van Helsing, and asking him to bring her peace. She closes her eyes and breathes her last. Lucy's funeral is arranged for the following day so that Lucy might be buried with her mother, and so that Arthur may return to Ring immediately after to continue arrangements for his own father's funeral. Rather than decay, Lucy's body appears more luminous and beautiful in death such that the men can scarcely believe she has passed. Van Helsing insists upon looking over Lucy's recent papers so as to prevent any critical or implicating information from being witnessed out of context. Once Mrs. Westenra's solicitor had been notified of his client's passing, Van Helsing and Seward check in on Lucy's presentation. Van Helsing places garlic flowers among Lucy's bouquets and a golden crucifix over Lucy's lips. Van Helsing stops by Seward's room and asks Seward to procure tools such that he might remove Lucy's head and heart. Seward objects to such desecration, but admits that he called Van Helsing because his methods are true. As Seward bids goodnight to Van Helsing, he is touched by the sight of one of Lucy's maids entering her burial chamber to pay final respects. The following morning, Van Helsing reveals that the maid had been robbing Lucy's corpse and had stolen the golden crucifix that had been placed over her lips. As a result, Van Helsing's ghastly desecration of Lucy would be postponed. Seward meets with Mr. Marquand, Mrs. Westenra's solicitor, who explains that Mrs. Westenra had all her affairs well in order, leaving everything to Arthur. Arthur arrives to bid farewell to Lucy, and breaks into tears on Seward's shoulder, disbelieving that she is truly dead. After the undertakers seal up Lucy's coffin, Van Helsing asks Arthur for permission to hold and read Lucy's private papers, to which Arthur acquiesces. For the remainder of the night, Van Helsing patrols the hallway beyond the burial chamber. Follow us on Twitter.com/CryptiCanticles, Facebook.com/DraculaRadioPlay, and at crypticcanticles.com

Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.06.30.546837v1?rss=1 Authors: Cirstian, R., Forde, N., Sotiropoulos, S., Andersson, J., Beckmann, C., Marquand, A. Abstract: Diffusion MRI is a neuroimaging modality used to evaluate brain structure at a microscopic level and can be exploited to map white matter fiber bundles in the brain. One common issue is the presence of artefacts such as acquisition artefacts (e.g. ringing), physiological artefacts (cardiac pulsation, breathing, motion), distortions (EPI, eddy currents) or post-processing (e.g. blurring, registration errors). These may lead to problems in downstream processing and can bias subsequent analyses. In this work we use normative modeling to create a semi-automated pipeline for detecting diffusion imaging artefacts and errors by modeling 24 image derived phenotypes using the UK Biobank dataset. We used several parameter types with different modelling complexities (DTI model: FA and MD; NODDI model: ICVF and ISOVF). Likewise, we selected several tracts of various sizes and modelling difficulties (corpus callosum, corticospinal tract, uncinate fasciculus and fornix). Our method is compared to two traditional quality control modalities: a visual quality control protocol performed on 500 subjects and quantitative quality control using other automated pipelines. The normative modelling framework proves to be comprehensive and efficient in detecting diffusion imaging artefacts arising from various sources as well as outliers resulting from inaccurate post-processing (i.e., erroneous registrations). This is an important contribution by virtue of this methods ability to identify the two problem sources (i) image artefacts and (ii) post-processing errors, which subsequently allows for a better understanding of our data and informs on inclusion/exclusion criteria of participants. A., Chakravarty, M., Diedrichsen, J., Marquand, A. F., Frens, M. A., Muetzel, R. L. Abstract: In the past, the cerebellum has been best known for its crucial role in motor function. However, increasingly more findings highlight the importance of cerebellar contributions in cognitive functions and neurodevelopment. Using large scale, population-wide neuroimaging data, we describe and provide detailed, openly available models of cerebellar development in childhood and adolescence, an important time period for brain development and onset of neuropsychiatric disorders. Next to a traditionally used anatomical parcellation of the cerebellum, we generated growth models based on a recently proposed functional parcellation. In both, we find an anterior-posterior growth gradient mirroring the age-related improvements of underlying behavior and function, which is analogous to cerebral maturation patterns and offers new evidence for directly related cerebello-cortical developmental trajectories. Finally, we illustrate how the current approach can be used to detect cerebellar abnormalities in clinical samples. Copy rights belong to original authors. N., Beckmann, C. F., Marquand, A. F. Abstract: In this study we dissect the heterogeneity that underlies traditional group-level analyses, and determine how individualised patterns of predicted activation relate to age, sex, and variations in acquisition parameters and task design choices. To this end we take advantage of six large open-access/shared datasets and collate a large representative sample of over 7500 participants from which we build a normative of task-evoked activation during a widely used emotional reactivity task, the Emotional Face Matching Task. This enables us to bind heterogeneous datasets to a common reference model and enables meaningful comparisons between them. We then apply this model to the naturalistic and clinically realistic MIND-Set cohort, which is a heterogeneous and highly comorbid sample containing individuals with one or more current diagnosis (affective and anxiety disorders, autism spectrum disorders and/or attention deficit hyperactivity disorder). This enables us to determine whether, and if so how, participants with mental illness and/or neurodivergence differ from the reference cohort, both at the group level and at the level of the individual and in terms of cross-diagnostic symptom domains in addition to diagnosis. We show that patients have, on average, a higher frequency of extreme deviations, and have unique spatial distributions depending on the DSM-IV diagnosis and the number of co-occurring diagnoses when models are constructed using the face greater than shapes task contrast. Models built using the face greater than baseline task contrast, have, by comparison, greater predictive value for individuals' functioning across four transdiagnostic domains. We demonstrate the application of the normative modelling framework to task-based functional neuroimaging data, discuss its potential to further our understanding of individual differences in brain function within reference populations, and further validate the clinical relevance of these models. Abstract: This work illustrates the use of normative models in a longitudinal neuroimaging study of children aged 6-17 years and demonstrates how such models can be used to make meaningful comparisons in longitudinal studies, even when individuals are scanned with different scanners across successive study waves. More specifically, we first estimated a large-scale reference normative model using hierarchical Bayesian regression from N=40,435 individuals across the lifespan and from dozens of sites. We then transfer these models to a longitudinal developmental cohort (N=5,985) with three measurement waves acquired on two different scanners that were unseen during estimation of the reference models. We show that the use of normative models provides individual deviation scores that are independent of scanner effects and efficiently accommodate inter-site variations. Moreover, we provide empirical evidence to guide the optimization of sample size for the transfer of prior knowledge about the distribution of regional cortical thicknesses. We show that a transfer set containing as few as 25 samples per site can lead to good performance metrics on the test set. Finally, we demonstrate the clinical utility of this approach by showing that deviation scores obtained from the transferred normative models are able to detect and chart morphological heterogeneity in individuals born pre-term. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2022.11.14.516460v1?rss=1 Authors: Rutherford, S., Barkema, P., Tso, I. F., Sripada, C., Beckmann, C. F., Ruhe, H. G., Marquand, A. F. Abstract: In this work, we expand the normative model repository introduced in (Rutherford, Fraza, et al., 2022) to include normative models charting lifespan trajectories of structural surface area and brain functional connectivity, measured using two unique resting-state network atlases (Yeo17 and Smith-10), and an updated online platform for transferring these models to new data sources. We showcase the value of these models with a head to head comparison between the features output by normative modeling and raw data features in several benchmarking tasks: mass univariate group difference testing (schizophrenia versus control), classification (schizophrenia versus control), and regression (predicting general cognitive ability). Across all benchmarks, we confirm the advantage (i.e., stronger effect sizes, more accurate classification and prediction) of using normative modeling features. We intend for these accessible resources to facilitate wider adoption of normative modeling across the neuroimaging community. Copy rights belong to original authors. T., Andreassen, O., Hinne, M., Beckmann, C. F., Marquand, A. F. Abstract: Normative modelling is an emerging technique for parsing heterogeneity in clinical cohorts. This can be implemented in practice using hierarchical Bayesian regression, which provides an elegant probabilistic solution to handle site variation in a federated learning framework. However, applications of this method to date have employed a Gaussian assumption, which may be restrictive in some applications. We have extended the hierarchical Bayesian regression framework to flexibly model non-Gaussian data with heteroskdastic skewness and kurtosis. To this end, we employ a flexible distribution from the sinh-arcsinh (SHASH) family, and introduce a novel reparameterisation that is more suitable for Markov chain Monte Carlo sampling than existing variants. Using a large neuroimaging dataset collected at 82 different sites, we show that the results achieved with this extension are better than a warped Bayesian linear regression baseline model on most datasets. We also demonstrate that the attained flexibility is essential for accurately modelling highly nonlinear relationships between aging and imaging derived phenotypes, which shows that the extension is important for pushing the field of normative modelling forward.

Abstract: In this paper, we propose a new framework for understanding and modelling neurobiological extreme atypicalities for individual participants. We combine the strength of normative models, to make predictions for individual patients, with multivariate extreme value statistics, which allows us to model the outer centiles accurately, enabling accurate estimation of risk and detection of atypicality. In general, most statistical methods focus principally on estimating the mean of the distribution and are not appropriate to capture the tails of the distribution. We believe that the tails carry most of the information for predicting psychopathy in neurological and psychiatric disorders and should therefore be modelled carefully. In the univariate case, we demonstrate how to fit a generalized Pareto distribution with the peaks over threshold method. For the multivariate case, we present a novel framework that allows us to capture the multivariate tails of the distribution fully. The model relies on the so-called tail pairwise dependence matrix (TPDM). The TPDM is similar to a covariance matrix, but specified for the tails. From the TPDM we can create a reduced basis of vectors that explain most of the structure underlying the extreme values. The method is similar to covariance principal component analysis, but uniquely made for the extreme values. To evaluate this framework, we use a range of neuroimaging-derived measures from the UK Biobank dataset, including structural, functional and diffusion image-derived phenotypes. We find that the first extreme principal component mostly consists of whole-brain volume and area measures and the second extreme principal component is mostly compromised of white matter tracts. We further demonstrate the link between extreme brain deviations in the individual from a reference cohort to behaviour. Copy rights belong to original authors. There is no doubt, to be human right now is not easy. It is almost impossible not to feel the shift underneath in the world order and the feeling is palpable. For us in Queensland and NSW, this is on top of catastrophic floods, and the pandemic. It is Ok to be human and feel the suffering. It is also OK, to escape for a few minutes into worlds that seem far removed from danger, clean-up, masks, and strife. This is the beauty of creating worlds for others to escape to. Rethinking Scripture Podcast - Episode 30: Rethinking Advent - The NativityDecember 21, 2021 - Host: Dr. Gregory HallWhen we think of the advent of the Christ, many of our perceptions are based scene at the nativity. Abstract: Machine learning predictive models are being used in neuroimaging to predict information about the task or stimuli or to identify potentially clinically useful biomarkers. However, the predictions can be driven by confounding variables unrelated to the signal of interest, such as scanner effect or head motion, limiting the clinical usefulness and interpretation of machine learning models. The most common method to control for confounding effects is regressing out the confounding variables separately from each input variable before machine learning modeling. However, we show that this method is insufficient because machine learning models can learn information from the data that cannot be regressed out. Instead of regressing out confounding effects from each input variable, we propose controlling for confounds post-hoc on the level of machine learning predictions. This allows partitioning of the predictive performance into the performance that can be explained by confounds and performance independent of confounds. This approach is flexible and allows for parametric and non-parametric confound adjustment. We show in real and simulated data that this method correctly controls for confounding effects even when traditional input variable adjustment produces false-positive findings. Abstract: The complexity of social interactions is a defining property of the human species. Many social neuroscience experiments have sought to map "perspective taking", "empathy", and other canonical psychological constructs to distinguishable brain circuits. This predominant research paradigm was seldom complemented by bottom-up studies of the unknown sources of variation that add up to measures of social brain structure; perhaps due to a lack of large population datasets. We aimed at a systematic de-construction of social brain morphology into its elementary building blocks in the UK Biobank cohort (n=~10,000). Coherent patterns of structural co-variation were explored within a recent atlas of social brain locations, enabled through translating autoencoder algorithms from deep learning. The artificial neural networks learned rich subnetwork representations that became apparent from social brain variation at population scale. The learned subnetworks carried essential information about the co-dependence configurations between social brain regions, with the nucleus accumbens, medial prefrontal cortex, and temporoparietal junction embedded at the core. Some of the uncovered subnetworks contributed to predicting examined social traits in general, while other subnetworks helped predict specific facets of social functioning, such as feelings of loneliness. Our population-level evidence indicates that hidden subsystems of the social brain underpin interindividual variation in dissociable aspects of social lifestyle. A., Counsell, S. J., Smith, S. M., Rueckert, D., Hajnal, J. V., O'Muircheartaigh, J., Edwards, A. D. Abstract: Objective: Preterm birth carries a significant risk for atypical development. While studies comparing group means have identified a number of early brain correlates of prematurity, they may "average out" effects significant in a single individual. To understand better the cerebral consequences of prematurity, we created normative "growth curves" characterizing neonatal brain development and explored the effect of preterm birth and related clinical risks in individual infants. Methods: We used Gaussian process regression to map typical volumetric development in 275 healthy term-born infants, modelling for age at scan and sex. We compared magnetic resonance images of 89 preterm infants (born 28.7-34 weeks gestational age) scanned at term-equivalent age to these normative charts and related deviations from typical volumetric development to both perinatal clinical variables and neurocognitive scores at 18 months. We then tested if this approach can be generalized to an independent dataset of 253 preterm infants (born 28-31.6 weeks gestational age) also scanned at term-equivalent age but using different acquisition parameters and scanner, who were followed-up at 20 months. Results: In both preterm cohorts, cerebral atypicalities were widespread and often multiple, but varied highly between individual infants. Deviations from normative brain volumetric development were associated with perinatal factors including respiratory support, nutrition and postnatal growth, as well as with later neurocognitive outcome. Conclusion: Group-level understanding of the preterm brain might disguise a large degree of individual differences. We provide a method and a normative dataset for clinicians and researchers to profile the individual brain. This will allow a more precise characterization of the cerebral consequences of prematurity and improve the predictive power of neuroimaging. Copy rights belong to original authors. On episode 90 of The Walking Dead Talk Through, Mark and I discuss The Walking Dead Season 9 Episode 8, titled Evolution. Mark liked this one more than I did. I would have appreciated filling in the Michonne vs. everyone else, or so it seems, mystery rather than spending all this time with Henry and the angsty teens. By the way, during the hiatus, sign up to our Facebook group to stay in touch!
Talking Through Evolution On this episode 90 of The Walking Dead Talk Through, Mark and I discuss The Walking Dead Season 9 Episode 8, which is titled Evolution. This was written by David Leslie Johnson-McGoldrick, and directed by Michael E. Satrazemis. Mark loved this episode, but I didn’t. Some choices were bothering me this time around. SPOILER ALERT!!! SPOILER ALERT!!! The rest of this blog post requires a SPOILER WARNING!!! If you haven’t watched this week’s episode, then you probably should stop now. Surprising death… of the character and his jingle!!! I am not afraid to admit that I enjoyed my Jesus jingle. I enjoyed it a lot more than the character, so now that he’s gone in spectacular fashion, I am sad to see it go. Don’t get me wrong. I am sad to see Jesus go, but he never lived up to his hype. He was just another character on the show. I know he was a much-loved character from the comic book, but in the show, he was underutilized. I never felt like I got to know him very well. He was always in the background. He went out in a great sequence, in a surprising turn. It’s sad that he actually lived up to some of his potential in his last scene. What happened? Would you please just tell us what happened??? We get more of this hinting that there’s a big rift between Alexandria and the other communities. Strike that… Not between Alexandria, but Michonne and the other communities. Everyone is giving her the cold shoulder this episode: Tara, Enid, Dianne, and even Carol. Yes, they talk, but notice how they didn’t hug? Whatever happened, I’m starting to feel manipulated, much like I did before we found out the fate of Madison in Season 4 of Fear. I just wish they would tell us what happened. Too much teenage angst for a mid-season finale! The trials and tribulations of Henry were things we had seen before, both from Fear S3E3 TEOTWAWKI when Alicia met with the kids of Broke Jaw Ranch, and TWD S5E12 Remember, with Carl meeting Enid and Ron, and then later with Aiden and Nicholas’ walker plaything. I really didn’t need to see this stuff in the mid-season finale. Instead of getting a lot more information about this Michonne vs. everyone else that keeps being hinted at, we get more of Henry. It’s not that I don’t think we should have seen that – it was a timing issue. It could have waited for the back half. Instead, we have 2 1/2 months to wait for any of that to be explained; I could have really used that to be explained a lot more, and I wasn’t waiting for Henry’s story or the other angsty teens. By the way, using cats as bait is not cool. That kid is a serial killer wanna-be!!! Thankfully, ratings are up for Evolution… This week, TWD got a 1.96 in the 18-49, with 5.089 million, up from 1.80 and 4.794 million last week. That’s down from the two episodes previous to that, but some of that can be explained because of the long holiday. Charts below.

Grand mansions were a major part of Gilded Age New York. The newly minted millionaires built themselves lavish residences, many of which had interiors inspired by the ancient world. Pompeii's houses were a particularly popular reference point. Also learn about how Henry G. Marquand and Lawrence Alma Tadema created a remarkable Greco-Pompeian room with a piano that would have made Elton John jealous.

We’ve hit 60 eps in the Next 100 Project and it’s a biggie. The 3rd of the original Star Wars flicks has 2 new (and underrated) villains and it ties up a lot of the loose ends. Problems? Sure, but this is a fabulous end to the series. (well, until the prequels did a number on all that). Anyway, we enjoyed the Jedi. You should enjoy some java from our longtime sponsor, Sparkplug Coffee. Also, we dedicate this episode to the late and very great Geoff Wall. The Force will be with him...always.

When Seattle book publisher Ed Marquand stumbled across a dearth of cheap real estate in a struggling small town not far from the big city, he thought he may have found a solution to the problem. But will Marquand be received as a knight in shining armor, or a colonizer come to conquer and pillage?

Mr. Moto is small in stature but strong and an expert in judo. He was the title character of a series of books, beginning with No Hero (1935; British title: Mr Moto Takes a Hand, reprint title: Your Turn, Mr. Moto), and of eight films between 1937 and 1939, in which he was portrayed by Peter Lorre. With the beginning of World War II, Mr. Moto fell out of favor with Americans, and no new books or movies about him appeared between 1942 and 1957. A dedicated and cold-blooded spy for Imperial Japan, Moto is not a conventional hero. He does not look for opportunities to commit violence but has no problem with killing people who obstruct his plans, and he would not hesitate to take his own life if necessary. But he is a master of concealing his true nature while under cover, and usually appears dull, naive, utterly harmless. He does not try to correct the bigoted attitudes of Westerners toward him and other Asians, and is not above encouraging such condescension. It often works to his advantage, leading Westerners to ignore or underestimate him.THIS EPISODE:May 20, 1951. NBC network. "A Force Called X07". Sustaining. Mr. Moto foils a communist plot to destroy part of New York with a portable atomic weapon. The music bridges have been deleted. The program may be dated March 26, 1951. While John P. Marquand (right) found literary success in his books such as The Late George Apley, he found commercial success in a series of detective/spy novels featuring a Japanese mystery man - Mr. I.A. Moto. This week I feature the agent that Peter Lorre made famous and also made Lorre successful. By the time radio found the novels, Moto had become a detective/agent fighting Communism in America.
