Abstract: Variation in cortical cytoarchitecture is the basis for histology-based definition of cortical areas, such as Brodmann areas. Single cell transcriptomics enables higher-resolution characterization of cell types in human cortex, which we used to revisit the idea of the canonical cortical microcircuit and to understand functional areal specialization. Deeply sampled single nucleus RNA-sequencing of eight cortical areas spanning cortical structural variation showed highly consistent cellular makeup for 24 coarse cell subclasses. However, proportions of excitatory neuron subclasses varied strikingly, reflecting differences in intra- and extracortical connectivity across primary sensorimotor and association cortices. Astrocytes and oligodendrocytes also showed differences in laminar organization across areas. Primary visual cortex showed dramatically different organization, including major differences in the ratios of excitatory to inhibitory neurons, expansion of layer 4 excitatory neuron types and specialized inhibitory neurons. Finally, gene expression variation in conserved neuron subclasses predicts differences in synaptic function across areas. Together these results provide a refined cellular and molecular characterization of human cortical cytoarchitecture that reflects functional connectivity and predicts areal specialization. In episode 250, Kestrel welcomes Natalie Shehata, a stylist focused on sustainability, to the show. Natalie currently works as the Retail Trainer for The Social Outfit, a Sydney-based fashion label who provides employment and training to people from refugee and new migrant communities. “We have these brands, and we have people kind of saying — ok, I need to tick this box and I need to do this and I need to make sure that I have women of color in our photo shoots and our editorial campaigns, and I need to make sure x, y and z. And that to me is why diversity is tokenistic, because it's not happening from the roots, it's not happening from the foundation, it's not happening from a system that was built by BIPOC and for BIPOC.” -Natalie In 2018, Natalie presented a speech at the Disposable Planet seminar for Eco Fashion Week Australia titled: “How the sustainable fashion space should focus on: representation, inclusivity and visibility.” While this was written almost 3 years ago, it continues to resonate strongly today, and has proven to make a powerful influence on the fashion community in Australia, specifically. One aspect Natalie highlights in this speech is why diversity can be very tokenistic. The following part of from that speech acknowledges some of the ideas we explore throughout this episode: “The communities most affected by our sustainable industry decisions are Black and Brown communities, yet they are not afforded the right to take part in the decision making process. It is the White privileged, resourced and elite groups who dictate the climate of fashion – now and for the future. When we're referring to the current climate of sustainable fashion and bringing the topic of visibility to media, we're faced with the over saturation at the moment of words like diversity - now is the time to acknowledge how powerful language is in communicating messages. Abstract: Abundant anatomical and physiological evidence supports the presence of at least three distinct types of relay glutamatergic neurons in the primate dorsal lateral geniculate nucleus (dLGN) of the thalamus, the brain region that conveys visual information from the retina to the primary visual cortex. Relay neuron diversity has also been described in the mouse dLGN (also known as LGd). Different types of relay neurons in mice, humans and macaques have distinct morphologies, distinct connectivity patterns, and convey different aspects of visual information to the cortex. To investigate the molecular underpinnings of these cell types, and how these relate to other cellular properties and differences in dLGN between human, macaque, and mice, we profiled gene expression in single nuclei and cells using RNA-sequencing. These efforts identified four distinct types of relay neurons in the primate dLGN, magnocellular neurons, parvocellular neurons, and two cell types expressing canonical marker genes for koniocellular neurons. Surprisingly, despite extensive documented morphological and physiological differences between magno- and parvocellular neurons, we identified few genes with significantly differential expression between transcriptomic cell types corresponding to these two neuronal populations. We also detected strong donor-specific gene expression signatures in both macaque and human relay neurons. Likewise, the dominant feature of relay neurons of the adult mouse dLGN is high transcriptomic similarity, with an axis of heterogeneity that aligns with core vs. shell portions of mouse dLGN. Together, these data show that transcriptomic differences between principal cell types in the mature mammalian dLGN are subtle relative to striking differences in morphology and cortical projection targets. Finally, we align cellular expression profiles across species and find homologous types of relay neurons in macaque and human, and distinct relay neurons in mouse. Abstract: Ca2+/calmodulin-dependent protein kinase II alpha (CaMKII) is an abundant neuronal signaling protein involved in synaptic plasticity and memory formation1,2. The central hub domain regulates the activity of CaMKII by organizing the holoenzyme complex into functional oligomers3-6. Recent findings have suggested that the hub is also an allosteric determinant of kinase activity7, and is thus an emerging target for therapies to correct CaMKII dysregulation8,9. However, pharmacological modulation of the hub domain has never been demonstrated. Here we show that stabilization of the CaMKII hub domain confers neuroprotection. By combining photoaffinity labeling and chemical proteomics using small molecule analogs of the natural metabolite {gamma}-hydroxybutyrate (GHB)10, we reveal that CaMKII is the selective target for GHB. We further find that these GHB analogs bind to the hub interior by solving a 2.2 [A] crystal structure of CaMKII with bound ligand. Using differential scanning fluorimetry, we show that binding of ligands to the hub interior increases the thermal stability of hub oligomers in a concentration-dependent manner. Moreover, we demonstrate the functional significance of this hub stabilization by showing substantial neuroprotective effects in cellular excitotoxicity assays and in a mouse model of cerebral ischemia. Together, our results reveal that CaMKII hub stabilization is the mechanism by which GHB provides endogenous neuroprotection and that small-molecule CaMKII-selective ligands have therapeutic potential. SusanShehata is a woman of many (MANY) talents but all of them boil down to this; Susan's work centers around helping people identify and unwind the limiting beliefs or the limiting stories - the core stories that keep us stuck. The first half of this episode has us talking about the many healing modalities Susan employs including space work, breath work, and core story work in which she's developed her own process including the simple system she's created to identify our stories around worth, lovability, belonging, safety, competency, power, and freedom and how those core stories become our subconscious operating system driving how we show up in the world. This is all absolutely fascinating stuff but it gets even better... Susan and I talk about her work as a trauma educator and how she's helping in the anti-racism front. Susan says - racism is one of the largest, if not the largest, collective and historical traumas. Historical because of course, it goes back so long and collective because it has not been resolved. So unlike some historical traumas that perhaps we can speak of in past tense, racism is obviously and unfortunately, alive and well. I don't want to give it all away but Susan gets personal with her own stories of technically being white but of Middle Eastern descent and how that's informed her own core stories around safety and belonging. What excites Susan? The fact that we are collectively having very hard conversations. Amen.
