Chris Romulo was born in Queens, not exactly the easiest of areas to grow up. He found his way into Muay Thai kickboxing simply because he got into a street fight, and ended up with a shattered eye socket, a broken nose and a bruised spirit. He didn't want to be afraid anymore. Romulo began his amateur career 1996 at age 21 . In 2003, Romulo began training under Jason Strout. That same year, they worked together to develop Church Street Boxing Gym's first Muay Thai program. Romulo's amateur career included five titles.As a professional fighter, Romulo's record was 13–3 [10 KOs] his accolades include:WKA super middleweight North American title in 2010. US National ChampionshipNorth American ChampionshipBronze medal in Bangkok in the World Cup.After retiring from  a 16 year fighting career, he now "fights" with his words through the "Champ Up" program as a youth empowerment speaker! “People get hung up on the ranking as opposed to the teaching methodology.  Upgrade your teaching methodology instead of getting more techniques.  Instead of collecting new techniques connect the basic techniques so that you can make them look advanced.” -Apolo LadraMy guest this episode is Tuhon Apolo Ladra who has trained in the martial arts for over 40 years.  He is a 7'th degree black belt in Tae Kwon Do, he has taught thousands of students and managed 57 schools.  He is a master instructor of Pekti Tirsia Kali under Grand Tuhon Leo Gaje, and has been inducted int the Black Belt Magazine's Hall of Fame as “Weapons Instructor of the Year”.  Tuhon Apolo is the founder of the iKali and the World Kali Association which is dedicated creating and standardizing curriculum and requirements for students, instructors and school owners while spreading the indigenous art of the Philippines, the Art of the Blade, worldwide.This episode is filled with insight.  We discuss teaching and training methods that are designed to develop students and instructors and uplift the arts.  We talk about Tuhon Apolo's traditional Tae Kwon Do roots and how he transitioned into building one of the largest Kali organizations in the world.  My key takeaway is that continuing education on training methods is what will uplift the arts.  Instead of chasing rankings and techniques, time and effort must be spent on developing your teaching ability so that you can create future high quality instructors.  We also discuss the advantages of teaching by concepts and principles so that students can learn how to find their own unique expression of movement instead of trying to copy their instructors.  Everyone has different attributes and limitations, empowerment lies and finding who you are and how you express yourself.  I thoroughly enjoyed this conversation and I hope you do too.

Fredia "The Cheetah" Gibbs is a former martial artist, kickboxer, model, and boxer, from Chester Pennsylvania. During her kickboxing career, she held ISKA, WKA, and WKF World Titles. Before her kickboxing career, she won three world titles in Tae Kwon Do and was an All-American in basketball and track. Fredia The Cheetah Gibbs made history when she became the first African-American female Kickboxing ISKA World Champion. She earned the name "The Most Dangerous Woman in the World" after an upset in 1994 in her fight against World Champion Valérie Wiet-Henin of France in the "Battle of the Masters" Pay Per View Event in San Jose, California. She went on to become one of the most dominant champions of all time and remains a significant historical figure in light and super lightweight kickboxing divisions. She competed from 1991–1997, amassing a record of 16 wins, 0 losses, and 15 KOs, and three world Titles. Her fight gear is displayed at the Sports Legend of Delaware County Museum, dedicated to preserving the history and memory of Delaware County Sports Legends. She is now an author, celebrity trainer, philanthropist, motivational speaker, and radio host. In 2016–2017 she was honored and participated in the Orange County Heritage Black History Parade and was nominated to attend the United States of Women Summit hosted by the White House. She was named one of the Top Ten Greatest African American Female Athletes of All-time. She is the first female athlete honored with a statue in Pennsylvania and the first female Martial Artist statue on the Planet! The EKN OutLap is a preview podcast show, geared at providing you the top stories, breaking news, and championship battles going into the weekend. This weekend, EKN will be trackside, minus the live portion. EKN Managing Editor David Cole will be at the World Karting Association Charlotte Motor Speedway Karting Challenge to cover the triple-header. The WKA programs for Road Racing, Manufacturers Cup Series, and Speedway Dirt Series will compete at the Concord, North Carolina facility, similar to what was done in the old North American Karting Championships event and what you see at Daytona KartWeek. The annual World Karting Association Daytona KartWeek was able to take place inside the Daytona International Speedway during its traditional December 28-30 time slot to close out racing for the 2020 calendar, beginning the 2021 season. Unfortunately, EKN was able to be trackside as WKA begins to celebrate its 50th season. Thanks to our new Face2Face, and the work of the staff at Margay Racing, EKN was able to connect with the three winners in the Ignite Karting Dash at Daytona program. Gill became a world champion in kickboxing at the age of 21.He formally retired from fighting in 2002. In 1991, he won the World Kickboxing Association (WKA) light middleweight and super welterweight full contact titles.[citation needed] The following year he won the WKA middleweight championship. He was the International Sport Karate Association freestyle champion of 1993. Standing tall at 6 ft 3", Gill has an impressive line-up of British, European and World titles. He is the first UK Asian to be a world champion in a contact sport and became four times World Kickboxing Champion, an achievement that has never been beaten

Unquestionably, the greatest kickboxing champion of all time, Don “The Dragon” Wilson is the only kick-boxer to win 11 world titles in 3 weight divisions (light-heavyweight, super-light-heavyweight, and cruiserweight) and for 6 sanctioning organizations (WKA, STAR, KCIK ,ISKA, PKD, AND IKF). Along the way, he defeated 11 other world champions, 12 number-one contenders and 15 champions on 4 continents. Don’s impressive career record consists of 71 wins, 47 by knockout, and 6 by kick-knockout, and he was also the first kung-fu practitioner to become a World Kickboxing champion. In 1996, Don was named the Greatest Competitor of All Time by Kickside magazine-Europe. As Don’s phenomenal kickboxing career was winding down, he came to Hollywood at the suggestion of his friend and fellow martial artist, Chuck Norris. After hooking up with agent Ray Cavaleri (who is still his agent), Don got a national commercial, and a stint on the soap opera “General Hospital”. Shortly thereafter came a fateful audition with the legendary filmmaker Roger Corman, who had discovered such talents as William Shatner, Jack Nicholson, and Robert DeNiro. Corman was so impressed with Don’s kickboxing career and his natural charm, that he immediately signed him to a non-exclusive seven movie contract, and Don starred in the huge hit, “Bloodfist”. Since Don was still fighting, Corman actually once insured Don’s face for $10 million with Lloyd’s of London. Don has established himself as a hard working Hollywood action star, having starred in 29 films since his film career began in 1989. He has had as many as five films released within a fourteen month period. His most recent project is a movie entitled “Paying Mr. McGetty.”

Das Leukocyten-spezifische Integrin LFA-1 spielt eine wichtige Rolle bei der Immunantwort, durch die Vermittlung dynamischer Zell-Zell- bzw. Zell-Matrix-Interaktionen. Die kontrollierte Adhäsion bzw. Deadhäsion von Leukocyten bedarf einer spezifischen Regulation des LFA-1-Integrins und die Aufklärung der molekularen Grundlagen dieser Vorgänge ist von großem Interesse. Cytohesin-1 war unmittelbar vor Beginn dieser Arbeit als cytoplasmatischer Regulationsfaktor der durch LFA-1 vermittelten Zelladhäsion identifiziert worden und seine spezifische Interaktion mit der cytoplasmatischen Domäne von CD18 konnte in vitro dokumentiert werden. Im Rahmen dieser Arbeit gelang es zunächst, die Assoziation von Cytohesin-1 und LFA-1 auch endogen, im intakten Zellverband, mittels Kolokalisationsstudien in der lymphoblastoiden B-Zellinie LCL-721, zu demonstrieren. Ferner konnte mit Hilfe von Mutationsanalysen die, für die Interaktion kritische Region in der cytoplasmatischen Domäne von CD18 lokalisiert werden. Sie befindet sich im aminoterminalen Bereich und umfaßt die Aminosäuren WKA(723 - 725). Die Mutation dieser Aminosäurereste nach TRG resultierte in einem vollständigen Interaktionsverlust mit Cytohesin-1. Die Inhibition der Cytohesin-1/CD18-Bindung konnte dabei sowohl durch Protein-Protein-Interaktionsanalysen in Hefe als auch durch biochemische Bindungsstudien in vitro dokumentiert werden, wobei jeweils Fusionsproteine der cytoplasmatischen Domäne von CD18 charakterisiert wurden. Funktionale Analysen der WKA(723-725)-Region von CD18 ergaben, daß die Mutation von WKA(723-725) nach TRG im intakten LFA-1-Molekül eine signifikante Reduktion der Integrin- Aktivität zur Folge hatte. Sowohl T-Zellklone als auch nicht hämatopoetische Zellen, wie HeLa, wiesen nach Expression von LFA-1(TRG), mit Hilfe rekombinanter Vaccinia- Viren, eine stark reduzierte Adhäsionsfähigkeit an immobilisiertes ICAM-1 auf. Ferner ergaben funktionale Studien mit HeLa-Zellen, die LFA-1 stabil exprimierten, daß Cytohesin-1 nur dann eine gesteigerte Adhäsion dieser Zellen an ICAM-1 induzierte, wenn sie Wildtyp-LFA-1 exprimierten. HeLa-Zellen, die LFA-1(TRG) exprimierten, ließen sich durch Cytohesin-1 zu keiner verstärkten Adhäsion aktivieren. Diese Ergebnisse demonstrierten die Bedeutsamkeit der Cytohesin-1/CD18-Interaktion für eine effiziente, durch LFA-1 vermittelte Zelladhäsion. Unklar war jedoch der Mechanismus, durch den Cytohesin-1 die Integrin/Liganden-Bindung regulierte. Studien mit dem Reporterantikörper 24 ließen darauf schließen, daß Cytohesin-1 durch die Bindung an CD18 eine Konformationsänderung in der extrazellulären Domäne des LFA-1-Integrins induzieren konnte, die möglicherweise die Affinität des Rezeptors modulierte. Diese Modulation der LFA-1-Konformation schien jedoch nicht hinreichend für eine stabile Bindung an ICAM-1 zu sein, wie eingehendere Analysen von Dr. W. Kolanus zeigten. Vielmehr erforderte eine effiziente Zelladhäsion zusätzlich die Guaninnukleotid-Austauschfunktion (GEF-Funktion) von Cytohesin-1, da die GEF-defekte Punktmutante, Cytohesin-1(E157K), nicht mehr in der Lage war, die Adhäsion von Jurkat E6-Zellen an ICAM-1 stabil zu induzieren. Biochemische Interaktionsstudien konnten dabei zeigen, daß die Mutante weiterhin fähig war, die cytoplasmatische Domäne von CD18 zu binden. Diese und weitere Ergebnisse von Dr. W. Nagel, die einen Zusammenhang zwischen der GEF-Funktion von Cytohesin-1 und dem „Spreading“ von adhärenten Jurkat E6-Zellen aufzeigten, legen die Vermutung nahe, daß Cytohesin-1 durch einen dualen Mechanismus in die LFA-1-Regulation involviert ist. Sowohl die direkte Interaktion von Cytohesin-1 und dem Integrin als auch seine GEF-Funktion stellen essentielle Faktoren für eine stabile Zelladhäsion, die durch LFA-1 vermittelt wird, dar. Welche funktionalen Mechanismen dabei durch den Guaninnukleotid-Austausch und der damit verbundenen Aktivierung einer GTPase induziert werden, ist noch unklar. Primär wäre eine Modulation des Aktin-Cytoskelettes und eine damit verbundene erhöhte laterale Mobilität der Integrine denkbar, die eine verstärkte Rezeptormultimerisierung und dadurch eine Aviditätsänderung des Integrins ermöglicht. Weitere Studien dieser Arbeit analysierten die Regulation von Cytohesin-1 selbst. Es konnte gezeigt werden, daß PI3-Kinase in die Kontrolle der Cytohesin-1-Funktion involviert war. Die Überexpression einer konstitutiv aktiven Form dieser Kinase (P110*) führte zu einer gesteigerten Adhäsion von Jurkat E6-Zellen an ICAM-1. Eine Inkubation dieser Zellen mit dem PI3-Kinase-spezifischen Inhibitor Wortmannin resultierte dagegen in einer signifikanten Reduktion der Zelladhäsion. Weitere funktionale Analysen, die die Zelladhäsion von Jurkat E6-Zellen nach Koexpression von P110* und der PH-Domäne von Cytohesin-1 untersuchten, sowie eingehendere Studien von Dr. W. Nagel, ermöglichten die Entwicklung eines Modells zur Regulation von Cytohesin- 1. Demzufolge führt die Aktivierung der PI3-Kinase zu einer verstärkten Rekrutierung von Cytohesin-1 an die Plasmamembran. Als Rekrutierungsmodul fungiert dabei die PHDomäne, die durch Bindung von PtdIns(3,4,5)P3, einem Produkt der PI3-Kinase, die Assoziation mit der Membran gewährleistet. Die Rekrutierung von Cytohesin-1 an die Plasmamembran führt zur Aktivierung von LFA-1 und der damit verbundenen stabilen Zelladhäsion an ICAM-1.
