Believing that her future was in the ministry, Kristen Carlone Hurley saw a future in divinity school with a husband and kids somewhere in the Northeast. When her parents blindsided her with the news of their divorce in just before her junior year and she was going through relationship issues of her own, her faith was shaken. She dealt with a period of depression and first put the divinity school on hold until she realized she needed another path. After regrouping, she took opportunities as they came, no matter how divergent from the original ideal. Ultimately where she landed diverged quite a bit.In this episode, find out from Kristen how sometimes exercising faith means knowing you'll find something good where it is, not necessarily where you thought it would be…on today's Roads Taken with Leslie Jennings Rowley. About This Episode's GuestKristen Carlone Hurley followed opportunities as they came about, ultimately finding herself in the financial sector. She lives in Florida with her husband, enjoying the sunshine and living her bliss. T., Townsend, K. L. Abstract: Telomerase reverse transcriptase (TERT) is expressed by quiescent adult stem cells (ASC) in numerous adult murine and human tissues, but has never been explored in the adult brain. Here, we demonstrate that TERT+ cells in the adult mouse brain represent a novel population of multipotent ASCs that are localized to numerous classical neuro/gliogenic niches (including the ventricular-subventricular zone, hypothalamus, and olfactory bulb), as well as more recently described regions of adult brain plasticity such as the meninges and choroid plexus. Using a direct-reporter mouse line, we found that TERT+ cells expressed known neural stem cell markers such as Nestin and Sox2, but not markers of committed stem/progenitor cells, nor markers of mature neuronal or glial cells. TERT+ ASCs rarely expressed the proliferation marker Ki67, and in vitro TERT+ cells lost TERT expression when activated by growth factors, together indicating a quiescent phenotype similar to what has been observed in other tissues. When cultured, TERT+ cells behaved like neural stem cells by forming neurospheres, which could proliferate and become more metabolically active once stimulated by growth factors. TERT+ cells were observed in numerous brain niches, particularly near the ventricles and cerebrospinal fluid barriers, but notably, TERT+ cells were never observed in the hippocampus. Lineage tracing of TERT+ cells in adult transgenic mice (mTERTrtTA::oTET-Cre::RosamTmG) revealed large-scale expansion of TERT+ progeny and differentiation to diverse cell types in multiple brain regions. For example, lineage-traced cells expressed markers of mature neurons, oligodendrocytes, astrocytes, ependymal cells, and choroid epithelial cells, thus demonstrating the striking multipotency of this stem cell population in basal tissue turnover of the adult brain. Together, these data demonstrate that TERT+ cells represent a novel population of multipotent stem cells that contribute to basal plasticity and regeneration in the adult mouse brain. Some career superintendents make their marks by hosting major tournaments or pushing the agronomic envelope, while others quietly go about their business of growing great grass while also navigating the potholes and pitfalls that invariably spring up over time. John Carlone, CGCS, has done that over 37 years as a golf course superintendent at two clubs in one of the toughest markets in the industry: Long Island, NY. Fresh out of the University of Rhode Island (where he had a notable roommate, Frank Rossi), Carlone went to work for Scott Niven at the Stanwich Club in 1983 before taking over the reins at Middle Bay Country Club in 1985. He stayed there until 1996 when he left for Meadow Brook Club on the north shore of Long Island, where he has been ever since. In this conversation with Peter McCormick, Carlone offers hard-earned lessons about family, dedication, staff, club politics, humility, swallowing one's pride when needed and "not mailing it in". A fascinating chat with a good friend of TurfNet for many years and our latest All Star of Turf.

Lorraine Carlone was married for more than 30 years to a malignant narcissist who terrorized her while she raised two daughters and did everything she could to try to please him and save her marriage. In this gripping podcast, she shares her story with poignant honesty because she is now on a mission to save others from being in similar toxic relationships. Lorraine was one of my actual clients and this is one of the most important interviews I have ever done. It should be required listening to anyone who feels trapped in an abusive relationship. She details how she tried to leave once without a plan and had to go back. Then how she made a plan and was actually able to leave. How she finally escaped; how she got stronger; how she negotiated and got exactly what she wanted, even from one the most difficult situations and seemingly unwinnable cases. Grab your pad, your pen, your tissues, then listen and listen again. More About Lorraine Carlone A native of Naples, Lorraine Carlone grew up spending her days in Pine Ridge Estates, riding her horse down to Vanderbilt Beach. She has seen every part of the sleeping fishing town grow, from back when nothing existed beyond Airport Road to becoming one of the nation's best places to live. Combining her knowledge of the Naples area with her background in construction and interior design, Lorraine's expertise is unparalleled in making the process seamless for her customers. Prior to becoming a real estate professional, Lorraine spent more than 15 years working in interior design and 20 years working in the construction industry. Upon graduating from The Art Institute of Ft. Lauderdale, she partnered with her mother in a custom drapery business before opening her own luxury decorating firm, Lorraine Carlone Interiors. Next, Lorraine worked alongside her husband in his electrical service and contracting company, serving as both the chief financial and chief operating officer. Under her guidance, the company grew from two employees to over 100, with revenues of more than $10 million. Lorraine spent 20 years educating herself in business finance and operations, becoming a lighting control engineer in the process. The business, which Lorraine and her husband sold several years ago, became one of the most esteemed electrical contracting companies in Southwest Florida. Becoming a licensed real estate professional was the next logical step for Lorraine in pursuing her dreams of helping people achieve their next step in life. Having spent more than 50 years in the Naples area, with 30 of those dedicating herself to design and construction, her knowledge is unmatched. Her expertise offers her customers peace of mind, and a confidence that it's not just a house that is being sold or purchased - it's a home. Whether you are a permanent resident, relocating to Naples or a seasonal visitor, Lorraine will dedicate herself to finding your family the perfect fit. The proud mom of two daughters, Lorraine enjoys early morning walks and spending late afternoons on the beach until sunset. She is active in the community, volunteering for the Shelter for Abused Women & Children, Collier County Honor Flight and Golden PAWS Assistance Dogs. An avid horseback rider, Lorraine specializes in equestrian estates and farms, with a highlighted specialty in hunter/jumper dressage. Fluent in Spanish, Lorraine is a member of Florida REALTORS®, the Naples Area Board of REALTORS® and the National Association of REALTORS®. This week we hear the teacher's perspective from “the other side of the camera,” as Dr. Carlone describes what it's been like to teach during the pandemic, what has been most challenging, what's been successful, and how she has grown professionally. The conversation also touches on topics including learning loss and social-emotional health, which have been prominent in recent education discussions. About the host: Dr. Wendy Oliver is a highly qualified Tennessee teacher and administrator who has pioneered digital learning across multiple states and school districts. She has authored digital teaching standards and developed software that allows teachers to self-assess their knowledge of digital instruction. "It's only an island if you look at it from the water". Amity Island's Police Chief Brody from the movie JAWS had his own way of rationalizing the insanity that can come with being the man in charge on a small rock surrounded by water. In the game-changing 1976 Blockbuster we're treated to a thumbnail sketch of some of the more mundane aspects of an island Police Chief's life. Block Island's Chief of Police, Vin Carlone, has a full plate as well. And while he hasn't had to hunt down a man-eating Great White Shark...yet, that task might seem more appealing (and possibly easier) than attending a Town Council meeting.Imagine being responsible for keeping the peace on an island where the summertime population can swell to over 20,000 people all looking for a good time. Keep in mind- everyone's definition of a "good time" is different. He has only 6 full-time officers to cover the beat. Even with an additional 6-7 auxiliary summertime officers (all retired cops over the age of 50) brought in to help during the peak season it's an impossible task. One would have to possess superhuman strength to handle it all. And the Chief is indeed strong- both physically and mentally. But every man has his limits. And Vin Carlone at the end of the day is not a superhero. He is but a man. A man who oftentimes finds himself at ideological odds with those he has sworn to protect. His relationship with the residents of Block Island can be complicated. But his feelings about Block Island are not. He loves this place. The headaches associated with being the Chief in a small town are far outweighed by the benefits of existing in such a close-knit community. In Episode Six we dive right in with Chief Vin Carlone, discussing how his tenure as head of the Detective Division across the water in Narragansett prepared him to deal with Block Island's seasonal quirks. Lessons that Vin learned in Narragansett were applied here in order to turn the tide on Block Island's reputation as "the place where anything goes".Marc recounts his first meeting with Vin and how that meeting led to a long partnership between Block Island's nightlife establishments and the Police force in an effort to reel in what some felt was an "out of control" party scene. But what exactly makes Chief Carlone tick? Rich and Marc explore the psyche of a veteran law enforcement official and discover that what can be misconstrued as a hard-boiled exterior is oftentimes a necessity rather than a choice. Vin explains the need to compartmentalize certain aspects of his job in order to maintain some semblance of normalcy in day to day life. Fair warning- the conversation gets heavy at times.Yet for all the good our Chief of Police has done for the community, it can be a thankless job. Vin discusses how he deals with the feeling that some of his efforts aren't necessarily appreciated. Spoiler alert- he doesn't. Chief Carlone sticks to his assertion that it's better to present the facts, accept what may be undue criticism and be disliked rather than play the politician in an attempt to make everyone happy. It's not all doom and gloom though. There's plenty of comedic relief that comes with the job. In one example, Vin recounts the story of an inebriated, birthday suit-clad individual who makes multiple bad decisions over the course of the tale.So hop in the passenger seat as we ride along with Vin Carlone, Block Island's Chief of Police. Don't make us hit you with the taser! Even if we have to slap the cuffs on ya'- reach up and hit "play", Cap'!

Caroline Basyn, Global CIO and GBS Officer at Mondelēz International, discusses the importance of setting a solid foundation before planning and executing a digital transformation. With an influx of data comes new problems to be solved. Instilling a cloud and data strategy, setting up a consumer-centric framework, and building innovation teams out of siloed departments are some of the solutions Basyn touches on. Once the foundation is set, deploying corporate culture changes, Agile workplaces, and AI and ML technology gets done quicker, cheaper, and with more longevity than a quick-fix solution. “I used to say quality is not the enemy of speed, but in a certain sense, you need to be able to marry both.”

Woody Allen's family and Frank Sinatra's family are both troubled and possibly related. Perhaps they could have used better childhoods and they wouldn't have turned out so weird. It's not a secret that bad childhoods often produce broken people, and childhoods are, on average, getting worse. See omnystudio.com/listener for privacy information. Put on your red hats and khakis, it's time for our Nu-Metal episode! We discuss this ever-polarizing sub-genre/moment in time with our good pal Peter Carlone of award-winning comedy duo Peter n' Chris, just in time to pick his wedding music! The three of us travel all the way back to the year 2000 (or slightly earlier), where combining misspelled words to make your band name and dressing in weird pants was the order of the day. Discussion includes: the electronic scrapings and Seinfeld bass on Mudvayne's 'L.D.50'; how Limp Bizkit's 'Significant Other' makes Fred Durst seem whiter than the cast of Full House; Korn's 'Follow The Leader' and that time Peter went to a Korn show with a bunch of grandmas; how Linkin Park's 'Hybrid Theory' is as awesome as it is annoying; and Devin loves to hate how much Fraz loves Papa Roach's 'Infest'. We also go through a master list of nu-metal bands, go deep on the crossover between Nu-Metal and Christian metal, and unleash a virtual army of homing pigeons. It's one for the ages, y'all. LOOK INTO DEEZ EYEZ!
