Frizzled receptors look like GPCRs — but the field is split on whether they actually are. Gunnar Schulte has spent 25 years building the case, one receptor at a time.Schulte is a professor at Karolinska Institute in Stockholm, where his lab investigates Wnt–frizzled signaling at the molecular level. His research maps G-protein coupling specificity across all 10 frizzled subtypes, develops conformational biosensors to detect receptor activation, and searches for small molecules that could finally make frizzled receptors pharmacologically tractable. In this conversation, he walks through the evidence — what the conformational data show, why the Wnt ligand problem has stalled the field for decades, and how a compound originally designed for Smoothened became the closest thing frizzled pharmacology has to a starting point. Why each frizzled subtype couples to a different G protein — and why that distinction changes how the field should think about targeting themWhy 19 Wnt ligands remain almost impossible to work with, and what the lipid modification problem costs drug discoveryHow SAG1.3, a Smoothened agonist, became the first small molecule to activate frizzled 6 as a partial agonistWhat conformational sensors reveal about frizzled activation — and why Schulte considers this his strongest argument for their GPCR identityHow disheveled and G proteins may compete for receptor access through conformational selectionDr. GPCR Ecosystem: https://www.ecosystem.drgpcr.com/Membership & Pricing: https://www.ecosystem.drgpcr.com/university-pricingWeekly News: https://www.ecosystem.drgpcr.com/gpcr-weekly-news

In this episode, we sit down with Lauren Schulte Wang, founder and CEO of The Flex Co., the brand behind the menstrual disc that has sold over 185 million units, holds 88% market share in its category, and is now carried in over 30,000 retail locations including Target, Walmart, and CVS. Lauren built Flex from scratch in 2016, creating an entirely new product category with zero search volume and a consumer behavior change challenge that she describes as one of the hardest jobs a marketer can have. She opens up about how obsessive customer listening shaped their entire go-to-market strategy, what it took to fight platform by platform to earn the right to advertise a period product on social media, why word of mouth is still their strongest acquisition channel after nearly ten years, and how reaching profitability in year seven gave Flex the agency to put customers above all else. Key Takeaways:// Changing human behavior is the hardest job in marketing. Flex isn't asking someone to switch brands, they're asking them to rethink something deeply personal. That challenge shaped every part of how they educate, nurture, and retain customers.// Retail shelf space is a billboard. Their target customer is in that aisle every single month. Lauren's strategy: build awareness through influencers and streaming TV, then let the shelf close the deal.// Word of mouth is still their number one acquisition channel, after ten years. Building a product people feel compelled to tell their friends about is not a soft metric. It is a growth strategy.// Profitability is about agency. Reaching profitability in year seven gave Flex the ability to make decisions that put customers first without being beholden to investor timelines or the next raise.// 88% market share and 3% awareness is the opportunity, not the problem. Most brands would panic at low awareness. Lauren sees it as the greatest runway ahead.Connect with Lauren: LinkedInLearn More: Website____Join the MHH Collective! In this episode of the Epigenetics Podcast, we talked with Tomohisa Toda from the Max-Planck-Zentrum für Physik und Medizin about his work on the long-term maintenance of neuronal identity, with a focus on epigenetic and RNA-based mechanisms in brain stability and aging. Dr. Toda describes how brain circuits are stabilized over time, why critical periods are temporally restricted, and how epigenetic regulation may help maintain established neural identity. This led him to postdoctoral work on neural stem cells and long-term maintenance. We cover his work on nuclear pore and nuclear lamina proteins, including NUP153 and Lamin B1. He explains that NUP153 is enriched in neural stem cells and appears to act as a platform for recruiting factors that help maintain the stem cell epigenome. For Lamin B1, we discuss its decline during aging, how its loss can lead to stem cell exhaustion, reduced adult neurogenesis, and age-related mood dysregulation in the hippocampus. We also discuss LINE-1 RNA, where we learn that reducing LINE-1 promotes neural progenitor differentiation. He explains that this effect is linked to the RNA sequence itself rather than retrotransposition, based on rescue experiments. Finally, we talk about his finding that a subset of postnatally born brain cells contains nuclear RNAs that remain detectable for up to two years. He describes their nuclear enrichment, possible association with heterochromatin, and ongoing work to understand their sequence features, modifications, and biological function.

Welcome to Season 3, Episode 13 of A Friend for the Long Haul - A Long Covid Podcast! In this episode, I got to connect with Heather Schulte. Heather is the powerhouse behind Stitching the Situation. From her website: "Stitching the Situation is a continuation of artist Heather Schulte's textile work, Situation Report, a daily cross-stitch documentation of the coronavirus case and death counts in the U.S. from Jan 20-June 25. The Situation Report panels began as the artist's way to record cases in the US, and translate them visually with stitch. "We discuss a unique art exhibition called "It Comes in Waves," centered around the impacts of COVID-19. Set in The People's Building, an art gallery located on the east side of Aurora, Colorado, Heather took me on a tour of the displayed art pieces, which include drawings, dye sublimation prints, and masks. The exhibition, organized by Heather, features a mix of personal stories and collective experiences related to living with Long Covid. Special attention is given to the ‘Stitching the Situation' project, which involves community participation in cross-stitching data and personal stories onto cloth. The discussion also highlights the incorporation of art from incarcerated individuals, collaborations with other artists, and the logistical challenges of managing such a significant project. I'm absolutely thrilled that this podcast is included in the exhibit. I called my portion "A Life Redacted," and it is made up of excerpts of episodes with words redacted from the transcripts. It Comes in Waves illustrates the power of art in documenting and processing collective trauma.
