BUFFALO, NY- June 5, 2024 – A new research paper was published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 10, entitled, “Serine racemase expression profile in the prefrontal cortex and hippocampal subregions during aging in male and female rats.” Aging is associated with a decrease in N-methyl-D-aspartate (NMDA) receptor function, which is critical for maintaining synaptic plasticity, learning, and memory. Activation of the NMDA receptor requires binding of the neurotransmitter glutamate and also the presence of co-agonist D-serine at the glycine site. The enzymatic conversion of L-serine to D-serine is facilitated by the enzyme serine racemase (SR). Subsequently, SR plays a pivotal role in regulating NMDA receptor activity, thereby impacting synaptic plasticity and memory processes in the central nervous system. As such, age-related changes in the expression of SR could contribute to decreased NMDA receptor function. However, age-associated changes in SR expression levels in the medial and lateral prefrontal cortex (mPFC, lPFC), and in the dorsal hippocampal subfields, CA1, CA3, and dentate gyrus (DG), have not been thoroughly elucidated. In this new study, researchers Linda Bean, Prodip K. Bose, Asha Rani, and Ashok Kumar from Indiana University School of Medicine, North Florida/South Georgia Veterans Health System, and the University of Florida aimed to determine the SR expression profile, including protein levels and mRNA, for these regions in aged and young male and female Fischer-344 rats. Their results demonstrate a significant reduction in SR expression levels in the mPFC and all hippocampal subfields of aged rats compared to young rats. In the 53rd episode of Hockey Te Charcha, we sat down with Shri Ashok Kumar, the son of the legendary Major Dhyan Chand and recipient of the Hockey India Major Dhyan Chand Lifetime Achievement Award at the Hockey India 6th Annual Awards 2023. Shri Ashok Kumar was part of the greatly revered side that won India's only World Cup title, against Pakistan in 1975. He was the one who scored to complete the 2-1 victory over Pakistan in the Final. Additionally, Ashok is a proud recipient of the prestigious Arjuna Award in 1974. In this episode, Ashok Kumar expressed his emotions upon receiving the Hockey India Major Dhyan Chand Lifetime Achievement Award. He took us through the nerve-wracking 1975 World Cup Final against Pakistan through his eyes and shared anecdotes from the life of his father, Major Dhyan Chand. Ashok Kumar also shed light on the hardships present in Indian Men's Hockey Team midfielder Vivek Sagar Prasad's battle while training under him and outlined his expectations from the current squad at the Paris 2024 Olympics.

Tom Blenkinsop was born  in Middlesbrough, brought-up in Marton and graduated from Teesside University with a BSc in Philosophy, Politics and Economics, and the University of Warwick with an MA in Continental philosophy.Tom joined  the Army Reserve in 2017 in the Royal Military Police in which he still serves today, and was an MP whilst training to be an RMP, something we explore in this episode.Tom worked as a constituency researcher for Ashok Kumar, Labour MP for Middlesbrough South and East Cleveland, from 2002 to 2008. He became a campaign manager for the Community Trade Union in 2008 and continued in the role until his election to Parliament in 2010.Tom was selected as the Labour candidate for Middlesbrough South and East Cleveland in April 2010, following Ashok Kumar's unexpected death the previous month. He was elected at the May 2010 general election. In his first Parliamentary term, he was a member of the Environment, Food and Rural Affairs Select Committee from 2010 to 2012, the Standards and Privileges Committee from 2010 to 2011 and the Treasury Select Committee in 2011. He joined the opposition front bench under the leadership of Ed Miliband, serving as a whip from 2011 to 2015.He was re-elected at the 2015 general election. During his second Parliamentary term, he was a member of the Energy and Climate Change Committee from 2015 to 2016, the Privileges Committee from 2015 to 2017, the Standards Committee from 2015 to 2017, and the Northern Ireland Affairs Committee from 2016 to 2017.After the calling of the 2017 general election, Blenkinsop announced that we would not seek re-election as an MP as he could not campaign for Labour whilst Corbyn served as party leader.  Again, this is something we discuss at length in the podcast.Blenkinsop returned to the Community union as a London-based project manager after leaving Parliament, and became a public affairs advisor for the Federation of Small Business in 2020. We thank Tom for sharing his story with us!Support the show✅Support The Show Help Us Grow! Pradyuman Thakur was just seven years old. Barun Thakur, ever the devoted dad, had driven both kids to the gate of their private school. In less than ten minutes, Pradyuman would be dead. He had been stabbed twice in the neck before being rushed to the hospital. Allegedly, the school did not tell his parents about Pradyuman's injuries or call the police. By noon the same day, the local police arrested Ashok Kumar - it was reported that the confession was tortured out of him and he was . A month later, the CBI arrested a sixteen-year-old student. Serine Racemase Upregulation Improves Learning and Synaptic Function

A new research paper was published in Aging (Aging-US) Volume 15, Issue 7, entitled, “Viral vector-mediated upregulation of serine racemase expression in medial prefrontal cortex improves learning and synaptic function in middle age rats.” An age-associated decrease in N-methyl-D-aspartate receptor (NMDAR)-mediated synaptic function contributes to impaired synaptic plasticity and is associated with cognitive impairments. Levels of serine racemase (SR), an enzyme that synthesizes D-serine, an NMDAR co-agonist, decline with age. In this new study, researchers Brittney Yegla, Asha Rani and Ashok Kumar from the University of Florida's McKnight Brain Institute predicted that enhancing NMDAR function via increased SR expression in middle age (when subtle declines in cognition emerge) may enhance performance on a prefrontal cortex-mediated task sensitive to aging. “We hypothesized that augmenting SR expression within mPFC glutamatergic neurons would improve attention and cognitive flexibility in middle-aged rats and facilitate synaptic responses in the mPFC. Thus, for this study, SR expression was upregulated in pyramidal neurons of the mPFC through lenti-viral technology to enhance NMDAR function and evaluate its impact on cognitive flexibility and NMDAR-mediated synaptic transmission in middle-age rats.” Middle-aged (~12 mo) male Fischer-344 rats were injected bilaterally in the medial prefrontal cortex (mPFC) with viral vector (LV), SR (LV-SR) or control (LV-GFP). Rats were trained on the operant attentional set-shift task (AST) to examine cognitive flexibility and attentional function. LV-SR rats exhibited a faster rate of learning compared to controls during visual discrimination of the AST. Extradimensional set shifting and reversal were not impacted. Immunohistochemical analyses demonstrated that LV-SR significantly increased SR expression in the mPFC. Electrophysiological characterization of synaptic transmission in the mPFC slices obtained from LV-GFP and LV-SR animals indicated a significant increase in isolated NMDAR-mediated synaptic responses in LV-SR slices. Thus, results of the current study demonstrated that prefrontal SR upregulation in middle age rats can improve learning of task contingencies for visual discrimination and increase glutamatergic synaptic transmission, including NMDAR activity. “The results from this study support the beneficial effects of the D-serine pathway involvement in NMDAR-mediated transmission and cognitive function, expanding the literature to emphasize its role in not only the hippocampus but also the PFC. The new outbreak of the virus put the spotlight on the garment district of the city - where the new outbreak may have emerged - much of which is characterised by extremely exploitative labour practices in small factories serving giant online retailers such as Boohoo and major high street brands. I spoke to Ashok Kumar about the reshoring of garment production in the UK, how Covid-19 has affected and changed the manufacturing of clothes in both the global north and south, and how fast fashion helps to prevent increased investment in technology and serves to maintain labour-intensive forms of production.

This podcast highlights the updated clinical practice guideline on adult sinusitis that is published as an April 2015 supplement to Otolaryngology - Head and Neck Surgery, the official journal of the American Academy of Otolaryngology - Head and Neck Surgery (AAO-HNS) Foundation. In the first of two podcasts for this supplement, Editor in chief John Krouse is joined by authors Ashok Kumar and Maggie Kramper in discussing key findings from the guideline in the diagnosis and treatment of adults with symptoms of sinusitis and their implications for primary care health providers and patients.   Sinusitis affects about 1 in 8 adults in the United States, resulting in more than 30 million annual diagnoses. More than 1 in 5 antibiotics prescribed in adults are for sinusitis, making it the fifth-most common diagnosis responsible for antibiotic therapy.  Many symptoms of sinusitis can overlap with other common conditions such as allergic rhinitis, and can complicate the diagnosis and treatment of these individuals.  Common symptoms of acute sinusitis include purulent nasal discharge accompanied by nasal obstruction, facial pain/pressure/fullness, or both. Patients and their health providers can utilize a variety of approaches in treating these symptoms, judiciously using antibiotics and other prescription and over-the-counter medications as indicated.  These guidelines discuss recommendations and options for clinicians treating adult sinusitis and their patients.
