With many organoid models emerging on the market there is growing need to develop methods to adapt organoids to the established work flows and assays that are the foundations of modern research.  A method was developed for processing, embedding, and sectioning Organ-Chips for FFPE workflows. Organ-Chips were treated with Histogel and trimmed to fit standard sized cassettes. Followed by processing and embedding as usual before proceeding to the microtome. The resulting slides have successfully been stained with hematoxylin and eosin and immunohistochemistry, as well as shown promising results for use on multiplexing platforms. Although the process is laborious and requires continued refinement the slides should be sufficient for highplex proteomics and transcriptomics methods and can be used in addition to FFPE tissues for research into immunology, oncology, and virology.

Welcome to The Nonlinear Library, where we use Text-to-Speech software to convert the best writing from the Rationalist and EA communities into audio. This is: Malaria Vaccine Research Help Needed, published by joshcmorrison on December 30, 2023 on The Effective Altruism Forum. We at 1Day Sooner posted recently about scoping a campaign to push for an accelerated rollout of the newly approved R21/Matrix-M malaria vaccine. The vaccine was recently prequalified by the WHO, a key step on the critical path to vaccine distribution, but much remains to be done. We greatly appreciate the more than a dozen people who reached out to help after our last post. Their work was invaluable for producing our December Malaria Vaccination Status Report, the development of which has been critical to improving our understanding of the problem. Our colleague Zacharia Kafuko's op-ed as well as Peter Singer's on the subject are also both good sources for further reading. We plan to publish a new status report every month and maintain a rolling public comment version to reflect our latest understanding of the issue and use as a sort of global workspace to share the most critical information about obstacles and enablers for widespread distribution. To make our research work for this more sustainable we're moving to a pool system where members sign up for at least four days out of the month where they will be assigned a 1-2.5 hour research or writing task to update and improve our status report document. Pool members will be paid $100 per pool day. (Here is a punch list of the type of goals we have for our next draft. here.). We are looking to add 5-10 new pool members for January beyond those who signed up last month. If you're interested in helping, please email ryan.duncombe@1daysooner.org. Questions and comments are very welcome. Thanks! Thanks for listening. To help us out with The Nonlinear Library or to learn more, please visit nonlinear.org

On today's episode, we revisit a conversation we had with two pioneers of the mRNA-based Covid-19 vaccines, Katalin Karikó, PhD, and Drew Weissman, MD, PhD. The two were just awarded the 2023 Nobel Prize in Physiology or Medicine for their groundbreaking research in messenger RNA (mRNA) technology. When we spoke with Dr. Kariko and Weissman in June 2022, they had just received the Ross Prize in Molecular Medicine. The Feinstein Institutes for Medical Research grants this honor to recognize promising careers in the fields of science and research. In this look back on that conversation, the history-making duo discusses what motivated them to endure their decades-long scientific struggles, plans to continue their mRNA research and what it means to be awarded the Ross Prize. Also on the show, we speak with Kevin Tracey, MD, president of the Feinstein Institutes for Medical Research, about the importance of supporting young scientists, early-stage research and the culture of science. Meet the experts Katalin Kariko, PhD, adjunct professor of Neurosurgery at the University of Pennsylvania and a senior vice president at BioNTech Drew Weissman, MD, PhD, the Roberts Family Professor of Vaccine Research in Penn's Perelman School of Medicine The award Drs. Kariko and Weissman received is one of six categories that are recognized each year. Atlantic Salmon and Maine's renewable wood pulp industry may sound like an unlikely duo at first, but Deborah Bouchard and Sarah Turner from the Aquaculture Research Institute (ARI) have recognized the potential this pairing could have for the future of environmental, economic, and social sustainability for the US aquaculture industry. Debbie and Sarah Bouchard are to researching a new generation of safer, more sustainable, and cheaper vaccines for finfish. Here is where Atlantic Salmon meets its unlikely match, wood pulp. This abundant polymer is biocompatible, biodegradable, quite versatile, and easily modified – making it a phenomenal candidate for drug delivery. Tune in to learn more and hear about their results so far! and Vibrio ordalii. This adjuvant, a component of vaccines to improve and prolong immuneresponse, showed no evidence of cellular damage and no effects on growth for the fish whileinducing a strong immune and antibody boosting response. With less side effects and cheaper tomanufacture than existing vaccines these preliminary results are exciting, highlighting the efficacyof nanocellulose adjuvants. ISAv and V. ordalii alone can cause farmers to lose 90% of their stocks, amounting to over $1billion annually. Development of this new, safe, and affordable vaccine using nanocellulose is notonly huge for Atlantic salmon farmers but other food production systems as well. This newgeneration of low cost, tunable vaccine formulation is paving a way toward more sustainableaquaculture, with potential applicability for other fish species and even terrestrial animals as well. Leveraging direct industry support from Cooke Aquaculture, Kennebec River Biosciences,Benchmark Animal Health LTD, this interdisciplinary team from UMaine's Aquaculture ResearchInstitute and the Chemical and Biomedical Engineering Department has immense experience andresources relating to fish health, immunobiology, and vaccine formulation. Results will be shareddirectly through ARI's web site, Cooperative Extension and Maine Sea Grant extensionprofessionals, national conferences, and peer-reviewed publications.
