Sabina Kaczanowska's research focus is to understand the role of the immune system in shaping the microenvironment of metastasis during cancer progression. Specifically, she is investigating how myeloid cells orchestrate the immunosuppressive program in the pre-metastatic niche and how we can apply this knowledge of myeloid biology to design new immunotherapeutic strategies for the treatment of solid tumors. She joined us on OsteoBites to discuss her recently published paper on Immune determinants of CAR-T cell expansion in solid tumor patients receiving GD2 CAR-T cell therapy.https://pubmed.ncbi.nlm.nih.gov/38134936/Chimeric antigen receptor T cells (CAR-Ts) have remarkable efficacy in liquid tumors, but limited responses in solid tumors. The NCI conducted a Phase I trial (NCT02107963) of GD2 CAR-Ts (GD2-CAR.OX40.28.z.iC9), demonstrating feasibility and safety of administration in children and young adults with osteosarcoma and neuroblastoma. Patient samples were evaluated by multi-dimensional proteomic, transcriptomic, and epigenetic analyses. The data uncover mediators of CAR-T biology and correlates of expansion that could be utilized to advance immunotherapies for solid tumor patients.https://clinicaltrials.gov/study/NCT02107963Dr. Sabina Kaczanowska received her Ph.D. in Molecular Microbiology and Immunology from the University of Maryland, Baltimore for her dissertation work on T-cell immunotherapy at the UM Marlene and Stewart Greenebaum Comprehensive Cancer Center. She joined the Tumor Microenvironment and Metastasis Section of the Pediatric Oncology Branch at the National Cancer Institute as a postdoctoral fellow in 2017 with Dr. Rosandra Kaplan. Dr. Kaczanowska was appointed as a Staff Scientist in the Pediatric Oncology Branch in 2022 to continue her work evaluating the responses of solid tumor patients to immunotherapy and translating new immuno-oncology approaches into the clinic.

Bioinformatics is a relatively new field of science that is very interdisciplinary in nature. Its practitioners use a mixture of biology, chemistry, physics, statistics, and computer science to develop methods and software aimed at helping integrate and understand biological and other data.  Our guest for this episode is Nikhil Ram Mohan, Staff Scientist at the Stanford University School of Medicine. He describes bioinformatics as the bridge to understanding biology. We learn about his international studies and path that brought him to this current role and field of study, and then dive into some of his recent work. Here he and his team analyze biobank samples using digital PCR (dPCR) and quantitative PCR (qPCR) and compare results from the two while correlating results with additional data available for each sample to determine if SARS-CoV-2 RNA detection and quantification in blood can serve to help predict potential for patient coinfection. Their work found that dPCR was able to detect SARS-CoV-2 in samples that were negative when evaluated by qPCR and that a series of biomarkers can help predict coinfection.   We also get to hear a bit of Nikhil's interesting personal story, which includes his undergraduate engineering studies in India and leaving his native country for the first time when he moved to the U.S. for graduate school.  We learn how he managed changes in culture, what he loves about teaching, and about him being a new father.  Visit the Absolute Gene-ius page to learn more about the guest, the hosts, and the Applied Biosystems QuantStudio Absolute Q Digital PCR System.

In this week's episode, I speak with Damien Desfontaines, also known by the pseudonym “Ted”, who is the Staff Scientist at Tumult Labs, a startup leading the way on differential privacy. In Damien's career, he has led an Anonymization Consulting Team at Google and specializes in making it easy to safely anonymize data. Damien earned his PhD and wrote his thesis at ETH Zurich, as well as his Master's Degree in Mathematical Logic and Theoretical Computer Science.Tumult Labs' platform makes differential privacy useful by making it easy to create innovative privacy and enabling data products that can be safely shared and used widely. Samuel Minot is a Staff Scientist with the Microbiome Research Initiative in the Vaccine and Infectious Disease Division of the Fred Hutch Cancer Research Center. He begins this conversation with a foundational question in microbiology: how do the microbes on us impact our health?  He then discusses the complexity of the interaction of bacteria and the viruses that infect them, also called phages. He offers examples of new discoveries constantly upending our picture of what viruses and bacteria can do and ways infectious viruses impact some bacteria and cause disease. As an example, he discusses cholera, which is harmful because of a satellite virus that infects a bacterium: the two together make the disease. He then lays the ground for why it is important to prioritize a gene-level study of our microbiome by describing the impossibility of culturing every bacterium. He describes what functional annotation is and how that concept allows him to identify genes that affect human health and work to understand data at this level.  He talks about the big impacts in his field, namely newer findings on how the microbiome influences the treatment of cancer. Studies show that the kinds of microbes in our gut relate to our immune response's handling of different treatments to fight the cancer. Dr. Michael Nestor, Ph.D., is Board Director, of the Government-University-Industry Research Roundtable ( GUIRR - https://www.nationalacademies.org/guirr/about ), within the Policy and Global Affairs Division, at the U.S. National Academy of Sciences (NAS). The GUIRR is charged with improving the research enterprise of the United States by successfully resolving the cross-sectoral issues that prevent the U.S. research enterprise from reaching its full potential. This mission is achieved by convening senior-most representatives from government, universities, and industry to frame the critical issues, followed – when appropriate – by the execution of activities designed to address specific cross-sectoral impediments to achieving a healthy, vibrant research enterprise. GUIRR provides a unique forum for dialogue among top government, university and industry leaders of the national science and technology enterprise. Prior to this role at NAS, Dr. Nestor was a Scientific Engagement Lead at Johnson & Johnson Innovation – JLABS in Washington, D.C. where he was responsible for external scientific engagement with regional academic research institutions, start-ups, investment partners, and portfolio management, sourcing and evaluating innovative companies with the aim to foster a productive life science ecosystem and bring value to the pipeline of the Johnson & Johnson Family of Companies. Dr. Nestor received his Ph.D. in Neuroscience from The University of Maryland, School of Medicine, and completed postdoctoral fellowships at the National Institutes of Health and The New York Stem Cell Foundation, where he was also a Staff Scientist. He was also an NIH Institutional Research and Academic Career Development Award (IRACDA) Fellow at Rutgers University, focused on teaching in minority-serving institutions and helping increase the participation of traditionally underrepresented groups in science. Dr. Nestor was also Director of Neural Stem Cell Research at The Hussman Institute for Autism where he led his own lab studying autism by creating brain organoids from human induced pluripotent stem cells. His lab developed a multiplexed high-throughput CRISPR and drug-screening platform. Dr. Nestor also served as Co-Chair of the Neural Stem Cell Working Group at the University of Maryland, School of Medicine and as a venture advisor to the UM Momentum Fund and the Abell Foundation. Further, he was an American Association for the Advancement of Science (AAAS), Science and Technology Policy Fellow in the Office of Science at The Department of Energy (DOE). At the DOE, he was focused on synthetic biology, artificial intelligence, and biosecurity initiatives. He also launched his own human stem cell consulting company, Synapstem. A few topics we discussed with Olga in this episode are:How Data Science and Analytics tell a patient's story The value in Glucose Monitoring The company NutriSense - “The value of data and what we call data science and analytics is to tell a story.”Telling a patient's story through Data Science and AnalyticsThe standard practice Olga uses when thinking about a particular question or problem is to look at it from a holistic picture, a pattern, a vignette, an individual person. So, “Data can be an amazingly objective and comprehensive tool for helping us understand what's going on.”She looks at their journey, their data over a period of time combining the entire data set and the rigorous statistical analyses to come up with a scientifically valid answer, then overlaying that with an individual's data to say, this is what it looks like for a single person, not just a sum of a bunch of data points.Keeping a record of your experience, your environment can be extremely helpful for figuring out which anecdotes to listen to and not.Olga mentions the value of a food diary. A lot of people with chronic disease struggle to understand what they eat may or may not contribute to their symptoms. She said you could anecdotally say, "Well, I have inflammatory bowel disease. It seems like when I eat food with emulsifiers, I have symptoms. I have flare ups." People think they would notice these patterns. Olga says the right thing to do is to start keeping that diary and note every time you eat food with emulsifiers, what are your symptoms like the next one or two days and so forth.Glucose MonitoringUsing glucose monitoring you can track morning, noon, and night metabolic characteristics in a person noticing how they change and evolve over time. As these continuous glucose monitors develop, researchers have started using them in academic settings on healthy individuals and research shows there's a lot going on under the surface for healthy people that they didn't really anticipate. One of the most surprising findings was that different people have different responses to the same food. This came out of a landmark study in 2015 out of a group in Israel. They had 800 people wear continuous glucose monitors for two weeks. They had them eat specific things and they saw that the same person can eat a banana and experience a high spike, but eat a cookie and have a much less dramatic increase in blood sugar. And then the reverse is true for someone else.“I think what we would hear when we start listening to our members is that maybe more traditional vital signs of your heart rate, your pulse, amount of oxygen, your blood, glucose is really a marker that's valuable for understanding where you're at today across many dimensions.”NutrisenseThe company is three years old and has three co-founders. AEON (the Astronomical Event Observatory Network) will be a collection of world-class telescope facilities which can be accessed on demand, at the touch of a button.  At the heart of the network, LCO is joining forces with the NOIRLab CSDC and the SOAR 4.1m and Gemini 8m telescopes to build a network for the LSST era.   BIO:  Rachel joined Las Cumbres Observatory (LCO) as a post-doctoral scholar in 2007, and has since become a Staff Scientist. She specializes in the detection and characterization of exoplanets, initially as a founder member of the SuperWASP transit survey, for which she received the RAS Group Achievement Award.    Since joining LCO, she began to study exoplanets via the transient phenomena of microlensing, and now leads the microlensing group there. In the course of this research, she developed automated Target and Observation Management software (TOMs) to make observations through the LCO Network. The U.S. Fish and Wildlife Service imposed a trade ban on 201 salamander species in 2016 in order to prevent the import of the the fungal pathogen Batrachochytrium salamandrivorans ('Bsal') which could be a major threat to the world's salamander hotspot of North America (and the U.S. in particular). However, the recent discovery that frogs can also carry Bsal has led scientists to urge the American government to ban the import of all salamander and frog species to the country. But what other policies or regulations could be enacted to prevent Bsal from wiping out this rich amphibian heritage? Today's episode is an encore of an interview I did with Dr. Lisa Horowitz last fall. Suicide is the second leading cause of death in youth 15-24. Dr. Horowitz talks about how pediatricians can incorporate the Ask Suicide Screening Questions (ASQ) screening tool into their practice to help identify children and adolescents at risk of suicide and to link them to coordinated care.    Dr. Lisa Horowitz is a Staff Scientist and Pediatric Psychologist at the Nation Institute of Mental Health Intermural Research Program at NIMH. The major focus of her research has been detection of suicide risk in the medical setting. She is the head PI in several ‘Suicide Prevention Protocols' using the ASQ Toolkit. Hosted by Richard Wiese—explorer extraordinaire and President of The Explorers Club—this episode features Nina Lanza, distinguished planetary scientist, Mars expert and Mars rover operator. Nina, a first-class Mars rover operator is currently a Staff Scientist at Los Alamos National Laboratory in the Space and Remote Sensing group (ISR-2)—a branch developing and applying remote sensing capabilities to problems of global security and related sciences. Nina received her Bachelor of Arts in Astronomy, at Smith College in Northampton, MA. She went on to earn her Masters in Earth and Environmental Sciences at Wesleyan University, and received her Doctorate in Earth and Planetary Sciences from the University of New Mexico. Nina's interest in space began young. “In 1986,” Nina says “my parents took me to an outreach event at a local university to observe Halley's comet, which was making a close approach to Earth. I was seven years old and had not thought much about space (at that age). But when I looked through the telescope for the first time and saw the comet, I knew from that moment on that I wanted to know what was out there!” As head of the Mars Curiosity Rover ChemCam engineering team at Los Alamos National Laboratory, Nina operates the Curiosity Rover's ChemCam. Nina controls the ChemCam instrument that is attached to the Curiosity rover—one of five NASA rovers sent to the red planet—using a rock-vaporizing laser that analyzes the Martian surface. The instrument shoots rocks with a laser and then her team analyzes them, while a separate team selects which of the Martian rocks to blast. “The microphone is not just to listen to what the aliens are saying,” Nina jokes. “It can help us further analyze the rock.” Nina's outgoing, fun-loving personality brings a life to science in a way that few others can. She is broadly interested in understanding the history of water on Mars and the potential for life in Martian environments; however, her most recent work focuses on manganese in the Martian environment and its implications for habitability and bio-signatures. In addition to her work with the Mars Curiosity rover, in 2015 and 2016, Lanza traveled to Antarctica to recover 569 meteorites from the ice. These ANSMET—The Antarctic Search for Meteorites—meteorite finds may have fallen into the Antarctic snow thousands of years ago. The region's katabatic winds scour away layers of ice, exposing these space rocks. Nina Lanza is currently living her dream of working on a spaceship with lasers on Mars and encourages young women to experience the joy of going into a scientific field, despite the occasional old-school attitudes and gender stereotypes. Nina recently received a generous grant from the renowned NYC-based, The Explorers Club that will fund a team of experts who will use cutting-edge technology in a beautiful, Mars-like Arctic desert to tell the story of how the search for the chemistry of life on Mars begins with fieldwork on Earth near the Haughton crater in northern Canada. Nina makes time every day to do some kind of physical activity, usually high-intensity interval training or yoga. She also sings in a small choral group and enjoys spending time with her son. Prior to starting up Indoor Environmental Engineering, Mr. Offermann was a Staff Scientist with the Building Ventilation and Indoor Air Quality Program at Lawrence Berkeley National Laboratory. He has been a recipient of research grants regarding building air quality and ventilation field studies, tracer gas techniques, contaminant emission rate measurements, and performance testing of air cleaning devices.    Mr. Offermann served as an expert witness for the U.S. Federal Trade Commission regarding the performance claims found in advertisements of air cleaners. He also provided consultation to the American Home Appliance Manufacturers (AHAM) during the development of AHAM Standard AC-1, which is used to measure the Clean Air Delivery Rates (CADR) for portable air cleaners. He was also a past chairman of ASHRAE TC2.3 and SPC-145 committees for gas phase air filters and a corresponding member of ASHRE TC2.4 and SPC-52.2 for particulate air filters. Mr Offermann will discuss the impact of air filtration on indoor airborne concentrations of SARS-CoV-2, including what types of air filtration work and what types pose potential health risks. Selwyn received his B.S. degree in chemistry from New York University, Albany, in 1974, and his Ph.D. degree in chemical physics from University of California, Berkley, in 1979.  From 1979 to 1982, he did postdoctoral research at the University of Canterbury, Christchurch, New Zealand, and at the U.S. Naval Research Laboratory, Washington, DC.  From 1982 to 1994, he worked first as a Staff Scientist with IBM and then as a Research Manager at the IBM Watson Research Center.  He co-chaired a NATO Advanced Research Conference in 1993 and is the recipient of two R&D100 Awards.  He has been with the Los Alamos National Laboratory (LANL), Los Alamos, NM, since 1994, and studies the chemistry and physics of plasmas used in materials processing.  He is also the President and founder of APJeT, Inc., a spin-off from LANL that is commercializing a licensed atmospheric pressure plasma technology for material processing applications.  Gary has since retired and started his own company, Greenthemetek, treating fabrics and currently has a mask treatment to prevent COVID19 in the FDA approval process.   Green Theme Technologies Inc. As a pediatrician screening for suicide, It's not about the ones you find positive for suicide, it's about the ones you may miss. Today's guest Dr. Horowitz is a Staff Scientist and Pediatric Psychologist at the Nation Institute of Mental Health Intermural Research Program at NIMH and for years she has been helping health care professionals not let kids slip through the cracks. She excels in Suicide Risk Detection and is head PI in several ‘Suicide Prevention Protocols' using the ASQ Toolkit.
